Phytotoxicity of Flumequine (F) on the aquatic duckweed, Lemna minor L., and plant drug uptake were evaluated by a simple ecotoxicological test. Flumequine, at all concentrations between 50 and 1000 μg L-1 tested, affected the plant growth: leaves and roots were damaged, but duckweed continued to grow on a five weeks period. Furthermore, increasing drug concentrations decreased the chlorophyll b content in plants. These effects depend on F uptake by plants, which is quite high (from 0.72 to 13.93 μg g-1 plant dry weight). Based on this activity, Lemna can be taken into consideration as a tool for in situ remediation of drug contaminated waters: the presence of Lemna significantly lower the F concentration in culture media on a five weeks period. Waves of cyclin synthesis and degradation regulate the activity of Cdc2 protein kinase during the cell cycle. Cdc2 inactivation by Wee1B-mediated phosphorylation is necessary for arrest of the oocyte at G2-prophase, but it is unclear whether this regulation functions later during the metaphase-to-anaphase transition. We show that reactivation of a Wee1B pathway triggers the decrease in Cdc2 activity during egg activation. When Wee1B is down-regulated, oocytes fail to form a pronucleus in response to Ca(2+) signals. Calcium-calmodulin-dependent kinase II (CaMKII) activates Wee1B, and CaMKII-driven exit from metaphase II is inhibited by Wee1B down-regulation, demonstrating that exit from metaphase requires not only a proteolytic degradation of cyclin B but also the inhibitory phosphorylation of Cdc2 by Wee1B. 相似文献

Marine sponges commonly host a repertoire of bacterial-associated organisms, which significantly contribute to their health and survival by producing several anti-predatory molecules. Many of these compounds are produced by sponge-associated bacteria and represent an incredible source of novel bioactive metabolites with biotechnological relevance. Although most investigations are focused on tropical and temperate species, to date, few studies have described the composition of microbiota hosted by Antarctic sponges and the secondary metabolites that they produce. The investigation was conducted on four sponges collected from two different sites in the framework of the XXXIV Italian National Antarctic Research Program (PNRA) in November–December 2018. Collected species were characterized as Mycale (Oxymycale) acerata, Haliclona (Rhizoniera) dancoi, Hemigellius pilosus and Microxina sarai by morphological analysis of spicules and amplification of four molecular markers. Metataxonomic analysis of these four Antarctic sponges revealed a considerable abundance of Amplicon Sequence Variants (ASVs) belonging to the phyla Proteobacteria, Bacteroidetes, Actinobacteria and Verrucomicrobia. In particular, M. (Oxymycale) acerata, displayed several genera of great interest, such as Endozoicomonas, Rubritalea, Ulvibacter, Fulvivirga and Colwellia. On the other hand, the sponges H. pilosus and H. (Rhizoniera) dancoi hosted bacteria belonging to the genera Pseudhongella, Roseobacter and Bdellovibrio, whereas M. sarai was the sole species showing some strains affiliated to the genus Polaribacter. Considering that most of the bacteria identified in the present study are known to produce valuable secondary metabolites, the four Antarctic sponges could be proposed as potential tools for the discovery of novel pharmacologically active compounds. 相似文献

Verocytotoxic Escherichia (E.) coli strains are responsible for swine oedema disease, which is an enterotoxaemia that causes economic losses in the pig industry. The production of a vaccine for oral administration in transgenic seeds could be an efficient system to stimulate local immunity. This study was conducted to transform tobacco plants for the seed-specific expression of antigenic proteins from a porcine verocytotoxic E. coli strain. Parameters related to an immunological response and possible adverse effects on the oral administration of obtained tobacco seeds were evaluated in a mouse model. Tobacco was transformed via Agrobacteium tumefaciens with chimeric constructs containing structural parts of the major subunit FedA of the F18 adhesive fimbriae and VT2e B-subunit genes under control of a seed specific GLOB promoter. We showed that the foreign Vt2e-B and F18 genes were stably accumulated in storage tissue by the immunostaining method. In addition, Balb-C mice receiving transgenic tobacco seeds via the oral route showed a significant increase in IgA-positive plasma cell presence in tunica propria when compared to the control group with no observed adverse effects. Our findings encourage future studies focusing on swine for evaluation of the protective effects of transformed tobacco seeds against E. coli infection. 相似文献

SummaryGlobe artichoke (Cynara cardunculus L. var. scolymus) provides a rich dietary source of bio-active compounds derived from phenylpropanoid metabolism, notably caffeoylquinic acids (CQAs) and flavonoids. Micropropagation techniques have been established for this species, but in vitro cultures have not yet been extended to generate an efficient system for the induction of callus tissue. In this study, we compared more than 100 combinations of media supplements (e.g., phytohormones, absorbers of polyphenols, and inhibitors of polyphenol oxidase), along with various light regimes, and three different genotypes of globe artichoke to define the optimal conditions for callus induction from leaf explants. This led to the elaboration of an in vitro culture protocol which resulted in a high frequency of callus induction after just 1 week in culture. The procedure used leaf explants from virus-free, meristem culturederived plantlets. Quantitative HPLC analysis revealed that, as in globe artichoke leaves, the predominant phenolic esters present in callus were mono- and di-caffeoylquinic acids (diCQA). The concentration of diCQA was three- to five-fold higher in calli than in leaves. The exposure of calli to UV-C light further enhanced the levels of CQAs. In vitro callus culture combined with UV-C irradiation may thus represent a viable production system for diCQA that is suitable for the synthesis of pharmacologically-active compounds. 相似文献

The quantification of biomass of woody plants is at the basis of calculations of forest biomass and carbon stocks. Although there are well-developed allometric models for trees, they do not apply well to shrubs, and shrub-specific allometric models are scarce. There is therefore a need for a standardized methodology to quantify biomass and carbon stocks in open forests and woodlands.Aims
To develop species-specific biomass estimation models for common shrubs, as well as a multispecies shrub model, for the subtropical semiarid Chaco forest of central Argentina.Methods
Eight shrub species (Acacia aroma, Acacia gilliesii, Aloysia gratissima, Capparis atamisquea, Celtis ehrenbergiana, Larrea divaricata, Mimozyganthus carinatus, and Moya spinosa) were selected, and, on average, 30 individuals per species were harvested. Their total individual dry biomass was related with morphometric variables using regression analysis.Results
Crown area as well as crown-shaped variables proved to be the variables with the best performance for both species-specific and multispecies shrub models. These allometric variables are thus recommended for standardized shrub biomass assessments.Conclusion

Alpine strawberry (Fragaria vesca L.) was grown in hydroponics with the nutrient film technique, in order to evaluate the effects of four buffer concentrations (1.3, 1.6, 1.9, 2.2 mS cm−1) and two cultural cycles (summer-spring versus autumn-spring) in terms of growth, yield and fruit quality (dry and optical residues, sugars, acids, antioxidants, mineral composition). The longer summer-spring cycle gave a correspondingly higher yield than the autumn-spring one. The 1.3 mS cm−1 nutrient solution was the most effective in terms of overall and spring production. However, the autumn and winter yields were not affected by the buffer EC. Fruit quality did not change with the cultural cycle, but the berries harvested in the spring had higher vitamin C and sucrose content and lower nitrate content compared with berries picked up in the winter. Fruit quality was also improved when the nutrient solution concentration increased. From the productive point of view, the cultural cycle choice should be made considering that 71% of the yield of the more productive summer-spring cycle derived from the spring harvest. Moreover, as regards the nutrient solution strength, 1.3 mS cm−1 EC should be preferred during the spring season, whereas the 2.2 mS cm−1 EC proved to be best in the winter in terms of fruit quality. 相似文献

Plants respond to insect attack by emission of volatile organic compounds, which recruit natural enemies of the attacking herbivore, constituting an indirect plant defence strategy. In this context, the egg parasitoid Trissolcus basalis is attracted by oviposition-induced plant volatiles emitted by Vicia faba plants as a consequence of feeding and oviposition by the pentatomid host Nezara viridula. However, this local tritrophic web could be affected by the recent invasion by the alien pentatomid bug Halyomorpha halys, an herbivore that shares the same environments as native pentatomid pests. Therefore, we investigated in laboratory conditions the possible impact of H. halys on the plant volatile-mediated signalling in the local tritrophic web V. faba–N. viridula–T. basalis. We found that T. basalis wasps were not attracted by volatiles induced in the plants by feeding and oviposition activities of H. halys, indicating specificity in the wasps’ response. However, the parasitoid attraction towards plant volatiles emitted as a consequence of feeding and oviposition by the associated host was disrupted when host, N. viridula, and non-associated host, H. halys, were concurrently present on the same plant, indicating that invasion by the alien herbivore interferes with established semiochemical webs. These outcomes are discussed in a context of multiple herbivory by evaluating the possible influences of alien insects on local parasitoid foraging behaviour. 相似文献
