Fusarium spp. is among the largest and most important pathogen groups that attack major grain crops including sorghum. The objectives of this study were to compare the virulence of Fusarium spp. associated with sorghum and examine the mode of resistance of genotypes to the disease. Eight Fusarium species – F. verticillioides, F. thapsinum, F. andiyazi, F. proliferatum, F. nyagamai, F. pseudoanthophilum, F. brevicatenulatum, and F. pseudonygamai – were studied using three sorghum genotypes under greenhouse condition. Three of these pathogens (F. verticillioides, F. thapsinum, and F. andiyazi) were selected for genetic analysis of resistance under field conditions. Sorghum genotypes with contrasting stalk rot reactions were selected for use in both the greenhouse and field experiments. Two weeks after flowering, plants were inoculated with liquid inoculum culture (5 × 104 conidia ml−1) of the different pathogens. Plants were harvested 4 weeks after inoculation and rated for disease severity on the basis of lesion length and number of nodes crossed by the lesion. Among the pathogens, F. thapsinum resulted in consistently higher disease scores in all genotypes under all environments. Likewise, genotype SC599 showed the greatest and most stable resistance as inbred as well as in hybrid combinations as shown by consistently high GCA for resistance to all pathogen species. We recommend that future screening exercises for Fusarium stalk rot resistance utilize F. thapsinum as the causal organism and include the resistant genotype SC599 as a control.  相似文献

A standardized extrusion cooking process was developed for production of a high protein weaning food based on peanuts, maize and soybeans. Major factors evaluated included the effects of blend formulation, extrusion temperature and feed moisture content on ease of extrusion and product quality characteristics. Results showed bulk density and hardness increased while expansion index decreased with increase in feed moisture content. At a fixed range of feed moisture content, product bulk density and firmness decreased while expansion index increased with increasing extrusion temperature. For ease of extrusion and best product quality in terms of sensory attributes and cooking properties, the following extrusion parameters were established for a blend formulation of 75% maize, 10% peanut and 15% soybean: feed particle size of 300–400 m extruded using a screw speed of 500 rpm, with a feed rate of 4.6 kg/min, feed moisture content of 16–18%, and extrusion temperature of 100 ,°C–105,°C. Pair-wise comparison of the sensory attributes of porridges prepared from milled samples of the weaning foods showed significant differences between extruded products and existing traditional counterparts, with very high scores for all sensory attributes of the extruded products, especially extruded raw (non-roasted) blend samples. In the Home-Use-Test, at least 92% of respondents in two out of the three major ecological zones of Ghana placed overall sensory and functional characteristics of extruded raw blend samples as highly acceptable. About 7% of respondents scored sensory and functional quality attributes as acceptable.  相似文献

The cytoplasmic membrane protein TonB spans the periplasm of the Gram-negative bacterial cell envelope, contacts cognate outer membrane receptors, and facilitates siderophore transport. The outer membrane receptor FhuA from Escherichia coli mediates TonB-dependent import of ferrichrome. We report the 3.3 angstrom resolution crystal structure of the TonB carboxyl-terminal domain in complex with FhuA. TonB contacts stabilize FhuA's amino-terminal residues, including those of the consensus Ton box sequence that form an interprotein beta sheet with TonB through strand exchange. The highly conserved TonB residue arginine-166 is oriented to form multiple contacts with the FhuA cork, the globular domain enclosed by the beta barrel.  相似文献

Glioblastoma, the most common and aggressive type of brain tumors, has devastatingly proliferative and invasive characteristics. The need for finding a novel and specific drug target is urgent as the current approaches have limited therapeutic effects in treating glioblastoma. Xyloketal B is a marine compound obtained from mangrove fungus Xylaria sp. (No. 2508) from the South China Sea, and has displayed antioxidant activity and protective effects on endothelial and neuronal oxidative injuries. In this study, we used a glioblastoma U251 cell line to (1) explore the effects of xyloketal B on cell viability, proliferation, and migration; and (2) investigate the underlying molecular mechanisms and signaling pathways. MTT assay, colony formation, wound healing, western blot, and patch clamp techniques were employed. We found that xyloketal B reduced cell viability, proliferation, and migration of U251 cells. In addition, xyloketal B decreased p-Akt and p-ERK1/2 protein expressions. Furthermore, xyloketal B blocked TRPM7 currents in HEK-293 cells overexpressing TRPM7. These effects were confirmed by using a TRPM7 inhibitor, carvacrol, in a parallel experiment. Our findings indicate that TRPM7-regulated PI3K/Akt and MEK/ERK signaling is involved in anti-proliferation and migration effects of xyloketal B on U251 cells, providing in vitro evidence for the marine compound xyloketal B to be a potential drug for treating glioblastoma.  相似文献

The living Laotian rodent Laonastes aenigmamus, first described in early 2005, has been interpreted as the sole member of the new family Laonastidae on the basis of its distinctive morphology and apparent phylogenetic isolation from other living rodents. Here we show that Laonastes is actually a surviving member of the otherwise extinct rodent family Diatomyidae, known from early Oligocene to late Miocene sites in Pakistan, India, Thailand, China, and Japan. Laonastes is a particularly striking example of the "Lazarus effect" in Recent mammals, whereby a taxon that was formerly thought to be extinct is rediscovered in the extant biota, in this case after a temporal gap of roughly 11 million years.  相似文献

Solutes have been added to strengthen elemental metals, generating usable materials for millennia; in the 1960s, solutes were found to also soften metals. Despite the empirical correlation between the "electron number" of the solute and the change in strength of the material to which it is added, the mechanism responsible for softening is poorly understood. Using state-of-the-art quantum-mechanical methods, we studied the direct interaction of transition-metal solutes with dislocations in molybdenum. The interaction increases dramatically with increasing electron number and strongly influences the mechanisms responsible for plasticity in these materials. Our quantitative model explains solution softening of metals by using changes in energy and stress scales of plasticity from solutes.  相似文献

Psychologists have repeatedly shown that a single statistical factor--often called "general intelligence"--emerges from the correlations among people's performance on a wide variety of cognitive tasks. But no one has systematically examined whether a similar kind of "collective intelligence" exists for groups of people. In two studies with 699 people, working in groups of two to five, we find converging evidence of a general collective intelligence factor that explains a group's performance on a wide variety of tasks. This "c factor" is not strongly correlated with the average or maximum individual intelligence of group members but is correlated with the average social sensitivity of group members, the equality in distribution of conversational turn-taking, and the proportion of females in the group.  相似文献

The fine particles serving as cloud condensation nuclei in pristine Amazonian rainforest air consist mostly of secondary organic aerosol. Their origin is enigmatic, however, because new particle formation in the atmosphere is not observed. Here, we show that the growth of organic aerosol particles can be initiated by potassium-salt-rich particles emitted by biota in the rainforest. These particles act as seeds for the condensation of low- or semi-volatile organic compounds from the atmospheric gas phase or multiphase oxidation of isoprene and terpenes. Our findings suggest that the primary emission of biogenic salt particles directly influences the number concentration of cloud condensation nuclei and affects the microphysics of cloud formation and precipitation over the rainforest.  相似文献

数值模拟在温室环境研究中应用日趋广泛,正确选择温室模拟模型对模拟结果的准确性非常重要。本研究以温室夜间各表面的温度为边界条件,采用CFD软件对12m跨度温室温度环境进行二维及三维空间的稳态模拟求解。结果表明温室中间横剖面温度在三维模拟及二维模拟中空间变化趋势相同且与测试结果相近:在温室前部及温室后上部模拟值与测试值的绝对误差在5%以内,在温室中部尤其是近土壤表面误差偏大。由温室中间及距山墙1m远两个横剖面的1m高温度线分布对比结果可见,温室长度大于其跨度两倍以上时,山墙对室内中部温度环境的影响不明显,模拟温室中部横剖面的温度用二维模型可行。  相似文献
