Fire blight caused by the gram-negative bacterium Erwinia amylovora is a major disease of pome fruit like apple (Malus domestica) or pear (Pyrus communis). Detection of the pathogen is hampered by low titres usually being present during initial flower infection and the brief time frame for analysis to decide upon subsequent countermeasures. Here we describe a loop-mediated isothermal amplification of DNA (LAMP) assay for genomic DNA of Erwinia amylovora, which relies on a highly specific primer design that excludes amplification from typical DNA sources of the orchard biological system and from sample handling. The assay enables the fast detection of down to approximately 20?cfu of pure Erwinia amylovora or 100?fg genomic DNA (corresponding to approximately 25 E. amyolovora equivalents) per reaction within 45?min. Fast and reliable detection of E. amylovora in orchard samples by naked eye is achieved through a visual colour change indication with hydroxynaphthol blue. The assay avoids use of complex technical devices and is thus suitable for field testing.  相似文献

Background

In the androdioecious nematode Caenorhabditis elegans virtually all progeny produced by hermaphrodite self-fertilization is hermaphrodite while 50% of the progeny that results from cross-fertilization by a male is male. In the standard laboratory wild type strain N2 males disappear rapidly from populations. This is not the case in some other wild type isolates of C. elegans, among them the Hawaiian strain CB4856.

Results

We determined the kinetics of the loss of males over time for multiple population sizes and wild isolates and found significant differences. We performed systematic inter- and intra-strain crosses with N2 and CB4856 and show that the males and the hermaphrodites contribute to the difference in male maintenance between these two strains. In particular, CB4856 males obtained a higher number of successful copulations than N2 males and sired correspondingly more cross-progeny. On the other hand, N2 hermaphrodites produced a higher number of self-progeny, both when singly mated and when not mated.

Conclusion

These two differences have the potential to explain the observed variation in male persistence, since they should lead to a predominance of self-progeny (and thus hermaphrodites) in N2 and, at the same time, a high proportion of cross-progeny (and thus the presence of males as well as hermaphrodites) in CB4856.  相似文献

In forest soils where a large fraction of total phosphorus (P) is in organic forms, soil micro-organisms play a major role in the P cycle and plant availability since they mediate organic P transformations. However, the correct assessment of organic P mineralization is usually a challenging task because mineralized P is rapidly sorbed and most mineralization fluxes are very weak. The objectives of the present work were to quantify in five forest Spodosols at soil depths of 0-15 cm net mineralization of total organic P and the resulting increase in plant available inorganic P and to verify whether net or gross P mineralization could be estimated using the C or N mineralization rates. Net mineralization of total organic P was derived from the net changes in microbial P and gross mineralization of P in dead soil organic matter. We studied very low P-sorbing soils enabling us to use lower extractants to assess the change in total inorganic P as a result of gross mineralization of P in dead soil organic matter. In addition, to enable detection of gross mineralization of P in dead soil organic matter, a long-term incubation (517 days) experiment was carried out. At the beginning of the experiment, total P contents of the soils were very low (19-51 μg g−1) and were essentially present as organic P (17-44 μg g−1, 85-91%) or microbial P (6-14 μg g−1; 24-39%). Conversely, the initial contents of inorganic P were low (2-7 μg g−1; 9-15%). The net changes in the pool size of microbial P during the 517 days of incubation (4-8 μg g−1) and the amounts of P resulting from gross mineralization of dead soil organic matter (0.001-0.018 μg g−1 day−1; 0.4-9.5 μg g−1 for the entire incubation period) were considerable compared to the initial amounts of organic P and also when compared to the initial diffusive iP fraction (<0.3 μg g−1). Diffusive iP corresponds to the phosphate ions that can be transferred from the solid constituents to the soil solution under a gradient of concentration. Net mineralization of organic P induced an important increase in iP in soil solution (0.6-10 μg g−1; 600-5000% increase) and lower increases in diffusive iP fractions (0.3-5 μg g−1; 300-2000% increase), soil solid constituents having an extremely low reactivity relative to iP. Therefore, soil micro-organisms and organic P transformations play a major role in the bioavailability of P in these forest soils. In our study, the dead soil organic matter was defined as a recalcitrant organic fraction. Probably because gross mineralization of P from this recalcitrant organic fraction was mainly driven by the micro-organisms’ needs for energy, the rates of gross mineralization of C, N and P in the recalcitrant organic fraction were similar. Indirect estimation of gross mineralization of P in dead soil organic matter using the gross C mineralization rate seems thus an alternative method for the studied soils. However, additional studies are needed to verify this alternative method in other soils. No relationships were found between microbial P release and microbial C and N releases.  相似文献

Consistent data demonstrate the positive response of sap-sucking insects to water-stressed plants, but there is a lack of information about the performance of chewing species, including whether their responses vary according to their feeding specializations. We tested the hypothesis that herbivores with distinct feeding strategies and host specialization will respond differently to two plant conditions, i.e. water deficit and kaolin treatment, where the latter is used to reduce abiotic and biotic plant stressors. We determined the development and reproduction of four major cotton pest species with different feeding strategies (chewing and sap sucking) and with specializations ranging from monophagous to polyphagous using well-watered or water-stressed cotton plants, which were treated or not treated with kaolin. Three chewing (lepidopteran) species, Alabama argillacea (Hüb.), Heliothis virescens (Fabr.) and Chrysodeixis includens (Walk), as well as one sap-sucking species, Bemisia tabaci (Gen.), were used as models. The neonate larvae of all three chewing species exhibited lower survival when fed the leaves of water-stressed plants. In addition, the neonate larvae fed leaves from water-stressed and kaolin-treated plants did not complete their development or they exhibited low survival. Older larvae also exhibited significantly lower survival on kaolin-treated plants. The effects on life history traits were more obvious in monophagous chewing species compared with polyphagous species when fed leaves from water-stressed plants. The performance of B. tabaci was superior on water-stressed and untreated kaolin plants. Thus, water-stressed and kaolin-treated plants negatively affected all chewing species, whereas water-stressed plants only positively affected the sap-sucking species.  相似文献

Self-actuating materials capable of transforming between three-dimensional shapes have applications in areas as diverse as biomedicine, robotics, and tunable micro-optics. We introduce a method of photopatterning polymer films that yields temperature-responsive gel sheets that can transform between a flat state and a prescribed three-dimensional shape. Our approach is based on poly(N-isopropylacrylamide) copolymers containing pendent benzophenone units that allow cross-linking to be tuned by irradiation dose. We describe a simple method of halftone gel lithography using only two photomasks, wherein highly cross-linked dots embedded in a lightly cross-linked matrix provide access to nearly continuous, and fully two-dimensional, patterns of swelling. This method is used to fabricate surfaces with constant Gaussian curvature (spherical caps, saddles, and cones) or zero mean curvature (Enneper's surfaces), as well as more complex and nearly closed shapes.  相似文献

Salinity has a two‐phase effect on plant growth, an osmotic effect due to salts in the outside solution and ion toxicity in a second phase due to salt build‐up in transpiring leaves. To elucidate salt‐resistance mechanisms in the first phase of salt stress, we studied the biochemical reaction of salt‐resistant and salt‐sensitive wheat (Triticum aestivum L.) genotypes at protein level after 10 d exposure to 125 mM–NaCl salinity (first phase of salt stress) and the variation of salt resistance among the genotypes after 30 d exposure to 125 mM–NaCl salinity (second phase of salt stress) in solution culture experiments in a growth chamber. The three genotypes differed significantly in absolute and relative shoot and root dry weights after 30 d exposure to NaCl salinity. SARC‐1 produced the maximum and 7‐Cerros the minimum shoot dry weights under salinity relative to control. A highly significant negative correlation (r2 = –0.99) was observed between salt resistance (% shoot dry weight under salinity relative to control) and shoot Na+ concentration of the wheat genotypes studied. However, the salt‐resistant and salt‐sensitive genotypes showed a similar biochemical reaction at the level of proteins after 10 d exposure to 125 mM NaCl. In both genotypes, the expression of more than 50% proteins was changed, but the difference between the genotypes in various categories of protein change (up‐regulated, down‐regulated, disappeared, and new‐appeared) was only 1%–8%. It is concluded that the initial biochemical reaction to salinity at protein level in wheat is an unspecific response and not a specific adaptation to salinity.  相似文献

Laser altimetry by the MESSENGER spacecraft has yielded a topographic model of the northern hemisphere of Mercury. The dynamic range of elevations is considerably smaller than those of Mars or the Moon. The most prominent feature is an extensive lowland at high northern latitudes that hosts the volcanic northern plains. Within this lowland is a broad topographic rise that experienced uplift after plains emplacement. The interior of the 1500-km-diameter Caloris impact basin has been modified so that part of the basin floor now stands higher than the rim. The elevated portion of the floor of Caloris appears to be part of a quasi-linear rise that extends for approximately half the planetary circumference at mid-latitudes. Collectively, these features imply that long-wavelength changes to Mercury's topography occurred after the earliest phases of the planet's geological history.  相似文献

The annual dynamics of fish assemblage in a pond-connecting canal of a South Moravian pond aquaculture facility was studied to reveal potential assemblage effects on pond stocks and vice versa. The species richness of fish assemblage and seasonal dynamics of its development were predominantly dependent upon the fish stock of the interconnected ponds (particularly due to escapees during the period of pond harvesting), which contributed to a considerable increase of commercial fish species occurrence. Using a seasonal parameter, the season proved to have a major impact on fish assemblage dynamics (r = −0.71, P < 0.001). Spring samples were dominated by commercial fishes of aquaculture origin (mainly Cyprinus carpio). Occurrence of several fish species originating from the wild (bleak, Alburnus alburnus, chub, Leuciscus cephalus and roach, Rutilus rutilus) was also reported. The pond canal was found to be an important reservoir site of persistent survival and potential subsequent spreading of some undesirable alien fishes such as topmouth gudgeon (Pseudorasbora parva) and gibel carp (Carrasius auratus). Moreover, more attention should be paid to the potential negative impacts of all fishes of aquaculture origin upon the natural ecosystem and to the adverse effects due to the immigration of wild fish into aquaculture pond farming units.  相似文献
