Despite its economic importance and recent genome release, the need for molecular tools for Hevea brasiliensis is high. In the frame of a disease resistance study, EST sequences were retrieved from public database or generated by sequencing SSH libraries. Sequences were trimmed and microsatellite motifs searched using an ad hoc bioinformatic pipeline, and pairs of primers for the amplification of candidate markers were generated. We found a total of 10 499 unigenes from both sources of sequences, and 673 microsatellites motifs were detected using the default parameters of the pipeline. Two hundred sixty‐four primer pairs were tested and 226 (85.6%) successfully amplified. Out of the amplified candidate markers, 164 exhibited polymorphism. Relationships based on dendrograms using simple matching index and diversity statistics based on EST‐SSRs were compared with Genomic SSRs, showing the potentialities of EST‐derived microsatellites for resistance studies but also for population genetics approaches. 相似文献

Protein quality was evaluated for mechanically separated chicken meat (MSC) and salmon protein hydrolysate (SPH), and for extruded dog foods where MSC or SPH partially replaced poultry meal (PM). Apparent total tract digestibility (ATTD) of crude protein (CP) and amino acids (AA) in the protein ingredients and extruded foods was determined with mink (Neovison vison). The extruded dog foods included a control diet with protein from PM and grain, and two diets where MSC or SPH provided 25% of the dietary CP. Nutrient composition of the protein ingredients varied, dry matter (DM) was 944.0, 358.0 and 597.4 g/kg, CP was 670.7, 421.2 and 868.9 g/kg DM, crude fat was 141.4, 547.8 and 18.5 g/kg DM and ash was 126.4, 32.1 and 107.0 g/kg DM for PM, MSC and SPH respectively. The content of essential AA (g/100 g CP) was more than 10.0 percentage units lower in SPH than in PM and MSC. The ATTD of CP differed (p < 0.001) between protein ingredients and was 80.9%, 88.2% and 91.3% for PM, MSC and SPH respectively. The ATTD of total AA was lowest (p < 0.001) for PM, and similar (p > 0.05) for MSC and SPH. In the extruded diets, the expected higher ATTD of CP and AA from replacement of PM with MSC or SPH was not observed. The ATTD of CP was determined to be 80.3%, 81.3% and 79.0% for the PM, MSC and SPH extruded foods respectively. Furthermore, the ATTD of several AA was numerically highest for the PM diet. Possibly, extrusion affected ATTD of the diets differently due to different properties and previous processing of the three protein ingredients. 相似文献

A new method for pest risk assessment and the identification and evaluation of risk‐reducing options is currently under development by the European Food Safety Authority (EFSA) Plant Health Panel. The draft method has been tested on pests of concern to the European Union (EU). The method is adaptable and can focus either on all the steps and sub‐steps of the assessment process or on specific parts if necessary. It is based on assessing changes in pest population abundance as the major driver of the impact on cultivated plants and on the environment. Like other pest risk assessment systems the method asks questions about the likelihood and magnitude of factors that contribute to risk. Responses can be based on data or expert judgment. Crucially, the approach is quantitative, and it captures uncertainty through the provision by risk assessors of quantile estimates of the probability distributions for the assessed variables and parameters. The assessment is based on comparisons between different scenarios, and the method integrates risk‐reducing options where they apply to a scenario, for example current regulation against a scenario where risk‐reducing options are not applied. A strategy has been developed to communicate the results of the risk assessment in a clear, comparable and transparent way, with the aim of providing the requestor of the risk assessment with a useful answer to the question(s) posed to the EFSA Plant Health Panel. The method has been applied to four case studies, two fungi, Ceratocystis platani and Cryphonectria parasitica, the nematode Ditylenchus destructor and the Grapevine flavescence dorée phytoplasma. The deleterious effects of heat stress on animal health are being increasingly recognized. This study aimed to determine hair cortisol (HC) and serotonin levels in lactating Holstein cows under heat stress conditions with different coat and hair‐cut color. Forty‐five multiparous lactating Holstein cows (days in milk = 130 ± 47, body weight = 753 ± 85 kg) were divided to two main groups of over 80% black coat color (BC) and over 85% white coat color (WC) visually observed based on registry certificates and subdividing to black hair sample (BH) and white hair samples (WH) in 2 × 2 factorial arrangements. Hair samples were taken from the forehead of the individuals. Higher HC levels were observed in BC than WC cows (P < 0.05). No differences were found in HC levels between BH and WH groups (P > 0.05). Serotonin levels showed no difference between BC and WC (P > 0.05). Interaction between coat color and hair color was not significant (P > 0.05). The cortisol levels in hair are not affected by pigmentation. However, pigmentation within the coat alters cortisol levels. In conclusion, white coat color retains less cortisol than the black coat. Therefore, white coats are preferable for dairy cows under heat stress conditions. 相似文献

We evaluated recruitment of larvae of catarina scallop, Argopecten ventricosus, in the area of Puerto Peñasco, NE Gulf of California. We moored artificial collectors in six sites from June 2007 to August 2008 and replaced them every 2 months. We used monthly (July 2002–September 2011) sea surface temperature (SST, °C) and surface chlorophyll‐a concentration (SSChl, mg m?3) Aqua/MODIS satellite data to describe seasonal environmental behaviour study area. Also, we recorded bottom temperature at each site every 4 h, and every 2 months measured sea surface salinity, temperature and dissolved oxygen. We used a repeated measures anova to evaluate differences in the number of recruited spat between main factors, and analysed the presence of multimodal spat shell size frequency distributions. Overall, spat recruitment was negatively correlated with seawater temperature and showed higher spat recruitment abundances throughout winter, which is the season with the highest surface chlorophyll a concentration. We estimated multimodal shell size frequency distributions characterized by more than one modal size. The natural collection of A. ventricosus spat on artificial collectors in the area can be successfully performed over a protracted period (November–December to May–June). The Korean rockfish, Sebastes schlegeli, is a valuable and intensively exploited species in Korea. We discuss the genetic diversity and genetic structure of four Korean rockfish populations using eight microsatellite loci. In total, 161 different alleles from 138 individuals were observed. Average allele number per locus ranged from 2.5 to 23 and allelic richness varied from 13.38 to 14.63 within a population. Despite a long history of stocking practices, we found very high levels of polymorphism (mean heterozygosity = 0.810), which is comparable to other congeneric species. No significant difference in genetic diversity and molecular genetic variance (FST and RST) was observed among four local samples (P > 0.1). Little indication of contemporary inbreeding (FIS= 0.051) or population structure (K = 1) was detected. This absence of differentiation may reflect high levels of gene flow along the coast of Korea. Our study demonstrates that rockfish in Korea should be managed as a single unit. Currently, the species does not appear to be genetically threatened, but the potential for a rapid loss of genetic diversity remains. This information on the genetic characteristics of Korean rockfish populations has important implications for fisheries management and conservation efforts, and will aid in the sustainable exploitation of the fishing resources and the preservation of biodiversity. 相似文献
