This study is part of an ongoing effort to develop animal models that provide milk and sufficient infant (offspring) plasma samples to fully describe a drug's pharmacokinetics to quantitate the risk to the nursing infant. Ciprofloxacin was administered to six healthy Holstein cows as a constant rate intravenous infusion (flow rate was weight adjusted) to achieve a steady‐state concentration of approximately 300 ng/mL for 7 days. Plasma and milk samples were collected from the cow at regular intervals over the course of the 7 days. The plasma and milk samples were analyzed for ciprofloxacin by high‐performance liquid chromatography. The milk was fed to calves, and calf plasma samples were analyzed to study the lactational transfer of ciprofloxacin from dam to nursing neonate. Remarkably, concentrations of ciprofloxacin in milk were 45 times higher than plasma drug concentrations in the dam. Approximately 6% of the administered dose was transferred to the milk, resulting in an average oral dose of 0.5 mg/kg to the calves with every feeding. The drug did not accumulate in the calves, and plasma concentrations were between one‐tenth and one‐fifth the plasma concentrations of the dam. 相似文献

Complementary DNA clones were isolated that represent the 5' terminal 2.5 kilobases of the murine Duchenne muscular dystrophy (Dmd) messenger RNA (mRNA). Mouse Dmd mRNA was detectable in skeletal and cardiac muscle and at a level approximately 90 percent lower in brain. Dmd mRNA is also present, but at much lower than normal levels, in both the muscle and brain of three different strains of dystrophic mdx mice. The identification of Dmd mRNA in brain raises the possibility of a relation between human Duchenne muscular dystrophy (DMD) gene expression and the mental retardation found in some DMD males. These results also provide evidence that the mdx mutations are allelic variants of mouse Dmd gene mutations. 相似文献

Protoplasts were isolated from leaves of Lotus corniculatus and Trifolium repens and from cotyledons of L. corniculatus. These protoplasts divided and produced colonies. The plating efficiency of protoplasts of both species was improved when agarose was used as a supporting medium. Plants were regenerated more regularly and in larger numbers from colonies of L. carniculatus than T. repens. The use of a culture line of T. repens that had been selected for its response in culture markedly increased the proportion of protoplast-derived cultures which regenerated shoots. The heterogeneity of nutrients in forest soils is governed by many biotic and abiotic factors. The significance of nutrient patchiness in determining soil processes remains poorly understood. Some saprotrophic basidiomycete fungi influence nutrient heterogeneity by forming large mycelial networks that enable translocation of nutrients between colonized patches of dead organic matter. The effect of mycophagous soil fauna on these networks and subsequent nutrient redistribution has, however, been little studied. We used a soil microcosm system to investigate the potential effects of a mycophagous collembola, Protaphorura armata, on nutrient transfer within, and nutrient loss from, the mycelium of a saprotrophic basidiomycete fungus, Phanerochaete velutina. A 15N label, added to central mycelium, was used to track nitrogen movement within the microcosms across 32 days. Although collembola grazing had little impact on δ15N values, it did alter the partitioning of 15N between different regions of mycelia. Less 15N was transferred to new mycelial growth in grazed systems than in ungrazed systems, presumably because collembola reduced fungal growth rate and altered mycelial morphology. Surprisingly, collembola grazing did not increase the mineralization of N from mycelium into the bulk soil. Overall, our results suggest that mycophagous soil fauna can alter nutrient flux and partitioning within fungal mycelium; this has the potential to affect the dynamics and spatial heterogeneity of forest floor nutrients. 相似文献

Cognitive functions dependent on the prefrontal cortex, such as the ability to suppress behavior (response inhibition) and to learn from complex feedback (probabilistic learning), play critical roles in activities of daily life. To what extent do different neurochemical systems modulate these two cognitive functions? Here, using stop-signal and probabilistic learning tasks, we show a double dissociation for the involvement of noradrenaline and serotonin in human cognition. In healthy volunteers, inhibition of central noradrenaline reuptake improved response inhibition but had no effect on probabilistic learning, whereas inhibition of central serotonin reuptake impaired probabilistic learning with no effect on response inhibition. 相似文献

Soil organic carbon(SOC) has primary importance in terms of soil physics, soil fertility and even of climate change control. One hundred soil samples were taken from an intensively cultivated Cambisol to quantify SOC redistribution triggered by soil erosion under a subhumid climate, by the simultaneous application of diffuse reflectance(240–1 900 nm) and traditional physico-chemical methods.The representative sample points were collected from the solum along the slopes at the depth of 20–300 cm with a mean SOC content of 12 g kg~(-1). Hierarchical cluster analyses were performed based on the determined SOC results. The spatial pattern of the groups created were similar, and even though the classifications were not the same, diffuse reflectance had proven to be a suitable method for soil/sediment classification even within a given arable field. Both organic and inorganic carbon distributions were found to be a proper tool for estimations of past soil erosion processes. The SOC enrichment was found on two sedimentary spots with different geomorphological positions. Soil organic matter composition also differed between the two spots due to selective deposition of the delivered organic matter. The components with low-molecular-weight reached the bottom of the slope where they could leach into the profile, while the more polymerised organic matter compositions were delivered and deposited even before on a higher segment of the slope in an aggregated form. This spatial difference appeared below the uppermost tilled soil layer as well, referring the lower efficiency of conventional ploughing tillage in soil spatial homogenisation. 相似文献
