The properties of 220 strains of Clostridium perfringens and Clostridium perfringens-like organisms were studied. A scheme was designed for the identification of these strains. Three duodenally cannulated, lactating Holstein cows were dosed with rare earth-labeled grain to evaluate effects of passage model, sampling site, and marker dosing time on digesta passage parameters. Cows were given ad libitum access to feed twice daily. Rare earth-labeled grain (applied by the 24-h immersion technique) was fed immediately before (Dy) or 2 h after (Yb) the morning feeding, and duodenal digesta and feces were sampled. Marker excretion curves were fit to a two-compartment, biexponential model, using curve peeling or to a series of two-compartment models, with one to six orders of gamma time-dependency in the fast compartment, using nonlinear regression. Passage estimates from the curve-peeled, biexponential model were similar to those from the best fit of the nonlinear models, which had three orders of gamma time-dependency. Ruminal passage rate of grain, averaged across models, sampling site, and dosing time, was .077/h. Estimates of time to first appearance of marker at the sampling site and mean retention times were longer (8 vs 1 h and 25 vs 17 h, respectively), but passage rates were similar, when determined from fecal compared with duodenal samples (P less than .05). Marker dosing time did not influence any of the parameters. It is concluded that both curve-peeling (linear regression) and nonlinear regression methods can be equally useful for evaluating passage kinetics of grain in dairy cows and that sampling site and time of marker dosing have little effect on passage parameter estimates. 相似文献

Rabies virus is able to infect in vitro a wide range of homeothermic and poikilothermic cells but little is known about its multiplication in arthropod cells. In this research the infection of rabies virus in Aedes pseudoscutellaris cells, a mosquito cell line susceptible to mosquito-borne viruses, was studied. After 60 days of incubation at 26 degrees C up to 70% of infected cells showed the synthesis of both viral nucleocapsid and envelope antigens, although viral yield and cell damage could not be detected. Research performed in order to investigate the role of membrane carbohydrate moieties in rabies virus-mosquito cell interaction suggested the participation of galactose and N-acetylglucosamine whereas sialic acid, known to be a rabies binding site in many homeothermic cell lines, was not involved. 相似文献

A sandwich enzymeimmunoassay (EIA) for pregnant mare serum gonadotropin (PMSG) using a microtiter plate was developed. Sensitivity of the assay to PMSG was 15.6 mIU/ml (0.2 ng/well). The PMSG levels in serum were measured with the EIA in superovulated and anti-PMSG rabbit antiserum treated mice and heifers. In mice, the PMSG blood level was measurable in the serum 4-6 days after intraperitoneal injection of 5-30 IU of PMSG. The administration of anti-PMSG antiserum at the same dose level as PMSG caused a rapid decrease in the PMSG blood level, declining to undetectable levels within 17 hours. In heifers, the PMSG level was measurable at 10-11 days after the injection of 2500 or 3000 IU of PMSG. When antiserum was injected 48 hours after the PMSG injection, the clearance rate of PMSG was affected by the route of the administration. The administration of 3000 units of anti-PMSG antiserum intravenously caused a rapid decline and the disappearance of circulating PMSG within 17 hours. When 3000 units of anti-PMSG antiserum was injected intra-muscularly, the PMSG blood level also decreased and became unmeasurable 24 hours after administration; however, it was still detectable for up to 17 hours. These results indicate that the administration of anti-PMSG antiserum at the proper timing and dosage could lead to successful superovulation through the improvement of hormonal conditions. 相似文献

We describe the spatial epidemiological features of the 6.8 million meat-juice serological tests that were conducted between 1995 and 2004 as part of the Danish swine Salmonella control programme. We investigated pig and farm density using edge-corrected kernel estimations. Pigs were aggregated at the county level to assess county-level risk, and then we investigated farm-level risk by giving farms a case or non-case label using a cut-off of 40% of pigs positive. Conditional probability surfaces, correcting for the underlying population at risk, were produced for each year of the study period using a novel kernel estimator with a spatially adaptive smoothing bandwidth. This approach improves on previous methods by allowing focussed estimation of risk in areas of high population density while maintaining stable estimates in regions where the data are sparse. Two spatial trends in the conditional probability of a farm being a case were evident: (1) over the whole country, with the highest risk in the west compared to the east; and (2) on the Jutland peninsula with the highest risk in the north and south. At the farm-level a consistent area of risk was the south-west of Jutland. Case farms tended to aggregate indicating spatial dependency in the data. We found no association between pig or farm density and Salmonella risk. We generated hypotheses for this spatial pattern of risk and we conclude that this spatial pattern should be considered in the development of surveillance strategies and as a basis for further, more detailed analyses of the data. 相似文献

Semen availability in ram semen processing facilities is of great importance for the genetic improvement of sheep. Accordingly, any method that would increase sperm viability in low viability ejaculates could be useful. In this study, the possibility of a glass beads filtration method by estimating the beads total surface provided (TSP) for adhesion of spermatozoa, was evaluated. Initially, two different TSP (102 and 154 cm(2)) achieved by various sizes of beads (1500, 2000 and 3000 microm) were tested and no significant difference in sperm viability improvement was noticed for the same TSP by different beads (p > 0.05). Next optimization tests were performed in which three different funnels were used for filtration at a standard TSP (154 cm(2)). The pear-shaped funnel was found to be the most appropriate for filtration, as semen volume recovery and sperm viability improvement were more pronounced (p < 0.05). Finally, filtration tests were conducted with pear-shaped funnels with different TSP (102 and 154 cm(2)) obtained by the aforementioned beads sizes (1500, 2000 and 3000 microm) in equal aliquots. Total surface provided of 102 cm(2) proved to be the more appropriate for filtration than 154 cm(2), as shown by the significant improvement of sperm viability (p < 0.01) and the significantly higher filtrate semen volume (p < 0.05). In conclusion, ram sperm viability improvement by more than 20% of its initial value and semen volume recovery by more than 60%, along with the fact that the total filtration time did not exceed 6 min in any case, suggest that through further development this method could be successfully used during ram semen processing. 相似文献
