The objective of this study was to evaluate the safety and efficacy of a purified porcine insulin zinc suspension for treating dogs with uncomplicated diabetes mellitus. Fifty-three dogs were treated for 60 days after an initial dose determination period. The means of the blood glucose concentrations during 12-hour glucose curves and the means of the blood glucose nadir concentrations during 12-hour glucose curves for all dogs were determined before beginning insulin therapy (time 0), at the end of the dose determination period (time 1), 30 days after time 1 (time 2), and 60 days after time 1 (time 3). Presence of polyuria, polydipsia, and ketonuria was determined at each time point. Adequacy of control of hyperglycemia was based on 12-hour blood glucose curves and improvement in clinical variables (results of physical examinations, historic information, polyuria, polydipsia, and ketonuria). Safety was evaluated by questionnaire, performance of physical examination, CBC, serum chemistry profile, and urinalysis. The means of the blood glucose concentrations during 12-hour glucose curves and the means of the blood glucose nadir concentrations during 12-hour glucose curves for all dogs at times 1, 2, and 3 were significantly lower compared with time 0 (P < .0001). There was a reduction in the proportion of dogs with polyuria, polydipsia, and ketonuria of 82, 86, and 80%, respectively. All of the dogs had adequate glycemic control at time 1, 66% at time 2, and 75% at time 3. At time 3, 66% of dogs required insulin injections q12h. Other than hypoglycemia, there were no important adverse effects of insulin administration. The insulin, was safe and efficacious for reducing blood glucose and clinical signs in dogs with diabetes mellitus.  相似文献

The veterinary school admission interview is a widely used selection tool, yet concerns persist about its reliability, validity, and cost. Relative to medicine, optometry, and dentistry schools, veterinary schools have been more likely to conduct panel interviews and to fix the interview's weight in selection decisions, strategies that increase interview validity. This article provides strategies for further increasing the veterinary school interview's validity. Interview reliability and validity studies point to key strategies the veterinary school admissions committee can implement before the interview: (1) establishing the interview's purpose(s); (2) conducting a "job" analysis to identify desirable candidate skills, knowledge, and attributes; (3) developing a structured and panel interview where interviewers, if possible, are blind to other admission data; (4) training interviewers; (5) setting a reasonable interview schedule; and (6) determining methods for analyzing applicant data. During the interview, interviewers should proceed through a structured series of steps: (1) open the interview with a specified agenda; (2) probe for information using structured questions and anchored rating scales; (3) close the interview to allow for candidate questions; and (4) evaluate the interview data. After the interview, the admissions committee should (1) analyze the interview data within and across interviewers and (2) analyze the data across all selection tools in order to assign relative weights to the selection tools.  相似文献

Four ruminally and duodenally cannulated steers (703.4 +/- 41 kg initial BW) were used in a 4 x 4 Latin square to evaluate the effects of field pea inclusion level on intake and site of digestion in beef steers fed medium-concentrate diets. Steers were offered feed ad libitum at 0700 and 1900 daily and were allowed free access to water. Diets consisted of 45% grass hay and 55% by-products based concentrate mixture and were formulated to contain a minimum of 12% CP (DM basis). Treatments consisted of (DM basis) 1) control, no pea; 2) 15% pea; 3) 30% pea; and 4) 45% pea in the total diet, with pea replacing wheat middlings, soybean hulls, and barley malt sprouts in the concentrate mixture. Experimental periods consisted of a 9-d dietary adjustment period followed by a 5-d collection period. Grass hay was incubated in situ, beginning on d 10, for 0, 2, 5, 9, 14, 24, 36, 72, and 98 h; and field pea and soybean hulls for 0, 2, 5, 9, 14, 24, 36, 48, and 72 h. Total DMI (15.0, 13.5, 14.1, 13.5 +/- 0.65 kg/d) and OM intake (13.4, 12.0, 12.6, 12.0 +/- 0.58 kg/d) decreased linearly (P = 0.10) with field pea inclusion. Apparent ruminal (17.5, 12.0, 0.6, 6.5 +/- 4.31%) and true ruminal CP digestibility (53.5, 48.7, 37.8, 46.2 +/- 3.83) decreased linearly (P < 0.10) with increasing field pea. Neutral detergent fiber intake (8.9, 7.9, 7.8, 7.0 +/- 0.3 kg/d) and fecal NDF output (3.1, 2.9, 2.6, 2.3 +/- 0.2 k/d) decreased linearly (P < 0.03) with increasing field pea. No effects were observed for microbial efficiency or total-tract digestibility of OM, CP, NDF, and ADF (P > or = 0.16). In situ DM and NDF disappearance rates of grass hay and soybean hulls decreased linearly (P < 0.05) with increasing field pea. Field pea in situ DM disappearance rate responded quadratically (P < 0.01; 5.9, 8.4, 5.5, and 4.9 +/- 0.52%/h, for 0, 15, 30, and 45% field pea level, respectively). Rate of in situ CP disappearance of grass hay decreased linearly (P < 0.01) with increasing field pea level. Field pea is a suitable ingredient for beef cattle consuming medium-concentrate diets, and the inclusion of up to 45% pea in by-products-based medium-concentrate growing diets decreased DMI, increased dietary UIP, and did not alter OM, NDF, or ADF digestibility.  相似文献

Effects of increasing level of field pea (variety: Profi) on intake, digestion, microbial efficiency, and ruminal fermentation were evaluated in beef steers fed growing diets. Four ruminally and duodenally cannulated crossbred beef steers (367+/-48 kg initial BW) were used in a 4 x 4 Latin square. The control diet consisted of 50% corn, 23% corn silage, 23% alfalfa hay, and 4% supplement (DM basis). Treatments were field pea replacing corn at 0, 33, 67, or 100%. Diets were formulated to contain a minimum of 12% CP, 0.62% Ca, 0.3% P, and 0.8% K (DM basis). Each period was 14 d long. Steers were adapted to the diets for 9 d. On d 10 to 14, intakes were measured. Field pea was incubated in situ, beginning on d 10, for 0, 2, 4, 8, 12, 16, 24, 36, 48, 72, and 96 h. Bags were inserted in reverse order, and all bags were removed at 0 h. Ruminal fluid was collected and pH recorded at -2, 0, 2, 4, 6, 8, 10, and 12 h after feeding on d 13. Duodenal samples were taken for three consecutive days beginning on d 10 in a manner that allowed for a collection to take place every other hour over a 24-h period. Linear, quadratic, and cubic contrasts were used to compare treatments. There were no differences in DMI (12.46 kg/d, 3.16% BW; P > 0.46). Ruminal dry matter fill (P = 0.02) and mean ruminal pH (P = 0.009) decreased linearly with increasing field pea level. Ruminal ammonia-N (P < 0.001) and total VFA concentrations (P = 0.01) increased linearly with increasing field pea level. Total-tract disappearance of OM (P = 0.03), N (P = 0.01), NDF (P = 0.02), and ADF (P = 0.05) increased linearly with an increasing field pea level. There were no differences in total-tract disappearance of starch (P = 0.35). True ruminal N disappearance increased linearly (P < 0.001) with increasing field pea level. There were no differences in ruminal disappearance of OM (P = 0.79), starch (P = 0.77), NDF (P = 0.21), or ADF (P = 0.77). Treatment did not affect microbial efficiency (P = 0.27). Field pea is a highly digestible, nutrient-dense legume grain that ferments rapidly in the rumen. Because of their relatively high level of protein, including field peas in growing diets will decrease the need for protein supplementation. Based on these data, it seems that field pea is a suitable substitute for corn in growing diets.  相似文献

The objectives of this study were to evaluate intestinal cellularity and vascularity in mature ewes in response to dietary restriction and pregnancy status and to quantify the response of these variables to increased nutrient demand of fetal growth. In Exp. 1, 28 mature Dorset x crossbred white-faced ewes (61.6+/-1.8 kg initial BW) were fed a pelleted, forage-based diet. Treatments were arranged in a 2 x 3 factorial, with dietary restriction (60% restriction vs. 100% maintenance for respective states of pregnancy) and pregnancy status (nonpregnant, NP; d 90 and 130) as main effects. Dietary treatments were initiated on d 50 of gestation and remained at 60 or 100% maintenance throughout the experiment. Nonpregnant ewes were fed dietary treatments for 40 d. In Exp. 2, four Romanov ewes were naturally serviced (Romanov fetus and Romanov dam; R/R); two Romanov embryos per recipient were transferred to four Columbia recipients (Romanov fetus and Columbia recipient; R/C), and three Columbia ewes were naturally serviced (Columbia fetus and Columbia dam; C/C). In Exp. 1, dietary restriction and pregnancy status interacted with regard to maternal jejunal DNA concentration (P < 0.01), with restricted ewes having a greater DNA concentration (mg/g; fresh basis) at d 130. Vascularity (percentage of total tissue area) in the jejunum was increased (P < 0.06) as a result of dietary restriction and pregnancy status. Total microvascular volume ofjejunal tissue was not altered by dietary restriction and increased (P < 0.01) at d 130 of pregnancy. In Exp. 2, R/R ewes had less (P < 0.09) DNA (g) in the jejunum compared with R/C and C/C ewes. Jejunal vascularity (%) was increased (P < 0.05) in R/R ewes compared with R/C or C/C ewes, whereas total jejunal microvascular volume remained unchanged. These data demonstrate intestinal vascular density responds to changes in diet and physiological state. In addition, pregnancy increased total jejunal microvascular volume.  相似文献

Two metabolism (4 x 4 Latin square design) experiments were conducted to evaluate the effects of corn condensed distillers solubles (CCDS) supplementation on intake, ruminal fermentation, site of digestion, and the in situ disappearance rate of forage in beef steers fed low-quality switchgrass hay (Panicum virgatum L.). Experimental periods for both trials consisted of a 9-d diet adaptation and 5 d of collection. In Exp. 1, 4 ruminally and duodenally cannulated steers (561 +/- 53 kg of initial BW) were fed low-quality switchgrass hay (5.1% CP, 40.3% ADF, 7.5% ash; DM basis) and supplemented with CCDS (15.4% CP, 4.2% fat; DM basis). Treatments included 1) no CCDS; 2) 5% CCDS; 3) 10% CCDS; and 4) 15% CCDS (DM basis), which was offered separately from the hay. In Exp. 2, 4 ruminally and duodenally cannulated steers (266.7 +/- 9.5 kg of initial BW) were assigned to treatments similar to Exp. 1, except forage (Panicum virgatum L.; 3.3% CP, 42.5% ADF, 5.9% ash; DM basis) and CCDS (21.6% CP, 17.4% fat; DM basis) were fed as a mixed ration, using a forage mixer to blend the CCDS with the hay. In Exp. 1, ruminal, postruminal, and total tract OM digestibilities were not affected (P = 0.21 to 0.59) by treatment. Crude protein intake and total tract CP digestibility increased linearly with increasing CCDS (P = 0.001 and 0.009, respectively). Microbial CP synthesis tended (P = 0.11) to increase linearly with increasing CCDS, whereas microbial efficiency was not different (P = 0.38). Supplementation of CCDS to low-quality hay-based diets tended to increase total DM and OM intakes (P = 0.11 and 0.13, respectively) without affecting hay DMI (P = 0.70). In Exp. 2, ruminal OM digestion increased linearly (P = 0.003) with increasing CCDS, whereas postruminal and total tract OM digestibilities were not affected (P > or = 0.37) by treatment. Crude protein intake, total tract CP digestibility, and microbial CP synthesis increased (P < or = 0.06) with increasing level of CCDS supplementation, whereas microbial efficiency did not change (P = 0.43). Ruminal digestion of ADF and NDF increased (P = 0.02 and 0.008, respectively) with CCDS supplementation. Based on this data, CCDS used in Exp. 2 was 86.7% rumen degradable protein. The results indicate that CCDS supplementation improves nutrient availability and use of low-quality forages.  相似文献

We investigated extraneural manifestations in scrapie-infected transgenic mice expressing prion protein lacking the glycophosphatydylinositol membrane anchor. In the brain, blood, and heart, both abnormal protease-resistant prion protein (PrPres) and prion infectivity were readily detected by immunoblot and by inoculation into nontransgenic recipients. The titer of infectious scrapie in blood plasma exceeded 10(7) 50% infectious doses per milliliter. The hearts of these transgenic mice contained PrPres-positive amyloid deposits that led to myocardial stiffness and cardiac disease.  相似文献

CASE DESCRIPTION: 4 racehorses were examined because of markedly abnormal behavior following administration of fluphenazine decanoate. CLINICAL FINDINGS: Clinical signs included restlessness, agitation, profuse sweating, hypermetria, aimless circling, intense pawing and striking with the thoracic limbs, and rhythmic swinging of the head and neck alternating with episodes of severe stupor. Fluphenazine was detected in serum or plasma from all 4 horses. The dose of fluphenazine decanoate administered to 3 of the 4 horses was within the range (25 to 50 mg) routinely administered to adult humans. TREATMENT AND OUTCOME: In 2 horses, there was no response to IV administration of diphenhydramine hydrochloride, but the abnormal behavior in these 2 horses appeared to resolve following administration of benztropine mesylate, and both horses returned to racing. The other 2 horses responded to diphenhydramine administration. One returned to racing. The other was euthanized because of severe neurologic signs, respiratory failure, and acute renal failure. CLINICAL RELEVANCE: Findings indicate that adverse extrapyramidal effects may occur in horses given fluphenazine decanoate. These effects appear to be unpredictable and may be severe and life threatening. Use of fluphenazine decanoate as an anxiolytic in performance horses is not permitted in many racing and horse show jurisdictions, and analytic procedures are now available to detect the presence of fluphenazine in serum or plasma.  相似文献

BACKGROUND: Phytophthora blight induced by Phytophthora capsici is responsible for serious yield loss in vegetable production in the United States and other countries. This study was conducted to evaluate the efficacy of Brassica cover crops used as soil amendments for managing Phytophthora blight of squash. RESULTS: In greenhouse studies, disease incidence on squash plants was significantly reduced by soil amendment with mustard shoots or roots used at 1 and 2.5% (plant tissue/soil, w/w). The shoots of canola used at 1 or 2.5% also suppressed disease, while the roots of canola or other crops did not reduce disease significantly. In field studies, soil amendments with mustard and canola provided the greatest disease reduction and increased squash yield significantly compared with the non‐treated control. Mustard and canola did not appear to be susceptible to P. capsici. CONCLUSION: The results indicated that some Brassica crops, particularly mustard and canola, had the potential to significantly reduce Phytophthora blight on squash when used as soil amendments. As P. capsici has a remarkable ability to develop resistance to chemical fungicides, use of effective Brassica cover crops could be a biorational alternative to fungicides and a valuable component in developing integrated disease management programs.
