CASE DESCRIPTION: 3 immature screw-tailed dogs were evaluated because of progressive pelvic limb paraparesis. CLINICAL FINDINGS: Each dog had marked ataxia and paresis of the pelvic limbs and a palpable deformity of the midthoracic portion of the vertebral column. Pain perception in the pelvic limbs was considered normal, and there was no evidence of fecal or urinary incontinence in any of the 3 dogs. Radiography and magnetic resonance imaging revealed hemivertebrae with severe dorsoventral stenosis of the vertebral canal resulting in spinal cord compression in 2 dogs and lateral compression in the other. TREATMENT AND OUTCOME: Each dog underwent decompressive surgery consisting of dorsal laminectomy or hemilaminectomy and vertebral stabilization by use of combinations of Kirschner wires or threaded external fixator pins plus polymethylmethacrylate bone cement. All dogs regained strong locomotor function with minimal residual pelvic limb ataxia. CLINICAL RELEVANCE: Little detailed information regarding surgical treatment of hemivertebrae in dogs is available; results of treatment in these 3 dogs suggest that spinal cord decompression and stabilization of the vertebral column can achieve a satisfactory, functional outcome.  相似文献

The mechanical properties of wheat bran and the contribution of each constitutive tissue on overall bran properties were determined on a hard wheat (cv. Baroudeur) and a soft wheat (cv. Scipion). Manual dissection allowed three different layers to be separated from wheat bran, according to radial and longitudinal grain orientations, which were identified by confocal laser scanning microscopy as outer pericarp, an intermediate strip (comprising inner pericarp, testa, and nucellar tissue), and aleurone layer, respectively. Tissue microstructure and cell wall composition were determined. Submitted to traction tests, whole bran, intermediate, and aleurone layers demonstrated elastoplastic behavior, whereas pericarp exhibited elastic behavior. By longitudinal orientation, pericarp governed 50% bran elasticity (elastic strength and rigidity), whereas, in the opposite orientation, bran elastic properties were mostly influenced by the other tissues. Regardless of test orientation, the linear force required to bran rupture corresponded to the sum of intermediate and aleurone layer strengths. According to radial orientation, the intermediate strip governed bran extensibility, but according to longitudinal orientation, all tissues contributed until bran disruption. Tissues from both wheat cultivars behaved similarly. A structural model of wheat bran layers illustrated the detachment of pericarp from intermediate layer within radial bran strips.  相似文献

Transport of contaminants from coal storage areas through the rooting zone has the potential to negatively affect shallow ground water and terrestrial vegetation. Reject coal piles represent a worst-case scenario for contamination from coal storage areas because of the long residence time of the coal. Preliminary hydrologic data were collected from an area of stressed vegetation adjacent to an inactive ash basin and reject coal pile in South Carolina. The water table is near the surface (< 60 cm) in this wetland area and water movement is primarily to the west, toward the Savannah River. Acid leachate migration from the reject coal pile has contaminated the water-table aquifer with sulfate (up to 22 200 mg L?1) and metals (Fe to 9560 mg L?1) Al to 1110 mg L?1) A shallow plume of contaminated ground water parallels the reject coal pile and is elongated in the direction of ground-water movement. The plume is migrating westward at a calculated velocity range of 6.3 to 10.4 m yr?1. Leachate migration from the reject coal has produced a highly acidic (pH down to 2.2) and highly saline (EC to 11.8 dS m?1) rooting zone and is most likely responsible for the stressed vegetation in the study area.  相似文献

A method was developed for determining the amount of seed and skin proanthocyanidin extraction into wines by direct measurement. This method was based upon the analysis of proanthocyanidin cleavage products after acid catalysis in the presence of excess phloroglucinol. On the basis of the analysis of proanthocyanidin extracts from grape tissues, two observations were made as follows: (i) the seed and skin proanthocyanidin extension subunit compositions were considerably different from each other, and (ii) their composition did not vary with extraction time. Thus, by comparing the proportional extension subunit composition of proanthocyanidins in wine relative to their proportional composition in corresponding grape seed and skin, it was possible to determine the contribution of each to wine. To provide additional information, the procedure was used to investigate seed and skin proanthocyanidin extraction during commercial-scale fermentations that had undergone 4 or 10 day low temperature prefermentation skin contact prior to the onset of fermentation. The results for both fermentations indicated that the proportion of skin tannin declined during fermentation and also showed that at the end of fermentation the amount and proportion of skin tannin were the same.  相似文献

Cereal anchorage failure, or lodging, is the permanent displacement of a crop from the vertical and results in significant annual yield losses globally. Several factors have been identified as contributors to this phenomenon but the precise mechanisms of failure are still largely unknown because of difficulties in observing these processes as they occur in situ. To identify potential soil management practices to minimize losses associated with cereal root failure, an understanding of the nature of root‐soil interactions attributed to lodging is needed. An experiment was conducted that involved field impregnation and subsequent thin sectioning of lodged and unlodged root‐soil complexes from contrasting soils, cereal crops, and management practices to elucidate the effects of lodging on soil structure and porous architecture. Using image analysis, size and distribution of pores in soils were quantified at both meso‐ (100–30 μm) and microscales (<30 μm). A significant effect of lodging on porosity was recorded whereby lodging reduced total porosity through compaction created by movement of the stem base, although this was variable among soil types. Pore‐size distributions comprehensively supported these trends since alteration in the relative frequency of pores within specific size classes was clearly observed. The effects of lodging were more pronounced at the mesoscale because the data were more susceptible to variations created by natural soil heterogeneity at the microscale. These data suggested that sideways movement of the subterranean stem within the soil is a significant factor which is likely to affect the propensity for a cereal plant to lodge, indicating soil strength in the upper part of the soil profile is crucial.  相似文献

Two genes of wheat low-molecular-weight glutenin subunits (LMW-GS), B16 and P73, were cloned and expressed in E. coli. They were homologous to proteins encoded respectively at Glu-B3 and Glu-D3 loci. The N-terminal and C-terminal halves of B16 (NB16 and B16C) and the two chimeras combining the halves of the two genes (B16-P73 and P73- B16) were also expressed. All these constructs were compared for their reactivity with IgE from 24 patients suffering from different forms of wheat allergies. The results confirmed that LMW-GSs bound IgE in all adult allergies tested. Strong differences in reactivity between all the constructs were observed. They were disease-dependent. In wheat-dependent exercise-induced anaphylaxis (WDEIA), the reactivity of the constructs depended partly on common epitopes with omega-5 gliadins but also on differences in molecule conformation. The presence of NB16 in the constructs greatly influenced their IgE reactivity.  相似文献

Interactions of proline-rich proteins (PRPs) with flavan-3-ols was studied using poly(L-proline) as a model protein by means of isothermal titration calorimetry (ITC). Several parameters were varied: (i) the galloylation and B-ring trihydroxylation of the flavan-3-ols (catechin, epicatechin, epicatechin gallate, and epigallocatechin gallate) and (ii) the degree of polymerization (monomers were compared to a mixture of oligomers with average degree of polymerization of 3.85). Large differences were observed between the flavan-3-ol monomers: no enthalpy change was measured when catechin and epicatechin were titrated by poly(L-proline), whereas thermodynamic parameters were determined in the case of galloylated monomers and mixture of oligomers. Stoichiometry ranged from 1 oligomer bound for each 12 proline units to 1 galloylated monomer for each 8 or 10 proline units. Association constants were in the range of 10(4)-10(5) M(-1), indicating a relatively high affinity of galloylated flavanols toward poly(L-proline), and the coexistence of both enthalpy- and entropy-driven phenomena was suggested. Finally, the binding of grape seed tannins to proteins was shown to be a cooperative process.  相似文献

In France, as in many other European countries, Mycosphaerella graminicola (Fuckel) Schr?ter in Cohn (anamorph Septoria tritici), the causal agent of wheat leaf blotch, is controlled by foliar applications of fungicides. With the recent generalization of resistance to strobilurins (QoIs), reliable control is mainly dependent upon inhibitors of sterol 14 alpha-demethylation (DMIs). To date, strains with reduced sensitivity to DMIs are widespread, but disease control using members of this class of sterol biosynthesis inhibitors has not been compromised. In this study, sensitivity assays based on in vitro effects of fungicides towards germ-tube elongation allowed the characterization of seven DMI-resistant phenotypes. In four of them, cross-resistance was not observed between all tested DMIs; this characteristic concerned prochloraz, triflumizole, fluquinconazole and tebuconazole. Moreover, the highest resistant factors to most DMIs were found only in recent isolates; according to their response towards prochloraz, they were classified into two categories. Molecular studies showed that DMI resistance was associated with mutations in the CYP51 gene encoding the sterol 14 alpha-demethylase. Alterations at codons 459, 460 and 461 were related to low resistance levels, whereas, at position 381, a valine instead of an isoleucine, in combination with the previous changes, determined the highest resistance levels to all DMIs except prochloraz. Mutations in codons 316 and 317 were also found in some isolates exhibiting low resistance factors towards most DMIs.  相似文献

In contrast to mammals and birds, fish display an amazing diversity of genetic sex determination systems, with frequent changes during evolution possibly associated with the emergence of new sex chromosomes and sex‐determining genes. To better understand the molecular and evolutionary mechanisms driving this diversity, several fish models are studied in parallel. Besides the medaka (Oryzias latipes Temminck and Schlegel, 1846) for which the master sex‐determination gene has been identified, one of the most advanced models for studying sex determination is the Southern platyfish (Xiphophorus maculatus, Günther 1966). Xiphophorus maculatus belongs to the Poeciliids, a family of live‐bearing freshwater fish, including platyfish, swordtails and guppies that perfectly illustrates the diversity of genetic sex‐determination mechanisms observed in teleosts. For X. maculatus, bacterial artificial chromosome contigs covering the sex‐determination region of the X and Y sex chromosomes have been constructed. Initial molecular analysis demonstrated that the sex‐determination region is very unstable and frequently undergoes duplications, deletions, inversions and other rearrangements. Eleven gene candidates linked to the master sex‐determining gene have been identified, some of them corresponding to pseudogenes. All putative genes are present on both the X and the Y chromosomes, suggesting a poor degree of differentiation and a young evolutionary age for platyfish sex chromosomes. When compared with other fish and tetrapod genomes, syntenies were detected only with autosomes. This observation supports an independent origin of sex chromosomes, not only in different vertebrate lineages but also between different fish species.  相似文献
