Production and functional characterization of recombinant bovine interleukin-8 as a specific neutrophil activator and chemoattractant

Interleukin-8, a member of the chemokine family of cytokines, is a potent neutrophil chemoattractant in many non-rodent species. In this study, recombinant bovine interleukin-8 (rbIL-8) was expressed in bacteria as a glutathione-S-transferase fusion protein. The fusion protein was purified by glutathione-Sepharose affinity chromatography and recombinant rbIL-8 was eluted by cleaving with thrombin. The purified rbIL-8 molecule was approximately 8 kDa and was confirmed as authentic IL-8 by Western analysis. Recombinant bovine IL-8 induced specific dose-dependent in vitro chemotaxis of neutrophils at doses as low as 1.0 ng/ml, and this activity was inhibited by pre-treatment of rbIL-8 with a monoclonal antibody to ovine IL-8. Neutrophils exposed to rbIL-8 developed pseudopodia and became elongated as determined by microscopic analysis and flow cytometry. Injection of 3.3 ng to 3.3 microg of rbIL-8 into the skin of a normal calf induced dose-dependent recruitment of neutrophils but not eosinophils. Intravascular margination of neutrophils was obvious at the injection sites from 15 to 60 min after administration of rbIL-8, and extravascular neutrophil numbers increased steadily from 1 to 18 h after injection. Neutrophils with morphologic features of apoptosis were detected in these lesions at 18 and 30 h after injection, and this correlated with reduction in the number of dermal neutrophils. These results confirm unequivocally that bovine IL-8 functions as a neutrophil, but not an eosinophil, chemoattractant in vitro and in vivo.     

Physiological and biochemical consequences of electroimobilisation in conscious sheep

An electroimmobilisation device has been developed to facilitate the automated shearing of sheep, but there is little information on its effects on the body. We have studied its effects on the cardiovascular system and on intermediary metabolism in sheep. Electroimmobilisation caused statistically significant increases in mean arterial pressure, heart rate, cardiac output, renal and hepatic and hindquarter glucose and lactate flux, organ and whole body oxygen flux, hindquarter blood flow and core temperature and decreases in arterial and posterior vena cava blood pH, renal and hepatic blood flow and PaCO2. Notably, no change occurred in PaO2. The metabolic changes demonstrated the capacity of sheep to respond to the increased muscular and cardiovascular work induced by electroimmobilisation. Pulmonary function was not compromised during electroimmobilisation as judged from blood gas changes, and the acid/base changes were rapidly reversed after electroimmobilisation. The recovery to control conditions for all perturbations generally took no longer than 30 min, consistent with a rapid and physiologically adequate reversal by the animal's homeostatic mechanisms.     

Alterations in the bovine bronchoalveolar lavage proteome induced by dexamethasone

Stressors such as transportation, weaning and co-mingling increase susceptibility to bacterial pneumonia in cattle and are associated with elevated levels of endogenous glucocorticoids. To determine the effect of glucocorticoids on the proteins expressed in the fluid lining the respiratory tract, bronchoalveolar lavage (BAL) was performed on cattle treated with dexamethasone or saline and proteins were resolved by two-dimensional electrophoresis (2-DE). Significant changes in expression were observed for 9 of the 363 detected spots, and the identities of these proteins were determined by mass spectrometry. Consistent with the initiation of an acute phase response, the expression of alpha-1-acid glycoprotein (orosomucoid) and alpha-1-antitrypsin was increased and alpha-2-HS-glycoprotein (fetuin) was decreased in the BAL fluid of dexamethasone-treated cattle. In addition, dexamethasone induced the expression of two hydrophobic ligand-binding proteins, adipocyte-fatty acid binding protein and odorant binding protein (OBP), as well as the proteins alpha-enolase, cofilin-1 and immunoglobulin J chain. OBP mRNA expression in bronchial biopsies was quantified by real-time RT-PCR and the 6-fold higher levels of expression observed in dexamethasone- versus saline-treated animals correlated with the changes observed in OBP protein level. These findings demonstrate glucocorticoid-dependent changes in the protein composition of the epithelial lining fluid of the respiratory tract, identifying proteins potentially integral to respiratory disease susceptibility.     

Organic Glasses: A New Class of Photorefractive Materials

The performance of amorphous organic photorefractive (PR) materials in applications such as optical data storage is generally limited by the concentration of active molecules (chromophores) that can be incorporated into the host without forming a crystalline material with poor optical quality. In polymeric PR systems described previously, performance has been limited by the necessity of devoting a large fraction of the material to inert polymer and plasticizing components in order to ensure compositional stability. A new class of organic PR materials composed of multifunctional glass-forming organic chromophores is described that have long-term stability and greatly improved PR properties.     

The Origin of the Great Bend of the Nile from SIR-C/X-SAR Imagery

The course of the Nile in northern Sudan follows a contorted path through Precambrian bedrock. Radar imagery shows that basement structures control the river's course in this region. Northward-flowing segments follow Precambrian fabrics, whereas east-west segments follow faults of much younger age. These faults may reflect recent uplift of the Nubian Swell and deflection of the river to the southwest to form the great bend of the Nile.
