This study evaluated the performance of an immunochromatographic test (ICT) for the diagnosis of canine brucellosis caused by Brucella canis, comparing its results with that of the rapid slide agglutination test with and without the use of 2‐mercaptoethanol and the agar gel immunodiffusion test (AGID). The microbiological culture, PCR and clinical examination were used as reference. According to the results obtained in clinical examination, blood culture, culture of semen and vaginal swab and PCR in blood, semen and vaginal swab, a total of 102 dogs were divided into three groups: B. canis‐infected dogs (Group 1), B. canis‐non‐infected dogs (Group 2) and dogs with suspected brucellosis (Group 3). The diagnostic sensitivity of RSAT, 2ME‐RSAT, AGID and ICT in Group 1 was, respectively, 75%, 37.5%, 27.8% and 89.58%. The diagnostic specificity of RSAT, 2ME‐RSAT, AGID and ICT in Group 2 was, respectively, 91%, 100%, 100%, and 100%. In dogs with suspected brucellosis, 9.67% were RSAT positive, none was positive by 2ME‐RSAT, 3.22% were AGID positive and 6.45% were ICT positive. The main drawback concerning canine brucellosis diagnosis is the lack of a highly sensitive serological assay to be used as a screening test to the rapid identification of infected animals. The ICT showed a high diagnostic specificity and a diagnostic sensitivity value greater than that observed in the RSAT, 2ME‐RSAT and AGID. However, 10.41% of infected dogs had negative results by ICT. These dogs were positive by microbiological culture and/or PCR, indicating active infection and consequently a higher potential of spreading Brucella. Global Oscillation Network Group data reveal that the internal structure of the sun can be well represented by a calibrated standard model. However, immediately beneath the convection zone and at the edge of the energy-generating core, the sound-speed variation is somewhat smoother in the sun than it is in the model. This could be a consequence of chemical inhomogeneity that is too severe in the model, perhaps owing to inaccurate modeling of gravitational settling or to neglected macroscopic motion that may be present in the sun. Accurate knowledge of the sun's structure enables inferences to be made about the physics that controls the sun; for example, through the opacity, the equation of state, or wave motion. This study was conducted to evaluate the influence of a vancomycin pretreatment on the ability of marker (nalidixic-acid resistant) Salmonella Enteritidis (SE(M)), field Salmonella Enteritidis (SE(E)), and marker Salmonella Typhimurium (ST(M)) strains to colonize within the intestinal and reproductive tracts and translocate to other organs of leghorn laying hens. In each of three trials, caged laying hens (76, 26, and 33 wk ofage) were divided into six groups designated to receive SE(M), SE(F), or ST(M), and half were pretreated with vancomycin (n = 11-12 hens). Vancomycin-treated hens received 10 mg vancomycin in saline/kilogram body weight orally for 5 days to inhibit Gram-positive bacteria within the intestines. On Day 6, all hens were concurrently challenged by oral, intravaginal, and intracolonal routes with Salmonella and placed into separate floor chambers by Salmonella strain. Two weeks postinoculation, all hens were euthanatized and the ceca, spleen, liver/gall bladder (LGB), upper (URT), and lower (LRT) reproductive tracts, and ovarian follicles were aseptically collected, and analyzed for Salmonella. Results did not differ for the three hen's ages and were therefore combined. The vancomycin pretreatment also had no significant effect on the colonization ability of SE(M), SE(F) or ST(M), and therefore results were combined within Salmonella strain. The marker strain of Salmonella Enteritidis was recovered from 21% of ceca, 4% of LGB, 9% of LRT, and 17% of the fecal samples. The field strain of Salmonella Enteritidis was recovered from 88% of ceca, 96% of spleen, 92% of LGB, 30% of LRT, 4% of URT, 13% of follicle, and 42% of the fecal samples. The marker strain of Salmonella Typhimurium was recovered from 100% of ceca, 74% of spleen, 91% of LGB, 30% of LRT, 9% of URT, 9% of follicle, and 100% of the fecal samples. Among ceca, spleen, LGB, and fecal samples, SE(F) and ST(M) colonization was significantly greater than SE(M) colonization. Overall prevalence of Salmonella in the reproductive tracts of challenged hens was relatively low, ranging from 4% to 30%. 相似文献

ABSTRACTTo determine the minimum age/size at which Southern Flounder, Paralichthys lethostigma, can safely be moved to outdoor rearing facilities in Texas, we examined survival of simulated temperature drops in two distinct life stages: premetamorphic larvae and two size classes of postmetamorphic juveniles (small = 9.8 ± 0.3 mm in TL; large = 19.7 ± 0.6 mm). Temperature was lowered by ?0.33°C/h to 4°C, 7°C, or 10°C, held for 48 h and then raised at +0.33°C/h back to normal rearing temperature. Fish were monitored daily for survival. Larger postmetamorphic flounder had high survival for all temperature treatments (89%–100% survival), whereas both premetamorphic larvae and smaller postmetamorphic juveniles had low survival (<30%) for all temperature treatments. 相似文献

The effect of melatonin implants administered during non‐breeding season in Rasa Aragonesa rams on sperm motility parameters and other reproductive traits was assessed. In a first experiment, two Rasa Aragonesa rams were implanted (with melatonin group M), remaining other two males as control group (C). Semen of each group was collected from 1 May to 23 June, twice or three times a week, and motility parameters were assessed using a computer‐assisted sperm analysis system. Melatonin increased the percentage of progressive motile spermatozoa, particularly during 46–75 days after melatonin implantation (p < 0.01). In experiment 2, M and C in vitro fertilization ability had been determined by zona‐pellucida binding assays, using spermatozoa from experiment 1, obtained 60–70 days after melatonin was implanted. A significantly higher number of spermatozoa attached per oocyte was observed in frozen‐thawed immature ovine oocytes incubated with sperm from M animals than in those incubated with sperm from the C group (p < 0.01). Finally, a field assay (experiment 3) was performed. In this case, five Rasa Aragonesa rams were implanted with melatonin and three remained as control group. Sperm doses from those animals were used for artificial insemination of 2608 Rasa Aragonesa ewes from 39 different farms at non‐breeding season. Fertility, litter size and fecundity were studied. Semen from melatonin implanted rams seemed to increase both fertility and fecundity in ewes inseminated with spermatozoa obtained 46–60 days after implantation (p < 0.1). Thus, melatonin treatment in rams during non‐breeding season modifies sperm motility parameters and seems to improve the fertilization parameters obtained. 相似文献

BACKGROUND: Four‐arm olfactometer bioassays were conducted to assess the behavioural responses of the adult maize weevil, Sitophilus zeamais (Motschulsky) (Coleoptera: Curculionidae), to harvested seeds of host plants, i.e. white maize, yellow maize (Zea mays L.) and winter wheat (Triticum aestivum L.) (Poaceae), and non‐host plant materials, i.e. alligator pepper, Aframomum melegueta (Rosk) K. Schum (Zingiberaceae), rhizomes of ginger, Zingiber officinale (Roscoe) (Zingiberaceae), and West African black pepper, Piper guineense Thonn and Schum (Piperaceae). Additional bioassays with host plant volatiles were conducted in the presence of three doses of non‐host plant materials. RESULTS: Both sexes of the weevil showed strong attraction to maize and wheat seed volatiles, but were significantly repelled (P < 0.001) by odours from A. melegueta, Z. officinale and P. guineense. Furthermore, S. zeamais avoided maize and wheat seeds presented in combination with the non‐host plant material at 10% (w/w) and 33% (w/w) levels. CONCLUSIONS: A. melegueta, Z. officinale and P. guineense have the potential for use in the protection of stored grains by resource‐poor farmers with local access to these plants. Wide-angle seismic data along the Mantle Electromagnetic and Tomography (MELT) arrays show that the thickness of 0.5- to 1. 5-million-year-old crust of the Nazca Plate is not resolvably different from that of the Pacific Plate, despite an asymmetry in depth and gravity across this portion of the East Pacific Rise. Crustal thickness on similarly aged crust on the Nazca plate near a magmatically robust part of the East Pacific Rise at 17 degrees15'S is slightly thinner (5.1 to 5.7 kilometers) than at the 15 degrees55'S overlapping spreading center (5.8 to 6.3 kilometers). This small north-south off-axis crustal thickness difference may reflect along-axis temporal variations in magma supply, whereas the across-axis asymmetry in depth and gravity must be caused by density variations in the underlying mantle. 相似文献

Seafloor diking-eruptive events represent the irreducible, quantum events of upper oceanic crustal accretion. They record events by which a large portion of the oceanic crust has formed through geological history. Since 1993, the U.S. Navy's real-time Sound Surveillance System has allowed location of ongoing acoustic signatures of dike emplacement and basalt eruptions at ridge crests in the northeast Pacific. These diking-eruptive events trigger a sequence of related, rapidly evolving physical, chemical, and biological processes. Magmatic volatiles released during these events may provide nutrients for communities of subseafloor microorganisms, some of which thrive in high-temperature anaerobic environments. Many of the organisms identified from these systems are Archaea. If microorganisms can thrive in the water-saturated pores and cracks within deep, volcanically active portions of our planet, other hydrothermally active planets may harbor similar life forms. 相似文献
