Dihydrodipicolinate synthase (DHDPS) (EC 4.2.1.52) and dihydrodipicolinate reductase (DHDPR) (EC 1.3.1.26) have attracted much recent attention as potential herbicide targets. DHDPS was feedback-inhibited by (S)-lysine; inhibition was reversible and uncompetitive with respect to both (S)-ASA and pyruvate. Homoserine lactone was a reversible non-competitive inhibitor of DHDPS with respect to both (S)-ASA and pyruvate. (R)-Cysteine sulfinic acid and (S)-glutamic acid were reversible uncompetitive inhibitors of DHDPS with respect to (S)-ASA. (S)-Aspartic acid was a reversible mixed-type inhibitor. Dipicolinic acid was a reversible competitive inhibitor of DHDPR with respect to the substrate (4S)-4-hydroxy-2,3,4,5-tetrahydro-(2S)-dipicolinic acid, as was isophthalic acid. Δ3-Tetrahydroisophthalic acid was a moderate inhibitor of both DHDPS and DHDPR. Phytophthora austrocedri is a recently invasive soilborne pathogen which is causing widespread mortality of Juniperus communis in northern Britain. The pathways by which a single genotype of P. austrocedri has spread to infect such a geographically dispersed range of woodland sites within a relatively short timeframe are unknown. This study examined the detectability of P. austrocedri in soil and water within infected J. communis woodland using qPCR to gain a better understanding of the pathogen's key mechanisms of spread. A Phytophthora metabarcoding method was also applied to investigate the wider diversity of Phytophthora species present in water at one of the sites. qPCR analyses of P. austrocedri in soil samples at a J. communis woodland exhibiting low‐to‐moderate levels of disease suggested a slow natural spread of the pathogen in soil, requiring high moisture conditions. However, the ubiquity of P. austrocedri DNA in soil samples collected across a heavily infected J. communis site suggests that once established at a site the pathogen can be spread readily in soil locally, most likely vectored by animal movements and/or human activities. The hypothesis that P. austrocedri is aerially transmitted in rainwater was not adequately proven, and an alternative hypothesis for the widespread distribution of the pathogen on J. communis in northern Britain is presented. Metabarcoding identified DNA from a diverse range of Phytophthora species in river and rainwater samples although the main target pathogen, P. austrocedri, was not amplified which disagreed with some of the qPCR findings. Possible reasons for this are discussed.  相似文献

In this study, an attempt was made to use vaginal electrical impedance to predict calving in a female white rhinoceros (Ceratotherium simum simum) and to determine the relationship between vaginal electrical impedance and hormonal profiles during pregnancy. The principle behind vaginal electrical impedance is that a change in the ionic balance of vaginal and cervical mucus occurs in response to changes in reproductive hormones. Three times weekly vaginal electrical impedance readings and fecal samples were collected from midgestation to calving (a 6-mo period). The extracted fecal samples were analyzed for immunoreactive estrogens, progestagens, and corticoids by RIA. Vaginal electrical impedance readings did not decrease before calving but remained consistent throughout the last 140 days of pregnancy. Fecal progestagens in the white rhinoceros decreased between day 17 and day 1 before calving, whereas estrogens increased between 4 and 2 mo before calving, with an additional increase occurring 1 mo before calving. Fecal corticoids increased 5 mo before calving, slowly declined, and increased again within 3 wk before calving. A decline in vaginal electrical impedance was noted 168 days before calving and remained at low levels for 4 wk. At the time of this decrease, the female became aggressive toward the male and began lactating. Fecal progestagens and estrogens did not change during this time; however, fecal corticoids increased as vaginal electrical impedance readings returned to normal along with her behavior and cessation of lactation. In summary, the use of vaginal electrical impedance could not predict parturition in the white rhinoceros. However, an anomaly occurred during pregnancy that was supported by vaginal electrical impedance readings, a change in female behavior, premature lactation, and a subsequent increase in fecal corticoids. The etiology of this physiological anomaly is unknown, yet it did not compromise pregnancy.  相似文献

Thiopurine methyltransferase (TPMT) produces inactive metabolites of azathioprine and, in humans, has a variable amount of activity. Humans with low TPMT activity commonly develop myelotoxicity when receiving azathioprine. Our study sought to characterize the distribution of TPMT activity in a population of dogs and to determine whether the pretreatment knowledge of red blood cell (RBC) TPMT activity could predict myelotoxicity in dogs receiving azathioprine. RBC TPMT activity was measured in 299 healthy dogs, and 9 dogs that represented a wide range of enzyme activity received azathioprine at a standard therapeutic dose for 30 days. TPMT activity in healthy dogs was log normally distributed and varied over an approximately 7-fold range. Geometric mean, minimum, and maximum RBC TPMT activities were 37.1, 16.3, and 115 nmol per gram of hemoglobin (gHb) per hour, respectively. TPMT deficiency was not identified. Two populations of TPMT activity in dogs were detected by statistical modeling (commingling analysis). Dogs with intermediate TPMT activity (14-38 nmol/gHb/h) receiving azathioprine had significantly lower neutrophil counts during week 4 than during weeks 0-3, whereas those with high activity (>39 nmol/gHb/h) did not have a significant change in neutrophil count. An analysis of TPMT activity in 6 dogs with a history of azathioprine-associated myelotoxicity in a clinical setting revealed either intermediate or high TPMT enzyme activity in all dogs, suggesting that TPMT activity, as measured in RBCs, is not the sole cause of severe azathioprine-associated myelosuppression in dogs.  相似文献

Complications of renal biopsies are well documented except for the change in renal function after a biopsy. Eighteen healthy, adult cats were divided into two groups (n = 9 cats/group). For the measurement of global and split renal function, Group 1 used the renal uptake of 99mTc-DTPA and Group 2 used the renal uptake of 99mTc-MAG3. Scintigraphic data were collected on days (-4), (-3), 0, 1, 2, and 4 post renal biopsy. Using ultrasound guidance, biopsies were taken from the right renal cortex on dO, before acquiring scintigraphic images. P - values less than 0.10 were considered significant due to the limited number of observations. The only statistically significant change (p = 0.08) in global renal function detected was by day following a unilateral renal biopsy. Cats imaged using 99mTc-MAG3 had discernible liver activity. A unilateral, ultrasound guided renal biopsy has minimal effect on renal function in normal, healthy sedated cats.  相似文献

Work on farms and in restaurants is characterized by highly gendered and racialized divisions of labor, low wages, and persistent inequalities. Gender, race, and ethnicity often determine the spaces where people work in the food system. Although some research focuses on gendered divisions of labor in restaurants and on farms, few efforts look more broadly at intersectional inequalities in food work. Our study examines how inequality is perpetuated through restaurant and farm work in the United States and, specifically, how gender and race/ethnicity influence where people work, their tasks and responsibilities, and their work experiences. In describing restaurant work, people in the restaurant industry typically refer to the front and back of the house to distinguish between different working spaces, jobs, and workers. We use this spatial metaphor of front and back of the house to analyze intersectional inequalities of food work in restaurants and on farms. The data derive from conversations with 63 restaurant and farm owners, managers, and workers in California and Pennsylvania. Our findings suggest that gendered and racialized bodies often define who works in the front and back of the “house,” and that owners and workers often naturalize gender and racial divisions of labor in food work. Despite these patterns, we found evidence of attempts to reduce these inequalities on farms and in restaurants.  相似文献

Many economically important cattle diseases spread between herds through livestock movements. Traditionally, most transmission models have assumed that all purchased cattle carry the same risk of generating outbreaks in the destination herd. Using data on bovine viral diarrhoea virus (BVDV) in Scotland as a case example, this study provides empirical and theoretical evidence that the risk of disease transmission varies substantially based on the animal and herd demographic characteristics at the time of purchase. Multivariable logistic regression analysis revealed that purchasing pregnant heifers and open cows sold with a calf at foot were associated with an increased risk of beef herds being seropositive for BVDV. Based on the results from a dynamic within-herd simulation model, these findings may be partly explained by the age-related probability of animals being persistently infected with BVDV as well as the herd demographic structure at the time of animal introductions. There was also evidence that an epidemiologically important network statistic, “betweenness centrality” (a measure frequently associated with the potential for herds to acquire and transmit disease), was significantly higher for herds that supplied these particular types of replacement beef cattle. The trends for dairy herds were not as clear, although there was some evidence that open heifers and open lactating cows were associated with an increased risk of BVDV. Overall, these findings have important implications for developing simulation models that more accurately reflect the industry-level transmission dynamics of infectious cattle diseases.  相似文献

HistoryA four year old male neutered Domestic Short Hair cat presented for general anaesthesia for hind limb orthopaedic surgery. The cat had been anaesthetized four days previously with propofol and isoflurane and made an uneventful recovery.Physical Examination and ManagementOn pre-anaesthetic examination the cat had a temperature of 38.9 °C and was otherwise healthy. After a premedication of acepromazine and pethidine, anaesthesia was induced with thiopental and maintained with isoflurane in oxygen 50% and nitrous oxide 50%. Increases in heart rate, respiratory rate, end tidal carbon dioxide tension and temperature were observed, occurring sequentially, from 110 to 175 minutes after anaesthetic induction. Despite ceasing all warming measures and attempting to cool the patient, body temperature continued to rapidly rise, reaching 42.5 °C and limb rigidity was observed. Isoflurane administration was stopped and esmolol was administered. Cardiac arrest occurred. Cardio-pulmonary cerebral resuscitation was commenced and a lateral thoracotomy was performed to allow cardiac compressions and internal defibrillation. Atropine, adrenaline, glucose and dopamine were administered and cold saline was instilled into the thoracic cavity.Follow-upResuscitation was unsuccessful and the cat died.ConclusionsA presumptive diagnosis of malignant hyperthermia was made. Malignant hyperthermia should be considered, even if prior exposure to volatile inhalational anaesthesia was uneventful, and prompt and aggressive therapy instituted.  相似文献
