Racemic gossypol and its related derivatives gossypolone and apogossypolone demonstrated significant growth inhibition against a diverse collection of filamentous fungi that included Aspergillus flavus, Aspergillus parasiticus, Aspergillus alliaceus, Aspergillus fumigatus, Fusarium graminearum, Fusarium moniliforme, Penicillium chrysogenum, Penicillium corylophilum, and Stachybotrys atra. The compounds were tested in a Czapek agar medium at a concentration of 100 μg/mL. Racemic gossypol and apogossypolone inhibited growth by up to 95%, whereas gossypolone effected 100% growth inhibition in all fungal isolates tested except A. flavus. Growth inhibition was variable during the observed time period for all tested fungi capable of growth in these treatment conditions. Gossypolone demonstrated significant aflatoxin biosynthesis inhibition in A. flavus AF13 (B(1), 76% inhibition). Apogossypolone was the most potent aflatoxin inhibitor, showing greater than 90% inhibition against A. flavus and greater than 65% inhibition against A. parasiticus (B(1), 67%; G(1), 68%). Gossypol was an ineffectual inhibitor of aflatoxin biosynthesis in both A. flavus and A. parasiticus. Both gossypol and apogossypolone demonstrated significant inhibition of ochratoxin A production (47%; 91%, respectively) in cultures of A. alliaceus.  相似文献

To study the mechanism of coffee melanoidin formation, green coffee beans were prepared by (1) removal of the hot water extractable components (WECoffee); (2) direct incorporation of sucrose (SucCoffee); and (3) direct incorporation of type II arabinogalactan-proteins (AGPCoffee). As a control of sucrose and AGP incorporation, lyophilized green coffee beans were also immersed in water (control). The original coffee and the four modified "in bean" coffee models were roasted and their chemical characteristics compared. The formation of material not identified as carbohydrates or protein, usually referred to as "unknown material" and related to melanoidins, and the development of the brown color during coffee roasting have distinct origins. Therefore, a new parameter for coffee melanoidin evaluation, named the "melanoidin browning index" (MBI), was introduced to handle simultaneously the two concepts. Sucrose is important for the formation of colored structures but not to the formation of "unknown material". Type II AGPs also increase the brown color of the melanoidins, but did not increase the amount of "unknown material". The green coffee hot water extractable components are essential for coffee melanoidin formation during roasting. The cell wall material was able to generate a large amount of "unknown material". The galactomannans modified by the roasting and the melanoidin populations enriched in galactomannans accounted for 47% of the high molecular weight brown color material, showing that these polysaccharides are very relevant for coffee melanoidin formation.  相似文献

This study presents the application of multivariate statistical tools for the evaluation of spatial variations and the interpretation of water quality data obtained in a monitoring program of Lis river basin surface water, Portugal. Twenty-seven physicochemical and microbiological parameters were determined in six water sampling campaigns at 16 monitoring sites during the period from September 2003 to November 2006. Correlation analysis, principal component analysis, and cluster analysis were performed to evaluate the main water pollution sources and to characterize the spatial distribution of water pollution profiles in river basin. The results achieved with the statistical methodologies led to distinguish natural and anthropogenic pollution sources. Additionally, monitoring sites with similar water pollution profile were identified, indicating that some monitoring locations can be changed to improve the spatial characterization of water quality in the river basin. CBO, CQO, P, and N were identified as significant variables affecting spatial variations, namely in the Lis river middle reach. Besides the identification of main pollution sources, the applied statistical tools were able to identify spatial patterns of water pollution in Lis river basin, which further helps in the reassessment of the number and location of monitoring sites.  相似文献

In the tropics,frequent nitrogen(N)fertilization of grazing areas can potentially increase nitrous oxide(N2O)emissions.The application of nitrification inhibitors has been reported as an effective management practice for potentially reducing N loss from the soil-plant system and improving N use efficiency(NUE).The aim of this study was to determine the effect of the co-application of nitrapyrin(a nitrification inhibitor,NI)and urea in a tropical Andosol on the behavior of N and the emissions of N2O from autotrophic and heterotrophic nitrification.A greenhouse experiment was performed using a soil(pH 5.9,organic matter content 78 g kg-1,and N 5.6 g kg-1)sown with Cynodon nlemfuensis at 60%water-filled pore space to quantify total N2O emissions,N2O derived from fertilizer,soil ammonium(NH4+)and nitrate(NO3-),and NUE.The study included treatments that received deionized water only(control,NI).No significant differences were observed in soil NH4+content between the UR and UR+NI treatments,probably because of soil mineralization and NO3-produced by heterotrophic nitrification,which is not effectively inhibited by nitrapyrin.After 56 d,N2O emissions in UR(0.51±0.12 mg N2O-N concluded that the soil organic N mineralization and heterotrophic nitrification are the main processes of NH4+and NO3-production.Additionally,it was found that N2O emissions were partially a consequence of the direct oxidation of the soil's organic N via heterotrophic nitrification coupled to denitrification.Finally,the results suggest that nitrapyrin would likely exert significant mitigation on N2O emissions only if a substantial N surplus exists in soils with high organic matter content.  相似文献

MyD88 is a key downstream adapter for most Toll-like receptors (TLRs) and interleukin-1 receptors (IL-1Rs). MyD88 deficiency in mice leads to susceptibility to a broad range of pathogens in experimental settings of infection. We describe a distinct situation in a natural setting of human infection. Nine children with autosomal recessive MyD88 deficiency suffered from life-threatening, often recurrent pyogenic bacterial infections, including invasive pneumococcal disease. However, these patients were otherwise healthy, with normal resistance to other microbes. Their clinical status improved with age, but not due to any cellular leakiness in MyD88 deficiency. The MyD88-dependent TLRs and IL-1Rs are therefore essential for protective immunity to a small number of pyogenic bacteria, but redundant for host defense to most natural infections.  相似文献

Purpose

The vegetation of the Campinaranas occurs in humid areas with hydromorphic sandy soils at the Amazon region. Thus, the determination and in situ monitoring of moisture content in Campinarana soils, besides the detection of subsurface layers are key measures for studying these soil–vegetation systems. Also, the application of ground penetrating radar (GPR) in deep sandy sedimentary sequence of Amazonia is a promising tool to enhance the knowledge on depositional and soil formation features.

Materials and methods

We studied representative soils of the Campinaranas at the National Park of Viruá, state of Roraima (Brazilian Amazonia), through the use of geophysical methods (soil moisture sensors and GPR). The study was applied in four sandy soils. Besides chemical and physical analysis of soils, soil moisture sensors were installed for monitoring during an entire hydrological year (2010/2011), and performed the calibration of sensors , coupled with imaging of the soil along transects with GPR.

Results and discussion

As a result of calibration of the soil moisture sensors we obtained a general equation with an R 2 greater than 0.9. There is an influence of soil moisture content and soil temperature in the distribution of vegetation types in Campinaranas. The use of GPR identified some determinants characteristics in these soils for the differentiating the Campinaranas, represented by spodic and C horizons.

Conclusions

The spodic horizons in soils under Forest Campinarana provided potential errors in the determination of soil moisture, requiring calibration data for the precise use of this device. The investigation of the soil through the GPR showed interesting results, which allowed continuous visualization of the main soil horizons along transects in the phytophysiognomies of Campinaranas.  相似文献

L-galactono-gamma-lactone dehydrogenase (GLDH) activity and vitamin C content as ascorbic acid (AA) plus dehydroascorbic acid (DHA) were evaluated in five potato tuber cultivars (Agata, Altesse, Franceline, Manon, and Monalisa). The effect of fresh-cutting and subsequent refrigerated storage of Manon potato under different atmospheres (air, 20% CO(2) + air, 100% N(2), and vacuum packaging) on GLDH activity and vitamin C content was also determined. GLDH from the five potato tuber cultivars showed typical inhibition kinetics by high substrate concentration in the synthesis of AA from its physiological precursor L-galactonic acid-gamma-lactone (GL). GLDH activity was not correlated with the corresponding vitamin C content in any potato tuber cultivar. GLDH from all the cultivars presented a major isoform with isoelectric point (IEP) 5, which changed to IEP = 4.3 after minimal processing. In addition, the GLDH-catalyzed synthesis of AA by the new isoform showed typical Michaelis kinetics, in which the enzyme became more efficient to catalyze the reaction. Whether the change in the isoform pattern was due to either post-translational modifications or de novo synthesis of a new isoenzyme remains unanswered. Fresh-cutting increased GLDH activity from 4.7-fold (vacuum packaging) to 11-fold (air) after 6 days. In addition, 100% of vitamin C content was retained in air and decreased in the rest of atmospheres after this storage period, following the sequence vacuum packaging (89%) > 100% N(2) (78%) > 20% CO(2) + air (63%). This tendency was correlated with the corresponding GLDH activity detected in each storage atmosphere, except in the case of 20% CO(2) + air. Vacuum packaging proved to be the best storage condition, because fresh-cut potatoes did not turn brown and retained 89% of initial vitamin C content.  相似文献

Effect of caffeic acid on the color of red wine   总被引:4,自引:0,他引:4  
The copigmentation effect of prefermentation additions of different doses of caffeic acid was investigated during the 1997 harvest. Microfermentation with the major red grape cultivars Listán negro and Negramoll, grown in the Canary Islands, was carried out with the same protocol. Visible and UV spectra were registered periodically. HPLC chromatograms were carried out. The color enhancement of cv. Negramoll wine varied between 13 and 75% (AU at 520 nm), and that of cv. Listán negro wine between 25 and 45% at the end of fermentation. During aging these values were enhanced to reach even >100% in some cases. An initial complex of the 1:1 type, where one molecule of caffeic acid associates with one molecule of anthocyanin, has been identified using the mathematical procedure of Brouillard et al. (J. Am. Chem. Soc. 1989, 111, 2604-2610). Caffeic acid seems to contribute to color stability and protection against oxidation. The importance of nonpigment composition in pigment extraction and color retention during and after fermentation is demonstrated.  相似文献

The effect of minimal processing on polyphenol oxidase (PPO), peroxidase (POD), phenylalanine ammonia-lyase (PAL), and phenolic compounds was studied in five potato cultivars (Agria, Cara, Liseta, Monalisa, and Spunta). Minimal processing caused an overall increase in PPO, POD, and PAL activities. The isoform pattern of PPO was the same for all of the cultivars before and after processing. No latent PPO was detected. The isoperoxidase pattern was approximately the same among cultivars. An increase in POD activity was related to the specific induction of an acidic isoperoxidase. PAL showed an induction pattern characterized by the presence of a maximum peak of activity after 4 days of processing for all of the cultivars. The sequence of browning susceptibility of potato cultivars was as follows: Monalisa > Spunta > Liseta > Cara > Agria. Browning development was only partially correlated to PAL activity (only during the first 4 days after wounding). However, this correlation could not explain the above sequence of browning susceptibility. Minimal processing caused an increase of chlorogenic acid, whereas tyrosine content remained unchanged. In summary, no significant correlation was found between either rate or degree of browning and any other biochemical and physiological attribute investigated (PPO, POD, hydrogen peroxide, ascorbic acid content, and initial phenolics content as well as total and individual phenolics accumulation).  相似文献

Polyphenols present in red table grape varieties Red Globe, Flame Seedless, Crimson Seedless, and Napoleon, and the white varieties Superior Seedless, Dominga, and Moscatel Italica were analyzed by HPLC-DAD-MS. The anthocyanins peonidin 3-glucoside, cyanidin 3-glucoside (and their corresponding p-coumaroyl derivatives), malvidin 3-glucoside, petunidin 3-glucoside, and delphinidin 3-glucoside were found. In addition, caffeoyltartaric acid, p-coumaroyltartaric acid, and the flavonols quercetin 3-glucuronide, quercetin 3-rutinoside, quercetin 3-glucoside, kaempferol 3-galactoside, kaempferol 3-glucoside, and isorhamnetin 3-glucoside were detected. Flavan-3-ols were also detected, and were identified as gallocatechin, procyanidin B1, procyanidin B2, procyanidin B4, procyanidin C1, catechin, and epigallocatechin. These phenolics were present only in the skin, as the flesh of these grape cultivars was almost devoid of these compounds. Anthocyanins were the main phenolics in red grapes ranging from 69 (Crimson Seedless) to 151 (Flame Seedless) mg/kg fresh weight of grapes, whereas flavan-3-ols were the most abundant phenolics in the white varieties ranging from 52 (Dominga) to 81 (Moscatel Italica) mg/kg fresh weight of grapes. Total phenolics ranged from 115 (Dominga) to 361 (Flame Seedless) mg/kg fresh weight of grapes. This means that a serving of unpeeled table grapes (200 g) could provide up to 72 mg of total phenolics (Flame Seedless). These results indicate that the intake of unpeeled table grapes should be recommended in dietary habits as a potential source of antioxidant and anticarcinogenic phenolic compounds.  相似文献
