Our main goal was to investigate if robust chemical fingerprints could be developed for three Argentinean red wines based on organic, inorganic, and isotopic patterns, in relation to the regional soil composition. Soils and wines from three regions (Mendoza, San Juan, and Co?rdoba) and three varieties (Cabernet Sauvignon, Malbec, and Syrah) were collected. The phenolic profile was determined by HPLC-MS/MS and multielemental composition by ICP-MS; (87)Sr/(86)Sr and δ(13)C were determined by TIMS and IRMS, respectively. Chemometrics allowed robust differentiation between regions, wine varieties, and the same variety from different regions. Among phenolic compounds, resveratrol concentration was the most useful marker for wine differentiation, whereas Mg, K/Rb, Ca/Sr, and (87)Sr/(86)Sr were the main inorganic and isotopic parameters selected. Generalized Procrustes analysis (GPA) using two studied matrices (wine and soil) shows consensus between them and clear differences between studied areas. Finally, we applied a canonical correlation analysis, demonstrating significant correlation (r = 0.99; p < 0.001) between soil and wine composition. To our knowledge this is the first report combining independent variables, constructing a fingerprint including elemental composition, isotopic, and polyphenol patterns to differentiate wines, matching part of this fingerprint with the soil provenance.  相似文献

Potato virus Y (PVY) strains were originally defined by interactions with different resistance genes in standard potato cultivars. Five distinct strain groups are defined that cause local or systemic hypersensitive responses (HRs) in genetic background with a corresponding N gene: PVY(O), PVY(N), PVY(C), PVY(Z), and PVY(E). The nucleotide sequences of multiple isolates of PVY(O) and PVY(N) differ from each other by ≈8% along their genomes. Additionally, complete genome sequences of multiple recombinant isolates are composed of segments of parental PVY(O) and PVY(N) sequences. Here, we report that recombinant isolate PVY-L26 induces an HR in potato 'Maris Bard' carrying the putative Nz gene, and is not recognized by two other resistance genes, Nc and Ny(tbr). These genetic responses in potato, combined with the inability of PVY-L26 to induce vein necrosis in tobacco, clearly define it as an isolate from the PVY(Z) strain group and provide the first information on genome structure and sequence of PVY(Z). The genome of PVY-L26 displays typical features of European NTN-type isolates with three recombinant junctions (PVY(EU-NTN)), and the PVY-L26 is named PVY(Z)-NTN. Three typical PVY(NTN) isolates and two PVY(N) isolates, all inducing vein necrosis in tobacco, were compared with PVY-L26. One PVY(NTN) isolate elicited HR reactions in Maris Bard, similar to PVY-L26, while two induced a severe systemic HR-like reaction quite different from the quasi-symptomless reaction induced by two PVY(N) isolates. 'Yukon Gold' potato from North America produced HR against several PVY(NTN) isolates, including PVY-L26, but only late and limited systemic necrosis against one PVY(N) isolate. Consequently, according to symptoms in potato indicators, both PVY(Z) and PVY(NTN) isolates appeared biologically very close and clearly distinct from PVY(O) and PVY(N) strain groups.  相似文献

Botrytis cinerea is a major crop pathogen infesting >220 hosts worldwide. A cryptic species has been identified in some French populations but the new species, B. pseudocinerea, has not been fully delimited and established. The aim of this study was to distinguish between the two species, using phylogenetic, biological, morphological, and ecological criteria. Multiple gene genealogies confirmed that the two species belonged to different, well-supported phylogenetic clades. None of the morphological criteria tested (spore size, germination rate, or mycelial growth) was able to discriminate between these two species. Sexual crosses between individuals from the same species and different species were carried out. Only crosses between individuals from the same species were successful. Moreover, population genetics analysis revealed a high level of diversity within each species and a lack of gene flow between them. Finally, a population survey over time showed that B. cinerea was the predominant species but that B. pseudocinerea was more abundant in spring, on floral debris. This observation could not be explained by temperature adaptation in tests carried out in vitro or by aggressiveness on tomato or bean leaves. This study clearly establishes that B. cinerea and B. pseudocinerea constitute a complex of two cryptic species living in sympatry on several hosts, including grapevine and blackberry. We propose several biological or molecular tools for unambiguous differentiation between the two species. B. pseudocinerea probably makes a negligible contribution to gray mold epidemics on grapevine. This new species has been deposited in the MycoBank international database.  相似文献

Purpose

At the global scale, gardening activities are often performed in urban areas with a historical background of pollution. In this study, a participatory program was developed with citizens concerned by gardening activities near a 50-year-old regulated lead recycling company, with the aim of co-constructing the tools for the assessment and management of potential sanitary risks induced by historic pollution with persistent (eco) toxic metals: lead and cadmium.

Materials and methods

Soils and vegetables (lettuce, leek, celery, carrot, chard, pumpkin, and celeriac) samples were collected from four kitchen gardens neighboring a 50-year-old secondary lead smelter. Both total and in vitro human bioaccessible metal concentrations in the cultivated plants were measured in relation to soil characteristics.

Results and discussion

The results showed that the soils of these gardens were slightly contaminated by metals (Pb, 77 to 236 mg kg?1; and Cd, 0.5 to 1 mg kg?1) in comparison with the natural geologic background. However, significant pollution of vegetables can occur especially with lead (Pb up to 9.8 mg kg?1 in lettuce) and certainly as a result of direct foliar transfer. The washing of plants before consumption is therefore recommended in the context of atmospheric fallout of ultrafine particles enriched with metals.

Conclusions

Metal bioaccessibility measure integrates the influence of metal type, plant type, and soil physico-chemical properties. Based on the results, it is proposed that human bioaccessible fraction of metals may also be currently taken into account as well as total metal quantities and bioaccumulation factors in risk assessment studies performed in gardens. Overall, this study has led to reflections and functional recommendations aimed at reducing human exposure and to finally developing sustainable gardening practices.

Intraspecific phenotypic diversity is the raw material for evolution, so understanding its origin and maintenance is critically important for conservation of biodiversity. Intraspecific diversity in a trait or a suite of traits can result from genetic diversity and/or phenotypic plasticity. The two are, however, not independent as plasticity has been shown to evolve. In this study, we evaluated the importance of phenotypic plasticity in generating morphological diversity in populations of small benthic Arctic charr in Iceland, using a rearing experiment with contrasting modes of feeding. We also examined the association between phenotypic plasticity in offspring groups generated by the contrasting feeding modes and important ecological variables characterising the natural habitats of the respective populations. Although the level of plasticity could not be related to any of the ecological measurements, clear differences in morphological reaction norms among populations suggest that plasticity is an important aspect of morphological diversity of the charr. It is not clear whether that plasticity is adaptive, but it is notable that reaction norms in all populations have similar reaction to the treatments.  相似文献

The importance of the amino acid L‐arginine (ARG) for conceptus growth and litter size has been demonstrated in various species. L‐arginine is part of embryo‐derived polyamines, a substrate for nitric oxide synthase and stimulates protein synthesis by the embryo. In the present study, we have investigated whether dietary L‐arginine supplementation stimulates early conceptus growth in mares. Warmblood mares with singleton pregnancies received either an arginine‐supplemented diet (approximately 0.0125% of body weight, n = 12) or a control diet (n = 11) from days 15 to 45 after ovulation. Diameter of the embryonic vesicle (days 14, 17, 20 of pregnancy) and size of the embryo respective foetus (length and maximal diameter, days 25–45 of pregnancy at 5‐day intervals) were determined by transrectal ultrasound. At foaling, weight and size of the foal and the placenta were determined. Blood for determination of equine chorionic gonadotrophin (eCG) and progestin concentrations was collected repeatedly. Neither eCG nor progestin concentration in plasma of mares differed between groups at any time. No effects of arginine treatment on diameter of the embryonic vesicle between days 14 and 20 of pregnancy were detected. Diameter of the embryo/foetus on days 40 to 45 of pregnancy strongly tended to be enhanced by arginine supplementation (p = 0.06). Weight and size of neither the foal nor placenta at birth differed between groups. In conclusion, L‐arginine supplementation was without negative effects on early equine embryos and may support embryonic growth at the beginning of placentation.  相似文献

A randomized controlled trial (RCT) was carried to evaluate the effect of a feed additive on Campylobacter contamination of broilers reared in commercial conditions. Twenty‐four broiler flocks naturally contaminated with Campylobacter were enrolled in the RCT: 12 were assigned to a control group (C) fed with a conventional finishing feed from 4 weeks of age to slaughter (around 35 days), and the other group of 12 flocks (S) was fed with a finishing feed supplemented with 250 ppm of a patented feed additive (an ion‐exchanged clay compound) previously proven to reduce Campylobacter contamination in broiler caeca under experimental conditions. Enumeration of Campylobacter colonies in caeca (8 per flock) was carried out following ISO standards before feed distribution and at slaughter. Before treatment, the caecal Campylobacter load tended to be lower in C flocks (7.1 ± 1.9 log CFU/g, CI95% [6.6–7.5]) than in S flocks (7.7 ± 1.0 log UFC/g, CI95% [7.5–7.9]) (= .05). At slaughter, the bacterial load was similar in the S (7.7 ± 1.0 log CFU/g, CI95% [7.5–7.9]) and C groups (7.5 ± 1.2 log CFU/g, CI95% [7.2–7.8]) (= .73). Therefore, the feed additive had no significant effect on the caecal Campylobacter load at slaughter under the tested conditions. The logistical constraints inherent in field trials and the natural variability of Campylobacter contamination in naturally infected broiler flocks make it difficult to reproduce experimental results in in situ farm conditions. RCT testing of an intervention strategy in commercial situation is therefore a key step in evaluating pre‐harvest interventions against food‐borne pathogens.  相似文献

Purpose

This study assesses the potential of two contrasted fragrant Pelargonium cultivars to induce pH and dissolved organic carbon (DOC) changes in the soil solution, Pb speciation, and their subsequent effects on rhizosphere phytoavailable Pb.

Materials and methods

Rooted plantlets were grown in special devices, floating on aerated nutrient solution in PVC tanks. This setup allows roots to be physically separated, through a mesh, from a 3-mm soil matrix layer that can be considered as rhizosphere soil. Two contrasted soils, each spiked with Pb-rich particles, emitted from a battery recycling industry, were used at total burdens of 500 and 1500 mg Pb kg?1 in addition to a control unspiked soil. Soil solution pH, phytoavailable Pb, DOC, Pb adsorption, precipitation on roots, and Pb phases in soil and plant were investigated.

Results and discussion

Attar of Roses (Attar) cultivar acidified its rhizosphere by 0.4 pH units in both spiked soils. Concolor Lace (Concolor) was unable to change soil solution pH on soil-1 and increased it by 0.7 units on soil 2. Concentrations of Pb in soil solution from Attar plants were always higher than those of Concolor ones. DOC contents of both unspiked soil-1 and soil-2 without plants were not significantly different. In the case of spiked samples, DOC contents in the rhizosphere soil were increased by three and two times for Attar and Concolor, respectively, compared to the unspiked soil without plant. Both cultivars were able to increase DOC contents, independent of soil type and level of contamination. Accumulation of Pb in shoots and roots was higher in Attar as compared to Concolor due to enhanced available Pb as a result of pH and DOC modifications of the rhizosphere soil. Significant amounts of Pb were adsorbed on roots of both cultivars. X-ray elemental analysis of precipitates on roots revealed the association of Pb with P in cylinder-like structures. Extended X-ray absorption fine structure (EXAFS) spectroscopy revealed that Pb was present, to a major extent in the inorganic form, mainly as PbSO4 in the soil, whereas it was complexed with organic species within plant tissues. The conversion of Pb into organic species could decrease toxicity, may enhance plant tolerance, and could increase translocation.

Conclusions

Plant-induced changes were responsible for the modification of lead phases within the soil. Immobile forms present in the source leaded particles as well as in the soils were converted into soluble species, ultimately improving the phytoavailable or soil solubilized Pb.
