1. Poultry on farms are sometimes required to be killed in an emergency, such as during a disease epidemic, yet none of the available methods are ideal. Whole-house carbon dioxide (CO2) administration has practical advantages, but gives rise to welfare concerns.

2. The study measured the body temperature, respiration, cardiac and brain activity (EEG) responses of 10 adult hens placed in tiered cages in a deep pit house while the entire flock (28,000 end-of-lay hens) was killed with CO2. Video and thermographic images were also recorded. Liquid CO2 was injected into the building producing a gaseous concentration of 45% within 19?min.

3. Those hens nearest the gas delivery site showed delayed respiratory, cardiac and EEG responses compared with those at more distant locations. Although sub-zero temperatures were recorded in the immediate vicinity of some birds, body temperatures indicated that they did not die of hypothermia.

4. EEG characteristics strongly associated with unconsciousness were used to determine an unequivocal time to loss of consciousness; this ranged from 6·0 to 10·5 (average 7·8) min after onset of gas injection. Distinctive cardiac and respiratory responses were seen following gas exposure; in particular, birds responded to inhalation of CO2 by deep breathing.

5. The primary welfare concern is the duration of unpleasant respiratory effects, such as deep breathing, while the birds were substantively conscious. However, the concentration of CO2 to which the birds were exposed while conscious would not have stimulated nasal and oral nociceptors. Time to death varied between 12·0 and 22·1?min after gas delivery.  相似文献

Selection criteria for yearling bulls commonly include indicators of fertility and carcass merit, such as scrotal circumference (SC) and intramuscular fat percentage (IMF). Genetic correlation estimates between ultrasound traits such as IMF and carcass marbling score (MS) with fertility traits SC and heifer pregnancy (HP) have not been reported. Therefore, the objective of this study was to estimate the genetic parameters among the indicator traits IMF and SC, and the economically relevant traits MS and HP. Records for IMF (n=73,051), MS (n=15,260), SC (n=43,487), and HP (n=37,802) were obtained from the Red Angus Association of America, and a 4-generation ancestral pedigree (n=10,460) was constructed from the 8,915 sires represented in the data. (Co)variance components were estimated using a multivariate sire model and average information REML to obtain estimates of heritability and genetic correlations. Fixed effects included contemporary group and the linear effect of age at measurement for all traits, and an additional effect of age of dam for both HP and SC. The random effect of sire was included to estimate additive genetic effects, which were assumed to be continuous for IMF, MS, and SC, but a probit threshold link function was fitted for HP. Generally moderate heritability estimates of 0.29 ± 0.01, 0.35 ± 0.06, 0.32 ± 0.02, and 0.17 ± 0.01 were obtained for IMF, MS, SC, and HP on the underlying scale, respectively. The confidence interval for the estimated genetic correlation between MS and HP (0.10 ± 0.15) included zero, suggesting a negligible genetic association. The genetic correlation between MS and IMF was high (0.80 ± 0.05), but the estimate for HP and SC (0.05 ± 0.09) was near zero, as were the estimated genetic correlations of SC with MS (0.01 ± 0.08) and IMF (0.05 ± 0.06), and for HP with IMF (0.13 ± 0.09). These results suggest that concomitant selection for increased fertility and carcass merit would not be antagonistic.  相似文献

Molecular analysis, serology and immunophenotyping for T lymphocytes and their subsets, B lymphocytes and monocytes were performed on dogs naturally infected with Leishmania infantum. Animals were categorised as asymptomatic dogs I (AD-I), with negative serology and positive molecular results, and asymptomatic dogs II (AD-II), with positive serology and positive molecular results, and these were compared to symptomatic dogs (SD) and control dogs (CD). AD-I exhibited immunophenotypic features similar to those of CD, including isotype profiles and concentrations of monocytes. Similar biomarkers were found in AD-II and SD, such as, higher levels of immunoglobulins IgG, IgG2, IgM and IgA and higher concentrations of eosinophils. High frequencies of T lymphocytes and CD4(+) T cells were observed in both AD-I and AD-II compared to SD, whereas CD8(+) T cells were higher only in AD-II compared with SD. Analysis of B lymphocytes revealed an increased frequency of this cell type only in AD-II animals compared with SD. Asymptomatic dogs appear to have a dichotomous infection spectrum that can influence the humoral and cellular immunological status during canine visceral leishmaniasis.  相似文献

This review assesses evidence from DNA analysis to determine whether there is sufficient genetic diversity within breeds to ensure that populations are sustainable in the absence of cross breeding and to determine whether genetic diversity is declining. On average, dog breeds currently retain approximately 87% of the available domestic canine genetic diversity. Requirements that breeding stock must be 'clear' for all genetic disorders may firstly place undue genetic pressure on animals tested as being 'clear' of known genetic disorders, secondly may contribute to loss of diversity and thirdly may result in the dissemination of new recessive disorders for which no genetic tests are available. Global exchange of genetic material may hasten the loss of alleles and this practice should be discussed in relation to the current effective population size of a breed and its expected future popularity. Genomic data do not always support the results from pedigree analysis and possible reasons for this are discussed.  相似文献

Breeding practices were analysed for 32 registered dog breeds representing very small registries (120 Central Asian shepherd dogs) through to very large registries (252,521 German shepherd dogs) in Australia. The vast majority (91%) of registered kennels in Australia that were sampled did not regularly employ either close breeding or popular sire usage in their kennels and the weighted mean inbreeding coefficient of Australian pedigree dogs was <5%. Australian breed mean inbreeding coefficients ranged from 0% (Central Asian shepherd dog) to 10.1% (Bichon Frise). Breed effective population sizes ranged from 26 (Ibizan hound) to 1090 (Golden retriever), comparable with other species of domesticated animals. The relatively low levels of inbreeding suggest that pedigree dog disorders are unlikely to arise frequently from the use of popular sires or close breeding in Australian registered dog breeds. It is possible that deleterious allele fixation might be driven by founder effects, genetic drift or adverse selection practices, which were not assessed in this analysis. European popular sire definitions should be revisited for rare breeds.  相似文献

Objective Scrapie, a transmissible spongiform encephalopathy (TSE) occurring naturally in sheep, characteristically shows a severe retinopathy that is well developed in the terminal phases of the disease. In this study, we set out to demonstrate similar retinal changes in our ruminant spiroplasmosis TSE model. Procedure The eyes from deer, sheep, and goats that were inoculated intracranially with the laboratory strain of spiroplasma (suckling mouse cataract [SMCA] strain of Spiroplasma mirum) or with Spiroplasma sp. isolated from the brains affected with scrapie or with chronic wasting disease were examined by light microscopy for pathologic changes and by immunocytochemistry for distribution of spiroplasma antigen. The eyes were also obtained from a research flock of sheep with terminal scrapie, from which the intraocular tissues were submitted aseptically for culture assay in M1D broth or as explants on bovine corneal endothelia (BCE). Results The eyes from the spiroplasmosis ruminant models showed retinopathy remarkably similar to eye lesions seen in sheep with scrapie. The spiroplasma antigen accrued in the ruminant model eye tissues, particularly in the retina, the vitreous humor, and the corneal endothelia. A Spiroplasma sp. grew out of the scrapie‐affected eyes both in the M1D broth and in the BCE cultures but did not expand. These new spiroplasma isolates differed immunologically from SMCA. Conclusion These data showed a clear association of spiroplasma with scrapie suggesting that these bacteria have a role in the pathogenesis of TSE and that the eye should be a research focus for future studies of TSE.  相似文献

Previously published multidisciplinary studies in the Miramichi and Bouctouche rivers (New Brunswick, Canada) noted significant changes in fish health parameters, including elevated tissue levels of organic contaminants and a wide range of physiological disturbances, in mummichog Fundulus heteroclitus (L.) from a site on the Miramichi River that received bleached kraft pulpmill and municipal effluent. The present study reports differences in the abundance of individual parasite species, as well as parasite infracommunity and component community composition, in mummichog from both rivers. These differences were evaluated in relation to host (size, condition, immune function, tissue organochlorine contaminant levels) and environmental (faecal coliform counts, salinity, temperature) data derived from the previously published studies. Overall, 18 parasite species were identified, the most common of which were Ascocotyle sp. larv., Ornithodiplostomum sp. larv., Posthodiplostomum sp. larv., and Proteocephalus filicollis (Rudolphi, 1802). There were broad differences in parasite community structure and composition between rivers and within rivers, the most prominent pattern being a pronounced difference between sites in the upper and lower estuary of each river that was likely driven by salinity. Mean infracommunity richness was also positively related to faecal coliforms (considered here as a surrogate measure of eutrophication via municipal sewage), and both were highest at the most polluted site. We noted no other significant relationships. Thus our data suggest that the parasite communities in these two estuaries were primarily structured by large upstream/downstream ecological gradients in salinity, and secondarily by eutrophication due to pollution by municipal and industrial effluents. Overall, our results highlight the value of coordinated multidisciplinary studies for understanding the factors that shape parasite abundance and community structure.  相似文献

采用Goldkey及DNA Star分析软件对发表的3个不同血清型的鸡大肠杆菌1型菌毛结构基因(pilA)进行同源性及抗原位点分析。结果显示,3种鸡大肠杆菌(O_1、O_(78)及O_(88))l型菌毛pilA基因相似性为91.36%~98.38%,所编码的氨基酸同源性为93.50%~97.84%;3种血清型鸡大肠杆菌(O_1、O_(78)及O_(88))1型菌毛蛋白二级结构在1~73位及150~185位氨基酸的二级结构基本相似,而111~150位氨基酸的α螺旋与β折叠的数量有所不同。菌毛蛋白亲水性、抗原性及抗原决定簇预测表明,来源于3种血清型大肠杆菌的1型菌毛蛋白在l~110位及150~184位氨基酸存在相似的二级结构及抗原位点,且不同血清型之间存在一定的共同抗原。  相似文献

Tunneling spectroscopy has been used to characterize the magnitude and temperature dependence of the superconducting energy gap (triangle up) for K(3)C(60) and Rb(3)C(60). At low temperature the reduced energy gap, 2triangle upkappaT(c) (where T(c) is the transition temperature) has a value of 5.3 +/- 0.2 and 5.2 +/- 0.3 for K(3)C(60) and Rb(3)C(60), respectively. The magnitude of the reduced gap for these materials is significantly larger than the value of 3.53 predicted by Bardeen-Cooper-Schrieffer theory. Hence, these results show that the pair-coupling interaction is strong in the M(3)C(60) superconductors. In addition, measurements of triangle up(T) for both K(3)C(60) and Rb(3)C(60) exhibit a similar mean-field temperature dependence. The characterization of triangle up and triangle up(T) for K(3)C(60) and Rb(3)C(60) provides essential constraints for theories evolving to describe superconductivity in the M(3)C(60) materials.  相似文献
