AbstractAIM: To evaluate resistance to anthelmintics containing abamectin, levamisole, and oxfendazole (AB-LEV-OX), derquantal and abamectin (DEQ-AB), moxidectin, and monepantel in naturally acquired gastrointestinal nematodes present on a sheep farm.METHODS: Faecal nematode egg count reduction tests (FECRT) were carried out on lambs that were approximately 7 months-old and infected with naturally acquired nematodes. Lambs were randomly allocated to one of five groups (n=15 per group): treatment with 2?mg/kg derquantel and 0.2?mg/kg abamectin; 0.2?mg/kg abamectin, 8?mg/kg levamisole HCl and 4.5?mg/kg oxfendazole; 2.5?mg/kg monepantel; 0.2?mg/kg moxidectin, or no treatment. Post-treatment samples were collected 12 days later. Abomasa and small intestines were collected from two slaughtered lambs from each of the DEQ-AB, AB-LEV-OX, moxidectin and control groups 15 days after treatment, for nematode counting.RESULTS: The FECRT demonstrated that efficacy was 90.3 (95% CI=84.2–94.1)% for AB-LEV-OX, 54.5 (95% CI=28.4–71.1)% for moxidectin, 99.2 (95% CI=97.4–99.8)% for DEQ-AB and 100% for monepantel, across all genera. For Trichostrongylus spp. efficacy was 85.5% for AB-LEV-OX and 46.7% for moxidectin. Haemonchus spp. were fully susceptible to all treatments. Post-treatment nematode counts indicated that the resistant Trichostrongylus spp. were from the small intestine.CONCLUSIONS: Anthelmintic resistance to both AB-LEV-OX and moxidectin was present in the Trichostrongylus genus on a commercial sheep farm. Monepantel and DEQ-AB were both effective against Trichostrongylus spp. based on FECRT results.CLINICAL RELEVANCE: This finding of resistance to an AB-LEV-OX triple-combination anthelmintic in the Trichostrongylus genus in sheep in New Zealand further limits anthelmintic treatment options available, and calls into question whether this combination is suitable for use as a quarantine treatment. 相似文献

Maiden heifers and lactating cows of known ovarian status and of several breeds were treated with a synthetic prostaglandin, cloprostenol, or a synthetic progestagen, norgestomet, at the start of an artificial insemination (AI) program. Animals in the cloprostenol treatment received 2 injections 10 days apart. Over the next 26 days those animals that showed oestrous behaviour were inseminated. Synchronisation rates and calving rates to insemination over the first 7 days were calculated. Those in the norgestomet treatment received an implant of norgestomet plus an injection of norgestomet and oestradiol valerate. The implant was removed 10 days later and the animals were given an injection of pregnant mare serum gonadotrophin (PMSG). They were inseminated at 48 h (maiden heifers) or 56 h (lactating cows) after implant removal. Calving rates to fixed-time insemination were recorded. After completion of the AI program the animals in both treatments were joined with bulls. Overall calving rates (AI plus bulls) were calculated. By day 7 of the program, 82% of the maiden heifers and 76% of the lactating cows in the cloprostenol treatment had been detected in oestrus. By day 21 the respective figures were 99% and 81% Norgestomet treatment had an immediate and a prolonged effect on ovarian activity in those females classified as having inactive ovaries at the start of the AI program. Calving rates of those females to fixed-time AI and overall were similar to those of the females with active ovaries in both treatments. Their calving rates to fixed-time insemination, and overall calving rates for the lactating females, were significantly higher than the corresponding values of their contemporaries treated with cloprostenol and inseminated on observed oestrus over 7 days. For those females classified as having active ovaries at the start of the AI program, calving rates to first insemination and overall were similar for both treatments. Overall calving rates of lactating cows of each breed were, with one exception, higher in the norgestomet treatment than in the cloprostenol treatment. Although norgestomet treatment was more expensive than cloprostenol treatment, the advantage in calf crop resulted in an overall monetary advantage to the norgestomet treatment. 相似文献

ABSTRACTEquine dentistry is a rapidly developing clinical specialty. It has benefitted from key advances in anatomical and physiological research, development of equipment and instrumentation, utilisation of standing sedation and anaesthesia protocols, a change towards minimally invasive surgical techniques, and the introduction of restorative and endodontic techniques translated from techniques used in human and canine dentistry. Anatomical research has provided further insight into the endodontic system of incisors and cheek teeth and how it changes throughout development with age. Studies of the periodontium have demonstrated a rich vascular supply and repair capacity. Routine dental visits are increasingly being performed utilising sedation and clinical instruments for routine examinations. Equipment has become more efficient, battery-operated and miniaturised giving benefits to both equine dental maintenance work and advanced techniques, assisting the transition to minimally invasive techniques, and the development of endodontic and restorative dentistry. Diagnosis has also benefitted from advances in equipment such as patient-side digital radiography systems, high definition oroscopy, and small diameter flexible fibrescopes that are capable of visualising inside a pulp canal. Dental units combining endodontic high- and low-speed drills, suction and air or water flush are becoming increasingly used and adapted for equine use. Sedative combinations and standing anaesthesia protocols have meant that revisions of traditional techniques, as well as novel techniques, can be performed with almost no requirement for general anaesthesia. Equine dentistry can only continue to advance in this way if there is early identification of dental disease through clinical oral examinations, leading to a system based on prophylaxis, as in human dentistry. This necessitates a change in attitude of the public and industry in general to a proactive approach, with early intervention based on examination and diagnostic findings of practitioners and not necessarily based on the clinical signs displayed by the patient. 相似文献

Objective
To evaluate cardiopulmonary and recovery characteristics of horses administered total intravenous anesthesia (TIVA) with xylazine and ketamine combined with midazolam or propofol.Study design
Randomized crossover study.Animals
A group of eight adult horses, aged 7–22 years, weighing 493–740 kg.Methods
Horses were administered xylazine (1 mg kg?1) intravenously (IV), and anesthesia was induced with ketamine (2.2 mg kg?1) IV. Anesthesia was maintained for 45 minutes via IV infusion of xylazine (0.016 mg kg?1 minute?1) and ketamine (0.03 mg kg?1 minute?1) combined with midazolam at 0.002 mg kg?1 minute?1 (MKX), propofol at 0.05 mg kg?1 minute?1 (PKXlow) or propofol at 0.1 mg kg?1 minute?1 (PKXhigh). Additional ketamine was administered if a horse moved spontaneously. Cardiopulmonary variables, blood gases, lactate concentration, packed cell volume and total solids were recorded before sedation (baseline), at 10, 20, 30 and 45 minutes during TIVA and 10 minutes after standing. Recovery variables and quantitative recovery scores were compared. Significance was set at p < 0.05.Results
Additional ketamine was required for 50% of MKX horses. Systolic arterial pressure was elevated in MKX at 20 minutes compared with baseline (p = 0.043), at 10 and 20 minutes compared with PKXhigh (p = 0.007, p = 0.024) and at 20 and 30 minutes compared with PKXlow (p = 0.009, p = 0.02). MKX horses (5/8) were hypertensive compared with PKXlow (1/8; p = 0.017). All horses became hypoxemic (PaO2 ≤80 mmHg; 10.7 kPa) during TIVA. Recovery variables did not differ among treatments.Conclusions and clinical relevance
PKXlow and PKXhigh had similar cardiopulmonary and recovery performance compared with MKX. PKX combinations provided superior quality of anesthesia to that of MKX. A combination of propofol, ketamine and xylazine administered as TIVA can be used in horses to provide anesthesia for short procedures. Supplemental oxygen is recommended. 相似文献20.
Tight junctions (TJs) in inter-Sertoli junctional areas and epididymal epithelia build up the blood–testis barrier (BTB) and the blood–epididymal barrier (BEB), respectively. In this study, the expression of occludin, an integral member of the TJs, was examined in testis and different regions of epididymis of Lepus sinensis coreanus , an Korean wild rabbit species. In testis, intense occludin immunoreactivity was found in the basally located inter-Sertoli junctional area together with diffused immunoreactivity of occludin in the cytoplasm of Sertoli cells. It can be suggested that occludin is one of the robust elements of BTB in seminiferous tubules of rabbit testis. In proximal and distal caput epididymis, occludin immunoreactivity was found in the lateral as well as apical contacts of epithelial cells. In corpus epididymis, intense occludin immunoreactivity was found in the basolateral as well as apical contacts of epithelial cells together with cytoplasmic signal. In cauda epididymis, occludin immunoreactivity in luminal epithelia was relatively strong but largely found in the cytoplasm. This suggests that intriguing regulatory mechanisms differentially recruit occludin to the TJ in the different regions of epididymal epithelia. The differences in the subcellular localization as well as expression levels of occludin among the epididymal segments may reflect differential paracellular permeability of epithelia along the epididymal tubules and be correlated with sperm maturation in rabbit. In Western blot, a major form of occludin was MW 62 kDa together with small fragments of MW 34–39 kDa in testis and epididymis, suggesting the peptide cleavage of occludin. This is the first report on the molecular nature of TJs in a wild rabbit testis and epididymis. 相似文献
