ABSTRACT

Aims: To determine if an ELISA for measurement of IgA in equine serum could be used to measure concentrations of IgA in foal faeces and to determine correlations with concentrations in the milk of the dam.

Methods: Faeces from 20 Welsh Cob and Welsh Pony foals and milk from their dams were collected within 12?hours (Day 0) and at 6 days after parturition (Day 6). On Day 6, faeces could not be collected from 2/20 foals, and milk samples could not be collected from 3/20 mares. An equine IgA ELISA validated for serum and plasma was used to measure concentrations of IgA in all samples in triplicate. The precision of the assay for each sample type was determined using modified CV.

Results: IgA was not detectable in 7/20 Day 0 faecal samples and in 2/18 Day 6 faecal samples. For samples with detectable IgA, the mean modified CV was 10.5 (95% CI?=?6.0–15.0)% for Day 0 faecal samples, and was 6.8 (95% CI?=?4.3–9.4)% for Day 6 faecal samples. Median concentrations of IgA in faeces on Day 0 were lower than concentrations on Day 6 (0.7?mg/g vs. 37?mg/g dry matter; p?=?0.003). Concentrations of IgA in milk and faeces on Day 6 were statistically correlated (r?=?0.59; p?=?0.006).

Conclusions and clinical relevance: The IgA ELISA showed acceptable precision when used to estimate concentrations of IgA in foal faeces during the first week of life, but IgA could not be detected in 37% of meconium samples collected on Day 0. This assay may be useful for investigation of the role of maternal milk IgA in the gastrointestinal tract of neonatal foals, but further assessment of both accuracy and precision of the ELISA is required.  相似文献

Abstract

AIM: To evaluate resistance to anthelmintics containing abamectin, levamisole, and oxfendazole (AB-LEV-OX), derquantal and abamectin (DEQ-AB), moxidectin, and monepantel in naturally acquired gastrointestinal nematodes present on a sheep farm.

METHODS: Faecal nematode egg count reduction tests (FECRT) were carried out on lambs that were approximately 7 months-old and infected with naturally acquired nematodes. Lambs were randomly allocated to one of five groups (n=15 per group): treatment with 2?mg/kg derquantel and 0.2?mg/kg abamectin; 0.2?mg/kg abamectin, 8?mg/kg levamisole HCl and 4.5?mg/kg oxfendazole; 2.5?mg/kg monepantel; 0.2?mg/kg moxidectin, or no treatment. Post-treatment samples were collected 12 days later. Abomasa and small intestines were collected from two slaughtered lambs from each of the DEQ-AB, AB-LEV-OX, moxidectin and control groups 15 days after treatment, for nematode counting.

RESULTS: The FECRT demonstrated that efficacy was 90.3 (95% CI=84.2–94.1)% for AB-LEV-OX, 54.5 (95% CI=28.4–71.1)% for moxidectin, 99.2 (95% CI=97.4–99.8)% for DEQ-AB and 100% for monepantel, across all genera. For Trichostrongylus spp. efficacy was 85.5% for AB-LEV-OX and 46.7% for moxidectin. Haemonchus spp. were fully susceptible to all treatments. Post-treatment nematode counts indicated that the resistant Trichostrongylus spp. were from the small intestine.

CONCLUSIONS: Anthelmintic resistance to both AB-LEV-OX and moxidectin was present in the Trichostrongylus genus on a commercial sheep farm. Monepantel and DEQ-AB were both effective against Trichostrongylus spp. based on FECRT results.

CLINICAL RELEVANCE: This finding of resistance to an AB-LEV-OX triple-combination anthelmintic in the Trichostrongylus genus in sheep in New Zealand further limits anthelmintic treatment options available, and calls into question whether this combination is suitable for use as a quarantine treatment.  相似文献

A 15-month-old, male neutered Staffordshire Bull Terrier cross was presented to its referring veterinarian collapsed and agonal. He was immediately intubated, manually ventilated, and treatment commenced for presumptive snake envenomation with two vials of Tiger/Multi-Brown Snake Antivenom (minimum 7000 units/vial). The dog was transferred to a referral hospital intubated. Additional diagnostics performed following arrival at the referral hospital included a urine snake venom detection kit test, which was positive for brown snake immunotype. Three additional vials of Tiger/Multi-Brown Snake Antivenom (minimum 7000 units/vial) were administered until the dog was extubated and able to stand. Venom-induced consumptive coagulopathy (VICC) was diagnosed based on prolonged clotting times and scleral haemorrhage. Paroxysms of right ventricular outflow tract (RVOT) origin ventricular arrhythmias were treated with lignocaine and sotalol. Four days after presentation, a new-grade IV/VI systolic heart murmur was auscultated, prompting an echocardiogram. An anechoic and compartmentalised mass measuring 43 mm × 19 mm was visualized within the right ventricular wall at the RVOT, immediately adjacent to the pulmonic valve. The mass was causing a RVOT obstruction. Its appearance was suggestive of an intramyocardial haematoma, most likely secondary to VICC. The dog remained cardiovascularly stable, and treatment consisted of supportive care. Recheck echocardiograms at 2 and 7 weeks after discharge revealed progressive improvement of the intramyocardial mass and resolution of the associated heart murmur. Although intramyocardial haematomas are rare, it should be considered as a differential in dogs that develop a newly diagnosed heart murmur and/or cardiac arrhythmia following brown snake envenomation.  相似文献

Summary. 2,4-D-amine, 2,4-D ethyl ester, 2.4-DB-potassium. MCPA-sodium, diquat and a 1:1 mixture of 2,4,5-T butyl ester and 2,4-D ethyl ester were applied to the tropical pasture legume Centrosema pubescens Benth. (centro) grown either alone or with Panicum maximum Jacq. (guinea grass). Even at 0.5 Ib/ac (total) the 2,4,5-T/2,4-D mixture caused severe damage to seedling and mature centro. 2,4-D-amine, 2,4-D ethyl ester and 2,4-DB-potassium were less damaging and there were only slight differences in response to these. Diquat at 0.25 Ib/ac desiccated centro seedlings, but most plants survived and grew satisfactorily. There was some evidence that the growth of mature centro was depressed least when the herbicides were applied during vegetative quiescence.
Application just after the start of flowering altered the flowering pattern but did not reduce seed yields significantly. 2,4-D-amine at 0.75 and 1–5 lb/ac and 2,4-D ethyl ester at 1.5 Ib/ac decreased the percentage germination of abraded seed. Diquat 0.25 Ib/ac applied at the start of flowering caused significant increases in top growth and seed pod numbers, possibly the result of modification of the normal flowering pattern.  相似文献

The objectives of the present study were to examine the spatial patterns of sward dry matter (DM) and nitrogen (N) yields in a grass silage field at first, second, and third cuts over a 3-year period; quantify their temporal stabilities with temporal stability maps; and assess the potential for site-specific management in each pasture-growing period using classified management maps. At cut 1, the spatial patterns of DM and N yields proved to be well defined and temporally stable and were likely to be due to differences in the net N mineralization rates across the field during spring. In contrast, at cut 2, the patterns of DM production were patchy and temporally unstable. It was concluded that, in principle, a simple site-specific approach to N fertilization would be possible in this field during spring at cut 1. At later harvests, the rationale for a site-specific approach to fertilizer management was less clear with logistics appearing to be more complex and less feasible.  相似文献

The cellular reactions of excised hypocotyl segments of 31 cultivars of Phaseolus vulgaris to infection by two races of Colletotrichum lindemuthianum were surveyed. Consistent differences in the extent of fungal development and host cell necrosis were used to establish six infection categories.
Extreme susceptibility (category 5) involved initial formation of infection vesicles in living epidermal cells and a prolonged period of intracellular biotrophic development prior to necrotrophic growth and production of spreading lesions. Extreme resistance (category 0) involved rapid death and browning of penetrated epidermal cells, without formation of infection vesicles or a detectable biotrophic phase. Fungal growth and host necrosis were limited to single epidermal cells. Intermediate reactions (categories 1 to 4) initially resembled category 5, but after a biotrophic phase of varying duration, infected cells and some adjacent uninfected cells died and became brown, and fungal growth ceased. Symptoms ranged from flecks (small groups of dead cells) to large areas of necrosis (limited lesions). Similar intermediate reactions also occurred in normally susceptible hypocotyls which were transferred from 17 to 25°C, or which retained cotyledons.
The finding that resistance is expressed at different stages of infection is discussed in relation to the regulation of cultivar specificity.  相似文献
