Previous studies have shown an increase in plasma cortisol in gilts over farrowing irrespective of environment suggesting that factor(s) associated with parturition itself cause physiological stress. Factors involved in mediating the hypathalomo–pituitary–adrenal (HPA) axis at parturition are not well understood. This study examines the effect of piglet expulsion on the pituitary–adrenal axis by measurement of plasma cortisol, adrenocorticotropic hormone (ACTH) and β-endorphin. The effect of farrowing environment in modulating the acute response to piglet expulsion is also investigated. Twelve second parity sows, with indwelling jugular catheters, were moved into either a farrowing crate or a straw-bedded pen 5 days before their expected parturition date (EPD). Blood samples were taken from each sow during a prefarrowing baseline period and then rapid samples (2.5 mins) were taken for 10 min following the birth of two piglets. No effect of environment was found on any of the hormonal variables which reinforces the hypothesis that the physiological stress seen in parturient pigs is due to some intrinsic factor of parturition. Plasma cortisol, ACTH and β-endorphin did not change significantly in the period following piglet expulsion suggesting that individual piglet expulsions do not play a major role in 'parturition stress'. There was however, an increase in plasma cortisol, ACTH and β-endorphin in response to increasing piglet number which is consistent with previous studies of general farrowing in which cortisol increased as farrowing progressed. Therefore this study reinforces the hypothesis that physiological stress increases with ongoing parturition although this does not appear to be a result of piglet expulsion. The potential role of other factors which may be involved in causing 'parturition stress' should be investigated. 相似文献

Rates of protein synthesis of mice with a major gene (hg) for rapid postweaning gain (line Ch) and their normal counterparts (line CH) were determined at 21, 31 and 42 d of age with an intraperitoneal injection of a flooding dose of 14C-leucine. A preliminary experiment demonstrated that the relationship between the specific activities of leucine in acid-soluble supernatants and carcass protein corresponded to the theoretical precursor-product relationship, indicating that the method is valid for estimating protein synthesis rates. Using this method at 21, 31 and 42 d of age, whole-body protein fractional synthesis rates (FSR) were 43.7, 32.7 and 29.1%/d and 41.9, 32.6 and 33.1%/d for lines CH and Ch, respectively. Although differences between lines were not significant, FSR decreased with age. Absolute synthesis rate (ASR), where ASR = (FSR) X (whole body protein), was greater (P less than .001) at 21, 31 and 42 d of age in line Ch as compared with CH, and increased (P less than .001) with age. The relative contributions of liver, gastrointestinal tract, heart-kidney-lung and remaining carcass to whole body protein ASR were not affected by line, but did change (P less than .05) with age. Whole body protein fractional breakdown rate (FBR), calculated as FSR minus whole body protein fractional growth, indicates that differences between lines CH and Ch whole-body FSR and(or) FBR exist only between 24 and 33 d of age, and that the maximum value of this difference probably does not exceed 10%. 相似文献

Fenugreek staggers has occurred in sheep in Victoria, as both an acute and a chronic syndrome. Signs included quadraparesis, a high stepping fore limb gait and a 'bunny-hopping' hind limb gait. Changes consistent with acute oedema were found in the brain and spinal cord of acute cases, and Wallerian degeneration in the peripheral nerves of chronic cases. Kangaroo gait occurred in ewes in New South Wales, and the clinical signs and microscopic changes were remarkably similar to those of fenugreek staggers. The effects of various daily durations of CO2 enrichment ‘on early-sown glasshouse tomatoes are outlined. Reducing the daily enrichment period in the pre-planting stage (late November to mid-January) had only marginal effects on total yields and · monetary values. Reductions in the post-planting stage (mid-January to mid-April) caused significant depressions in yields, roughly proportional to the · reduction in enrichment time. For a given reduction in the duration of daily enrichment, delaying the start of enrichment in the morning was more detrimental than ending it prematurely before sunset.Varying the frequency of water applications during the CO2 enrichment period, from every second to every eighth day, had little effect, with no significant interactions between enrichment duration and watering frequency.Daily durations of CO2 enrichment somewhat shorter than the full sunrise-sunset periods, during the pre-planting stage, may not significantly reduce the total returns from the crop, but the consequent monetary saving would be quite small. A total of 1,355 random samples taken from bovine serums submitted for brucellosis testing in Victoria were submitted to the microscopic agglutination test for the presence of antibody to 12 serovars of Leptospira interrogans . The most common reaction obtained was to serovar hardjo , although the percentage of reactors varied from 24.8% in the metropolitan area to 56.3% in north-eastern Victoria (mean 40.6%). A total of 86.3% of farms from which 3 or more samples were taken had at least one reactor to serovar hardjo . The prevalence of antibody to other serovars was tarassovi (7.8% of reactors), ballum (3.7%), pomona (2.4%), autumnalis (1.8%) and bataviae (1.2%). Two-week dietary administration of 2500 ppb aflatoxin was sufficient to cause a decrease in bursal weights and a reduction in the number of splenic leukocytes in chicks. No significant effects on weight gain or feed efficiency were evident. The chicks also had elevated heterophil-to-lymphocyte ratios, suggesting a heightened reaction to stress. This effect could be blocked by dietary administration of the antioxidant butylated hydroxytoluene (BHT) at a concentration eight-fold over that normally present to preserve control feed. The relationship between postweaning DNA accumulation and body weight was calculated for a reference growing steer. Accumulation of DNA (hyperplasia) was calculated from patterns of protein accretion derived from literature data and protein-to-DNA ratios (Pro/DNA) measured in samples of muscles from chuck, round and plate, hides, intestines, rumens and livers of steers slaughtered at body weights ranging from 150 to 700 kg. Amounts of protein relative to DNA found in muscles from chuck, round and plate were used to estimate Pro/DNA changes in carcass. Corresponding values from intestines, rumen and liver were used to estimate Pro/DNA patterns for viscera. This ratio increased in carcass until animals reached body weights of approximately 300 kg and leveled off thereafter. No cell enlargement was evident in viscera. Thus, postweaning protein accretion in carcass results from both cell enlargement (hypertrophy) and DNA accumulation (hyperplasia) until body weights of 300 kg are attained, subsequent growth appeared to be hyperplastic in nature. A putative serine protease with a potential role in the plant biotic and abiotic stress response was purified from wheat leaf apoplastic fluid and partially characterized. Following two-dimensional electrophoresis a protein of Mr = 75 k and a pI of 4.2 to 4.5 was observed. This protein displayed in-gel protease activity and was specifically inhibited by phenylmethanesulfonyl fluoride and partially inhibited by Ca2+ and Zn2+, but not by E-64 or leupeptin. An internal tryptic fragment of 13 amino acids was identified by MALDI QqTOF MS/MS, and this peptide showed a high level of homology (85–100 % identity) to a highly conserved region of known plant subtilisin-like proteases. We demonstrated that the protease activity increased until a late stage of wheat leaf development and increased in response to heat shock. In both cases Rubisco large subunit was degraded with time. Protease activity was also increased during biotic stress. Leaves challenged with leaf rust (Puccinia triticina), showed an approximately three fold increase in protease activity during an incompatible interaction, compared to activity in mock-inoculated leaves and to leaves in a compatible leaf rust interaction. These results suggest that the expression of this serine protease could be involved in the defense response against both abiotic and biotic stresses. 相似文献
