The performance of insect-resistant transgenic poplar trees (Populusnigra) expressing a Cry1Ac gene from Bacillus thuringiensis subsp. Kurstaki HD-1 against poplar defoliators was evaluated in the field at the Manas Forest Station in Xinjiang Uygur
Autonomous Region during1994–1997. The results showed that the average percentage of highly damaged leaves on the transgenic
trees was 10% while that on the control trees in nearby plantations reached 80–90%. The average number of pupae per m2 of soil at 20cm depth in transgenic poplar plantation was 18 which was only 20% of that found in the non-transgenic control
field. The number of pupae and the leaf-damage on transgenic trees described above are all far below the threshold set for
chemical protection measures. The non-transformed poplar trees grown in the same plantation with the transgenic trees were
also protected indicating that cross protection occurred between these two kinds of trees. Insect-resistant transgenic poplar
trees have a potential application value in afforestation.

ABSTRACT Production of cacao in tropical America has been severely affected by fungal pathogens causing diseases known as witches' broom (WB, caused by Moniliophthora perniciosa), frosty pod (FP, caused by M. roreri) and black pod (BP, caused by Phytophthora spp.). BP is pan-tropical and causes losses in all producing areas. WB is found in South America and parts of the Caribbean, while FP is found in Central America and parts of South America. Together, these diseases were responsible for over 700 million US dollars in losses in 2001 (4). Commercial cacao production in West Africa and South Asia are not yet affected by WB and FP, but cacao grown in these regions is susceptible to both. With the goal of providing new disease resistant cultivars the USDA-ARS and Mars, Inc. have developed a marker assisted selection (MAS) program. Quantitative trait loci have been identified for resistance to WB, FP, and BP. The potential usefulness of these markers in identifying resistant individuals has been confirmed in an experimental F(1) family in Ecuador. 相似文献

Summary Seasonal effects of liming, irrigation, and acid precipitation on microbial biomass N and some physicochemical properties of different topsoil horizons in a spruce forest (Picea abies L.) were measured throughout one growing season. The highest biomass N was recorded in autumn and spring in the upper soil horizons, while the lowest values were obtained in summer and in deeper horizons. The clearest differences between the different soil treatments were apparent in autumn and in the upper horizons. Liming increased the microbial biomass N from 1.7% of the total N content to 6.8% (Olf1 layer) and from 1% to 2% of the total N content in the Of2 layer. . An increase in cation exchange capacity was observed down to the Oh layer, while soil pH was only slightly higher in the Olf1 and Of2 layers after liming. The effects of irrigation were less marked. The microbial biomass N increased from 1.7% of total N to 4.8% in the Olf1 layer and from 1% to 2% of total N in the Of2 layer. In the Olf1 layer an increase in C mineralization was observed. Acid precipitation decreased the microbial biomass N in the upper horizons from 4.8% of total N to 1.8% in the Olf1 layer and from 2% to 0.5% in the Of2 layer. 3 male sheep (phi 48.3 kg) were fed a semisynthetic diet containing acetyl urea as sole protein source and 15N-14C labelled acetyl urea (urea-C labelled) by intraruminal tube. A half life period of 4 hrs was established for the removal of labelled acetyl urea from the TCE-soluble portion of the ruminal fluid. The degree of 14C labelling in ruminal proteins was very low whereas the extent of 15N labelled protein synthesis was quite marked reaching a maximum between the 18th and 24th hour of experiment. The steepest rise of 15N incorporation into ruminal proteins was found to occur between 8 to 12 hrs after start of the experiment, i.e. at the time of peak level of 15N returned from 15N urea via the rumino-hepatic circulation. 23.3% of the amount of 14C activity administered (mean of all 3 experimental animals) was excreted through respiration. The curve patterns of both isotopes in the TCE soluble portion of the ruminal fluid were similar to that of the degasified TCE soluble portion of the blood blasma. At the peak time (8 hrs) a concentration of the nitrogen isotope of about 4 atom% excess of 15N was observed. The level of 14C labeling in blood plasma proteins was insignificant when compared with that of 15N labelling. The ratio at the peak time was 1:10; the same ratio was found for ruminal proteins. From this it can be concluded that the process of labelling of blood plasma proteins proceeds mainly through microbial protein synthesis. Sheep I and III excreted an average of 60.6% of 14C activity and 57.0% of the administered excess of 15N in the urine. 6 hrs after the beginning of the experiment 81% of the amount of urinary 14C activity was found to occur as acetyl urea; after 48 hrs this amount had decreased to 50%. All experimental sheep excreted a urinary sediment consisting mainly of acetyl urea. The level of faecal 14C excretion (1.4%-2.9% of the amount administered) was considerably lower than that of 15N excretion (9.1%--15.6% of the administered dose). The TCE soluble fraction of the faeces contained up to 2% of the 14C dose and 3% of the 15N dose. The true digestibility data of 15N from 15N acetyl urea varied between 96.4% and 98.2%. The ornithine transcarbamylase-deficient sparse fur mouse is an excellent model to study the most common human urea cycle disorder. The mutation has been well characterized by both biochemical and enzymological methods, but its exact nature has not been revealed. A single base substitution in the complementary DNA for ornithine transcarbamylase from the sparse fur mouse has been identified by means of a combination of two recently described techniques for rapid mutational analysis. This strategy is simpler than conventional complementary DNA library construction, screening, and sequencing, which has often been used to find a new mutation. The ornithine transcarbamylase gene in the sparse fur mouse contains a C to A transversion that alters a histidine residue to an asparagine residue at amino acid 117. 相似文献

Paramphistome infections are very common in ruminants and may induce clinical signs, but little is known about effective treatments. In this study, the efficacy of oxyclozanide against Calicophoron daubneyi was studied in goats and its activity tested against immature stages (10 days post-infection) at a dose of 22.5mg/kg bodyweight (BW) and against adult stages using two doses (15 and 22.5mg/kg BW). There was a reduction (82%) in the number of immature worms (compared to controls) but the result was not statistically significant. When tested against adult stages, however, oxyclozanide reduced the worm burdens by 95.6% and 95.9% at doses of 15 and 22.5mg/kg BW, respectively, with no significant difference between the two doses. The experiment demonstrated that oxyclozanide is highly effective in reducing the number of adult paramphistomes in goats. 相似文献
