Twelve anestrous, postpartum beef cows were used to determine the effect of calf removal on the effect of naloxone on serum luteinizing hormone (LH) concentrations. On d 1, six cows were injected iv with saline and six with 200 mg naloxone dissolved in saline. Blood samples were taken at 15-min intervals for 2 h before and 2 h after naloxone or saline administration. At the beginning of blood sampling, calves were removed from three cows in each treatment. At 48 h after calf removal (d 3), all cows were injected iv with 200 mg naloxone and blood samples were collected as on d 1. On d 1, naloxone treatment increased (P less than .01) serum LH concentrations from 1.2 +/- .3 ng/ml at time 0 to 4.3 +/- .6 ng/ml and 4.7 +/- .8 ng/ml at 15 and 30 min, respectively. Injection of saline had no effect on serum LH concentrations. Forty-eight-hour calf removal increased (P less than .01) serum LH concentrations in five of six cows (1.7 +/- .8 vs 4.4 +/- 1.2 ng/ml). Naloxone treatment failed to increase serum LH concentrations in these cows. Injection of naloxone increased (P less than .01) serum LH concentrations in the one cow that did not exhibit an LH increase after calf removal and in six cows whose calves were not removed (1.4 +/- .2 vs 4.4 +/- .5 ng/ml). The present study provides additional evidence that endogenous opioids regulate LH in the postpartum beef cow. We hypothesize that suckling stimulates an opioid inhibition of LH secretion and removal of the suckling stimulus removes the opioid inhibitory tone.  相似文献

The design of a t-piece ileal cannula and collecting device suitable for long term implantation in adult cockerels, is described. An inner retaining plate sutured to the serosal surface of the ileum prevents expulsion of the cannula while an outer retaining plate prevents the cannula from moving into the abdominal cavity. The cannulae, which were machined from PVC rod, were implanted in 11 cockerels, 10 of which survived in good health for 12 months. Reasons for the failures with earlier designs of cannulae are discussed.  相似文献

Wildlife, domesticated animals and humans are exposed daily to myriad chemicals present in our environment. The risk posed by these chemicals to one species is often determined by extrapolation from data gathered from another species. Several extensive studies have examined the capability of the liver to biotransform xenobiotics in animals commonly used in toxicity testing and in livestock. The present paper is a compilation of these data into a single source to permit comprehensive examination of inter-species variation in rates of hepatic biotransformation. Several substrates were studied for each enzyme system, including cytochrome P-450-dependent monooxygenases, epoxide hydrolases, UDP-glucuronosyltransferases, N-acetyltransferases, glutathione S-transferases and sulfotransferases. The numerous differences in substrate specificity for an individual enzymatic pathway reflect the apparent multiplicity of these enzymes in all 11 species studied. Several hundred- to several thousand-fold differences between species in enzymatic activities for certain substrates under well-defined conditions emphasize the need for caution and the risk of error in extrapolation of xenobiotic metabolism from one species to another. In spite of these uncertainties, knowledge of the rate of biotransformation may help us predict the fate of new chemicals in various species.  相似文献

A single-dilution indirect solid-phase radioimmunoassay (IRIA) was developed for the detection of low levels of anti-pseudorabies immunoglobulin G in swine sera. The assay derived increased sensitivity from the use of a second amplifying antibody. The IRIA was examined for its stoichiometry, amplification by secondary antibody, advantage of a single-dilution assay vs an end-point titration, sensitivity, and specificity. The assay had a near linear dose-response relationship with positive sera (serum-neutralization titer less than or equal to 1:16) and lacked a dose response with negative sera. With addition of the secondary antibody, the IRIA was enhanced 8.5-fold in net specific binding, and the end-point titer was amplified 32-fold. The single-dilution assay was proved to be a feasible test, compared with end-point titration. Anti-pseudorabies virus titers were at least 128-fold higher by IRIA than those by serum-neutralization test. Evidence indicated that there may be minimal or no cross-reactivity of IRIA antigen with anti-infectious bovine rhinotracheitis sera. The single-dilution IRIA was a rapid and sensitive test for anti-pseudorabies virus immunoglobulin G in swine sera.  相似文献

The objective of this study was to determine whether exposure of swine jejunum to crude culture filtrates containing Escherichia coli pig-specific, mouse-negative, heat-stable enterotoxin (STb) induces structural alterations in the jejunal mucosa of pigs. Two ligated intestinal loops in each of twelve 3-week-old pigs were exposed for 2 hours to sterile E coli culture filtrates from each of the following strains: 431 (STa-producing), 1261 (STa and STb-producing), and 1790 (STb-producing); recombinant strain HB101-pRAS-1 (STb-producing); the nontoxigenic K-12 variant HB101; or trypticase soy broth. Formalin-fixed sections from these loops were examined for sloughed cells around villi, and a lesion score was determined, indicating a change in villous epithelium from columnar to cuboidal or squamous cell types or to discontinuous epithelium. Villous lengths and crypt depths also were determined. For loops exposed to culture filtrates containing STa and STb or containing only STb, lesion scores and numbers of sloughed cells were greater (P less than 0.05) and villous length was shorter (P less than 0.01) than in loops not exposed to toxin. For loops exposed to culture filtrates containing STa, lesion scores, villus lengths, and numbers of sloughed cells were not different from those of loops not exposed to toxin. Therefore, exposure of swine jejunum to STb induced structural alterations in intestinal mucosa (ie, loss of villous absorptive cells and partial atrophy of villi) that were consistent with those causing compromised absorptive capacity.  相似文献

Crystalluria results from oversaturation of urine with crystallogenic substance. However, oversaturation may occur as a result of in vitro as well as in vivo events. The microscopic appearance of crystals only represents a tentative identification of their composition because variable conditions associated with their formation, growth, and dissolution may alter their appearance. Definitive identification is dependent on physical methods such as optical crystallography, x-ray diffraction, and electron microscopic analysis.  相似文献

Six series of experiments, involving 72 gilts in groups of 4 were carried out to investigate the effects of transportation, vibration and noise, and proximity of sexually mature adult breeding stock on attainment of first estrus in gilts of different ages. The animals grew normally during the periods of observation. Gilts aged 120 days (n = 12) at the start of the observations did not display estrus in response to exposure to vibration and noise or a 2 hr road journey. Similarly aged groups housed near adult breeding stock, but not exposed to vibration and noise, displayed estrus during the first 30 days of observation. All gilts aged 140 days (n = 12) displayed estrus within 5 days of being housed adjacent to adult breeding stock. Others, not housed near adult stock, but exposed to vibration and noise, or a road journey, did not consistently display estrus. When the observations were repeated on gilts aged 160 days (n = 12), the observations were similar. It was concluded that proximity of adult breeding stock and not transportation per se was the critical factor in inducing estrus behavior in normally growing gilts.  相似文献

A total of 174 dogs from an area hyperenzootic for Dirofilaria immitis were grouped into 4 age categories and necropsied; information was obtained on adult D immitis infections and on the presence of microfilariae. Serum samples from these dogs were examined by an enzyme-linked immunosorbent assay (ELISA) for antibody to adult D immitis and by an indirect fluorescent antibody test (IFAT) for antibody to microfilarial surface antigens. In dogs less than or equal to 5 months of age, necropsy demonstrated no evidence of infection; however, positive serologic results indicated that some of these dogs had prepatent infections. The percentage of dogs with ELISA titers (positive) increased with age, as did the percentage of dogs with adult D immitis infections. The IFAT results were positive in some dogs in each age category. Sera from all 29 dogs with occult infections were positive by ELISA. Sera from 6 of 20 dogs with occult dual-sex heartworm infections and 1 of 9 dogs with occult single-sex heartworm infections were positive by IFAT. For diagnosing occult dirofilariasis, the ELISA had a positive predictive value which increased with age of the dog to a maximum of 65.0% in dogs greater than or equal to 12 months of age; ELISA had a negative predictive value of 100% in all age groups. In contrast, positive and negative predictive values for the IFAT decreased with age of the dog to 60% and 37.5%, respectively, in dogs greater than or equal to 12 months of age.  相似文献

Zeranol implants were administered to 250 crossbred heifer calves at 1, 6 or 9 mo of age to evaluate growth, reproduction and calving performance. Heifers were assigned to eight treatment groups with 25 animals per group. Two additional groups of 25 heifers each were used to study the effects of multiple implants at two levels of nutrition on heifer performance. Implants at 1 mo of age (branding) increased heifer weights at 6 mo of age (weaning) by 5 kg (P = .08). Heifers receiving a combination of two implants gained faster (P less than .05) from weaning to breeding (6 to 13 mo) than controls or heifers implanted three times. Implants at either 6 or 9 mo increased (P less than .05) precalving pelvic areas (247 vs 241 cm2 and 248 vs 240 cm2 over controls, respectively). Implants did not affect the percent of heifers reaching puberty prior to breeding season. Conception rates in 62 d of breeding were comparable for implanted and control heifers (93 vs 96%), with the exception of heifers receiving implants at both 1 and 6 mo of age (56%). Calf birth weight, dystocia score, cow rebreeding rate and calf weaning weight were not affected by implant treatments. Heifers that received three implants and were fed at a high nutritional level (gained .62 vs .49 kg/d for regular level after weaning) tended (P greater than .10) to reach puberty at a higher rate prior to breeding and to have a higher total conception rate than implanted heifers on the regular nutrition level.  相似文献
