Crystallinity is an important property of woody materials; it responds to tree growth traits, structure, and chemical composition, and has a significant effect on Young’s modulus, dimensional stability, density, and hardness, etc. The ability of near-infrared (NIR) spectroscopy coupled with multivariate analysis to rapidly predict the crystallinity of slash pine (Pinus elliotii) plantation wood was investigated. The results showed that the NIR data could be correlated with the X-ray diffraction (XRD)-determined crystallinity of slash pine wood by use of partial least squares (PLS) regression, producing excellent coefficients of determination, r 2, and root mean square error of calibration, RMSEC. The use of either reduced spectral ranges or the selection of certain wavelengths consistent with known chemical absorptions did not have any detrimental effect on the quality of PLS models allowing the use of inexpensive, small, and portable spectrometers. These studies show that NIR spectroscopy can be used to rapidly predict the crystallinity of slash pine wood.  相似文献

The rehabilitation or intensified use of Imperata grasslands will require a much better understanding of their area, distribution, and characteristics. We generated estimates of the area of Imperata grasslands in tropical Asia, and suggested a typology of Imperata grasslands that may be useful to define the pathways toward appropriate land use intensification. We conclude that the area of Imperata grasslands in Asia is about 35 million ha. This about 4% of the total land area. The countries with the largest area of Imperata grasslands are Indonesia (8.5 million ha) and India (8.0 million ha). Those with the largest proportion of their surface area covered with Imperata are Sri Lanka (23%), the Philippines (17%), and Vietnam (9%). Laos, Thailand, Myanmar, and Bangladesh evidently all have similar proportions of their land area infested with Imperata (about 3 to 4%). Malaysia (< 1%), Cambodia (1%), and the southern part of China (2%) have but a minor proportion of their total land area in Imperata. The species was found widely distributed on the full range of soil orders. It occupied both fertile (e.g. some of the Inceptisols and Andisols) and infertile soils (Ultisols and Oxisols) across a wide range of climates and elevations. Imperata lands fall into four mapping scale-related categories: Mega-grasslands, itmacro-grasslands, meso-grasslands, and micro-grasslands. The mega-grasslands are often referred to as sheet Imperata. They are the large contiguous areas of Imperata that would appear on small-scale maps of say 1:1,000,000. We propose that this basic typology be supplemented with a number of additional components that have a key influence on intensification pathways: land quality, market access, and the source of power for tillage. The typology was applied in a case study of Indonesian villages in the vicinity of Imperata grasslands. We propose an international initiative to map and derive a more complete and uniform picture of the area of the Imperata grasslands. This should include selected studies to understand conditions at the local level. These are critical to build the appreciation of change agents for the indigenous systems of resource exploitation, and how they relate to local needs, values and constraints.  相似文献

Fourteen and 22 each of Salmonella Enteritidis and Salmonella Typhimurium (S. Typhimurium) were isolated from animals from 1983 to 1999 in Korea and tested for their antibiotic resistance patterns, phage types and resistance gene patterns. S. Typhimurium isolates were highly resistant to streptomycin, sulfisoxazole and tetracycline, 95, 95 and 86%, respectively. The incidence of multiple antibiotic resistance (resistant to more than two drugs tested) of S. Typhimurium isolates was extremely high (100%) comparing to S. Enteritidis isolates (21%). Two of the five ACSSuT (ampicillin, chloramphenicol, streptomycin, sulfisoxazole and tetracycline) resistant type S. Typhimurium isolates were phage type definitive type 104 (DT104).For the detection of resistance related genes in S. Enteritidis and S. Typhimurium isolates, particularly ACSSuT type S. Typhimurium, antibiotic resistance genes, cmlA/tetR, bla(PSE-1) and bla(TEM), and genus Salmonella specific gene, sipB/C, were amplified using four pairs of primers in a hot-start multiplex polymerase chain reaction (PCR). Two Korean isolates of S. Typhimurium DT104 showed bla(TEM) amplicons instead of bla(PSE-1) for the ampicillin resistance and they were susceptible to florfenicol. The multiplex PCR used in this study was useful in characterization of multiple drug resistant Salmonella isolates, especially ACSSuT type S. Typhimurium, and identification of beta-lactamase gene distribution among Salmonella isolates.  相似文献

Adipose tissue-derived mesenchymal stem cells (Ad-MSCs) are a promising source of cells for bone tissue engineering. Matrigel is a basement membrane extract containing multiple extracellular components. This mixture may promote the osteogenic differentiation of MSCs and provide a more appropriate microenvironment for transplanted cells. Here, we investigated the effect of Matrigel on the osteogenic potential of Ad-MSCs. Canine Ad-MSCs were cultured in 2D and 3D matrices and implanted into subcutaneous pouches of dogs either with or without Matrigel. Culture mineralization, cell adhesion efficiency, cell proliferation, osteoid matrix production and alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase activities were quantified and compared. Ad-MSCs grown in 2D cultures with Matrigel showed higher levels of calcium deposition and ALP activity than those grown in the absence of Matrigel under osteogenic conditions. In 3D cultures, the cells cultivated with Matrigel showed greater attachment, proliferation and osteogenic differentiation than those grown without Matrigel. In vivo, Ad-MSCs implanted with Matrigel showed higher osteogenic potential than those without Matrigel. In conclusion, these data suggest that the use of Matrigel can increase the osteogenic potential of canine Ad-MSCs.  相似文献

Soil health is important for the sustainable development of terrestrial ecosystem. In this paper, we studied the relationship between soil quality and soil microbial properties such as soil microbial biomass and soil enzyme activities in order to illustrate the function of soil microbial properties as bio-indicators of soil health. In this study, microbial biomass C and N contents (Cmic & Nmic), soil enzyme activities, and soil fertility with different fertilizer regimes were carried out based on a 15-year long-term fertilizer experiment in Drab Fluvo-aquic soil in Changping County, Beijing, China. At this site, 7 different treatments were established in 1991. They were in a wheat-maize rotation receiving either no fertilizer (CK), mineral fertilizers (NPK), mineral fertilizers with wheat straw incorporated (NPKW), mineral fertilizers with incremental wheat straw incorporated (NPKW+), mineral fertilizers plus swine manure (NPKM), mineral fertilizers plus incremental swine manure (NPKM+) or mineral fertilizers with maize straw incorporated (NPKS). In different fertilization treatments Cmic changed from 96.49 to 500.12 mg kg^-1, and Nmic changed from 35.89 to 101.82 mg kg^-1. Compared with CK, the other treatments increased Cmic & Nmic, Cmic/Corg (organic C) ratios, Cmic/Nmic, urease activity, soil organic matter (SOM), soil total nitrogen (STN), and soil total phosphorus (STP). All these properties in treatment with fertilizers input NPKM+ were the highest. Meantime, long-term combined application of mineral fertilizers with organic manure or crop straw could significantly decrease the soil pH in Fluvo-aquic soil (the pH around 8.00 in this experimental soil). Some of soil microbial properties (Cmic/Nmic, urease activity) were positively correlated with soil nutrients. Cmic/Nmic was significantly correlated with SOM and STN contents. Little is known about the innate defense mechanisms of the male reproductive tract. We cloned a 385-base pair complementary DNA and its genomic DNA named Bin1b that is exclusively expressed in the caput region of the rat epididymis and that is responsible for sperm maturation, storage, and protection. Bin1b exhibits structural characteristics and antimicrobial activity similar to that of cationic antimicrobial peptides, beta-defensins. Bin1b is maximally expressed when the rats are sexually mature and can be up-regulated by inflammation. Bin1b appears to be a natural epididymis-specific antimicrobial peptide that plays a role in reproductive tract host defense and male fertility.  相似文献

The histologic changes in the respiratory tracts of chickens were evaluated after hatchery fumigation with 40% formaldehyde vapors and vaccination against infectious bronchitis virus with live attenuated vaccine (Massachusetts serotype). One-day-old chickens were housed in four isolation units in controlled environmental conditions, fed and watered ad libitum, and separated into four groups: 1) fumigated and vaccinated birds (FV group); 2) nonfumigated and vaccinated birds (NFV group); 3) fumigated and nonvaccinated birds (FNV group); and 4) control group (C group). All birds were tested to be free from Mycoplasma gallisepticum and Mycoplasma synoviae. After necropsy on the first, eighth, and twenty-sixth days after birth, samples from tracheal upper portion and lungs were conventionally processed for light, scanning, and transmission electron microscopy. Tissue response was monitored by microscopic examination of trachea and lung. On the first day of observation, fumigated and vaccinated birds (FV group) showed extensively damaged tracheal epithelium with exfoliated areas and some active glands with electrodense granules, and in the lung, the primary bronchi epithelium had disorganized cilia and abundant lymphocytes, with emphysematous areas in tertiary bronchus. On day 8 after vaccination, cubical and cylindrical tracheal cell proliferation was observed, and on day 26, ciliated columnar epithelium was almost regenerated with heterophil corion infiltration, and hyaline cartilage nodules appeared in parabronchi. The nonfumigated and vaccinated birds (NFV) revealed less injury on the epithelial surface and a more rapid response to epithelial regeneration than the in only fumigated animals (FNV). The control group did not show remarkable morphologic changes. Postvaccinal and fumigation effects on the upper respiratory tract were temporary, whereas in lungs, increased emphysema, cartilage nodules in the interchange zone, and general lymphocyte infiltration had caused intensive injury.  相似文献
