Statistical algorithms for detecting safety signals are beginning to be applied to Animal Health Pharmacovigilance (PV) databases. How these signal detection algorithms (SDAs) perform in an animal health PV database is the subject of this report. Statistical methods and SDAs were assessed against a set of known signals in order to identify which SDAs were most appropriate for signal detection using the Elanco Animal Health PV database. A reference set of adverse events that should signal was created for 31 products across four species. Nine SDAs based on five disproportionality statistical methods were evaluated against the reference set. The performance metrics were sensitivity, precision, specificity, accuracy, and F score. For bovine and porcine products, the Observed‐to‐Expected (O/E) SDA was the closest in terms of geometric distance to 100% sensitivity and 100% precision. For canine and feline products, the Information Component (IC) SDA was geometrically closest to 100% sensitivity and 100% precision. Principal Component Analysis confirmed that the O/E and IC SDAs were unique performers with respect to one another and other SDAs. The performance of the SDAs was dependent on the choice of the statistical method with differences seen between animal species.  相似文献

The objective of this research was to evaluate the nitrogen, phosphorus and potassium (NPK) fertilizer effect on the development of ‘Valencia’ sweet orange nursery trees budded on Rangpur lime rootstocks in a greenhouse using containers. The experiment was a complete (1/5) 53 factorial randomized blocks design. Treatments comprised five concentrations (g per plant) of N (1.25; 6.25; 11.25; 16.25; 21.25); K (0.42; 3.75; 6.22; 9.34; 12.45); and P (0.19; 0.89; 1.59; 2.29; 2.99). Carbon dioxide assimilation rate (A), transpiration rate (E), stomata conductance (gs), and internal carbon dioxide (CO2) concentration (Ci) and water use efficiency (WUE). Reducing sugar, sucrose and starch were evaluated. Response functions were adjusted and nutrient rates for maximum yield were presented. The results indicated that the high levels of N (>16.25 g per plant) interferes negatively on photosynthesis. Potassium at intermediate fertilization levels (from 3.75 to 6.22 g per plant) had positive effects on photosynthesis. But P had little interference on photosynthesis. Carbohydrates levels were not related to nutrients fertilized.  相似文献

Chlorophyll meter readings below a threshold value would indicate the necessity to supply nitrogen (N) to the crop. This study aimed to determine threshold values for chlorophyll meter (SPAD) and nitrogen (N) rate effects on SPAD leaf measurements of potato cultivars through the crop cycle. Five N rates (0, 50, 100, 200, and 300 kg ha?1) were evaluated in a randomized block design, with four repetitions. SPAD index was measured on the oldest (OL) and on the fourth leaf from the apex (FL) at 7, 14, 21, 28, 35, and 49 days after the emergence of 90% of the plants in the plots. SPAD values decreased with plant age and were influenced by N rates and leaf position. At 21 DAE, the SPAD threshold values on FL were 43.0, 44.6, 46.5, and 50.0 for ‘Agata’, ‘Monalisa’, ‘Atlantic’, and ‘Asterix’, respectively. The corresponding values were 41.9, 43.5, 49.9, and 49.9 on OL. Plant age and leaf position should be standardized for the assessment of SPAD threshold values to diagnose nitrogen status of potato cultivars.  相似文献

Juvenile channel catfish Ictalurus punctatus were fed casein–gelatin‐based semi‐purified test diets with graded amounts of arginine (ARG, 0.5%, 1%, 2% and 4% of dry diet) for a 6‐week period, to evaluate the effects of dietary ARG supplementation on performance indicators, levels of plasma ARG, ornithine, citrulline, glutamine and glutamate, as well as selected innate immune parameters. Incremental dietary ARG resulted in significant improvements in weight gain, feed efficiency and protein efficiency ratios, protein retention, protein deposition and arginine deposition. Fish fed the 4% ARG diet had significantly higher values for all performance indicators, although these values were not different from those of fish fed the 2% ARG diet with the exception of protein retention, protein deposition and arginine deposition. Conversely, fish fed the 0.5% ARG diet had significantly lower values for all analysed plasma amino acids. Similarly, both phagocyte superoxide anion production and neutrophil respiratory burst were significantly decreased in fish fed the 0.5% ARG diet. These results demonstrate that the indispensability of dietary ARG is not only related to optimized protein utilization for growth but also its supplementation to fish diets may significantly impact several aspects of the immune system, including phagocyte function and thus improve overall fish health.  相似文献

The evaluation of a wide variety of immunostimulatory compounds in the diet of cultured fish has received heightened attention in recent years. However, two organic acids, polyhydroxybutyrate (PHB) and potassium diformate (KDF), have been researched to only a limited extent with aquatic species but have been shown to have various positive effects on terrestrial animals. Two algae extracts, carrageenan and alginic acid, also have been shown to elicit immunostimulation in some fish. Therefore, this study was conducted with red drum, Sciaenops ocellatus, as a model marine species to study the effects of organic acids and algae extracts as feed supplements by evaluating several humoral and cellular immune responses. A 7‐week feeding trial was conducted with disease‐free juvenile red drum (average initial wt. 2.6 ± 0.2 g). Semipurified diets were formulated to be isocaloric and contain 40% crude protein. Based on previous studies with other fish species, experimental diets were produced by supplementing the basal diet with KDF at 0.6%, PHB at 2%, alginic acid at 1% or carrageenan at 0.5% by weight in place of cellulose. Fish were stocked into 110‐L aquaria operated as a recirculating system with each diet assigned to three replicate aquaria containing 15 fish. At the end of 7 weeks, weight gain and feed efficiency were significantly (P < 0.0001) reduced in fish feed PHB compared to the basal diet and both algae extracts. The greatest phagocytic activity was found in fish fed the diet containing PHB. Total immunoglobulin level was higher in fish fed the diet supplemented with carrageenan. Goblet cell proliferation was greatest in the posterior end of the gastrointestinal tract but not different among dietary treatments. Organic acids and algae extracts evaluated in this study produced variable immunological responses in red drum with carrageenan showing the greatest potential as an immunostimulant.  相似文献

BackgroundIncreased concentration of haptoglobin (Hp) in serum is associated with survival of critically ill humans and horses. High serum activity of liver‐derived enzyme is associated with sepsis in children and foals.Hypothesis/ObjectivesInvestigate whether admission serum Hp and glutamic dehydrogenase (GLDH) are associated with systemic inflammatory response syndrome (SIRS) and survival of sick calves.AnimalsOne hundred two calves.MethodsRetrospective cross‐sectional study. Electronic medical records from all calves <30 days of age admitted to a teaching hospital for 8 years were reviewed. The signalment, clinicopathological findings, the presence of SIRS, final diagnosis, hospitalization time and outcome were recorded. A Cox proportional hazard ratio (HzR) were calculated to assess the association between clinicopathological variables and survival to discharge.ResultsSerum Hp concentrations were similar between SIRS (0.29 g/L; range, 0.05‐3.6) and non‐SIRS calves (0.22 g/L; range, 0‐4.2; P = .62). GLDH activity was similar between SIRS (12 U/L; range, 1‐1025) and non‐SIRS calves (9 U/L; range, 2‐137; P = .2). Absent suckle reflex (HzR: 6.44, 95% CI: 1.44‐28.86), heart rate (HR) < 100 beats per minute (bpm; HzR: 12.2; 95% CI: 2.54‐58.62), HR > 140 bpm (HzR: 3.59, 95% CI: 1.05‐12.33), neutrophil count <1.7 × 109/L (HzR: 7.36; 95% CI: 2.03‐26.66) and increased gamma‐glutamyl transferase activity (every 50‐unit, HzR: 1.12; 95% CI: 1.03‐1.21) were predictive of nonsurvival.Conclusions and Clinical ImportanceThe use of Hp and GLDH for prediction of survival in sick calves cannot be recommended at this time.  相似文献

At present, Foot-and-Mouth Disease (FMD) has been successfully controlled in most territories of South America, where only Ecuador and Venezuela remain as endemic countries. In this context, the precise characterization of circulating viruses is of utmost importance. This work describes the first molecular epidemiology study performed with the complete VP(1)-coding region of 114 field isolates of FMD virus (FMDV) type O, collected in the Andean countries mainly during 2002-2008. Sequences were aligned and compared to isolates responsible for emergencies in the Southern Cone of the continent between the years 2000 and 2006, and to other representative type O viruses worldwide. The results showed that FMD type O viruses isolated in South America and analyzed up to date are placed in 11 different lineages within the Euro SA topotype. Five of these lineages included viruses circulating in Ecuador and Venezuela during 2002-2008. The last emergencies reported in already-free areas in the Andean region, showed close relationships with viruses circulating in these endemic countries. Andean lineages showed a clear separation from the unique lineage containing viruses responsible for the emergencies in the Southern Cone, reflecting the different livestock circuits and providing evidence that support the ecosystem dynamics in the region. A wide geographical dissemination of the same strain in short time intervals has been observed, pointing to animal movements as the most significant risk parameter. This fact, together with an important generation of viral variants in areas under weak control strategies, reinforce the need of stronger official controls, as well as for establishing multinational cooperative measures in the border areas.  相似文献

The moxidectin (MXD) is an antiparasitic drug used in domestic animals. The mechanism of action, in mammals, involves GABA, a neurotransmitter with an important role in the sexual behavior control. Presently, the effects of 0.2 mg/kg therapeutic dose were studied on sexual behavior, sexual motivation, penile erection and central GABA levels. Sexual behavior results showed increased latencies to the first mount and intromission as well as in inter-intromission interval; a reduction in total mounts was detected on the drug post-treatment. No difference was observed between sexual motivation of control and experimental animals. MXD treatment reduced penile erection and hypothalamic GABA levels. The results suggest that MXD reduced sexual behavior and penile erection by an action on the hypothalamic GABA system. Probably, the lack of effects in the motivational test and the increased mount and intromission latencies as well as decreased total mounts could be explained as a consequence of reduced male rat erection process.  相似文献
