One kidney was collected from each of 32 fattening pigs at an abattoir in southern Vietnam in 2001 in order to demonstrate infecting Leptospira serovar and to associate renal macro- and microscopic findings with the presence of renal leptospires. Leptospires were demonstrated in 22 (69%) of the investigated kidneys by immunofluorescence. Multifocal interstitial nephritis (MFIN) and gross renal lesions (white spots) were each demonstrated in 24 (75%) kidneys. Leptospira interrogans serovar bratislava was isolated from one kidney. There was no association between presence of leptospires and MFIN (P=0.19), respectively and white spots (P=0.98), respectively. These data suggest that Leptospira infection is common among fattening pigs in the study area and that these animals may be considered as an occupational human health hazard. It is also suggested that the presence of white spots is an unreliable indicator of the presence of renal leptospires.  相似文献

An innovative in vitro hydroponic culture system used in potato (Solanum tuberosum L.) microtuber production is described in this paper. In vitro potato plantlets, 6–8 cm in height, derived from meristems of potato tubers cultured on 1/2 Murashige and Skoog (MS) nutrient medium after 30 days culture were cut into 1.5 cm stem node segments and used as explants. These stem nodes were cultured in a novel system called in vitro hydroponic culture system containing 1/2 MS medium supplemented with 0.5 μM naphthaleneacetic acid (NAA), 0.3 μM gibberellic acid (GA3), 3.7 μM adenine sulfate, 10% coconut water, 0.5 g/l activated charcoal, 80 g/l sucrose with or without 8 g l−1 agar. Liquid medium was distributed to the carrier substrates in each storey of the system with the aid of capillary robes. In the present paper, the effects of porous material used as substrate carrier and the number of storeys involved in the culture system on microtuber formation and their morphological characteristics are reported. Cotton layer substrate is more stable for organogenesis of potato microtubers. Microtubers, 3.19 mm in diameter and 49.82 mg in weight, could be harvested from a one-storey in vitro hydroponic culture system containing filter paper as substrate. However, microtubers cropped from three-storey in vitro hydroponic culture system with cotton layer were bigger and weightier than those from three-storey system containing filter paper. The above results of the in vitro hydroponic system examined in this study might open up a new approach in producing potato and other hygrophilous microtuber.  相似文献

We report the development of a Diversity Arrays Technology (DArT) marker panel and its utilisation in the development of an integrated genetic linkage map of white lupin (Lupinus albus L.) using an F8 recombinant inbred line population derived from Kiev Mutant/P27174. One hundred and thirty-six DArT markers were merged into the first genetic linkage map composed of 220 amplified fragment length polymorphisms (AFLPs) and 105 genic markers. The integrated map consists of 38 linkage groups of 441 markers and spans a total length of 2,169 cM, with an average interval size of 4.6 cM. The DArT markers exhibited good genome coverage and were associated with previously identified genic and AFLP markers linked with quantitative trait loci for anthracnose resistance, flowering time and alkaloid content. The improved genetic linkage map of white lupin will aid in the identification of markers for traits of interest and future syntenic studies.  相似文献

The aim of this study was to examine whether a morphological approach is efficient for selecting high‐quality porcine embryos produced by in vitro fertilization (IVF) under high polyspermy conditions. Frozen‐thawed Meishan epididymal spermatozoa showing moderate and high polyspermy were subjected to IVF (1 × 105 sperms/ml). Under conditions of moderate polyspermy, 4‐cell embryos selected at 48 hr after IVF (single selection) and 8‐cell embryos selected at 79 hr after IVF from the collected 4‐cell embryos (double selection) showed high developmental competence. Likewise, 4‐ and 8‐cell embryos produced by IVF under high polyspermy conditions also showed high competence for development to blastocysts. However, blastocysts derived from high polyspermy conditions had significantly fewer cells than those produced under moderate polyspermy conditions. Furthermore, the frequency of nuclear and chromosomal abnormalities in 4‐ and 8‐cell embryos produced under conditions of high polyspermy was significantly (p < .05) higher in comparison to moderate polyspermy conditions. These findings suggest that although high polyspermy affects the frequency of nuclear and chromosomal anomalies in porcine IVF embryos, subsequent selection based on morphological features of 4‐ and 8‐cell embryos even under high polyspermy conditions, could be an alternative option for selecting porcine IVF embryos with high development ability.  相似文献

This study aimed to examine the effects of ethanol-soluble components (Es) in soybean meal (SBM) on gut content transit, bile acid (BA) and pancreatic digestive enzyme secretions, nutrient apparent digestibility coefficients (ADC), liver and intestinal morphologies, and growth performance of pompano (Trachinotus blochii). The SBM was extracted with aqueous ethanol, then the supernatant and residue were separated and dried to produce ethanol-extracted SBM (ESBM) and the Es. Four experimental diets were formulated with fish meal (FM), ESBM and SBM as main dietary protein sources. The diets were denoted as follows: FMD (FM diet), SBMD (SBM diet), ESBMD (ESBM diet) and ESBM + EsD (ESBM plus Es diet). Thirty-five fingerling pompano with an initial body weight (BW) of 18.3 g were allocated to each of 12 polyvinyl chloride tanks (1000-L holding capacity), resulting in triplicate tanks per dietary treatment. For 10 weeks, the fish were hand-fed the experimental diets to apparent satiation twice daily. The results showed that the final BW, weight gain and specific growth rate were significantly lower, while the feed conversion ratio was higher in the SBMD and ESBM + EsD groups than in the ESBMD and FMD groups (p < 0.05). Fish fed SBMD and ESBM + EsD showed accelerated gastric transit, slowed intestinal mobility, and lowered secretions of BAs and pancreatic digestive enzymes as compared to those fed ESBMD and FMD. Morphological abnormalities in mucosal folds of the posterior intestine, but not the liver, were clearly observed in the SBMD and ESBM + EsD groups. These results indicated that the Es in SBM inhibited the digestive system, leading to decreased nutrient digestibility and growth performance in pompano. The findings of the present study suggested that removal of the Es would effectively improve the nutritional quality of SBM and enhance growth performance of pompano fed a SBM-based diet.  相似文献

Understanding of animal growth is important for the improvement of management and feeding practices; however, little is known about the growth curve in Vietnamese indigenous chicken. This study was performed to determine the most appropriate models for describing the growth curve of Vietnamese Mia chicken. The study evaluated the performances of the Logistic, Gompertz, Richards, and Bridges models of body weights in 224 Mia chickens. Models were fitted using minpack.lm package in R software and Akaike’s information criterion and Bayesian information criterion were used for model comparison. Based on these criteria, the Gompertz and Bridges were the best models for males and females, respectively. Estimated asymmetric weights (α) were ranged from 2,241.91 ± 14.74 (g) (Logistic) to 2,623.86 ± 30.23 (g) (Gompertz) for males and from 1,537.36 ± 10.97 (g) (Logistic) and 1,958.36 ± 72.92 (g) (Bridges) for females, respectively. The age at the inflection point was estimated from 9.32 to 10.5 weeks and from 8.51 to 9.86 weeks for males and females, respectively. In conclusion, the Gompertz model is the most suitable model for describing the growth curve of Mia chicken. The parameters obtained from growth models could help define feeding programs to meet nutritional needs from hatching to the age of maximum growth, reproduction programs, and marketing strategies.  相似文献

In the present study, we fabricated two-component extracellular matrix protein patterned substrates with fibronectin (FN) and laminin (LN) because of our interest in the mechanism of axonal regeneration and injury in the central and peripheral nervous systems. The authors investigated how the patterning order and method of attachment affected the spatial distribution and biological activity of the immobilized proteins. Micro-contact printing (μCP) techniques in concert with reactive surface chemistry were used to modify glass substrates with one- and two-component films of FN and LN, including micrometer-scale patterns of FN and LN. The composition and spatial distributions of both proteins on the patterned surfaces were characterized by x ray photoelectron spectroscopy, epi-fluorescence microscopy, atomic force microscopy, and time-of-flight secondary-ion mass spectrometry. The authors also characterized the biological activity of the top-most protein layer in a two-layer protein system as well as the ability of the top-most protein layer to mask the biological activity of an underlying protein layer using a fluorescence-based enzyme-linked immunosorbent assay. The order of protein deposition significantly affected the relative biological activity of the upper-most and underlying immobilized proteins. As a result of these optimization studies, maximum biological activity per surface protein was achieved by first immobilizing FN from solution, followed by μCP of LN on the FN. Addition of μCP LN films was able to mask ~84% of the underlying FN activity, whereas μCP FN films were only able to mask ~27% of the underlying LN activity.  相似文献

Large particles containing nitric acid (HNO3) were observed in the 1999/2000 Arctic winter stratosphere. These in situ observations were made over a large altitude range (16 to 21 kilometers) and horizontal extent (1800 kilometers) on several airborne sampling flights during a period of several weeks. With diameters of 10 to 20 micrometers, these sedimenting particles have significant potential to denitrify the lower stratosphere. A microphysical model of nitric acid trihydrate particles is able to simulate the growth and sedimentation of these large sizes in the lower stratosphere, but the nucleation process is not yet known. Accurate modeling of the formation of these large particles is essential for understanding Arctic denitrification and predicting future Arctic ozone abundances.  相似文献

Economic evaluations are critical for the assessment of the efficiency and sustainability of animal health surveillance systems and the improvement of their efficiency. Methods identifying and quantifying costs and benefits incurred by public and private actors of passive surveillance systems (i.e. actors of veterinary authorities and private actors who may report clinical signs) are needed. This study presents the evaluation of perceived costs and benefits of highly pathogenic avian influenza (HPAI) passive surveillance in Vietnam. Surveys based on participatory epidemiology methods were conducted in three provinces in Vietnam to collect data on costs and benefits resulting from the reporting of HPAI suspicions to veterinary authorities. A quantitative tool based on stated preference methods and participatory techniques was developed and applied to assess the non‐monetary costs and benefits. The study showed that poultry farmers are facing several options regarding the management of HPAI suspicions, besides reporting the following: treatment, sale or destruction of animals. The option of reporting was associated with uncertain outcome and transaction costs. Besides, actors anticipated the release of health information to cause a drop of markets prices. This cost was relevant at all levels, including farmers, veterinary authorities and private actors of the upstream sector (feed, chicks and medicine supply). One benefit associated with passive surveillance was the intervention of public services to clean farms and the environment to limit the disease spread. Private actors of the poultry sector valued information on HPAI suspicions (perceived as a non‐monetary benefit) which was mainly obtained from other private actors and media.  相似文献
