More than 50% of the annual worldwide harvest of mussels is produced in Europe. The mussel cultivation in Germany is based on an extensive on-bottom culture and depends entirely on natural resources for food, spat and space. Due to stakeholder conflicts and a lack of spat availability, mussel farmers tend to move offshore where space is not limited and adequate settlement guaranteed. Newcomers – the offshore wind farmers – are covering large areas in the German Bight which in contrast give the opportunity to use these areas in a multifunctional way by accepting mussel cultivation within the wind farms. This study compiles the basic data for offshore mussel cultivation in close vicinity to a designated offshore wind farm in the open sea of the German Bight and employs different case-scenario calculations to illustrate the impact of changing parameter values on overall profitability or non-profitability of this activity. Primary focus is placed on the production of consumer mussels but seed mussel cultivation is also taken into consideration. We show that production of consumer mussels with longline technology is sufficiently profitable even under the assumption of substantial cost increases. This is especially true, if existing capacities could be used. The cultivation of seed mussels depends on the possibility of using existing equipment. A substantial increase of seed mussel prices to at least 0.6 €, given the main cost categories remaining constant, turns this alternative into substantial profitability. This study concludes with providing some recommendations on how favorable terms or actions could further improve profitability of offshore mussel cultivation. Altogether, our results are intended to shed some light on business management topics that future offshore mariculture operators such as traditional mussel farmers should follow in order to be efficient.  相似文献

A calcium-selective electrode with solid ion exchanger was prepared from a solution of the calcium salt of a dialkylphosphoric acid in collodion. Accurate identification of mastitis pathogens is often compromised when using conventional culture-based methods. Here, we report a novel, rapid assay tested for speciation of bacterial mastitis pathogens using high-resolution melt analysis (HRMA) of 16S rDNA sequences. Real-time PCR amplification of 16S rRNA gene fragment, spanning the variable region V5 and V6 was performed with a resulting amplicon of 290bp. First, a library was generated of melt curves of 9 common pathogens that are implicated in bovine mastitis. Six of the isolates, Escherichia coli, Streptococcus agalactiae, Klebsiella pneumoniae, Streptococcus uberis, Staphylococcus aureus and Mycoplasma bovis, were type strains while the other 3, Arcanobacterium pyogenes, Corynebacterium bovis and Streptococcus dysgalactiae, were bovine mastitis field isolates. Four of the type strains, E. coli, S. agalactiae, K. pneumoniae and S. aureus, were found to be of human origin, while the other 3 type strains were isolated from bovine infections. Secondly, the melt curves and corresponding amplicon sequences of A. pyogenes, E. coli, S. agalactiae, S. dysgalactiae, K. pneumoniae, S. uberis and S. aureus were compared with 10 bovine mastitis field isolates of each pathogen. Based on the distinct differences in melt curves and sequences between human and bovine isolates of E. coli and K. pneumoniae, it was deemed necessary to select a set of bovine strains for these pathogens to be used as reference strains in the HRMA. Next, the HRMA was validated by three interpreters analyzing the differential clustering pattern of melt curves of 60 bacterial cultures obtained from mastitis milk samples. The three test interpreters were blinded to the culture and sequencing results of the isolates. Overall accuracy of the validation assay was 95% as there was difficulty in identifying the streptococci due to heterogeneity observed in the PCR amplicons of S. uberis. The present study revealed that broad-range real-time PCR with HRMA can be used as a powerful, fast and low-cost tool for the differentiation of clinically important bacterial mastitis pathogens.  相似文献

The health of cultivated blue mussels from an offshore and a nearshore sites was compared by assessing lysosomal membrane stability (LMS) and accumulations of lipofuscin and neutral lipids in the digestive glands. In addition, the condition index, parasite infestation rates, growth performance and energy status of the mussels were assessed during 10?months of sampling. Abiotic and biotic site characteristics were investigated by water analysis for each sampling cycle. Further, data of contaminant loads of the softbody and pollutant concentration of the water column, suspended particles and sediments from both sites were analysed. Results show that positive offshore effects due to dilution of contamination were not evident for all parameters analysed. Condition indices were high, and parasites were not present in offshore mussels. In contrast, the integrative health parameter LMS and growth performance were slightly, but not significantly, better in the nearshore cultivated mussels. These findings correspond with the higher contaminant concentrations in the softbodies of mussels and the water column from the offshore testing site. Both sites showed low LMS values and high accumulations of lipofuscin and neutral lipids, indicating that environmental conditions in the German Bight are generating stress for blue mussels even in the offshore areas. Exposure to fluvial transport points to a comparable probability for high contamination loads similar to nearshore areas. We therefore recommend adding a minimum salinity threshold to the definition of offshore aquaculture to exclude areas under fluvial influence.  相似文献

以麦秸纤维为增强材料、聚丙烯为基体物质、马来酸酐接枝聚丙烯(MAPP)为改性剂,制备麦秸纤维-聚丙烯复合材料。利用DMA、DSC、TG和SEM,探讨了MAPP的添加量(质量百分比1%、2%、5%、10%)和麦秸纤维形态(9、9~28、28~35、35目)对麦秸纤维 聚丙烯复合材料的热力学性能和结晶性能的影响。结果表明:①当MAPP的添加量为2%时,麦秸纤维-聚丙烯复合材料的储能弹性模量减小;当MAPP的添加量增加到5%、10%时,复合材料的储能弹性模量增加。②在麦秸纤维-聚丙烯体系内添加MAPP后,麦秸纤维 聚丙烯复合材料的结晶温度提高约1℃,结晶度增加了4%~8%;麦秸纤维以28~35目的形态作为聚丙烯基体的增强材料时,其复合材料的结晶温度为122.7℃,结晶率达到45.8%。③麦秸纤维-聚丙烯复合材料的热分解峰温分别为355和460℃。④麦秸纤维以纤维束的形态分布在基体聚丙烯中起增强作用,在整个体系内,麦秸纤维局部团聚且断裂明显。添加MAPP后,有利于基体物质在麦秸纤维表面的均匀覆盖。   相似文献

Aquaporins (AQPs) are essential membrane protein channels for the transport of water across membranes. Fluid movement in the epididymis is important for modulation of the luminal environment, in which sperm mature and reside. This study was designed to understand the morphology and localization of AQPs in ram efferent ducts (ED) and epididymis. For this purpose, the epididymis of seven animals were removed for histologic and immunohistochemical analyses. AQP1 immunoreactivity was observed in the apex of the ED, and AQP9 was found adjacent to the nuclei of the epithelial cells of the ED. The epithelial lining of ram epididymis is pseudostratified columnar and presents principal, basal, apical and narrow cells. In the initial segment (IS), a moderate reaction for AQP1 was observed in the apical cytoplasm of epithelial cells. An intense reactivity for AQP1 was noted over the microvilli of principal cells and in spermatozoa in the caput. In the corpus and cauda, AQP1 was noted only over the endothelial cells of vascular channels located in intertubular spaces. A weak‐to‐moderate reaction for AQP9 was observed in the nuclei of epithelial cells in the IS, caput and corpus of the epididymis. In the cauda, an intense reaction to AQP9 was observed in the epithelial border. In the IS, caput and corpus, the reactivity for AQP9 differed from those observed in domestic animals. The cauda showed a pattern similar to that previously described. These results indicate that AQPs 1 and 9 have reversed locations and roles in rams, suggesting activity variations related with fluid and solute absorption throughout the epididymis.  相似文献
