Shrimp farming is a fast‐expanding activity that has supported the growth in the supply of these crustaceans to consumers around the world. However, the end product is vulnerable to contamination at all stages of the process, including the rearing tanks, where current practices prioritize to raise stocking densities and the minimization of water renewal. It is thus important to evaluate the potential of these systems for the proliferation of undesirable microorganisms, which may render the product unfit for human consumption. In the present study, the presence of coagulase‐positive Staphylococcus, Salmonella spp., Vibrio spp., and total and thermotolerant coliforms was verified in biofloc tank and conventional pond systems used for the rearing of Litopenaeus vannamei in Pernambuco, Brazil, and the results were compared with the legislation regulating the marketing of fresh shrimp. Samples were collected from two biofloc tanks with a density of 375 shrimp m?2, and two conventional ponds with 12 shrimp m?2. None of the samples tested positive for either Salmonella spp. or coagulase‐positive Staphylococcus, which is consistent with the legislation. While no standards are defined legally for Vibrio spp. or coliforms, very low concentrations were recorded in both systems, in comparison with other studies. While some variation in bacterial contamination was observed over the rearing process, the end product of both systems was fit for human consumption. The results of the study indicate that, while the water is not renewed in the biofloc system, the development of undesirable microorganisms can be controlled, with no adverse effects for the end product.  相似文献

Atrazine is the triazinic herbicide most found in the rural aquatic environments due to its extensive use and its stability in such places. The mutagenicity and the genotoxicity of different concentrations of the Atrazine herbicide were determinated by the micronucleus test and the comet assay, using Oreochromis niloticus as test-system. The tested concentrations of Atrazine herbicide were 6.25, 12.5 and 25 μg/L, both for the micronuclei test and for the comet assay. The results showed a significant rate of micronuclei and nuclear abnormalities for all the tested concentrations of Atrazine herbicide. For the comet assay, we also observed results significantly different from the control in 6.25, 12.5 and 25 μg/L concentrations. Due to these results, we could infer that such herbicide may be dangerous to the lives of those organisms exposed to it.  相似文献

Sewage sludge (SS) is a by-product of wastewater treatment resulting from human and industrial activities. This waste is problematic because it is difficult to manage due to its volume and environmental impacts; therefore, new technologies for treatment are needed to make its final disposal feasible. Such technologies include the pyrolysis process that produces biochar, which can be used for agricultural purposes. The present study reports a two-year field experiment with SS biochar produced under different pyrolysis temperatures and its application to soil in combination with mineral fertilizer to evaluate its effect on soil chemical properties and on corn nutrition and yield. To conduct this evaluation, 15 Mg ha?1 of SS biochar combined or not with mineral fertilizer (NPK) was applied to the soil in the 2015 and 2016 cropping seasons. SS biochar increased soil chemical attributes, especially phosphorus, magnesium, cation exchange capacity and base saturation. Biochar resulted in a greater absorption of nutrients by plants and higher corn yields. However, SS biochar alone was incapable of increasing K uptake by corn. It was concluded that the application of SS biochar can replace mineral fertilizers (nitrogen, phosphorus and micronutrients) for corn production.  相似文献

The chemical composition and biological properties of Portuguese wild mushrooms ( Cantharellus cibarius, Hypholoma fasciculare, Lepista nuda, Lycoperdon molle, Lycoperdon perlatum, Ramaria botrytis, Tricholoma acerbum) were evaluated in order to assess these products as sources of nutrients and nutraceuticals. The analyzed mushrooms contain very useful phytochemicals such as phenolics, tocopherols, ascorbic acid, and carotenoids. All of the species proved to have antioxidant activity (measured by four different methods), being more significant for R. botrytis (EC 50 values < 1 mg/mL). Lycoperdon species were resistant to all of the tested microorganisms, and the other samples revealed antimicrobial activity selectively against Gram-positive bacteria, with very low minimal inhibitory concentration, in some cases, even lower than the standard. The combination of bioactive compounds and rich nutritional composition (high contents in protein and carbohydrates, low content in fat with the precious contribution of unsaturated fatty acids and the absence of trans fatty acids) in the mushroom makes it a very special food.  相似文献

The harvesting of non-timber forest products has been proposed as an alternative to timber harvesting that can increase rural income while having a reduced impact on forest structure. However, surprisingly little is known about the biological consequences of harvesting these products. We conducted a 3-year experiment in which we simulated the stem harvesting of the Amazonian plant Ischnosiphon polyphyllus, which is used by traditional and indigenous populations in the Amazon for the construction of baskets, mats, and other handicrafts used in manioc cultivation. We found that plant mortality is limited in all except the most extreme harvesting treatments. However, we also found that plants recuperate extremely slowly from experimental harvesting. These results suggest that current harvesting strategies may not be conservative enough to ensure long-term population survival.  相似文献

The genus Psidium includes important fruit crops. However, there are very few studies focusing on its reproductive biology, which limits the establishment of breeding programs. The present work investigated the reproductive biology of Psidium guajava and Psidium cattleianum in terms of compatibility of crossings between these two species aiming at interspecific hybridization because the latter species is an important source of resistance against the nematode Meloidogyne enterolobii. Several types of crosses were performed to understand the reproductive biology of these species, including the compatibility of intra- and interspecific crossings, using assisted in vivo germination of pollen grains on the stigma. In addition, the in vitro germination of both Psidium species was studied at different stages of fruit development to rescue young seeds to improve the chances of obtaining the hybrids. No fruits of 270 pollinations were obtained on guava buds at the pre-anthesis stage, regardless of the source of the pollen grain and the cultivar used as female genotype. Microscopic analyzes demonstrated the germination of pollen grains and pollen tube growth at crosses between guava cv. ‘Pedro Sato’ (P. guajava) and Psidium cattleianum. High germination percentages of Psidium cattleianum seeds were obtained in MS medium without sucrose or containing 15 g/L of this carbohydrate.  相似文献

This research is part of a project aimed at verifying the potential of a specifically assessed wooded riparian zone in removing the excess of combined nitrogen from the Zero River so as to reduce nutrient inputs into the Venice Lagoon. Among the specific objectives of the project, there was the determination of change in the composition of the microbial populations of soil of the wooded riparian strip. The composition of the bacterial communities collected at different depths inside and outside the riparian strip was determined by combined approaches involving cultivation (CFU), microscopic approaches (CTC test), and DNA-based techniques (ARDRA and DGGE). The size of the living population was the same inside and outside the experimental strip, with a minor percentage of culturable bacteria. Higher numbers of metabolically active bacteria and higher bacterial diversity were detected in the internal soil, with deeper soil layers showing reduced diversity, thus indicating that soil management within the riparian strip effectively supports the viability of bacterial communities. Total operational taxonomic units (OTUs) and percentage of single OTUs were also found to be always higher in the internal soil samples for all soil layers, with the percentage of Firmicutes increasing and Actinobacteria decreasing with depth. The increasing soil organic carbon inputs due to the contribution of the growing plants were found to support bacterial diversity in all soil layers. DNA-based analysis also indicated a clear effect of the applied treatments on soil bacterial diversity and a well-defined separation of the bacterial communities related to the different soil layers of the riparian strip.  相似文献

Infection with the gastrointestinal nematode Haemonchus contortus causes considerable losses in the sheep industry. In this study, we evaluated the effect that climate has on third-stage larvae (L3) of H. contortus in terms of their migration from sheep feces to Brachiaria decumbens grass, as well as their distribution among the forage plants. Fecal samples containing H. contortus L3 was deposited on the soil among the herbage at an initial height of 30 cm. Sample collection began 24h after contamination and was performed on alternate days over 13 days. The L3 were recovered and quantified in three strata (heights) of grass (0-10 cm, 10-20 cm and >20 cm) as well as in the remaining feces and a superficial layer of soil, collected from beneath the feces. In order to obtain results under different environmental conditions, fecal samples containing H. contortus L3 were deposited on pasture in January (summer), in April (autumn), and July (winter). In all of the periods, the L3 were able to migrate from the feces to the herbage. However, rains, accompanied by high relative humidity and high temperatures, apparently favored migration. The highest L3 recovery rate in the pasture was in the summer observation period, which had the highest number of days with measurable precipitation, high relative humidity (>68.2%), and the highest temperatures at the soil level (minimum and maximum means of 19°C and 42°C, respectively). Under those conditions, larvae began to reach the upper stratum of the grass (>20 cm) by 24h after the deposition of fecal matter, the number of larvae having reached that stratum peaking at seven days after deposition. In the autumn observation period, there was no rainfall in the first five days post-contamination. During that period, high numbers of larvae were found in the fecal samples demonstrating that feces can act as a reservoir of larvae in the absence of rain. Except for two days in the summer observation period, when most of the L3 were recovered from the tops of blades of grass, L3 where located predominantly at the base of the herbage. In conclusion, rainfall favors the migration of L3 from feces to herbage. In addition, larval migration up and along blades of grass can occur relatively rapidly when the temperature is high.  相似文献

Gluten is an important protein complex for baking products found in wheat, rye, barley, and some oat varieties. However, some people need to avoid these grains and their products because they result in gluten‐related disorders. The only treatment for these individuals is to engage in a gluten‐free diet. The objective of this work was to verify if the gluten content of several commercial food products sold in Brazil complied with their labeling. The Méndez ELISA R5 sandwich method was used to analyze 437 samples, and of these, 70% were labeled as gluten‐free, 26% as containing gluten, and 4% not labeled in relation to gluten. The results indicated that 89% of the products labeled as gluten‐free were correctly labeled and 11% were not, which represented a risk for celiac people.  相似文献

High‐density lipoprotein (HDL) is the main lipoprotein in the follicular fluid, and it has anti‐inflammatory, antioxidant and cryoprotectant properties. The anti‐inflammatory potential and antioxidant potential are derived from its lipid composition, especially the apolipoprotein AI (ApoAI) and paraoxonase 1 (PON1). The aim of this study was to evaluate the effect of HDL during in vitro maturation (IVM) on oocyte maturation and early bovine embryo development. For this, cumulus–oocyte complexes (COCs) were obtained from bovine ovaries collected at a local slaughterhouse. COCs (n = 2,250) were allocated into three groups (n = 50 COCs/group) according to the addition of HDL protein (HDL‐P) during IVM for 22 hr: 0 (control), 50 and 150 mg/dl. After IVM, COCs were inseminated (in vitro fertilization) and cultivated for 7 days. Total cholesterol concentration, total protein, triglycerides and ApoAI concentrations on IVM medium increased proportionally to HDL‐P addition. However, PON1 activity was not detected in any treatment. The addition of HDL‐P did not affect nuclear maturation rate, endogenous reactive oxygen species and glutathione levels in COCs (p > 0.05). The highest HDL‐P concentration (150 mg/dl) decreased cleavage and blastocyst rate (p < 0.05). Moreover, the HDL‐P 150 mg/dl group had lower cellular count/blastocyst than the 50 mg/dl group (p < 0.05). However, the addition of HDL‐P did not affect relative gene expression of evaluated genes. In conclusion, the complex HDL/ApoAI obtained from human plasma, in the absence of PON1 activity during in vitro oocyte maturation, decreased initial embryo development.  相似文献
