Eight plant species were subjected to artifical acid rains of pH 2.5, 2.0, 1.5, 1.0 and 0.5 in order to determine the threshold for and symptoms of damage. The plants were Erechtites, Robinia, Pinus, Quercus, Carya, Liriodendron, Acer and Corpus from the Coweeta Hydrologic Laboratory near Franklin, North Carolina Droplets of pH 2.0 produced brown necrotic spots on all species except Pinus while droplets of pH 1.0 produced necroses on leaves of all species examined. The size of necrotic spots increased with increasing acidity. Comparison of these results with the literature suggests that developing leaves are more easily damaged than are the mature leaves used in this study. The volume weighted average rainfall pH for Coweeta is 4.6 with observations ranging from 3.2 to 5.9. Results of this study suggest that a 10-fold increase in acidity from pH 3.2 to 2.2 in a single spring or summer storm could bring damage or death to mature leaves of dominant flowering plants in the Southern Appalachians. 相似文献

Climate and soil conditions in South Florida along with an extensive canal system facilitate movement of agricultural pesticides into surface waters. In a two-year study (2002-2004) of the currently used pesticides in South Florida, atrazine, endosulfan, metolachlor, chlorpyrifos, and chlorothalonil were the most frequently detected in the canals and in Biscayne Bay, with average concentrations of 16, 11, 9.0, 2.6, and 6.0 ng/L, respectively. Concentrations of atrazine and chlorpyrifos were highest near corn production. Chlorothalonil and endosulfan concentrations were highest near vegetable production, with no clear trend for metolachlor, which is used on multiple crops. Concentration data were used to calculate an aquatic life hazard potential for the planting period (November) versus the harvest period (March). This analysis indicated that a higher hazard potential occurs during harvest, primarily from the use of endosulfan. These data will also serve to document canal conditions prior to implementation of the Comprehensive Everglades Restoration Plan (CERP). 相似文献

From 1999 to 2002, the variations in carbon flux due to management practices (shrub removal, thinning) and climate variability were observed in a young ponderosa pine forest originated from clear-cutting and plantation in 1990. These measurements were done at the Blodgett Forest Ameriflux site located in the Sierra Nevada Mountains of California. Thinning in spring 2000 decreased the leaf area index (LAI) by 34% and added 496 g C m−2 of wood and leaf debris at the soil surface. Total ecosystem respiration was not significantly affected by thinning (1261 g C m−2 in 1999 and 1273 g C m−2 in 2000), while canopy photosynthesis decreased by 202 g C m−2. As a result the ecosystem shifted from a net sink of CO2 in 1999 (−201 g C m−2) to a small net source in 2000 (13 g C m−2). Woody and leaf debris resulting from thinning only accounted for maximum 1% and 7% of the total respiration flux, respectively. Thinning did not affect the relative proportion of the different components of respiration to an observable degree. Low soil water availability in summer 2001 and 2002 decreased the proportion of soil respiration to the total respiration. It also imposed limitations on canopy photosynthesis: as a result the ecosystem shifted from a sink to a source of carbon 1 month earlier than in a wetter year (1999). The leaf area index and biomass of the stand increased rapidly after the thinning. The ecosystem was again a sink of carbon in 2001 (−97 g C m−2) and 2002 (−172 g C m−2). The net carbon uptake outside the traditionally-defined growing season can be important in this ecosystem (NEE = −50 g C m−2 in 2000), but interannual variations are significant due to differences in winter temperatures. 相似文献

The oxidative stability of long-chain polyunsaturated fatty acid (PUFA) and docosahexaenoic acid (DHA)-containing fish and algae oils varies widely according to their fatty acid composition, the physical and colloidal states of the lipids, the contents of tocopherols and other antioxidants, and the presence and activity of transition metals. Fish and algal oils were initially much more stable to oxidation in bulk systems than in the corresponding oil-in-water emulsions. The oxidative stability of emulsions cannot, therefore, be predicted on the basis of stability data obtained with bulk long-chain PUFA-containing fish oils and DHA-containing algal oils. The relatively high oxidative stability of an algal oil containing 42% DHA was completely lost after chromatographic purification to remove tocopherols and other antioxidants. Therefore, this evidence does not support the claim that DHA-rich oils from algae are unusually stable to oxidation. Addition of ethylenediaminetetraacetic acid (EDTA) prevented oxidation of both fish and algal oil emulsions without added iron and at low iron:EDTA molar concentrations. EDTA, however, promoted the oxidation of the corresponding emulsions that contained high iron:EDTA ratios. Therefore, to be effective as a metal chelator, EDTA must be added at molar concentrations higher than that of iron to inhibit oxidation of foods containing long-chain PUFA from either fish or algae and fortified with iron. 相似文献

Food ingredients containing alpha-1,6-galactoside bonds elicit gastrointestinal disturbances in monogastric animals, including humans. Pretreatment of such ingredients with alpha-galactosidase (EC 3.2.1.22) has the potential to alleviate this condition. For this purpose, a thermostable alpha-galactosidase from Thermoanaerobacterium polysaccharolyticum was purified by a combination of anion exchange and size exclusion chromatographies. The enzyme has a monomeric molecular weight of approximately 80 kDa; however, it is active as a dimer. The optimum temperature for enzyme activity is 77.5 degrees C. Approximately 84 and 88% of enzyme activity remained after 36.5 h of incubation at 70 and 65 degrees C, respectively. Optimum activity was observed at pH 8.0, with a broad range of activity from pH 5.0 to 9.0. Different transition metals had weak to strong inhibitory effects on enzyme activity. The K(m) and V(max) of the enzyme are 0.29-0.345 mM and 200-232 micromol/min/mg of protein, respectively. Importantly, enzyme activity was only slightly inhibited by 75-100 mM galactose, an end product of hydrolysis. Enzyme activity was specific for the alpha-1,6-galactosyl bond, and activity was demonstrated on melibiose and soy molasses. 相似文献

The sorption of Cd(II) on illite was studied at initial Cd concentrations from 1 to 1000 μg 1?1, with a constant solid phase concentration of 0.5 g 1?1. Percentage sorbed at equilibrium increased sharply from pH 6.5 to 9.0, with a shift toward increased adsorption at lower initial concentrations, indicating specific, non-equivalent sites. Freundlich log-log plots were linear with a slope of 0.83. Susquehanna River sediments (silt/clay fraction) showed similar behavior, sorbing Cd somewhat more strongly than illite, as did mont-morillonite, while kaolinite sorbed Cd less strongly. Sorption on illite was not altered appreciably by removing the Fe- or Mn-oxide coating with dithionite, by removing organic matter with H2O2, or by using a carbonate-free medium. Equilibration time, desorption at lower pH's, and the effect of citrate in shifting the sorption to higher pH's were also studied. Fungus-growing termites (Isoptera, Macrotermitinae) play an important role in tropical ecosystems in modifying soil physical properties. Most of the literature regarding the impact of termites on soil properties refers to termite epigeous mounds. In spite of their abundance and activity in African savannas, few studies deal with the properties of underground nest structures (fungus-comb chambers) built by subterranean Macrotermitinae termites. We tested whether these termites significantly modify the soil physico-chemical properties within their nests in a humid tropical savanna and whether these effects are different for two termite species with differing building behaviour. Termite-worked soil material was collected from fungus-comb chamber walls of two widespread species: Ancistrotermes cavithorax, which builds diffuse and ephemeral nests and Odontotermes nr pauperans, which most often builds concentrated and permanent nests for a comparatively much longer period of time. Neither species influenced soil pH but both significantly modified soil texture and C-N content in their nest structures. A strong impact on clay-particle size was also detected but no significant differences in clay mineralogy. Thus Odontotermes has a greater effect on soil properties, that could be explained by its building behaviour and the concentration in space of its nest units. Therefore, spatial pattern and life-span of fungus-comb chambers should be an important parameter to be considered in the functional role of subterranean Macrotermitinae termites in the savanna. 相似文献

Concentrations of dissolved urea were monitored in several Chesapeake Bay tributaries from 1998 to 2002. Urea is a commonly used agricultural fertilizer and is also a breakdown product of poultry manure, which is used as an additional source of fertilizer throughout the watershed. Two trends were apparent. First, in several of the tributaries, seasonal peaks in ambient urea concentration coincided with the periods of the year (early spring and mid summer) when agricultural applications are most common. Second, highest annual mean concentrations (up to 2.6 μg atom N L?1), as well as highest individual measurements (up to 24 μg atom N L?1), were found for those tributaries with the most intensive agricultural and poultry operations. Peak urea concentrations were significantly higher than those which could be attained from in situ sources and regeneration. These elevated concentrations are of concern because this form of nitrogen has been shown to be a preferred form of nitrogenous nutrient for many phytoplankton, including some dinoflagellates which form harmful algal blooms. These results demonstrate that urea from land based sources can contribute to anthropogenic eutrophication. 相似文献

Permethrin is the most popular synthetic pyrethroid insecticide in agriculture and public health. For the development of the enzyme-linked immunosorbent assay (ELISA) to evaluate human exposure to permethrin, the glycine conjugate (DCCA-glycine) of a major metabolite, cis/trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane-1-carboxylic acid (DCCA), of permethrin was established as the target analyte. Four different types of the cis- and trans-isomers of immunizing haptens were synthesized as follows: N-(cis/trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane-1-carbonyl)glycine (hapten 3), N-(cis/trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane-1-carbonyl)-4-amino-l-phenylalanine (hapten 5), N-(N-(cis/trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane-1-carbonyl)glycine)amino-6-(2,4-dinitrophenyl)aminohexanoic acid (hapten 9), and N-(cis/trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane-1-carbonyl)glycine-4-oxobutanoic acid (hapten 24). Sixteen polyclonal antibodies produced against each cis- or trans-hapten-thyroglobulin conjugate as immunogens were screened against numerous hapten-bovine serum albumin conjugates as coating antigens. Six ELISAs with both a heterologous hapten structure and a heterologous hapten configuration (cis/trans or trans/cis) between antibody and coating antigen showed a high sensitivity for the target analyte. The IC50 was 1.3, 2.1, and 2.2 microg/L for the trans-target analyte and 0.4, 2.3, and 2.8 microg/L for the cis-target analyte. The immunizing haptens, except for hapten 5, provided the target specific antibodies. Molecular modeling of the haptens supported the selection of reasonable immunizing haptens that best mimicked the target analyte. Hapten 5 was suitable as a coating antigen rather than as an immunogen since it had a different geometry. Very low cross-reactivities were measured to permethrin, its free metabolite (DCCA), PBA-glycine conjugate, and glycine. The ELISA will be optimized for the detection of total cis/trans-DCCA-glycine in human urine samples. 相似文献

Molecular breeding is becoming more practical as better technology emerges. The use of molecular markers in plant breeding
for indirect selection of important traits can favorably impact breeding efficiency. The purpose of this research is to identify
quantitative trait loci (QTL) on molecular linkage groups (MLG) which are associated with seed protein concentration, seed
oil concentration, seed size, plant height, lodging, and maturity, in a population from a cross between the soybean cultivars
‘Essex’ and ‘Williams.’ DNA was extracted from F2 generation soybean leaves and amplified via polymerase chain reaction (PCR) using simple sequence repeat (SSR) markers. Markers
that were polymorphic between the parents were analyzed against phenotypic trait data from the F2 and F4:6 generation. For the F2 population, significant additive QTL were Satt540 (MLG M, maturity,
r2 = 0.11; height, r2 = 0.04, seed size, r2= 0.06], Satt373 (MLG L, seed size, r2 = 0.04; height, r2 = 0.14), Satt50 (MLG A1, maturity r2 = 0.07), Satt14 (MLG D2, oil, r2 = 0.05), and Satt251 (protein r2 = 0.03, oil, r2 =0.04). Significant dominant QTL for the F2 population were Satt540 (MLG M,height, r2 = 0.04; seed size, r2 = 0.06) and Satt14 (MLG D2, oil, r2 = 0.05). In the F4:6 generation significant additive QTL were Satt239 (MLGI, height, r2 = 0.02 at Knoxville, TN and r2 = 0.03 at Springfield, TN), Satt14 (MLG D2, seed size, r2 = 0.14 at Knoxville, TN), Satt373 (MLG L, protein, r2 = 0.04 at Knoxville, TN) and Satt251 (MLG B1, lodging r2 = 0.04 at Springfield, TN). Averaged over both environments in the F4:6 generation, significant additive QTL were identified as Satt251 (MLG B1, protein, r2 = 0.03), and Satt239 (MLG I, height, r2 = 0.03). The results found in this study indicate that selections based solely on these QTL would produce limited gains (based
on low r2 values). Few QTL were detected to be stable across environments. Further research to identify stable QTL over environments
is needed to make marker-assisted approaches more widely adopted by soybean breeders.
