Summary The use of soil. naturally infested with Heterodera avenae, to select resistant heterozygotes in backcross progenies of wheat, was tested for reliability. Selfed progenies from plants selected as resistant were cultured monoxenically in test tubes with nematodes hatched from single cysts, while backcross progenies from the same parent plants were grown in pots of naturally infested soil. Cyst counts were made after two months' growth. The results showed that over 50% of the backcross lines, screened in previous generations with naturally infested soil, had been erroneously selected as resistant. The test tube cultures clearly differentiated lines carrying resistance from those which were susceptible and corroborated results from pot tests.  相似文献

A twin‐screw extrusion study was performed in replicated trials to produce vegetable‐based feeds for juvenile yellow perch. Two isocaloric (3.06 kcal/g) experimental diets were balanced to contain 20 and 40% distillers dried grains with solubles (DDGS) and a constant amount (20%) of fermented high‐protein soybean meal (PepSoyGen) as the fishmeal protein replacers; crude protein content was targeted at 40%. A fishmeal‐based diet was used as a control. Extrusion conditions included conditioner steam (0.11–0.16 kg/min), extruder water (0.11–0.19 kg/min), and screw speed (230–300 rpm). Increasing DDGS from 0 to 40% led to a considerable rise in bulk density, lightness L*), yellowness (b*), and unit density but to decreases in water activity (aw) and expansion ratio by 12.6, 14.4, 23, 21, 31, and 13%, respectively. The lowest unit density of 791.6 kg/m3 and highest bulk density of 654.5 kg/m3 were achieved with diets containing 20 and 40% DDGS, respectively; changes in DDGS content did not affect extrudate moisture, absorption index, or thermal properties. Raising DDGS from 0 to 40% resulted in an increase in water solubility and redness (a*) by 13.4 and 35%, respectively. All extrudates had high durability (>98%), and low aw of less than 0.5. Overall, this study yielded viable feeds for yellow perch.  相似文献

This paper describes the successful use of two tagging systems, both produced by Northwest Marine Technology Inc., on larval and postlarval giant freshwater prawns, Macrobrachium rosenbergii. Visible implant (VI) elastomer tags (a coloured liquid that solidifies under the epidermis) were used on stage XI larval prawns (mean weight 0.01 g) and postlarval prawns (mean weight 0.07 g). VI alphanumeric tags (small biocompatible plastic labels also inserted under the epidermis) were tested on postlarval prawns (from a weight of 0.5 g). Tags were inserted using clove oil as anaesthetic, and survival, mortality and growth rates of tagged animals were compared with those of controls that were handled but not anaesthetized or marked. Twenty per cent of the larval prawns (the smallest of the group) died just after tagging, but thereafter the remaining prawns survived well, as did all the tagged postlarval prawns. Visibility of the VI elastomer tags in larval prawns deteriorated with time, though 79% of marks were still visible to the naked eye 70 days after tagging. VI elastomer tags in the postlarval group remained clearly visible for up to 100 days. Visibility of the VI alphanumeric tags fell shortly after tagging, but remained adequate thereafter. Moult rates in control and tagged animals were the same in larvae with VI elastomer tags and postlarvae with VI alphanumeric tags, but the moult rate in the postlarval prawns given elastomer tags was slower than in controls. Rates of growth were similar in tagged (elastomer and alphanumeric) and control postlarval prawns, once the size‐dependent mortality of tagged larval prawns was taken into account. We conclude that VI elastomer tags could be used to mark small numbers of individual larval and immediately postlarval prawns for periods of several months, and that VI alphanumeric tags could be used to mark an unlimited number of individuals from a size of approximately 0.5 g.  相似文献

Quadruplicate groups of juvenile red drum (initial mean weight 2.7 g; 20 fish per replicate) were fed experimental diets containing 35% crude protein and graded levels of lysine for eight weeks. Lysine concentration in the basal diet was 1.2% and was supplied by a combination of peanut meal and shrimp-head meal. The basal diet was supplemented with lysine-HCl to provide 1.6, 2.0, and 2.4% lysine. Each of these diets contained an essential amino acid (EAA) premix. Two additional diets were formulated to contain 1.2 and 2.4% lysine without the EAA premix. Weight gain and feed efficiency (FE) data indicated between 1.2 and 1.6% dietary lysine was adequate. However, serum lysine concentrations indicated 1.6-2.0% lysine was required. Fish fed 1.2 or 2.4% lysine, without the EAA premix, exhibited reduced weight gain and feed efficiency. Results indicated that red drum were able to utilize crystalline amino acids when incorporated into diets containing intact protein and, when lysine was adequate, the proteins were deficient in at least one other essential amino acid. It is recommended that a dietary lysine level of 5.7% of the dietary protein be used in formulating red drum diets.  相似文献

Factors controlling the movement of fish larvae from coastal spawning environments to estuarine nursery areas are important to fish recruitment. In this paper, the role of physical processes in larval transport to estuarine nursery areas in the Aransas Pass region, Texas, is examined using a circulation model coupled with a fixed‐depth particle transport model. Two phases of transport are examined: transport on the shelf to the tidal inlet and transport through the inlet to estuarine nursery areas. Observed pulsing in the supply of red drum (Sciaenops ocellatus) larvae to the tidal inlet is significantly correlated with modeled particle supply. This pulsing is not correlated with a specific physical process, but results from the interaction of several factors affecting water movement, including low‐frequency variations in water level and wind forcing. Simulations suggest that the primary spawning region for red drum larvae that utilize nursery habitat in the Aransas Pass region is located north of the inlet. Patterns in the trajectories of particles that successfully enter the inlet reveal that they move alongshelf in the nearshore region and then move into the inlet, rather than moving directly across the shelf to the inlet. The approach path of particles outside the inlet determines the spatial transport patterns for inlets with branched channels and multiple bays. This study demonstrates that physical processes play an important role in determining larval supply to a tidal inlet.  相似文献

Natural products chemistry is the discipline that lies at the heart of modern pharmacognosy. The field encompasses qualitative and quantitative analytical tools that range from spectroscopy and spectrometry to chromatography. Among other things, modern research on crude botanicals is engaged in the discovery of the phytochemical constituents necessary for therapeutic efficacy, including the synergistic effects of components of complex mixtures in the botanical matrix. In the phytomedicine field, these botanicals and their contained mixtures are considered the active pharmaceutical ingredient (API), and pharmacognosists are increasingly called upon to supplement their molecular discovery work by assisting in the development and utilization of analytical tools for assessing the quality and safety of these products. Unlike single-chemical entity APIs, botanical raw materials and their derived products are highly variable because their chemistry and morphology depend on the genotypic and phenotypic variation, geographical origin and weather exposure, harvesting practices, and processing conditions of the source material. Unless controlled, this inherent variability in the raw material stream can result in inconsistent finished products that are under-potent, over-potent, and/or contaminated. Over the decades, natural product chemists have routinely developed quantitative analytical methods for phytochemicals of interest. Quantitative methods for the determination of product quality bear the weight of regulatory scrutiny. These methods must be accurate, precise, and reproducible. Accordingly, this review discusses the principles of accuracy (relationship between experimental and true value), precision (distribution of data values), and reliability in the quantitation of phytochemicals in natural products.  相似文献
