This report describes a case of septic peritonitis and gall bladder rupture in German shepherd dog that developed 7 d after surgical treatment for gastric dilation-volvulus. Histological examination confirmed gall bladder necrosis, secondary to an acute ischemic event. Postoperative acute necrotizing cholecystitis has not been a previously reported complication in dogs.  相似文献

Plant cell cultures in which the appropriate P450 cDNA is introduced are expected to metabolise certain pesticides in large quantities. Two species of human P450 (CYP1A1 and CYP1A2) were introduced into tobacco cells (Nicotiana tabacum L) by Agrobacterium-mediated transformation. The transgenic plant cell cultures were selected by combination of kanamycin-resistance, 7-ethoxycoumarin O-de-ethylase activity, PCR and Western blot analysis. For metabolism studies, 14C-labelled atrazine was used as a model substance. The metabolites de-ethylatrazine and de-isopropylatrazine were found in the control culture as well as in the transgenic culture, whereas the non-phytotoxic metabolite de-ethyl-de-isopropylatrazine was found only in the transgenic cell cultures. The results showed that both foreign enzymes CYP1A1 and CYP1A2 catalyse N-dealkylation of atrazine. However, CYP1A2 exhibited a higher conversion rate than CYP1A1. In a time-course study the enzyme CYP1A2 catalysed predominantly N-de-ethylation followed by de-isopropylation. The extent of metabolism was considerably higher than in non-transformed cell cultures. The transgenic cell cultures can therefore be suitable tools for the production of large quantities of primary oxidised pesticide metabolites.  相似文献

OBJECTIVE: To evaluate the effect of dietary fat and energy density on body weight gain, body composition, and total energy expenditure (TEE) in neutered and sexually intact cats. ANIMALS: 12 male and 12 female cats PROCEDURE: Male cats were castrated (castrated male [CM]) or underwent no surgical procedure (sexually intact male [IM]). Female cats underwent ovariectomy (spayed female [SF]) or laparotomy and ligation of both uterine tubes without ovary removal (sexually intact female [IF]). Cats were fed either the low-fat (LF) or high-fat (HF) diet for 26 weeks, with the final allocation consisting of 8 groups: IF-LF IF-HE SF-LF, SF-HF IM-LF, IM-HF, CM-LF, and CM-HF. Mean food intake for each group was recorded daily, and body weight was monitored weekly throughout the study. Body composition and TEE were measured before surgery in week 0 and at the end of the study (week 26) by isotope dilution (double-labelled water). RESULTS: N eutered cats gained significantly more body fat and body weight (53.80+/-5.79%) than sexually intact cats (27.11+/-5.79%) during the study. Body weight gain of neutered cats fed the HF diet was greater than those fed the LF diet. Following correction for body composition, TEE was similar in all groups and no pattern towards increased food intake was evident. CONCLUSIONS AND CLINICAL RELEVANCE: Weight gain in neutered cats was decreased by feeding an LF, low energy-dense diet. To prevent weight gain in cats after neutering, a suitable LF diet should be fed in carefully controlled meals rather than ad libitum.  相似文献

OBJECTIVE: To determine whether adenosine pretreatment attenuates free radical production and muscle damage in ischemic and reperfused canine skeletal muscle. ANIMALS: 9 healthy mixed-breed dogs. PROCEDURE: Dogs were anesthetized, and both gracilis muscles were isolated, leaving only the major vascular pedicle intact. Saline (0.9% NaCl) solution was injected into the artery supplying the control flap, whereas adenosine (10 mg) was injected into the contralateral artery. Ischemia was induced in both flaps for 4 hours. alpha-Phenyl-N-tert-butylnitrone was administered IV to each dog 1 hour prior to reperfusion. Following 15 minutes of reperfusion, effluent blood samples from each muscle flap were obtained and processed for spin-trapping electron paramagnetic resonance (EPR) spectroscopy. Muscle biopsy specimens were obtained for histologic evaluation, and dogs were euthanatized. RESULTS: EPR spectra of strong intensity were obtained from analysis of 5 of 9 paired samples. Signals identified were characteristic of oxygen- and carbon-centered free radical adducts. Signal intensity of spectra from adenosine-treated flaps was significantly less than that of control flaps; mean signal attenuation was 36% in the adenosine-treated group. Histologic evaluation of muscle flaps did not reveal significant differences between groups. CONCLUSIONS AND CLINICAL RELEVANCE: Treatment of canine muscle flaps with adenosine prior to a period of ischemia reduced but did not completely attenuate free radical production after reperfusion. However, adenosine pretreatment did not affect histologic abnormalities.  相似文献

A comparison is made between the properties of permafrost and those of fragipans. The permafrost characteristics are summarized from published data on active permafrost and from personal observations on fossil permafrost. The study of the fragipans mainly concerns the silty sediments of Weichselian age which occur in Belgium and northern France.The distribution and the physical, macromorphological and micromorphological properties of fragipans can all satisfactorely be explained by the growth of segregated ice during permafrost aggradation. Particularly striking is the relationship of both permafrost and fragipans, to drainage conditions: the quantity of segregated ice is highest in moist or poorly drained soil and it is at such places that the best developed fragipans are observed.Certain arguments that can be stated against the hypothesis of fragipan genesis by permafrost are discussed: (1) the development of a platy structure by freeze-thaw or by wet-dry cycles, (2) the discontinuous distribution of the fragipan, (3) the development of the fragipan by pedogenetic weathering and migration processes. It is concluded that only the permafrost hypothesis can explain the complete set of properties of the fragipan.  相似文献

Several dysgenic traits may occur within the Drosophila melanogaster species as a result of crosses between different strains. Crossing two mutually interacting categories, named inducer and reactive, may lead, among other abnormalities, to a specific kind of female sterility that has proved useful for investigating the genetic factors involved in the interaction. The reactive state appears to result from a cytoplasmic state ultimately controlled by a chromosomal polygenic system. The inducer character is determined by a chromosomal factor that exhibits all characteristics of a transposable element. Overall, the data contribute to clarification of mutator activities in D. melanogaster and open new opportunities to investigate unusual genetic mechanisms.  相似文献

Thehe broad sprectrum protective effect of the non-protein amino acid -aminobutyric acid (BABA) against numerous plant diseases has been well-documented in the literature. Here, we present an overview of BABA-induced protection in various pathosystems. Contriidictory reports concerning the mechanism of action underlying this type of protection incited us to take advantage of Arabidopsis/pathogen interactions as model systems to investigate the action of BABA at the genetic and molecular level. We present evidence that the protective effect of BABA is due to a potentiation of natural defense mechanisms against biotic and abiotic stresses. In order to dissect the pathways involved in potentiation by BABA describe the use of a mutational approach based on BABA-induced female sterility in Arabidopsis.  相似文献

The effects on rumen kinetics after feed and water had been deprived for 72 hr were studied using four fistulated Bos indicus steers. The animals were assigned in a 2 × 4 crossover design with two treatments: feed and water ad libitum (control) and no feed and water for 72 hr (deprived) with four steers per treatment over two time periods. Feed and water deprivation caused decreases in the numbers of cellulolytic bacteria (1.4 vs. 0.4 cfu × 106/ml; p = .001), live (23.7 vs. 0.8 × 109/ml; p = .001), dead (12.7 vs. 0.5 × 109/ml; p = .001) and total bacterial counts (36.4 vs. 1.4 × 109/ml; p = .001) at day 0, compared with the control treatment. However, the deprived group had greater numbers of cellulolytic bacteria (2.7 vs. 50.1 cfu × 106/ml; p = .001), live (18.3 vs. 42.2 × 109/ml; p = .001), dead (6. 5 vs. 19.1 × 109/ml; p = .001) and total bacterial counts (24.8 vs. 61.3 × 109/ml; p = .001) from rumen fluid on day 4, compared with the control treatment. The numbers of protozoa in rumen fluid from the deprived group were less than (551.2 vs. 2.4 × 103/ml; p = .001) the control group on day 0. However, the deprived treatment had fewer protozoa in rumen fluid than the control treatment on day 4 (p = .001) and day 9 (p = .001). Volatile fatty acids and in vitro gas production as functional measurements of rumen fluid followed the same trend as the bacterial and protozoa populations. These results indicate that feed and water deprivation would have a negative but transient effect on the rumen kinetics of Bos indicus steers.  相似文献
