Satellite cells activity contributes to postnatal muscle growth. Herein, we have studied the respective influence of insulin and triiodothyronine (T3) on the proliferation and differentiation of primary bovine satellite cells isolated from Semitendinosus muscle of Montbéliard steers. Under basal conditions, satellite cells proliferated until the fifth day of culture, began to fuse into myotubes and expressed differentiation markers such as connectin, myogenin, and myosin heavy chain (MHC) isoforms. Insulin behaved as an effective mitogen. Moreover, it promoted extensive myotube formation and enhanced differentiation as shown by an increase in the accumulation of differentiation markers. Maximal differentiation occurred with insulin physiological range concentrations. A delay in the stimulation of differentiation was registered with a high dose that promoted maximal proliferation. Conversely, T3 decreased cell proliferation in a dose-dependent manner. In addition, fusion and biochemical differentiation (accumulation of connectin, MyoD1, myogenin, and myosin heavy chain isoforms) were also enhanced. Bovine satellite cells seemed to respond differentially to insulin and T3 for proliferation. Interestingly, both hormones displayed a myogenic influence. Our observations suggest that both hormones could influence bovine satellite cells in vivo and contribute to the regulation of postnatal muscle growth.  相似文献

Rice Root Genetic Architecture: Meta-analysis from a Drought QTL Database   总被引:3,自引:0,他引:3  
During the last 10 years, a large number of quantitative trait loci (QTLs) controlling rice root morphological parameters have been detected in several mapping populations by teams interested in improving drought resistance in rice. Compiling these data could be extremely helpful in identifying candidate genes by positioning consensus QTLs with more precision through meta-QTL analysis. We extracted information from 24 published papers on QTLs controlling 29 root parameters including root number, maximum root length, root thickness, root/shoot ratio, and root penetration index. A web-accessible database of 675 root QTLs detected in 12 populations was constructed. This database includes also all QTLs for drought resistance traits in rice published between 1995 and 2007. The physical position on the pseudo-chromosomes of the markers flanking each QTL was determined. An overview of the number of root QTLs in 5-Mb segments covering the whole genome revealed the existence of “hot spots,” The 32 trait × chromosome combinations comprising six or more QTLs were subjected to a meta-QTL analysis using the software package MetaQTL. The method enabled us both to determine the likely number of true QTLs in these areas using an Akaike information criterion and to estimate their position. The meta-QTL confidence intervals were notably reduced and, for the smallest ones, encompassed only a few genes.  相似文献

Horizontal ocean transport can influence the dynamics of higher‐trophic‐level species in coastal ecosystems by altering either physical oceanographic conditions or the advection of food resources into coastal areas. In this study, we investigated whether variability in two North Pacific Current (NPC) indices was associated with changes in productivity of North American Pacific salmon stocks. Specifically, we used Bayesian hierarchical models to estimate the effects of the north‐south location of the NPC bifurcation (BI) and the NPC strength, indexed by the North Pacific Gyre Oscillation (NPGO), on the productivity of 163 pink, chum, and sockeye salmon stocks. We found that for salmon stocks located in Washington (WA) and British Columbia (BC), both the BI and NPGO had significant positive effects on productivity, indicating that a northward‐shifted bifurcation and a stronger NPC are associated with increased salmon productivity. For the WA and BC regions, the estimated NPGO effect was over two times larger than the BI effect for pink and chum salmon, whereas for sockeye salmon the BI effect was 2.4 times higher than the NPGO. In contrast to WA and BC stocks, we found weak effects of both horizontal ocean transport processes on the productivity of salmon stocks in Alaska. Our results indicated that horizontal transport pathways might strongly influence population dynamics of Pacific salmon in the southern part of their North American ranges, but not the northern part, suggesting that different environmental pathways may underlie changes in salmon productivity in northern and southern areas for the species under consideration.  相似文献

The stock recovery rate, that is the ratio of the exploitable wood stock at the end of a felling cycle over the exploitable wood stock at the beginning of this cycle, is a key parameter used in the management plans of the natural forests in central Africa. Estimating this rate requires a model of forest dynamics. Forest managers usually use a formula that is based on a simple model that assumes constant vital rates. A generalization of this formula is based on matrix models of population dynamics. The stock recovery rate at the end of the k th felling cycle can be simply computed using matrix models. The asymptotic stock recovery rate (that is the limit as k tends to infinity) is the asymptotic growth rate (that is the dominant eigenvalue) of a transition matrix that includes harvest. The estimate of the stock recovery rate can be completed by its confidence interval using bootstrap methods. When applied to sapelli (Entandrophragma cylindricum, Meliaceae), a major timber species in central Africa, it turns out that a few thousands observations are required to estimate the stock recovery rate with an accuracy of at least 10%. The number of observations available on an experimental site in the Central African Republic does not permit to do better than an accuracy of about 45% at level 95%. This does not permit to conclude whether the asymptotic stock recovery rate is greater or less than one. As a conclusion, in management plans in central Africa, stock recovery rates should be given together with an indication of the variability of their estimate (standard error or confidence limits).  相似文献

From June to December, we determined the effects of variations in biochemical composition on delta(13)C of tree rings of 2-year-old oaks (Quercus petraea (Matt.) Liebl.) growing under semi-natural conditions, and the dependence of these effects of water stress during the growth season. Percent abundance, carbon concentration and delta(13)C were measured in holocellulose, lignin, extractive-free wood, starch and a water-soluble fraction. Relative concentrations of lignin and holocellulose in the extractive-free wood varied little during the season or in response to water stress, indicating that these compounds could not quantitatively explain the variations in whole-wood delta(13)C. Among all sampled tree rings, the relative concentration of each structural compound (holocellulose and lignin) accounted for less than 5% of the delta(13)C variability in whole wood. Variations in holocellulose and extractive-free wood delta(13)C between tree rings were almost identical (r > 0.95), whereas variations in lignin delta(13)C were less well correlated to these compounds. Whole-wood delta(13)C had a slightly altered isotopic signal compared with that of the structural compounds because of the presence of the extractive component. These results showed that variations in lignin delta(13)C and lignin concentration have little influence on extractive-free wood delta(13)C and whole-wood delta(13)C. Rather, holocellulose influences delta(13)C the most. Thus, we confirmed that, for climatic reconstruction from tree rings, removal of extractives by soxhlet is generally sufficient and sometimes unnecessary. Our findings also indicate that, in the case of rapid and severe water stress, the structural component did not accurately record the associated increase in delta(13)C because of dilution with previously formed organic matter and cessation of trunk growth. The effect of drought on carbon isotope ratios was more pronounced in the extractive compounds, making them good water stress indicators but only on a scale of days to months.  相似文献

Morphological and physiological seed characteristics of the hyacinth bean (Lablab purpureus) were investigated in a set of 18 different germplasm accessions, from wild over semi-domesticated forms to landraces and current cultivars. The objective of this study was to improve the understanding of the domestication of this tropical legume crop. Wild accessions were easily distinguished by morphological characters, such as small, greyish-brown, mottled seeds. Cultivated or semi-domesticated forms showed a much wider variation in size, colour and shape. Seed mass of cultivated accessions reached up to almost 10 times that of wild accessions. Most wild accessions showed a greater spread of germination and larger proportions of hard seeds, up to about 70%, than most cultivated and so-called semi-domesticated accessions. Cluster analysis applied separately to morphological and germination data and a subsequent discriminant analysis did not help integrating the morphological variability observed. The presumed occurrence of semi-domesticated accessions within the germplasm tested as well as the crop's origin from Africa alone or both Africa and Asia are debated on the basis of seed germination and hard seed coat dormancy.  相似文献

An incubation experiment with organic soil amendments was carried out with the aim to determine whether formation and use of microbial tissue (biomass and residues) could be monitored by measuring glucosamine and muramic acid. Living fungal tissue was additionally determined by the cell-membrane component ergosterol. The organic amendments were fibrous maize cellulose and sugarcane sucrose adjusted to the same C/N ratio of 15. In a subsequent step, spherical cellulose was added without N to determine whether the microbial residues formed initially were preferentially decomposed. In the non-amended control treatment, ergosterol remained constant at 0.44 μg g−1 soil throughout the 67-day incubation. It increased to a highest value of 1.9 μg g−1 soil at day 5 in the sucrose treatment and to 5.0 μg g−1 soil at day 33 in the fibrous cellulose treatment. Then, the ergosterol content declined again. The addition of spherical cellulose had no further significant effects on the ergosterol content in these two treatments. The non-amended control treatment contained 48 μg muramic acid and 650 μg glucosamine g−1 soil at day 5. During incubation, these contents decreased by 17% and 19%, respectively. A 33% increase in muramic acid and an 8% increase in glucosamine were observed after adding sucrose. Consequently, the ratio of fungal C to bacterial C based on bacterial muramic acid and fungal glucosamine was lowered in comparison with the other two treatments. No effect on the two amino sugars was observed after adding cellulose initially or subsequently during the second incubation period. This indicates that the differences in quality between sucrose and cellulose had a strong impact on the formation of microbial residues. However, the amino sugars did not indicate a preferential decomposition of microbial residues as N sources.  相似文献

Two new density estimators for k-tree distance sampling are proposed and their performance is assessed in simulated distance sampling from 22 stem maps representing a wide range of natural to semi-natural forest tree stands with random to irregular (clustered) spatial distribution of trees. The new estimators are model-based. The first (Orbit) computes density as the inverse of the average of the areas associated with each of the k-trees nearest to a sample location. The area of the k-th tree is obtained as a prediction from a linear regression model while the area of the first is obtained via a Poisson probability integral. The second (GamPoi) is based on the expected distribution of distance to the k nearest tree in a forest where the local distribution of trees is random but the stem density varies from sample location to sample location as a gamma distribution. In a comprehensive assessment with 17 promising reference estimators, a subset composed of Morisita’s, Persson’s, Byth’s, Kleinn’s, Orbit, and GamPoi was significantly better, in terms of relative root mean square error (RRMSE), than average. GamPoi emerged as the better estimator for sample sizes larger than or equal to 30. For smaller sample sizes, both Kleinn’s and Morisita’s appear attractive.  相似文献

Endoscopic biopsy specimens were obtained from the cardiac, fundic, and pyloric stomach regions of 28 semicaptive cheetahs (Acinonyx jubatus) to look for signs of helicobacteriosis and gastritis. Impression smear cytology and urease tests were positive in 23 and 26 of the cheetahs, respectively. Histopathology showed mild or no lesions in 27 cheetahs; only one cheetah had moderate gastritis. Gastritis was uncommon in these semicaptive cheetahs, and stress could play a major role in the development of helicobacteriosis-associated gastritis in the captive cheetah.  相似文献

Herpes simplex virus-1 (HSV-1) encephalitis (HSE) is the most common form of sporadic viral encephalitis in western countries. Its pathogenesis remains unclear, as it affects otherwise healthy patients and only a small minority of HSV-1-infected individuals. Here, we elucidate a genetic etiology for HSE in two children with autosomal recessive deficiency in the intracellular protein UNC-93B, resulting in impaired cellular interferon-alpha/beta and -lambda antiviral responses. HSE can result from a single-gene immunodeficiency that does not compromise immunity to most pathogens, unlike most known primary immunodeficiencies. Other severe infectious diseases may also reflect monogenic disorders of immunity.  相似文献
