Douglas fir (Pseudotsuga menziesii (Mirb.) Franco) needle-litter was given treatments of varying biocidal severity including air-drying, oven-drying, fumigation with methyl bromide plus chloropicrin. autoclaving, and a control. The litter was inoculated with a small amount of fresh litter homogenate and monitored for CO2 evolution rate and extractable enzymatic activity during a 77-day incubation. Enzymatic activities assayed included cellulase. xylanase, mannase, amylase. invertase, β-glucosidase, protease, polyphenoloxidase and peroxidase. The concentrations of extractable ninhydrin-reactive compounds, glucose, and reducing substances were also determined.
Air-drying produced a small, initial stimulation in respiration but had no effect on any enzymatic activity throughout the 77-day incubation except for an initial decrease in polyphenoloxidase and peroxidase activities. Oven-drying produced a substantial loss of all enzymatic activities initially but all carbohydrase and protease activities recovered to greater than control levels by 7 days; after a 1-day lag, oven-drying produced a pronounced stimulation in the rate of CO2 evolution. Autoclaving produced a 2-day lag in a less pronounced (compared to oven-drying) respiratory stimulation: initially all enzymatic activities were destroyed and only protease and β-glucosidase activities recovered to control levels. Fumigation produced a respiratory stimulation after a 9-day lag (probably due to residual chloropicrin) which coincided with a pronounced increase in cellulase. xylanase, mannase and amylase activities: initially, no activities except polyphenoloxidase and peroxidase were adversely affected by fumigation.
During the entire incubation, the activities of cellulase. xylanase. mannase. amylase and invertase were highly correlated with each other regardless of treatment. After 77 days, the rates of CO2 evolution of all samples were correlated with the level of amylase and cellulase activity (r = 0.86 and 0.83, respectively). This observation points to the value of enzymatic assays for following the processes which release substrate for microbial assimilation from plant litter. The close association of polysaccharidase and protease activities with the flush of CO2 evolution following biocidal treatment, particularly oven-drying, indicates that the decomposition of carbohydrates and proteins released from microbial biomass or plant litter is a mechanism for this often-observed respiratory stimulation. 相似文献
Interest in native species is growing across the tropics as reforestation of degraded lands becomes more widespread. In this study four tree species native to Panama - Cedrela odorata, Pachira quinata, Samanea saman, and Tabebuia rosea - were grown on rural farms at two dry tropical sites in Panama for up to five years. Survivorship and growth data at these “on-farm” trials are compared to data recorded from nearby experimental or “species selection” trial sites and evaluated in terms of soil fertility and management. Participant farmers were also asked about their interest in planting trees in general as well as their interest in 61 species grown in the species selection trial.Although, on-farm survivorship was variable and generally lower than that found on the species selection trial, one species (S. saman) experienced high and consistent survivorship. High survivorship combined with growth data from farms at both sites for this species suggests it would be a good candidate for extension projects working with rural farmers. Survivorship of other species appears more sensitive to farmer management and/or local site conditions. Generally lower growth on the Los Santos farms as compared to the species-selection trial is attributed to the lower soil fertility (plant available P) at the on-farm sites compared to the species selection trial. In contrast, only one species - P. quinata - had a growth variable found to be significantly lower between the on-farm and species selection trial sites in Rio Hato. C. odorata, P. quinata T. rosea can all be used in on-farm conditions with consideration to specific site and management conditions.By 2009, approximately 80% of the farmers planting trees still wished to participate in tree planting activities. All of the farmers no longer wishing to continue with the project expressed slow growth rates of trees as a principal reason. All but one of these farmers had growth rates for his/her trees markedly below those of the species selection trial nearby. Some farmers wishing to continue had very high mortality rates (>70% for all species), suggesting non-tangible benefits for participating in project activities. Other species that were not tested on-farm but grew well in the species-selection trials and were of interest to local farmers are discussed.As long as specific site and management conditions are carefully considered, data from species selection trials can be useful in informing tree planting projects with rural farmers; however, care should be taken to manage expectations. 相似文献
Concern about acidification in upland areas has brought about the need to model the stream hydrochemical response to deposition and land-use changes and calculate critical loads. Application of dynamic models such as MAGIC are preferable to steady-state methods, since they are able to produce an estimate of the time scale required to meet some water chemistry target given a reduction in acid deposition. These models typically consider annual changes in stream chemistry at one point. However, in order to protect biota from 'acid episodes', quantification of temporal variability needs to encompass event responses; in addition spatial variability across the catchment also needs to be considered. In this paper, modelling of both spatial and temporal variability is combined in a new framework which enables quantification of catchment hydrochemical variability in time and space. Both low and high flow hydro-chemical variability are quantified in terms of statistical distributions of ANC (Acid Neutralisation Capacity). These are then input as stochastic variables to an EMMA (End-Member Mixing Analysis) model which accounts for temporal variability and ANC is hence predicted as a function of time and space across the whole catchment using Monte-Carlo simulation. The method is linked to MAGIC to predict future scenarios and may be used by iteration to calculate critical loads. The Livestock Gross Margin Insurance for Dairy Cattle is a federally reinsured insurance program that enables US dairy producers to establish minimum levels of milk income net of feed cost. Given the structure of this program there are an infinite number of possible contract designs based on the choice of deductible level and proportion of production insured. Adding to this complexity, producers vary in their risk preferences, which affect the incentive to insure their margin. It is unclear as to how producers may adopt this program for revenue risk management. This paper investigates the interplay between producer risk preferences, contract design and the subsidization of premium in determining program coverage. We undertook this analysis within an expected utility framework. Optimal contracts under different rates of constant relative rate of risk aversion and subsidies were analyzed using a nonlinear optimization model. We found that total optimal coverage increased significantly with the level of risk of aversion at lower deductibles but as deductible level increased, the level of risk aversion had a lesser impact on total optimal coverages. As expected, at the same deductible and risk aversion levels, inclusion of a premium subsidy increased the total optimal coverage. 相似文献
Coevolution of mammals and their gut microbiota has profoundly affected their radiation into myriad habitats. We used shotgun sequencing of microbial community DNA and targeted sequencing of bacterial 16S ribosomal RNA genes to gain an understanding of how microbial communities adapt to extremes of diet. We sampled fecal DNA from 33 mammalian species and 18 humans who kept detailed diet records, and we found that the adaptation of the microbiota to diet is similar across different mammalian lineages. Functional repertoires of microbiome genes, such as those encoding carbohydrate-active enzymes and proteases, can be predicted from bacterial species assemblages. These results illustrate the value of characterizing vertebrate gut microbiomes to understand host evolutionary histories at a supraorganismal level. 相似文献
Vertebrate limb outgrowth is driven by a positive feedback loop involving Sonic Hedgehog (Shh), Gremlin, and Fgf4. By overexpressing individual components of the loop at a time after these genes are normally down-regulated in chicken embryos, we found that Shh no longer maintains Gremlin in the posterior limb. Shh-expressing cells and their descendants cannot express Gremlin. The proliferation of these descendants forms a barrier separating the Shh signal from Gremlin-expressing cells, which breaks down the Shh-Fgf4 loop and thereby affects limb size and provides a mechanism explaining regulative properties of the limb bud. 相似文献
Undoped, high-quality diamond is, under almost all circumstances, one of the best insulators known. However, diamond covered with chemically bound hydrogen shows a pronounced conductivity when exposed to air. This conductivity arises from positive-charge carriers (holes) and is confined to a narrow near-surface region. Although several explanations have been proposed, none has received wide acceptance, and the mechanism remains controversial. Here, we report the interactions of hydrogen-terminated, macroscopic diamonds and diamond powders with aqueous solutions of controlled pH and oxygen concentration. We show that electrons transfer between the diamond and an electrochemical reduction/oxidation couple involving oxygen. This charge transfer is responsible for the surface conductivity and also influences contact angles and zeta potentials. The effect is not confined to diamond and may play a previously unrecognized role in other disparate systems. 相似文献
Meiosis is a specialized cell division in which two chromosome segregation phases follow a single DNA replication phase. The budding yeast Polo-like kinase Cdc5 was found to be instrumental in establishing the meiosis I chromosome segregation program. Cdc5 was required to phosphorylate and remove meiotic cohesin from chromosomes. Furthermore, in the absence of CDC5 kinetochores were bioriented during meiosis I, and Mam1, a protein essential for coorientation, failed to associate with kinetochores. Thus, sister-kinetochore coorientation and chromosome segregation during meiosis I are coupled through their dependence on CDC5. 相似文献
