Toxigenic strains of Pasteurella multocida were readily differentiated from non-toxigenic strains by an agarose overlay method using bovine turbinate cells or bovine lung cells. Cells which were young and densely confluent were best suited to this assay. The incubation period required to distinguish toxigenic strains was dependent on the confluence of the monolayers, which was affected by the seeding rate, cell passage level and growth time prior to overlay. The agarose overlay method correctly identified 11 of 11 reference strains of Pasteurella multocida, and visible cytotoxic changes were present in the monolayers after 48 to 65 h. Outbreaks of the enzootic form of atrophic rhinitis in 2 New South Wales piggeries were associated with the isolation of toxigenic type D strains of P. multocida.  相似文献

Urine (U) and serum (S) were obtained every 2 hours during a 12- or 24-hour period from eight healthy 96-hour-old pony or horse foals. Dams' milk samples were obtained concurrently. Urine volume was measured during this 12- or 24-hour period. The mean amount of urine produced was 148 +/- 20 ml/kg/day. Baseline urinalyses were evaluated on all foals at two days of age, before any manipulation. Urine generally was dilute (less than 1.008) but the specific gravity was as high as 1.027 in one normal foal. Continuous (12 or 24 hour) urinary catheterization resulted in bacteriuria but not white blood cells in the urine. Prolonged catheterization did not cause foals to become febrile or exhibit clinical signs of cystitis or other illness. Urinary electrolyte excretion, urinary electrolyte clearances, and fractional electrolyte excretions (FE) were measured. When compared with normal values reported in adult horses, excretion, clearance, and FE were similar for sodium (Na) but higher for potassium (K), phosphorus (P), and calcium (Ca). There were no significant differences between data collected during different time periods, and it was concluded that the use of single sample urine/serum estimates of fractional excretion in the neonatal foal was an appropriate indicator of the renal handling of electrolytes, and when viewed in conjunction with urinalysis and other serum parameters, a valuable aid to evaluating renal function.  相似文献

OBJECTIVE: To determine the prevalence of Salmonella spp in the cecal-colon contents of cull (market) dairy cows at slaughter because of potential public health ramifications. DESIGN: Survey study. SAMPLE POPULATION: Cecal-colon contents collected from 5,087 cull (market) dairy cows at slaughter at 5 slaughter establishments across the United States. PROCEDURE: During 2 periods of the year, winter (January and February) and summer (July through September), 5 cull (market) cow slaughter establishments in the United States--west (WE), southeast (SEE), central (CE), north central (NCE), and south central (SCE)--establishments were visited, and cecal-colon contents of cull dairy cows were obtained at the time of slaughter. Samples were examined by microbiologic culture at a single laboratory for Salmonella spp. RESULTS: Salmonella spp were detected in 23.1% of cecal-colon content samples from cull dairy cows across the 5 slaughter establishments. The highest site prevalence (54.5%) was detected at the WE during the summer period, whereas the lowest was found at the CE during the summer (4.3%) and at the NCE during the winter (4.5%). Considerable variation in the daily prevalence of Salmonella spp was found, particularly at the WE and the SCE. Salmonella spp were isolated from 93% of cecal-colon contents collected on a summer day at the WE. CONCLUSIONS AND CLINICAL RELEVANCE: Results strongly suggest that there is a high prevalence of Salmonella spp in cull dairy cows at slaughter, which could burden Hazard Analysis Critical Control Point programs implemented in slaughter establishments. Procedures to reduce Salmonella load at the dairy farm and during transport to slaughter could reduce the risk of spread during the slaughter process.  相似文献

Copper is required in trace amounts for many body functions. The prominent effects of Cu deficiency or Cu toxicosis differs greatly between animal species. Along with iron, Cu is necessary for the transfer of O2 via a cascade of enzymes so that energy may be available for vital body functions without overheating of the tissues through rapid oxidation. As a part of lysyl oxidase, Cu has an obligate function in the maturation of all connective tissue (including elastic tissue and bone) maintaining the form and integrity of all body organs. As a constituent of tyrosinase, Cu is involved in the formation of melanin, thus preventing albinism. Copper also is involved in the myelination of nerve fibers and the production of neutrophils, enkephalins, lipoproteins, and cholesterol. Copper must be properly sequestered to prevent toxicosis. Copper is stored primarily as metallothioneins and as superoxide dismutase and is transported primarily as ceruloplasmin or as low molecular weight proteins, peptides, and amino acids.  相似文献

BACKGROUND

Stable flies [Stomoxys calcitrans (L.)] are economically important pests of cattle and other livestock. As an alternative to conventional insecticides, we tested a push–pull management strategy using a coconut oil fatty acid repellent formulation and an attractant-added stable fly trap.

RESULTS

In our field trials we found that weekly applications of a push–pull strategy can reduce stable fly populations on cattle as well as a standard insecticide (permethrin). We also found that the efficacy periods of the push–pull and permethrin treatments following on-animal application were equivalent. Traps with an attractant lure used as the pull component of the push–pull strategy captured sufficient numbers of stable flies to reduce on-animal numbers by an estimated 17–21%.

CONCLUSIONS

This is the first proof-of-concept field trial demonstrating the effectiveness of a push–pull strategy using a coconut oil fatty acid-based repellent formulation and traps with an attractant lure to manage stable flies on pasture cattle. Also notable is that the push–pull strategy had an efficacy period equivalent to that of a standard, conventional insecticide under field conditions. © 2023 The Authors. Studies of the interactions between the metabolism and the function of the red cell have shown the importance of the red cell's metabolism in contributing to the maintenance of adequate oxygen delivery. Some of the phosphorylated intermediates of glycolysis, especially DPG, are now known to reduce the affinity of hemoglobin for oxygen. Current evidence indicates that this phenomenon is due to the effects of the binding of DPG to the , beta-chains of deoxyhemoglobin. It appears that increases in red cell concentrations of DPG commonly occur during hypoxia, and that these increases (as well as normal physiological variation) significantly enhance oxygen transport. Artificial manipulation of erythrocyte metabolism may soon prove to be of great clinical usefulness in the treatment of a great variety of disorders which limit oxygen transport.  相似文献

Drosophila melanogaster acquired the potato blackleg bacterium,Erwinia carotovora var.atroseptica from laboratory cultures and infected potato plants and readily transmitted it to healthy wounded plants under greenhouse conditions.D. melanogaster also carried the bacterium both internally and externally. Transmission to the nearest plants occurred most often. The possibility of at least short term survival of the bacterium inside the body of the insect is postulated.  相似文献

The objective of this research was to evaluate a variety of stability and maturity indices for yard trimmings compost produced in the Puget Sound region of western Washington State. Compost samples were collected periodically during a 133-d composting cycle at a commercial composting facility, showing that indices of compost respiration rate were sensitive indicators of compost quality. All respiration rate indices identified a period of high respiration rates during active composting (first 27 d), and a period of relatively stable respiration rates during the latter part of curing (70 to 133 d). Chemical tests of compost solids showed less promise as maturity indicators, but provided valuable information on final compost quality. Mature yard trimmings compost had a C:N of 12, an NH4-N to NO3-N ratio of less than 4, a cation exchange capacity (CEC) of 400 cmol per kg of compost-C, and a pH between 6.5 and Seed germination tests and sensory tests (color and odor) were of limited value in assessing compost maturity. Fully-cured compost produced with forced aeration had a Solvita CO2 test value of 6 to 7 and a respiration rate via the alkaline trap method of 2 mg CO2-C g compost-C?1 d?1. It reheated less than 2°C in an insulated Dewar flask in a 7 d incubation. Further evaluation and calibration of respiration test protocols for compost quality assurance testing programs are recommended.  相似文献

This study assessed the involvement of lipopolysaccharide (LPS) in the non-typhoidal Salmonella encephalopathy (NTSE) caused by a unique isolate of Salmonella enterica serovar Saint-paul (SstpNPG). NTSE was prevented by genetic (deletion of murE) or pharmacologic (polymyxin) disruption of LPS on SstpNPG although the disruption of LPS did not deter brain penetration of the strain. This is the first study to demonstrate that LPS is involved in the manifestations of NTSE.  相似文献
