ABSTRACT: Helicobacter (H.) suis has been associated with chronic gastritis and ulcers of the pars oesophagea in pigs, and with gastritis, peptic ulcer disease and gastric mucosa-associated lymphoid tissue lymphoma in humans. In order to obtain better insight into the genes involved in pathogenicity and in the specific adaptation to the gastric environment of H. suis, a genome analysis was performed of two H. suis strains isolated from the gastric mucosa of swine. Homologs of the vast majority of genes shown to be important for gastric colonization of the human pathogen H. pylori were detected in the H. suis genome. H. suis encodes several putative outer membrane proteins, of which two similar to the H. pylori adhesins HpaA and HorB. H. suis harbours an almost complete comB type IV secretion system and members of the type IV secretion system 3, but lacks most of the genes present in the cag pathogenicity island of H. pylori. Homologs of genes encoding the H. pylori neutrophil-activating protein and γ-glutamyl transpeptidase were identified in H. suis. H. suis also possesses several other presumptive virulence-associated genes, including homologs for mviN, the H. pylori flavodoxin gene, and a homolog of the H. pylori vacuolating cytotoxin A gene. It was concluded that although genes coding for some important virulence factors in H. pylori, such as the cytotoxin-associated protein (CagA), are not detected in the H. suis genome, homologs of other genes associated with colonization and virulence of H. pylori and other bacteria are present.  相似文献

Pyranose oxidase (P2O) improves wheat flour dough stability and bread quality. We related its effect on dough spread behavior to that on dough and bread crumb structure. Increasing P2O addition levels gradually reduced dough flow. High P2O addition levels further increased dough strength, significantly increased dough cell wall thickness, and decreased bread loaf volume. Taken together, affecting dough spread behavior impacts dough and bread (crumb) structure, and dough structure largely determines bread crumb structure.  相似文献

Differences in hardness of wheat cultivars have been related to differences in interactions between the starch granule surface and the gluten protein matrix that are mediated by the proteins puroindoline (PIN) A and B. We examined whether or not PINs and (polar) lipids are associated at the starch granule surface, and, if so, how they interact with the starch granule surface itself. Starch was isolated from a soft wheat cultivar containing both wild‐type PINs and incubated with peptidases or lipases, or in extraction media (typically used for defatting). Protein, PIN, and lipid levels revealed that PINs and lipids are tightly associated together at the starch granule surface. Our results imply that PINs need lipids for binding to the granule surface but not vice versa.  相似文献

A simulation model for the production of the Nile tilapia (Oreochromis niloticus niloticus L.) for mixed‐ or mono‐sex culture and for poly‐culture with the African catfish (Clarias gariepinus Burchell 1822) or African snakehead (Parachanna obscura Günther 1861) is presented. The model is based on an exponential decay model used in population dynamics and follows an individual‐based approach. The model consists of a tilapia sub‐model representing population dynamics, growth and recruitment and a predator sub‐model representing the predation process and population dynamics and growth of the used predator. The model was calibrated with data on mixed‐ or mono‐sex culture of Nile tilapia and for poly‐culture with the African catfish or African snakehead obtained in Congo Brazzaville and validated with similar data from the Philippines, Thailand and the Ivory Coast. The model visualized major underlying processes in tilapia farming and aspects for further improvement of the model; growth is one of the most sensitive input parameters and should be quantitatively related to feeding level and feed quality; length at first maturity and quantification of the recruitment of Nile tilapia has a relatively large influence and recruitment should be related to the length of the females; prey–predator relations are too coarse and should be more fine‐tuned with the relation between prey size and predator size. Incorporation of these features would provide the basis of a model that can serve as a predictive and decision‐making support tool.  相似文献

In 1922, the colonial government introducedAcacia mearnsii in the tobacco-growing region of Wonosobo. Soon this species was accepted by the local people who developed an agroforestry system based on a rotation ofA. mearnsii and agricultural crops.Now, the growing of the black wattle is becoming less popular. To study the prospects for this agroforestry system 143 farmers from 5 villages were interviewed. The functions of this agroforestry system are the production of fuelwood and bark (which contains tannin for the leather industry), soil conservation and soil improvement. The following factors affectA. mearnsii growing: population density (which affects farm size and demand for domestic fuelwood), tobacco-processing (which requires fuel), topography and commercialization. If current trends continue, the contribution of the functions of this agroforestry system to welfare will not be sufficient to compete successfully with subsistence and cash crops like potatoes and tobacco. Fat, one of the three major ingredients of sugar‐snap cookies, affects dough properties, changes in dough dimensions during baking, and in the end, the properties of the baked product. We studied the effect of reducing fat levels (from 15.8 to 8.7% on a dough basis) on dough and cookie properties and related it to the SDS extractable protein (SDSEP) levels. Reducing fat levels increased dough elasticity (from 0.19 to 0.60 MPa) and dough intrinsic hardness (from 8.6 to 27.5 N·cm3/g). Because no differences in dough SDSEP levels were noticed when fat levels were reduced, the increased dough elasticity and hardness were related to a more pronounced physical gluten entanglement. Reducing fat levels in the recipe decreased the SDSEP levels of the baked cookies, indicating more protein cross‐linking during baking with lower fat levels. Our data show that the dispersed fat phase interferes with the formation of a gluten network during baking. Reducing the fat level in the recipe increased the intrinsic cookie break strength (from 39.5 to 100.3 N·cm3/g), which was related to more gluten cross‐linking.  相似文献

Norwegian wild reindeer Rangifer tarandus tarandus are divided into 23 virtually isolated populations, primarily due to the abandonment of traditional migration and movement corridors caused by the development of infrastructures. By conducting a nation-wide, interdisciplinary pre-post study on a temporal scale spanning centuries, we modelled current reindeer movements with respect to archaeological findings to quantify long-term changes in area use related to anthropogenic disturbance. The location of 3,113 pitfall traps and hunting blinds, built 600–2000 years ago and used until 350–400 years ago, testified the location of traditional movement corridors. Current movement routes were delineated using Brownian Bridge Movement Models based on 147 reindeer GPS-monitored during 10 years. Using Path Analysis we quantified direct, indirect and total effects of different infrastructures within multiple scales (1, 5, and 10 km-radius buffers) on the current probability of use of ancient movement corridors. Tourist cabins and roads had the strongest long-term direct effects at most scales: 1 tourist cabin and 1 km road within a 1 km-radius buffer would lead, respectively, to complete area abandonment, and to a 46 % decrease in the probability of use. Power lines and private cabins had significant indirect effects on area use through their effect on roads, while hiking trails and, in particular, hydroelectric dams had highly variable effects, not significant at a nation-wide scale. Finally, we provide a flexible tool to estimate the potential long-term direct and cumulative effects of different types of infrastructures at the desired spatial scale to be used for the development of future sustainable land management plans.  相似文献

In Tigray (Northern Ethiopia), soil moisture has been identified as the most limiting factor in agricultural production; on the other hand, loss of rain water through runoff as well as the induced soil loss has been determined as a critical problem in the region in the last two to three decades. To alleviate the above paradox, the government has mobilized communities and resources for the construction of physical soil and water conservation structures (stone bunds, terraces) in almost all land uses. However, yield improvement was mainly concentrated within the vicinity of the structures and runoff continued to overtop the structures, as no measures for in situ soil conservation were taken. The terwah system, consisting of traditional ploughing followed by making every 1.5–2 m contour furrows, and permanent raised beds with contour furrows at 60–70 cm interval treatments, were considered and evaluated as practices that could increase the efficiency of in situ water utilization and soil conservation. An experiment was started in Gum Selasa, which is one of the drought prone areas in Tigray, whereby runoff volume and sediment load were measured after every rain event. Permanent raised beds with contour furrows at 60–70 cm interval significantly (P < 0.05) reduced runoff volume, runoff coefficient and soil loss as compared to traditional ploughing: 255, 381 and 653 m3 ha−1 runoff was recorded from permanent bed, terwah and traditional ploughing, respectively during the whole cropping season. The above runoff induced 4.7 t ha−1 soil loss from permanent bed, 7.6 t ha−1 from terwah and 19.5 t ha−1 from traditional ploughing. Overall, contour furrows and permanent raised beds can be part of the ongoing intensification process which includes physical soil and water conservation, slope reforestation, irrigation development and agro forestry in crop lands. Moreover, the use of permanent raised beds if combined with crop mulching and crop diversification is an important component for the development of sustainable conservation agriculture practices in the region.  相似文献

Helicobacter (H.) suis colonizes the stomach of pigs and is the most prevalent gastric non-H. pylori Helicobacter species in humans. Limited information is available on host immune responses after infection with this agent and it is unknown if variation in virulence exists between different H. suis strains. Therefore, BALB/c and C57BL/6 mice were used to compare colonization ability and gene expression of various inflammatory cytokines, as determined by real-time PCR, after experimental infection with 9 different H. suis strains. All strains were able to persist in the stomach of mice, but the number of colonizing bacteria at 59 days post inoculation was higher in stomachs of C57BL/6 mice compared to BALB/c mice. All H. suis strains caused an upregulation of interleukin (IL)-17, which was more pronounced in BALB/c mice. This upregulation was inversely correlated with the number of colonizing bacteria. Most strains also caused an upregulation of regulatory IL-10, positively correlating with colonization in BALB/c mice. Only in C57BL/6 mice, upregulation of IL-1β was observed. Increased levels of IFN-γ mRNA were never detected, whereas most H. suis strains caused an upregulation of the Th2 signature cytokine IL-4, mainly in BALB/c mice. In conclusion, the genetic background of the murine strain has a clear impact on the colonization ability of different H. suis strains and the immune response they evoke. A predominant Th17 response was observed, accompanied by a mild Th2 response, which is different from the Th17/Th1 response evoked by H. pylori infection.  相似文献
