AIMS: To determine efficacy of a minimally invasive (MI) surgical approach using a human MI lumbar retractor for canine lumbosacral dorsal laminectomy and partial discectomy and to compare this technique to the standard open surgical (OS) approach.

METHODS: Lumbosacral dorsal laminectomy and partial discectomy was performed on 16 large-breed canine cadavers using either a standard OS (n=8) or MI (n=8) approach. Skin and fascial incision length, procedure time, and intraoperative complications were recorded. Postoperatively specimens were evaluated for laminectomy and discectomy dimensions, and visible damage to the cauda equina and exiting nerve roots.

RESULTS: Median length of skin and fascial incisions in the OS group were longer than in the MI group (p<0.001). Median laminectomy length was similar between both approaches (p=0.234) but width was greater for the MI than OS approach (p=0.002). Both approaches achieved similar partial discectomy width (p=0.279). Overall surgical time was longer for MI approaches compared to OS, with a median of 18.5 (min 15.5, max 21.8) minutes for MI compared to 14.6 (min 13.1, max 16.9) minutes for OS (p=0.001).

CONCLUSIONS: The MI approach reduced incision lengths while retaining comparable laminectomy and discectomy dimensions. For this in vitro model the MI approach required more time to complete, but this difference may not be relevant in clinical cases.

CLINICAL RELEVANCE: Dogs undergoing lumbosacral dorsal laminectomy are commonly large-breed dogs. The traditional open approach requires a large skin incision and soft tissue dissection, especially in overweight animals. A MI approach accomplishing the same surgical result while minimising soft tissue trauma could reduce post-operative pain and recovery time, and may lower wound-related complications. Clinical studies are needed to confirm postoperative benefit and assess operating times in vivo.  相似文献

Two concurrent pond studies were initiated in April 2011 to evaluate feeds containing solvent‐extracted distillers dried grains with solubles (SE‐DDGS) in both 28 and 32% protein catfish feeds. Channel catfish, Ictalurus punctatus, were stocked in multiple‐batch production ponds at the University of Arkansas at Pine Bluff (UAPB) and hybrid catfish, ♀ Ictalurus punctatus × ♂ Ictalurus furcatus, in single‐batch production ponds at the National Warmwater Aquaculture Center (NWAC), Mississippi State University, Stoneville, Mississippi, USA. The experimental design was a 2 × 2 factorial with two protein levels (32 and 28% protein), with and without 20% SE‐DDGS. Fish were fed once daily to apparent satiation, and managed similarly to a commercial farm for 186 d at UAPB and 172 d at NWAC. For channel catfish, two‐way ANOVA showed significant effects of both protein level (28% being better than 32%) and protein source (20% SE‐DDGS diets being better than traditional diets) on yield, survival, total diet fed, and feed conversion ratio (FCR), due primarily to effects on the understocked fingerlings. Partial budget analysis suggested that the 28% protein diet with the 20% SE‐DDGS was preferred economically for the prevailing feed costs. For hybrid catfish, two‐way ANOVA did not show significant differences in total diet fed, yield, survival, and processing yields, but use of SE‐DDGS resulted in a lower FCR. Results showed that both channel and hybrid catfish can utilize 20% SE‐DDGS in the diet without adversely affecting production or processing characteristics.  相似文献

The effects of lipopolysaccharide (LPS) on plasma cortisol and the expression of MyoD and myostatin (MSTN) mRNAs were evaluated in channel catfish. In addition, the effect of dexamethasone (Dex) on MyoD and MSTN mRNAs was examined. For the LPS injection experiments, juvenile channel catfish were injected intraperitoneally with 1.5 mg/kg LPS or sterile PBS. Blood was collected at 1, 3, 12, and 24 h post-injection for cortisol determination, and muscle samples were collected at 3, 12, and 24 h for mRNA analysis. For the Dex injection experiment, fish were injected with 1.0 mg/kg Dex or saline and muscle samples were collected at 12 and 24 h. There was no effect of LPS on plasma cortisol at any of the time points measured. Injection with LPS increased the abundance of MyoD mRNA at 3 and 12 h, and decreased the abundance of MSTN mRNA at 24 h. There was no effect of Dex injection on the abundance of MyoD mRNA. However, Dex injection decreased the abundance of MSTN mRNA at 12 h post-injection. These results suggest that LPS regulates the expression of MyoD and MSTN independently of an increase in plasma cortisol, and that the regulation of MyoD in the channel catfish differs from mammals in response to inflammatory stimuli. These results also confirm that exogenous glucocorticoids decrease the expression of MSTN as shown in other fish species.  相似文献

Diagnosis of paratuberculosis (Johne's disease) is stymied by the lack of 1 diagnostic tool that can be used to detect both subclinically and clinically infected animals. At present, fecal culture remains the single diagnostic test that can detect infection in both disease states provided the animals actively shed Mycobacterium paratuberculosis in their feces. Yet, fecal culture has a disadvantage associated with the protracted incubation period of 8-16 weeks before results are available. Detection of nucleic acids specific to M. paratuberculosis in fecal samples is a technique that can circumvent the culture method. This study describes a rapid, simple, and effective method to extract DNA from fecal samples and modification of a polymerase chain reaction assay for optimal sensitivity of detection. An evaluation of 1,000 well-characterized fecal samples was performed by the Colorado Department of Agriculture (Denver, CO) and the National Animal Disease Center (Ames, IA) to determine the sensitivity, specificity, and reproducibility of the new method. Results from this study show that the sensitivity of detection was highly dependent on the load of bacteria in the fecal sample with 81% detection of samples containing >70 colony-forming units (cfu)/g of feces and a 45% detection rate for samples containing less than 1 cfu/g. Similarly, reproducibility of the technique between the 2 laboratories (n = 250 samples) was much higher (75%) for the fecal samples containing high levels of M. paratuberculosis and reduced to 25% for samples with less than 1 cfu/g. An overall specificity of 83% was obtained for known negative samples. The method described here is rapid, simple, and inexpensive compared with other techniques. In addition, this method can detect animals that are shedding less than 1 cfu/g.  相似文献

Accurate prediction of meat yield in live animals may allow more efficient genetic improvement of meat yield in farm-raised catfish. An initial trial with 30 channel catfish demonstrated significant correlations among weight-adjusted residuals for muscle area measured from transverse ultrasound images and transverse sections at five locations along the trunk musculature (r = 0.30 to 0.70). Relationships of weight-adjusted residuals for three meat yield traits (carcass, whole fillet, and shank fillet) with weight-adjusted residuals for 15 external body shape measurements and five transverse ultrasound measurements of muscle area were determined for 51 female and 91 male channel x blue catfish backcross hybrids. Compared to males, females had smaller heads; deeper, wider, shorter bodies; larger ultrasound muscle area; and higher meat yield. Correlations between carcass traits and body shape and carcass traits and ultrasound measurements were generally higher for females than for males. Correlations among carcass traits and ultrasound muscle area were typically higher than correlations among carcass traits and external body shape in both sexes. A single ultrasound measurement explained 40 to 50% and 16 to 23% of the variation in meat yield traits of females and males, respectively. The best three-variable model using ultrasound and body shape traits explained 48 to 56% and 31 to 38% of the variation in meat yield traits in females and males, respectively. Differences between males and females for the variability in meat yield traits explained by the models may be related to sexual dimorphism for body shape and fillet yield observed in catfish. Ultrasound has potential for predicting meat yield in live fish, but improved prediction accuracy is needed. Differences in meat yield traits between males and females and among individuals within sexes suggest that selecting for fish with smaller heads and deeper, shorter body shape posterior to the visceral cavity will increase meat yield in catfish.  相似文献

Shiga toxins (Stx) produced by Escherichia coli cause systemic vascular damage that manifests as edema disease in swine and hemolytic uremic syndrome in humans. In vitro, Stx inhibit protein synthesis and, depending on circumstances, induce necrosis, apoptosis, or both. The mechanism of in vivo Stx-mediated vascular damage is not known. The ability of Stx to cause apoptosis of vasculature in vivo was studied in pigs with edema disease that was produced by oral inoculation with Stx-producing E. coli. Arterioles of ileum and brain were evaluated by terminal dUTP nick-end labeling (TUNEL) assay for DNA fragmentation in myocytes (10 infected pigs, 5 control pigs) and by transmission electron microscopy for ultrastructural changes characteristic of apoptosis (17 infected pigs, 8 control pigs). In comparison with controls, increased numbers of TUNEL-positive arterioles were detected in 6/10 (60%) subclinically affected pigs 14-15 days after inoculation. Ultrastructurally, lesions in myocytes consisted of lysis (necrosis), with cytoplasmic debris and nuclear fragments contained between intact basement membranes. Endothelial cell changes ranged from acute swelling to necrosis and detachment from basement membrane. Subclinically affected pigs (n = 14) tended to have changes predominantly in myocytes, whereas pigs with clinical illness (n = 3) more commonly had changes in endothelial cells. The arteriolar lesions and clinical signs of edema disease are attributed to the effects of Stx on vasculature. Therefore, our findings suggest that the Stx-induced arteriolar lesions seen in this study were primarily necrotic, not apoptotic. We suspect that necrosis was the principal cause of the DNA fragmentation detected.  相似文献

Strategic supplementation of P4 may be used to increase conception rates in cattle, but timing of supplementation in relation to ovulation, mass of supplementary P4 and formulation of the P4‐containing supplement has not been determined for beef cattle. Effects of supplementation of long‐acting progesterone (P4) on Days 2 or 3 post‐ovulation on development, function and regression of corpus luteum (CL) were studied in beef cattle. Cows were synchronized with an oestradiol/P4‐based protocol and treated with 150 or 300 mg of long‐acting P4 on Day 2 or 3 post‐ovulation (6–7 cows/group). Colour‐doppler ultrasound scanning and blood sample collection were performed from Day 2–21.5. Plasma P4 concentrations were greater (p < 0.05) from Day 2.5–5.5 in the Day 2‐treated groups and from Day 3.5–5.5 in the Day 3‐treated cows than in the control group. CL area and blood flow during Day 2–8.5 did not differ (p > 0.05) among groups, suggesting no effect of P4 treatment on luteal development. The frequency of cows that began luteolysis before Day 15 was greater (p < 0.04) in cows treated with 300 mg than in the controls, but there were no differences between non‐treated and 150 mg‐treated cows. The interval from pre‐treatment ovulation to functional and structural luteolysis was shorter (p < 0.01) in the combined P4‐treated groups than in the control cows. In conclusion, was showed for the first time that long‐acting P4 supplementation on Day 2 or 3 post‐ovulation increases P4 concentrations for ≥3 day, has no effect on luteal development, but anticipates the beginning of luteolysis in beef cattle.  相似文献

Three 28% protein feeds (premium, standard, or a low-cost diet) were fed to channel catfish in a pond study to compare growth, production, and costs as a follow up to similar work with 32% protein feeds. Twelve 0.1-ha earthen ponds at the University of Arkansas at Pine Bluff (UAPB) Aquaculture Research Station were stocked with carryover fish (0.39 kg mean weight) at 5,860 fish/ha and fingerling catfish (mean individual weight of 15.4 g) at 12,500 fish/ha. Total yield (gross and net) did not differ between fish fed the premium as compared to the standard diet, but that of the premium diet was significantly higher than that of fish fed the low-cost diet. The feed conversion ratio (FCR) did not differ between fish fed the premium and standard diets, but was lower than the FCR of fish fed the low-cost diet. There were no differences found in processing yields among treatments. Given the lower cost of the 28% standard diet and yields equivalent to those of the 28% premium diet, the 28% standard diet was economically preferable to the other diets tested.  相似文献
