The air-coupled ultrasonic inspection method is a widely applied non-destructive measuring technique in the wood-based panel industry. The technology is mainly applied to detect panel delaminations by analyzing the transmitted signal. Recent research deals with the use of ultrasonic techniques not only for the qualitative but also for the quantitative characterization of wood-based panels. To achieve a fundamental understanding of the behavior of ultrasonic waves in wooden panels, it is necessary to study the mechanisms that affect ultrasonic transmission and velocity during testing. Impedance and attenuation effects have been examined in previous studies. This article focuses on the interferences of ultrasonic waves. The interferences can be detected in experiments where the ultrasonic transmission is tested against the panel thickness. The results are verified with a mathematical model that explains the interferences due to multiple reflections inside the tested panels. By fitting the experimental data to the model predictions, the ultrasonic velocity and attenuation can be determined. So far, interference effects have not been considered for the non-destructive testing of wood-based panels. Patterns of geographical diversity, and the relationship between agro-morphological traits and fatty acid composition were
assessed for 193 safflower (Carthamus tinctorius) accessions representing forty countries. Accessions were assigned to eight groups based on geographical proximity. Cluster
and Principal Component analyses were performed to assess patterns of diversity among the accessions and to select the most
distant accessions from each of eight groups for analysis of randomly amplified polymorphic DNA (RAPD) markers. There was
a large amount of diversity for agro-morphological traits, fatty acid composition, and RAPD markers. Most correlations among
different traits were rather low. Plant height showed a positive correlation with days to flowering (r = 0.63**). Palmitic acid was positively correlated with stearic acid and oleic acid values, and negatively correlated with
linoleic acid (P < 0.01). Oleic acid and linoleic acid showed a strong negative correlation (r = −0.89**). The first three principal components together explained 59% of the variation, however, neither principal component
analysis (PCA) nor marker analysis revealed a clear relationship between diversity pattern and geographical origin. Accessions
from some geographical regions tended to group together, such as accessions from South Western Asia, Central Western Europe,
and the Mediterranean region. The correlation between the morphological matrix and the genetic matrix based on RAPD markers
was not significant (r = 0.027). Wide diversity in safflower germplasm indicates a considerable potential for improving this crop for both agronomic

Estimating the fatty acid composition of the oil in intact-seed rapeseed (Brassica napus L.) by near-infrared reflectance spectroscopy 总被引:7,自引:0,他引:7
The objective of this work was to evaluate the potential of near-infrared reflectance spectroscopy (NIRS) as a rapid method
to estimate the fatty acid composition of the oil in intact-seed samples of rapeseed. A total of 549 samples (3 g intact seed)
from selected mutant and breeding lines were scanned by NIRS, and 220 of them were selected and scanned again by using two
different adapters, which reduced the sample size to 300 and 60 mg, respectively. Selected samples were analysed by gas liquid
chromatography and calibration equations for individual fatty acids were developed. Calibrations for oleic, linoleic, linolenic,
and erucic acid were highly accurate, with values of r2 in cross validation from 0.95 to 0.98 (samples of 3 g), from 0.93 to 0.97 (300 mg), and from 0.84 to 0.96 (60 mg). Calibrations
for palmitic and stearic acid were less accurate, with values of r2 in cross validation always lower than 0.8, probably because of the narrow range available for these fatty acids. The accuracy
of the calibration equations for eicosenoic acid was very low (r2 = 0.69 in 3 g samples), although improved equations were developed (r2 from 0.78 to 0.91) when the relationship between erucic and eicosenoic acid was taken into account. We conclude that NIRS
is a powerful technique to estimate the fatty acid composition of the oil in rapeseed, provided that samples covering a wide
range of fatty acid levels are available, with the advantage that such estimation is possible with few additional costs when
NIRS is used for the determination of other seed quality traits.

A germplasm collection consisting of 1475 entries from 21 species of Brassica, including 36 lower taxa, was evaluated for the fatty acid composition of the seed oil. A total of 358 entries representing the taxonomic variability in the collection were selected and analysed by gas-liquid chromatography (GLC). The remaining 1117 entries were analysed by near-infrared reflectance spectroscopy (NIRS), after developing multi-species calibration equations. The results demonstrated that NIRS is an effective technique to assess variability for oleic, linoleic, linolenic and erucic acid in intact-seed samples of multiple Brassica species, provided that calibration equations be developed from sets containing large taxonomic and chemical variability. Some fatty acid ratios were used to estimate the efficiency of the different biosynthetic pathways. Two well-defined patterns were observed. The first one was characterised by high elongation efficiency and accumulation of high levels of erucic acid. The highest erucic acid content (>55% of the total fatty acids) was found in the cultivated species B. napus L., B. oleracea L., and B. rapa L., and in the wild species B. incana Tenore, B. rupestris Raf., and B. villosa Bivona-Bernardi, the three latter belonging to the B. oleracea group (n=9). The second pattern was characterised by high desaturation efficiency, resulting in the accumulation of high levels of the polyunsaturated linoleic and linolenic acid (up to more than 55%). The highest levels of these fatty acids were found in samples of B. elongata Ehrh., especially of the var. integrifolia Boiss. Using molecular-dynamics (MD) simulations the densities and self-diffusion coefficients of a range of liquid monoacid triacylglycerides (TAGs) have been studied as a function of temperature and, for the first time, pressure. While offset by their ambient properties, the response of the TAGs to temperature and pressure is qualitatively similar. Application of pressure was found to significantly increase densities and reduce diffusion of the TAG molecules, suggesting that it may have as much a role in processing and crystallizing TAGs as supercooling does. A solution of glycerol tripalmitate and glycerol trihexanoate was also studied, showing that application of pressure should lead to a significant decrease in the saturation point of the solution, which is an important consideration for processing TAGs. Different solid/liquid interfaces of glycerol tripalmitate have also been investigated. Although crystal growth could not be observed, dissolution of one interface was seen in the MD simulations. The results suggest that over moderate distances the melting of TAGs may be cooperative in nature, rather than involving dissolution of individual TAG molecules. 相似文献

Lawsonia intracellularis is the causative agent of proliferative enteritis in pigs (PPE). This bacterium is difficult to culture from clinical samples and antemortem demonstration is therefore usually performed by PCR on faecal samples. The aim of this study was to elucidate the frequency of L. intracellularis infection in pig herds in Estonia using PCR, histopathological methods and electronmicroscopical studies. The frequency of demonstration of L. intracellularis was highest in 9-12 weeks old pigs (68.1%). It was more frequent in growing pigs with enteritis on small farms where the system of "all-in all-out" was not practiced and where standards of hygiene were poor. Gross and histopathological studies demonstrated that characteristic macroscopic changes associated with PPE were localised to the distal jejunum and ileum.Thickened longitudinal and circumferential folds occurred in the mucosa of the affected regions of the bowel. Samples from pigs aged 4 to 20 weeks exhibited the most intensive inflammatory changes. The distal part of the jejunum, ileum and the upper third of proximal colon and cecum wall were visibly thickened with reduced luminal diameter. Hyperplasia of lymphoid tissue and, in many cases, pseudomembranous or fibrinous inflammation was found. L. intracellularis was detected in 56 young pigs using histopathological methods. Additionally, in 8 of these pigs intracellular bacteria were demonstrated in ilial epithelial cells by transmission electronmicroscopical (TEM) investigation. On the basis of these TEM investigations it was concluded that L. intracellularis causes disturbances of normal growth, differentiation and apoptosis of the epithelial cells of ileum. 相似文献

Since differences in the virulence of Mycoplasma (M.) hyopneumoniae strains have been described, the isolation of field strains followed by genotypic and phenotypic characterisation has become a major goal in epidemiological studies. The aim of this study was to compare various M. hyopneumoniae isolates from different pig herds and numerous pigs within the same herd. Therefore, pigs of 109 herds located in North-Western Germany were sampled either on-farm or during necropsies. Overall, 52 isolates of M. hyopneumoniae were recovered from 45 pigs originating from 21 herds. The identity of cultures was confirmed by PCR targeting the 16S-23S intergenic spacer region. Typing of isolates was achieved by random amplified polymorphic DNA (RAPD) analysis and multi-locus analysis of variable number of tandem repeats (VNTR) and demonstrated a high degree of heterogeneity of M. hyopneumoniae isolates. Differences among isolates recovered from animals of the same herd or even from the same pig revealed a grouping into different genotypic clusters. This outcome was observed with both methods. It was concluded that more than one strain of M. hyopneumoniae might be present in a pig herd and even in a single pig, suggesting high genetic heterogeneity between isolates of the same epidemiological source. These factors should be considered when applying nucleic amplification techniques for characterising M. hyopneumoniae strains to specify the epidemiology of infection and to evaluate virulence factors triggering the corresponding disease. 相似文献

A renewed interest in glucosinolates (GSLs) as compounds with biocidal and anticarcinogenic activity demands evaluation of the available variability in germplasm collections. The objective of the present study was to evaluate a germplasm collection of the genus Brassica for total content and profile of seed GSLs. A total of 1708 entries from 20 Brassica species were nondestructively analysed by near-infrared reflectance spectroscopy. The total GSL content and the concentrations of sinigrin, progoitrin, gluconapin, glucoerucin, glucoiberin, and 4-hydroxyglucobrassicin were estimated by means of previously developed calibration equations. One hundred and fifty entries, having either high GSL content or potentially interesting GSL profiles, were selected and further analysed by high-performance liquid chromatography. The collection contained great variability for GSL content and profile. Very high GSL contents (>200 mol g-1) were measured in accessions of B. montana, B. nigra, and B. oleracea. The greatest intraspecific variability occurred in B. oleracea, where six contrasting GSL profiles were identified. The detected variability might be useful for the development of Brassica crops containing high GSL content and specific GSL profiles. 相似文献

Erysipelas was diagnosed in a layer breeder flock in Sweden in 2002. Although vertical transmission of Erysipelothrix rhusiopathiae has not been previously described in chickens, the potential of erysipelas infection to adversely affect hatching eggs was of concern. To clarify the possible impact of erysipelas on hatching eggs and their progeny, an experiment was done using 200 hatching eggs collected from the infected flock. The eggs were incubated for 21 days, and the egg shells, infertile eggs, dead-in-shell embryos, and a sample of day-old hatched chicks and blood samples from 5-day-old chicks were cultured for E. rhusiopathiae. In addition, after 28 days of grow-out, the male chickens were euthanatized and cultured for the bacterium, and the remaining female chickens were placed as a backyard flock and observed over a 4-mo period. Bacteriological test results of the above-mentioned samples were negative for E. rhusiopathiae. Mortality rates were not excessive, and no clinical symptoms of erysipelas were observed during the period of observation. The result of the investigation suggests that in layer breeder chickens, E. rhusiopathiae is not vertically or egg transmitted and that the disease outbreak in the parent stock had no adverse impact on the quality of hatching eggs in terms of increased embryo mortality. 相似文献
