In 1979 a field trial was started to study the feasibility of maedi-visna control in sheep by half-yearly serological testing (by ELISA) with culling of sero-positive ewes and their progeny. In 13 commercial flocks, with a mean initial incidence of serological reactors of 17%, the sero-positive ewes and all their progeny, those of preceding years included, were culled after each half-yearly test. The percentage of sero-positive sheep decreased gradually and at the end of the second year, at the 5th test, all flocks were sero-negative. Also the 6th and 7th test did not yield sero-positive sheep. At the 8th test, however, 3 sero-positive ewes were detected in one of the flocks. A definite conclusion as to the source of infection could not be drawn. The following flock test was negative. In 2 other commercial flocks, which had a mean initial incidence of sero-positive sheep of 53%, those sero-positive and only their suckling lambs were culled. Here too, a gradual decrease in the incidence of sero-positive sheep was observed at the 2nd and 3rd test, but at the 4th test a sharp increase occurred. The programme was continued and a decrease followed until 0% was reached at the 7th test (end of third year). Age analysis of the sero-positive sheep which caused this peak revealed that the majority had been born before the start of the trial. This suggests that a 'second wave' of sero-positive sheep may be prevented and a quicker result obtained if progeny of preceding years are culled as well.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

In recent years, there has been renewed interest in primary hyperaldosteronism, particularly because of its possible role in the progression of kidney disease. While most studies have concerned humans and experimental animal models, we here report on the occurrence of a spontaneous form of (non-tumorous) primary hyperaldosteronism in cats. At presentation, the main physical features of 11 elderly cats were hypokalemic paroxysmal flaccid paresis and loss of vision due to retinal detachment with hemorrhages. Primary hyperaldosteronism was diagnosed on the basis of plasma concentrations of aldosterone (PAC) and plasma renin activity (PRA), and the calculation of the PAC:PRA ratio. In all animals, PACs were at the upper end or higher than the reference range. The PRAs were at the lower end of the reference range, and the PAC:PRA ratios exceeded the reference range. Diagnostic imaging by ultrasonography and computed tomography revealed no or only very minor changes in the adrenals compatible with nodular hyperplasia. Adrenal gland histopathology revealed extensive micronodular hyperplasia extending from zona glomerulosa into the zona fasciculata and reticularis. In three cats, plasma urea and creatinine concentrations were normal when hyperaldosteronism was diagnosed but thereafter increased to above the upper limit of the respective reference range. In the other eight cats, urea and creatinine concentrations were raised at first examination and gradually further increased. Even in end-stage renal insufficiency, there was a tendency to hypophosphatemia rather than to hyperphosphatemia. The histopathological changes in the kidneys mimicked those of humans with hyperaldosteronism: hyaline arteriolar sclerosis, glomerular sclerosis, tubular atrophy and interstitial fibrosis. The non-tumorous form of primary hyperaldosteronism in cats has many similarities with "idiopathic" primary hyperaldosteronism in humans. The condition is associated with progressive renal disease, which may in part be due to the often incompletely suppressed plasma renin activity.  相似文献

Fungi are key to the functioning of soil ecosystems, and exhibit a range of interactions with plants. Given their close associations with plants, and importance in ecosystem functioning, soil-borne fungi have been proposed as potential biological indicators of disturbance and useful agents in monitoring strategies, including those following the introduction of genetically modified (GM) crops. Here we report on the impact of potato crop varieties, including a cultivar that was genetically modified for its starch quality, on the community composition of the main phyla of fungi in soils, i.e. Ascomycota, Basidiomycota and Glomeromycota in rhizosphere and bulk soil. Samples were collected at two field sites before sowing, at three growth stages during crop development and after the harvest of the plants, and the effects of field site, plant growth stage and plant cultivar (genotype) on fungal community composition assessed using three phylum-specific T-RFLP profiling strategies and multivariate statistical analysis (NMDS ordinations with ANOSIM test). In addition, fungal biomass, arbuscular mycorrhizal colonization of roots and activities of extracellular fungal enzymes (laccases, Mn-peroxidases and cellulases) involved in degradation of lignocelluloses-rich organic matter were determined. Fungal community compositions, densities and activities were observed to differ significantly between the rhizosphere and bulk soil. The most important factors determining fungal community composition and functioning were plant growth stage for the rhizosphere communities and location and soil properties for the bulk soil communities. The basidiomycetes were the most numerous fungal group in the bulk soils and in the rhizosphere of young plants, with a shift toward greater ascomycete numbers in the rhizosphere at later growth stages. There were no detectable differences between the GM cultivar and its parental cultivar in terms of influence on fungal community structure of function. Fungal community structure and functioning of both GM- and parental cultivars fell within the range of other cultivars at most sampling moments.  相似文献

The structure of the soil in the arable layer is controlled by tillage, soil biota activities and weathering, whereas the structure below this layer is mainly the result of the activities of soil biota. Organic farmers tend to minimise the depth of the main tillage operation to encourage soil biota to create a soil structure with continuous biopores and a well crumbled topsoil. The best main tillage operation for preventive weed control, especially important in organic farming, is mouldboard ploughing. The shallow ploughing experiments described in this paper were conducted to ascertain the minimum ploughing depth for an ecologically accountable, sustainable tillage system with good weed control, good land qualities (in terms of workable days, aeration and soil moisture conditions) and finally with good yields. The “ecoplough” used for shallow ploughing was developed by Rumptstad Industries to meet the requirements of relatively shallow ploughing with good soil inversion for weed control. The plough has seven or eight bottoms for ploughing depths of 0.12–0.20 m, a working width of 2.1 m and a working speed of 1.7 m s−1. Its width is such that the tractor with wide low-pressure tyres runs on top of the land.

After using the plough for 6 years on Luvisols (>200 gkg−1<2 μm) in the IJsselmeer polders and on Luvisols (120–160 gkg−1<2 μm) in polders near the northern coast of The Netherlands, it was found that compared with conventional ploughing, shallow ploughing required less energy and labour and produced a relatively smooth surface. The latter facilitates the preparation of a seedbed consisting of relatively fine, strong, stable and moist aggregates. Organic matter, soil biota and nutrients were concentrated higher in the profile, influencing the workability of the soil, the growth of weeds and the growth of crops. Most of the yields were similar to yields after conventional ploughing, but weed populations increased when ploughing depth was <0.2 m. It was concluded that for organic farming on “active” soils (soils subject to shrink/swell with >200 g kg−1<2 μm), shallow ploughing seems to be the best reduced tillage system. It has several advantages. The main factor determining the minimum ploughing depth is control of weeds, especially of perennials.  相似文献

Biotic soil parameters have so far seldom played a role in practical soil assessment and management of grasslands. However, the ongoing reduction of external inputs in agriculture would imply an increasing reliance on ecosystem self-regulating processes. Since soil biota play an important role in these processes and in the provision of ecosystem services, biological soil parameters should be an integral part of soil assessment. The general objective of the current study is to investigate to what extent biotic soil parameters provide additional value in soil quality assessment of grassland on sandy soils. We measured abiotic and biotic soil parameters together with process parameters underlying ecosystem services in 20 permanent production grasslands. Cross-validated stepwise regression was used to identify abiotic and biotic soil parameters that explained the soil ecosystem services soil structure maintenance, water regulation, supply of nutrients, and grassland production, respectively.Process parameters underlying the ecosystem service soil structure maintenance such as bulk density and the percentage of sub-angular blocky elements were mainly influenced by SOM and its qualities. The correlations between penetration resistance at 0-10 cm and the percentage of soil crumbs with earthworms suggested a relationship to earthworm activity. Parameters underlying the service of water regulation showed no clear relationship to biotic soil parameters. Water infiltration rate in the field was explained by the penetration resistance at 10-20 cm. Process parameters underlying the service of nutrients’ supply such as the potentially mineralizable C and N were mainly determined by soil total N. The potential C and N mineralization were more related to biotic soil parameters, whereby each parameter was the other’s antithesis. The grassland production without N fertilization viz. the nitrogen supply capacity of the soil measured as N yield, was mainly explained by soil organic matter (SOM) and soil moisture, and to a lesser extent by soil total N. One gram of SOM per kg of dry soil corresponded to 3.21 kg N yield ha−1, on top of a constant of 15.4 kg N ha−1. The currently applied calculations in the Dutch grassland fertilization recommendation, underestimated in 85% of the production grasslands, the measured nitrogen supply capacity of the soil by on average 42 kg N ha−1 (31%). This legitimizes additional research to improve the currently applied recommendations for sandy soils. The response of N yield to N fertilization ranged from 35 to 102%. This wide range emphasizes the importance of a better recommendation base to target N fertilizer. The response of N yield to N fertilization was predicted by the total number of enchytraeids, the underlying mechanism of which needs further investigation on different soil types. This knowledge can be important for the optimal use of fertilizer and its consequences for environmental quality.  相似文献

Interactions between Serratia plymuthica A30 and a blackleg‐causing biovar 3 Dickeya sp. were examined. In a potato slice assay, S. plymuthica A30 inhibited tissue maceration caused by Dickeya sp. IPO2222 when co‐inoculated at a density at least 10 times greater than that of the pathogen. In glasshouse experiments, population dynamics of the antagonist and of the pathogen in planta were studied by dilution plating and confocal laser scanning microscopy (CLSM) using fluorescent protein‐tagged strains. Pathogen‐free minitubers were vacuum‐infiltrated with DsRed‐tagged Dickeya sp. IPO2222 and superficially treated during planting with a water suspension containing GFP‐tagged S. plymuthica A30. A30 reduced the blackleg incidence from 55% to 0%. Both the pathogen and the antagonist colonized the seed potato tubers internally within 1 day post‐inoculation (dpi). Between 1 and 7 dpi, the population of A30 in tubers increased from 101 to c. 103 CFU g?1 and subsequently remained stable until the end of the experiment (28 dpi). Populations of A30 in stems and roots increased from c. 102 to c. 104 CFU g?1 between 7 and 28 dpi. Dilution plating and CLSM studies showed that A30 decreased the density of Dickeya sp. populations in plants. Dilution plating combined with microscopy allowed the enumeration of strain A30 and its visualization in the vascular tissues of stem and roots and in the pith of roots, as well as its adherence to and colonization of the root surface. The implications of these finding for the use of S. plymuthica A30 as a biocontrol agent are discussed.  相似文献

The first half of this paper discusses the development of sustainability law. It takes as its basis that environmental law plays a central role in the achievement of sustainable development and that environmental lawyers and those from associated disciplines must come to terms with the imperatives of the internationally accepted concept of sustainable development. The second half of the paper deals with the implications of sustainability law for the teaching of environmental law, including the need to further liberate environmental law from the confines of law schools and lawyers, and to make it part of the common discourse of government, business and communities at large. A number of current initiatives in environmental legal education and training are canvassed, focussing on the Asia Pacific region.  相似文献
