Steers were generated from Angus (A), Beefmaster (BM), Brangus (BA), Gelbray (GB), and Simbrah (SB) sires mated to cows of their breed and to Brahman x Hereford F1 cows (except A) to characterize their carcass traits, composition, and palatability. The 290 steers (48 A, 48 BM, 36 BA, 31 GB, and 46 SB) were slaughtered at an equal fatness end point as determined by real-time ultrasound and visual evaluation. Angus steers had lighter (P < 0.01), more youthful (P < 0.01) carcasses with a higher (P < 0.05) quality grade, more (P < 0.01) fat thickness, and a larger (P < 0.01) longissimus area/100 kg than BM-, BA-, GB-, and SB-sired steers. Angus steers also had a lower (P < 0.01) specific gravity, a higher (P < 0.01) percentage fat and less (P < 0.05) lean in the 9th to 11th rib, and steaks aged for 10 d were more tender (P < 0.01) than steaks from Brahman-derivative sired steers. The BM- and BA-sired steers had lighter (P < 0.01), more youthful (P < 0.05) carcasses, and smaller (P < 0.01) longissimus area than GB- and SB-sired steers. The 9th to 11th rib section from the BM- and BA-sired steers had less lean and more bone (P < 0.01) than GB- and SB-sired steers. The BA-sired steers had more (P < 0.01) marbling and a higher (P < 0.05) quality grade than BM-sired steers. The SB-sired steers had heavier (P < 0.01) carcasses than the GB-sired steers. There were no differences in shear force for steaks aged for 3 d for any of the breed types, but with 10 d of aging, steaks from Angus steers were more tender, possibly indicating that steaks of Brahman-derivative breeds aged at a slower rate than those from Angus.  相似文献

Infections of the tick-borne ehrlichial pathogen, Anaplasma marginale, in cattle have been controlled, in part, by administration of low doses of tetracycline. Recently, a cell culture system was developed for A. marginale using a tick cell line derived from embryonic Ixodes scapularis. This study was designed to determine the effect of tetracycline on A. marginale propagated in a tick cell culture assay. Various concentrations of tetracycline (0, 0.01, 0.10, 1.0, 5, 10, 20 or 100 microg/ml) were added in medium to cultures 48h after cell monolayers were inoculated with A. marginale. A. marginale growth in the drug treated and control cultures was subsequently evaluated by indirect ELISA at 7 days post-infection (PI) and daily by light and electron microscopy (LM and EM). Infectivity of the culture-derived A. marginale was determined by inoculation of susceptible cattle with treated and untreated control cultures. Tetracycline doses of 5, 10, 20 and 100 microg/ml resulted in significant inhibition of A. marginale growth as determined by ELISA. Morphologic deterioration of Anaplasma, as determined by LM and EM, occurred in cultures treated with the same drug concentrations. A. marginale replication, inhibited in cultures treated on days 2-6 PI with 20 microg/ml tetracycline, was not apparent 96 days after antibiotic removal. Infected cell cultures treated with medium containing 20 microg/ml tetracycline proved to be non-infective when inoculated into susceptible splenectomized calves. All parameters studied herein demonstrated that tetracycline killed A. marginale in cultured tick cells. The Anaplasma-tick cell culture drug assay therefore, would be useful for screening and evaluating novel antibiotics for control of anaplasmosis.  相似文献

Anaplasma marginale is a tick-borne hemoparasite of cattle worldwide. The Virginia isolate of A. marginale was propagated previously in a cell line derived from embryos of the tick, Ixodes scapularis. The cultured Anaplasma (VA-tc) was passaged continuously for over 4 years and retained its infectivity for cattle and antigenic stability. We report herein the continuous in vitro cultivation of a second isolate of A. marginale derived from a naturally infected cow in Oklahoma (OK-tc). Blood from the infected cow was subinoculated into a splenectomized calf and blood collected at peak parasitemia was frozen, thawed and used as inoculum on confluent tick cell monolayers. Colonies of Anaplasma were apparent in low numbers at 9 days post exposure (PE) and infection in monolayers reached 100% by 4-5 weeks PE. Cultures were passaged by placing supernatant onto fresh tick cell monolayers at a dilution of 1:5 or 1:10. By the third passage development of the OK-tc was similar to that of the VA-tc and a 1:5 dilution resulted in 100% infection in 10-12 days. Inoculation of OK-tc into a splenectomized calf caused clinical anaplasmosis and Dermacentor ticks that fed on this calf transmitted the organism to a second susceptible calf. Major surface proteins (MSPs) 1-5 of the OK-tc were compared with homologous proteins present on VA-tc and the erythrocytic stage of the Oklahoma isolate. The MSPs 1, 2, 4, 5 were conserved on the OK-tc but there was evidence for structural variation in MSP3 between the cultured and erythrocytic stage of Anaplasma. MSP2 and MSP3 were the major proteins recognized by serum from infected cattle. Two-dimensional gels also identified positional differences between VA-tc and OK-tc in MSP2 and MSP3. The OK-tc may have potential to be used as antigen for development of an improved vaccine for anaplasmosis in the South Central United States.  相似文献

Ticks transmit pathogens that cause diseases which greatly impact both human and animal health. Vaccines developed against Boophilus spp. using Bm86 and Bm95 tick gut antigens demonstrated the feasibility of using vaccines for control of tick infestations. These vaccines also reduced transmission of tick-borne pathogens by decreasing exposure of susceptible hosts to ticks. The recently discovered tick antigens, 64P putative cement protein and subolesin involved in the regulation of tick feeding and reproduction, were also shown to reduce tick infestations. These antigens, together with the TROSPA receptor for Burrelia burgdorferi OspA were effective against tick-borne pathogens by reducing the infection levels in ticks and/or the transmission of the pathogen. Development of a vaccine targeted at both the tick vector and pathogen would contribute greatly to the control of tick infestations and the transmission of tick-borne diseases. These results have demonstrated that tick vaccines can be developed for control tick infestations and show promise for the prevention of the transmission of tick-borne pathogens.  相似文献

Bovine anaplasmosis is a tick-borne hemolytic disease of cattle that occurs worldwide caused by the intraerythrocytic rickettsiae Anaplasma marginale. Control measures, including use of acaricides, administration of antibiotics and vaccines, have varied with geographic location. Our research is focused on the tick-pathogen interface for development of new vaccine strategies with the goal of reducing anaplasmosis, tick infestations and the vectorial capacity of ticks. Toward this approach, we have targeted (1) development of an A. marginale cell culture system to provide a non-bovine antigen source, (2) characterization of an A. marginale adhesion protein, and (3) identification of key tick protective antigens for reduction of tick infestations. A cell culture system for propagation of A. marginale was developed and provided a non-bovine source of A. marginale vaccine antigen. The A. marginale adhesion protein, MSP1a, was characterized and use of recombinant MSP1a in vaccine formulations reduced clinical anaplasmosis and infection levels in ticks that acquired infection on immunized cattle. Most recently, we identified a tick-protective antigen, subolesin, that reduced tick infestations, as well as the vectorial capacity of ticks for acquisition and transmission of A marginale. This integrated approach to vaccine development shows promise for developing new strategies for control of bovine anaplasmosis.  相似文献

Infections of Ichthyophonus hoferi, a cosmopolitan parasite of marine fish, have recently been reported in rockfish, Sebastes spp., from the north‐eastern Pacific. Because I. hoferi also infects Pacific herring, Clupea pallasi Valenciennes, and salmonids in this region, we wanted to determine if Ichthyophonus parasites from rockfishes, Pacific herring and chinook salmon, Oncorhynchus tshawytscha (Walbaum), were the same. Small subunit ribosomal deoxyribonucleic acid sequence data revealed two haplotypes that were fixed among host species in geographic sympatry, one from rockfish and the other from both Pacific herring and salmon. These isolated populations of Ichthyophonus could be part of the same species that are ecologically separated because of host behaviours, or they could be distinct species that are host specific. Dietary patterns of the hosts indicate that ecological separation among hosts is possible, but the presence of distinct species may better explain the observed Ichthyophonus haplotype association with host species.  相似文献

Phosphodiesterase-4 (PDE 4) enzyme inhibitors have been shown to have anti-inflammatory properties in various animal disease processes and therefore could be effective drugs for the treatment of equine airway diseases. The purpose of this study was to evaluate the efficacy and adverse effects of the PDE 4 inhibitor L-826,141 in horses with heaves. In a blinded parallel design, horses with heaves exposed daily to moldy hay were given a placebo for 14 days and then administered either L-826,141 (n = 6; loading dose of 1 mg/kg IV followed by 0.5 mg/kg IV q48h) or dexamethasone (n = 6; 0.04 mg/kg IV q24h) from days 15 to 29 (study 1). Pulmonary function and bronchoalveolar (BAL) cytology were evaluated weekly from baseline (day 0) to 29 days. In study 2, horses were treated with L-826,141 (1.0 mg/kg IV q24h) for 8 days. Although ex vivo lipopolysaccharide-induced tumor necrosis factor (TNF)-alpha and LTB4 production by fresh blood were inhibited up to 90% after repeated administrations of L-826,141, this treatment failed to improve lung function. In contrast, dexamethasone (positive control) treatment resulted in significant improvement in lung mechanics and airway function in all horses. Neither drug had a significant effect on BAL total cell counts and differential cytology. Administration of the PDE 4 inhibitor L-826,141 for up to 14 days to horses with heaves was not associated with an improvement in airway function or inflammation. These findings suggest that the PDE 4 enzyme is not a key mediator of lung inflammation in heaves.  相似文献

A study was conducted to compare Brangus, Beefmaster, Gelbray, and Simbrah breed influences for economically important traits. Brangus (9), Beefmaster (12), Gelbray (10), and Simbrah (7) sires were used in purebred and crossbred (Brahman x Hereford F1 cows) matings to generate calves (326) in eight breed groups. Beefmaster cows were of similar size (448 kg), Brangus and Gelbray cows were 11% heavier (501 and 503 kg), and Simbrah cows were 21% heavier (548 kg) compared to Brahman x Hereford F1 cows (452 kg). Calves sired by Brangus and Beefmaster bulls had lower birth weights (35 vs 38 kg; P < 0.05), preweaning growth rates (0.87 vs 0.91 kg x d(-1); P < 0.01), and weaning weights (206 vs 219 kg; P < 0.01) than Gelbray- and Simbrah-sired calves. Birth weights, preweaning ADG, and weaning weight and hip heights were similar between Brangus- and Beefmaster-sired calves. Simbrah-sired calves had greater preweaning growth rates (0.94 vs 0.88 kg x d(-1); P < 0.05), weaning weights (227 vs 211 kg; P < 0.01), and adjusted 205-d hip heights (126 vs 122 cm; P < 0.05) than Gelbray-sired calves. Straightbred Angus steers were introduced in the postweaning portion of the study. Steer calves were placed on feed at an average age of 14.5 mo. Steers were removed from the feedlot upon attaining a targeted 10 mm of backfat. Feedlot ADG did not differ among sire breeds. Brahman-derivative sired steers required an additional 54 d on feed (P < 0.01) and were 86 kg heavier (P < 0.01) at harvest than Angus steers. Continental-Brahman steers spent an additional 25 d on feed (P < 0.05) and were 35 kg heavier (P < 0.01) at harvest than British-Brahman steers. Simbrah-sired steers were 52 kg heavier (P < 0.01) at harvest than Gelbray-sired steers when fed for a similar number of days (211 vs 203 d). However, straightbred Simbrah steers required an additional 12 d on feed (P < 0.01) and weighed 47 kg more (P < 0.01) than Simbrah-sired crossbred steers. The economic value of the heavier calf weaning weights may be offset by the attendant larger cow size of the Continental-Brahman compared to the British-Brahman breeds. Similarly, the heavier weights of Continental-Brahman compared to British-Brahman steers, when harvested at a prescribed level of fatness may be viewed as a benefit, but the increased number of requisite days in the feedlot is a disadvantage.  相似文献

Carcass data from 1,494 straightbred and rotational crossbred steers were collected over four generations. Mating systems included straightbreds (Angus [A], Brahman [B], Charolais [C], and Hereford [H]); two-breed rotations (A-B, C-B, and H-B); three-breed rotations (A-B-C, A-B-H, and B-C-H); and a four-breed rotation (A-B-C-H). Steers were randomly allocated to one of four postweaning treatments that varied in length of grazing and feeding periods. Treatment and breed group (four straightbreds and seven rotational combinations) significantly influenced hot carcass weight (HCWT), retail yield (RY), longissimus muscle area (LM), fat thickness (FT), marbling score (MS), USDA quality grade (QG), and Warner-Bratzler shear force (WBS). Feeding for longer periods resulted in greater (P < .05) HCWT, RY, FT, and MS, higher QG, and lower (P < .05) WBS values. Among the straightbreds, C was heavier and larger for HCWT, RY, and LM (P < .01), whereas A and H had greater (P < .01) FT and MS. The B was similar to H for HCWT, RY, and LM and to C for FT but ranked last (P < .01) for MS and WBS. Three- and four-breed rotational mating systems were superior (P < .05) to the two-breed rotation for HCWT, RY, and LM but were similar for FT, MS, and WBS. Rotational combinations exceeded (P < .05) the straightbreds for all carcass traits except MS.  相似文献
