Defining protective responses to pathogens: cytokine profiles in leprosy lesions   总被引:134,自引:0,他引:134  
The immunological mechanisms required to engender resistance have been defined in few infectious diseases of man, and the role of specific cytokines is unclear. Leprosy presents clinically as a spectrum in which resistance correlates with cell-mediated immunity to the pathogen. To assess in situ cytokine patterns, messenger RNA extracted from leprosy skin biopsy specimens was amplified by the polymerase chain reaction with 14 cytokine-specific primers. In lesions of the resistant form of the disease, messenger RNAs coding for interleukin-2 and interferon-gamma were most evident. In contrast, messenger RNAs for interleukin-4, interleukin-5, and interleukin-10 predominated in the multibacillary form. Thus, resistance and susceptibility were correlated with distinct patterns of cytokine production.  相似文献

Energetic young pulsars and expanding blast waves [supernova remnants (SNRs)] are the most visible remains after massive stars, ending their lives, explode in core-collapse supernovae. The Fermi Gamma-Ray Space Telescope has unveiled a radio quiet pulsar located near the center of the compact synchrotron nebula inside the supernova remnant CTA 1. The pulsar, discovered through its gamma-ray pulsations, has a period of 316.86 milliseconds and a period derivative of 3.614 x 10(-13) seconds per second. Its characteristic age of 10(4) years is comparable to that estimated for the SNR. We speculate that most unidentified Galactic gamma-ray sources associated with star-forming regions and SNRs are such young pulsars.  相似文献

Background

The division of labor in social insect colonies involves transitions by workers from one task to another and is critical to the organization and ecological success of colonies. The differential regulation of genetic pathways is likely to be a key mechanism involved in plasticity of social insect task behavior. One of the few pathways implicated in social organization involves the cGMP-activated protein kinase gene, foraging, a gene associated with foraging behavior in social insect species. The association of the foraging gene with behavior is conserved across diverse species, but the observed expression patterns and proposed functions of this gene vary across taxa. We compared the protein sequence of foraging across social insects and explored whether the differential regulation of this gene is associated with task behaviors in the harvester ant, Pogonomyrmex occidentalis.

Results

Phylogenetic analysis of the coding region of the foraging gene reveals considerable conservation in protein sequence across insects, particularly among hymenopteran species. The absence of amino acid variation in key active and binding sites suggests that differences in behaviors associated with this gene among species may be the result of changes in gene expression rather than gene divergence. Using real time qPCR analyses with a harvester ant ortholog to foraging (Pofor), we found that the brains of harvester ant foragers have a daily fluctuation in expression of foraging with mRNA levels peaking at midday. In contrast, young workers inside the nest have low levels of Pofor mRNA with no evidence of daily fluctuations in expression. As a result, the association of foraging expression with task behavior within a species changes depending on the time of day the individuals are sampled.

Conclusions

The amino acid protein sequence of foraging is highly conserved across social insects. Differences in foraging behaviors associated with this gene among social insect species are likely due to differences in gene regulation rather than evolutionary changes in the encoded protein. The task-specific expression patterns of foraging are consistent with the task-specific circadian rhythms observed in harvester ants. Whether the molecular clock plays a role in regulating foraging gene expression (or vice versa) remains to be determined. Our results represent the first time series analysis of foraging gene expression and underscore the importance of assaying time-related expression differences in behavioral studies. Understanding how this gene is regulated within species is critical to explaining the mechanism by which foraging influences behavior.  相似文献

Experiments from a range of ecosystems have shown that insectivorous birds are important in controlling the populations of their invertebrate prey. Here, we report on a large field experiment testing the hypothesis that management for enhancing recreational values in suburban woodlands affects the intensity of bird predation on canopy-living arthropods. Bird exclosures were used in two types of management (understory clearance and dense understory) at two foraging heights in oak Quercus robur canopies and the experiment was replicated at two sites.  相似文献

Below-ground (bg) shoot emergence rates of Sonchus arvensis are dependent on temperature and root weight. However, it is unknown to what extent this is due to a root depletion rate that depends on initial root weight, or due to differences in resource allocation to fine root and bg shoot growth. To resolve this, we retrieved data from an experiment in which plants were grown in the dark at constant temperature (4°C, 8°C, and 18°C) and harvested prior to or at shoot emergence. A dynamic mass-balance model, in which biomass of the initial root was allocated to bg shoot and fine root daily growth, and where respiration took place from all tissues, was used. The relative depletion rate of root biomass (RDR; d?1) and fraction of the depleted biomass allocated to bg shoots (SFRR) were estimated and calibrated to observed biomass. The RDR increased with initial root weight and temperature and SFFR was highest for light roots and lowest for heaviest roots, whereas the rest was allocated to fine root biomass. The length-to-biomass ratio of bg shoots decreased with initial root weight. Under between-year weather variations (2004–2010), the reduction in root biomass during the coldest April–May was simulated to be over 12 days delayed compared with the warmest spring. The influence of biomass allocation on bg shoot elongation of heavier roots was thus stimulated by a larger fraction of root biomass being depleted, but counteracted by a smaller fraction of it allocated into bg shoot elongation, compared with lighter roots. The complexity of shoot emergence based on root depletion estimates may be a reason why predictions based on only an accumulated root weight-specific temperature sum, as proposed by a previous study, are expected to be less uncertain than those based on root depletion estimates.  相似文献

The leaves and stems of the Rooibos plant ( Aspalathus linearis) are used for the production of an herbal tea known for its health promoting properties, which have been linked to its flavonoid content but which is substantially reduced by the traditional processing method employed. Selected food-grade fungi were screened for their potential to improve the yield of soluble matter extracted from rooibos plant material. Fungal cocktails of hydrolyzing enzymes enhanced either the yield of soluble solids ( Lentinula edodes and Rhizopus oryzae cultured in yeast peptone-wheat straw medium) or the yield in antioxidants from fermented rooibos ( R. oryzae cultured in potato dextrose or yeast peptone-wheat straw medium). When applied to green rooibos, L. edodes (cultured in yeast peptone-wheat straw medium) enhanced the release of soluble solids as well as color formation, leading to semifermented rooibos with a relatively high aspalathin content, compared to fermented rooibos.  相似文献

Cytological analysis of an underdeveloped chicken embryo at 6 days of incubation revealed a triploid-diploid mosaic condition. Of the 30 metaphases observed, 19 were triploid and 11 diploid. The triploid cells were 3A-ZZZ and diploid cells 2A-ZZ, as determined for the six largest pairs of chromnosomes.  相似文献

Dentistry in exotic carnivores is becoming more common as their level of care is enhanced. These cases present many interesting challenges, but adaptation of currently accepted dental procedures, use of newly developed instrumentation, and educated improvisation can handle almost any situation that a practitioner might encounter. It must be kept in mind, however, that there are virtually no medications, medicaments, or drugs that are approved for treatment of the various exotic carnivores. Therefore, the treatments that are discussed in this article should all be considered as "off label" use, and be reviewed closely before being applied to patients.  相似文献
