The effects of five environmental factors on variation in the rate of C2H2 reduction in two cowpea (Vigna unguiculata L. Walp.) and two soybean (Glycine max L. Merrill) cultivars were examined at two stages of growth in the field. Diurnal changes in C2H2 reduction, estimated as μmoles C2H4 produced·g?1 nodules·h?1, over a 30 h period were compared with changes in soil, canopy and air temperatures, global radiation, and vapour pressure deficit. Although the environmental factors showed one maximum and one minimum during a diurnal cycle, the C2H2 reduction rate in nodulated cowpea roots showed two peaks: one between 0600–1200 and another between 1800–2400 and two minima: one between 1200–1600 and another between 2400–0600. Variation in C2H2 reduction in soybean nodules did not show any definite pattern. Vapour pressure deficit appeared the most likely factor influencing the decline in C2H2 reduction between 1200–1600.The rate of C2H2 reduction in cowpeas was greater at the pre-flower than at the early pod-fill period; the rate in soybeans was not greatly different at early flower or early pod-fill.  相似文献

Soil Protozoa (primarily small naked amoebae and flagellates) were counted under control conditions and with stresses of nitrogen, water, or nitrogen and water under field conditions (Ecosystem Stress Area, Pawnee Site, northeastern Colorado, USA) in the summer of 1974. Protozoan populations were also measured in soil cores removed from the field and incubated under a wet-dry cycle. Protozoan numbers were higher in the top 1 cm of soil and overlying litter than at a depth of 5–6 cm in all treatments. After rainfall totaling 35 mm, the control and fertilized only treatments showed population increases with the fertilized only treatment showing the greater change. Protozoa showed marked responses to the addition of water to soil cores, with the largest numbers appearing after peak CO2 evolution. Numbers of active (trophic) forms ranged from about 20.000. g?1 dry soil in the control treatment under dry conditions to over 100,000. g?1 dry soil in the irrigated plus fertilized plot. There were few (<5%) cystic (dormant) forms in all treatments contrary to earlier studies in mesic climates. A rough estimate of protozoan production is presented.  相似文献

Amprolium, a coccidiostat added to poultry feed, occurs in excreta at concentrations of 204 μ g?1 and investigations were made of the effect of this quantity of amprolium on the biochemistry of soil to which manure had been applied.Greenhouse experiments showed that 0.8 μg g?1 amprolium was found in soil pots 80 days after treatment with manure at the equivalent of 56.1 t ha?1 and was detectable 20 days following treatment at the equivalent of 11.2 t ha?1.Laboratory experiments indicated that amprolium was differentially adsorbed to two complexing media, soil and manure. Since amprolium was a constituent of treated manure, it was expected that the amprolium manure-soil system would offer various sites for adsorption of amprolium. Mixing amprolium with soil and with soil plus untreated manure yielded approximately the same effect on amprolium adsorption based on recoveries in water solution, and as methanol extractable. However, total recovery of amprolium from treated manure added to soil was only a fraction of the above, indicating the high complexing capacity of the manure.No effect on soil respiration was observed by either pure amprolium or amprolium as a constituent of treated manure. The higher rate of manure application caused greater respiration due to the presence of more readily oxidizable organic matter, but the respiration pattern attributed to the manure component was not unlike the respiration pattern of the Guelph loam soil.  相似文献

Reduction of N2O in moist soil was inhibited completely by 10?2 atm C2H2 and partially by 10?5 atm C2H2. The effect of C2H4 was 104 times less than that of C2H2. Denitrification of NO?3 occurred in anaerobically or aerobically incubated waterlogged soil and in anaerobic but not in aerobic moist soil. In the absence of C2H2 there was transient accumulation of N2O. In the presence of C2H2 there was stoichiometric conversion of NO?3 to N2O. Some kinetics of the reduction of N2O and of NO?3 to N2O are presented. Denitrification of 1 μg added NO?3-N.g? could be measured within 1 h. Stoichiometries of production of N2O from NO?2 and NO?3, respectively, and production of CO2 attributable to denitrification were consistent with reported energy yields. Reduction of C2H2 to C2H4 occurred immediately following complete denitrification of added NO?3. The incubation of soil in the presence and in the absence of C2H2 thus permits assay of both denitrification and N2 fixation and provides information on the mole fraction of N2O in the products of denitrification.  相似文献

A method for vital staining of fungal mycelium with fluorescein diacetate (FDA) is described. In experiments with pure cultures, good correlation was obtained between relative staining efficiency, growth rate, and respiration. FDA thus appears to be a true vital stain, in that it stains only metabolically active mycelia.In fresh soil suspensions stained with FDA, brightly fluorescent hyphae and portions of hyphae were observed. The applicability of the method for measurement of active mycelium in the soil is discussed.  相似文献

The dry matter and crude protein production of the perennial ryegrass varieties S24, S23, Premo, Houba, Barlenna, Melle and Sceempter Pasture at annual nitrogen application rates of 212, 425, 637 and 1062 kg ha?1 were studied for the years 1972–74 inclusive. The DM and CP response curves for the variety Melle were steeper than those for other varieties tested and levelled off at a higher N application rate. The results suggest that Melle is a variety with a genotype capahle of utilizing N more efficiently at higher rates than the other varieties tested.  相似文献

Grass silage made in late May from S24 perennial ryegrass was offered to twelve Ayrshire cows in a 16-week feeding experiment. The silage had a DM content of 25.3%, a pH of 3.91, and contained 16.8% CP and 68.4% digestible organic matter in the DM. The silage was fed ad libitum and was the sole feed in the control treatment. In the other three treatments the silage was supplemented with a cube offered at rates of 0.8, 1.4 and 2.0 kg per 10 kg milk. The cube contained 82.2% groundnut, plus molasses and minerals, and had 37.9% CP in the DM. The daily intake of silage DM was 11.4 kg per cow on average over the four treatments which did not differ significantly. The mean daily milk yields were 14.8 kg per cow in the control treatment, and 16.5, 18.2 and 18.4 kg in the 0.8, 1.4 and 2.0 kg supplement treatments respectively. The SNF, CP and lactose contents of the milk were Wghest on the 1.4 kg supplement treatment. It is concluded that with a high digestibility silage, there is little evidence to support the feeding of more than 1.4 kg of high-protein cubes per 10 kg milk in the declining phase of lactation.  相似文献

A medium quality barn-dried hay and a poor quality field-cured hay were each supplemented with 3 levels of concentrate in the diet of young beef cattle, averaging 313 kg liveweight. The supplement was given on predetermined hay concentrate metabolizable energy (ME) ratios based on the determined ME of the hays and an assumed ME of 3·0 McalsAg DM for the supplement. The hay concentrate ratios used were 1:1, 1:2 and 1:3, the aim being to provide a reasonable rate of growth (0·7 kg/day) with the poorer hay. It was necessary to include urea in the concentrate mixtures used on the 1:1 and 1:2 ratios with the field-cured hay. The urea appeared to have an adverse effect on the intake and utilization of DM on the 1:1 ratio. The range of mean intakes of supplement DM was 3·02 to 4·41 kg/day for the barn-dried hay and 2·23 to 4·61 for the field-cured hay. Mean daily liveweight gains with the barn-dried hay were 0·76, 0·89 and 0·85 kg for the 1:1, 1:2 and 1:3 ratios, respectively, with no significant difference between ratios. The mean daily gains with the field-cured hay were 0·39, 0·77 and 0·75 for the three ratios, the mean gain on the 1:1 ratio being significantly lower than those of the other ratios. The ME conversion ratios were not significantly affected by either ratios or hay type, except on the 1:1 ratio with field-cured hay. There were significant differences between the barn-dried hay and field-cured hay diets in the digestibilities of DM, organic matter, crude protein and energy; the mean coefficients for DM being 74·5% and 65·6%, respectively.  相似文献

Acidophilic actinomycetes, growing between pH 3.5 and 6.5, were isolated from 17 natural soils and mine wastes. Most isolates were Streptomyces spp. and selected ones were compared with some established neutrophilic species by assessment of overall similarity. Acidophilic and neutrophilic strains clustered separately, differing in a number of cultural and physiological characters, in addition to pH requirements. The possible importance of acidophilic actinomycetes in soil is discussed.  相似文献
