Toll-like receptors (TLRs) are a class of pattern recognition receptors that play a pivotal role in the innate and adaptive immune systems. Studies have shown that TLR variants play roles in various human infectious diseases. The aim of the present study was to investigate whether the functional genetic variations at positions C632T, G1409A, A1475C, G1550A and G1596A in TLR1 and at A2700G and A3156G in TLR9 confer susceptibility or resistance to bovine tuberculosis (bTB). Genotyping of the TLR1 and TLR9 gene polymorphisms was performed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and single-strand conformation polymorphism (PCR-SSCP) in 586 Chinese Holstein cows (154 infected with bTB, 432 non-infected). The frequencies of the GH and HH genotypes at TLR1-G1596A differed significantly between bTB-infected and non-infected animals [p=0.001 for both GH and HH; GH odds ratio (OR)=2.43 95% confidence interval (CI) (1.47-4.03); and HH OR=1.49 95% CI (0.85-2.62)]. There was a trend toward an increased relative risk of tuberculosis (TB) incidence in the CD genotype at the TLR1-A1475C locus [p=0.056, OR=1.59 95% CI (0.98-2.58)]. The present study suggests that variants in the TLR1 gene are associated with susceptibility to bTB, whereas no significant association can be inferred from the polymorphisms in the TLR9 gene.  相似文献

The levels of oxidative stress markers are an important indicator of the physiological state of the parasite and its host. In the present study levels of lipid peroxidation, glutathione S transferase, glutathione, superoxide dismutase and catalase were determined in the Clinostomum complanatum progenetic metacercaria, obtained from the fish peritoneum (a hypoxic habitat). The in vivo transformed ovigerous adult worms were obtained from the aerobic environment of the buccopharyngeal region of experimentally infected chickens. Levels of antioxidant molecules were also determined in the blood of experimentally infected chickens. An increase in the levels of lipid peroxidation, and a significant decrease in the levels of glutathione S transferase, glutathione, superoxide dismutase and catalase was observed in the infected host as compared to the controls. In the ovigerous worms, the levels of lipid peroxidation, glutathione S transferase, glutathione, superoxide dismutase were found to be significantly less than the levels observed in the progenetic metacercaria. Since the establishment of worm in the buccal cavity of the avian host would lead to its exposure to oxygen and the haematophagous nature of the parasite also exposes it to the free radicals in the host blood, the progenetic metacercaria has evolved to produce excess free radical scavenging molecules reserved to combat the oxidative stress encountered within the microhabitat of the definitive host.  相似文献

Two completely randomised experiments compared the impact of different spices, each at 30 mg/g substrate DM, on the in vitro dry matter disappearance (IVDMD), methane, ammonia and volatile fatty acids (VFA) from either wheat flour (wheat) (experiment 1) as highly or hay based mixtures (experiment 2) as moderately fermentable substrates. Experiment 1 tested the effects of adding cinnamon, clove, coriander, cumin and turmeric, individually, on the rumen fermentation during their in vitro incubations with wheat for 24 h. All spices reduced methane (P < 0.05) from wheat as compared to the spice-free wheat (control). Coriander reduced methane more followed by turmeric, cumin and cinnamon than the control. The IVDMD of wheat did not differ for most spices except cinnamon where IVDMD was reduced (P < 0.05) as compared to the control. Whilst, rumen ammonia was greatest for cumin (P < 0.01) followed by coriander and turmeric, it was lowest for clove (P < 0.01). The spices did not affect the pH and total VFA of rumen fluid for wheat. However, the presence of clove reduced acetic acid (P < 0.05). Experiment 2 tested the effect of only three (coriander, cumin and turmeric) spices individually or together on the in vitro fermentation of hay based mixtures during 96 h of incubation. Whilst all the individual spices or their mixture reduced methane from the hay based mixtures (P < 0.05), the extent of this reduction was highest for turmeric, second highest for coriander and lowest for cumin. It appeared that most spices modified methane emission from wheat as a single ingredient or hay based mixtures without having detrimental effect on diet disappearance or fermentation in vitro. However, the extent of this modification depended on the type of a spice and substrate. Therefore, careful selection of a spice to suit specific feeds would be essential before their in vivo use to modify the fermentation efficiency of ruminant diets.  相似文献

An investigation of radioactivity and some heavy metal distribution in soil and sediment of the river basin (Bendimahi River, Van-Turkey) was conducted in two seasons of 2005. The samples of soil and sediment were collected from the basin and investigated for concentrations of some heavy metal and natural radioactivity. Concentrations of Pb, Cr, Mn, Fe, Co, Cu, Zn and Cd have been determined by Atomic Absorption Spectrometry (AAS), gross-alpha and gross-beta activity concentrations have been obtained by using gas-flow proportional counter in nuclear spectroscopic system. Correlation analysis was made for radioactivity and heavy metal concentrations and the Pearson correlation coefficients between gross-alpha and gross-beta radioactivity and heavy metal were determined.  相似文献

This paper presents the effects of particle size and mixing ratio on the properties including physical, mechanical, and decay resistance of wood plastic composites (WPCs). In addition, it also presents the effects of immersion temperatures on water absorption (WA) and thickness swelling (TS) of the WPCs. WPCs with a thickness of 6 mm were fabricated from Albizia richardiana King & Prain wood particles and recycled polyethylene terephthalate (PET) by the flat-press method. To prepare the WPCs, two different wood particle sizes (0.5–1.0 and 1.01–2.0 mm) were used along with four different mixing ratios (w/w). Subsequently, the physical properties include density, moisture content, WA, and TS, and mechanical properties include modulus of elasticity (MOE) and modulus of rupture (MOR) of the produced WPCs was evaluated. Furthermore, decay resistance was evaluated by the weight loss percentage method. Moreover, the effects of immersion temperatures on WA and TS of WPCs after 24 h of immersion in water at three different temperatures, i.e., 25, 50, and 75 °C were investigated. Results showed that the wood particle size had impact on WPC’s density (only 6% decreased with the increase of particle size); however, the density decreased by 29% when the wood particle content increased from 40 to 70%. The WA and TS gradually increased with the increase of particle content and decrease of particle size. In addition, WA and TS increased proportionately with increasing immersion temperature from 25 to 75 °C. Furthermore, the highest MOE (2570 N/mm2) was found for the WPCs fabricated from large wood particles having the ration of 50:50 (wood particle:PET). For decay resistance, WPCs consisted of larger particles and higher PET content showed greater resistance against decay. Therefore, it is comprehensible that fabrication of the WPCs from 50% large particles and 50% PET is technically feasible and further improvement of WPC performance like enhancement of MOE and reduction of density using coupling agent and agricultural waste fibers, respectively, in the WPC formulation is recommended.  相似文献

Gmelina arborea Roxb. is a fast-growing, deciduous tree species native to Bangladesh. It has been widely planted since 1992 for commercial production of pulpwood in hilly areas of Bangladesh. The purpose of this study was to derive well-fitting allometric models for estimation of aboveground biomass, and carbon and nutrient (nitrogen, phosphorus and potassium) stocks in G. arborea to ensure sustainable production and management of this species. Eight linear models with 64 regression equations were tested for estimation of aboveground biomass, carbon and nutrient stocks in different parts (leaves, branches, bark and stem) of the plant. The best-fitting allometric models were selected in accordance with various relevant statistical criteria.  相似文献

In this review we demonstrate the interaction of the blueprint of an individual (the genome, genomic DNA), its phenotype and the environment. The phenotype consists of quantitative (e.g. growth, milk yield) or functional characteristics e.g. fitness, longevity, fertility and disease resistance. The latter characteristics influence the welfare of an animal substantially. As only the genetically determined part of a particular characteristic is transferred from one generation to the next, it is important to know what the genetic variants (alleles) of the parents at one or more gene loci are. New methods in molecular biology have made it possible to localize and characterize important genes which help to breed more efficient and healthy animals. The exact characterization of the phenotype is vital in identifying genes with major effects and therefore the cooperation with experts from veterinary medicine, biochemistry, and biology is indispensable. As well as an overview of available genetic tests in farm animals, we show various examples how to identify the molecular basis of a particular phenotype and how to use the results in practical breeding programs. Genetic diagnosis enables the breeder to identify undesired alleles early and hinders therefore its uncontrolled distribution in the population. In the long term this leads to a smaller number of affected animals and depending on the disease it may help to prevent animals from suffering.  相似文献

Cercospora heliotropii-bocconii is a fungal pathogen of the ephemeral annual weedHeliotropium europaeum. The effects of wetness period and inoculum concentration on disease severity were studied under controlled conditions. The fungus was grown on different artificial culture media and carrot juice agar with 5 g l–1 yeast extract was found to be the most suitable medium for conidial production under artificial conditions. Abundant disease symptoms only occurred after 8 h of wetness at 20°C. The minimum incubation period before disease symptoms appeared was 8 days following a wetness period of at least 40 h. Inoculum concentration of 1×104 conidia per ml killed plants in less than one month and reduced seed production by two thirds. These results suggest that this pathogen has the potential to reduce plant survival and seed bank replenishment of this annual weed species.  相似文献

Ganoderma boninense causes severe losses to oil palm in South East Asia. The disease typically manifests itself as basal stem rot, but there remains controversy over the route of infection and source of inoculum. Using isolates differing in aggressiveness, infection via roots was confirmed; it was also shown that large inoculum provided as Ganoderma -infested palm- or rubber-wood blocks (12 × 6 × 6 cm) is necessary for soil infection of seedlings after 6–8 months. Smaller blocks (3 × 3 × 3 cm) produced rapid (≤ 3 months) infection of roots and lower stem when physically attached to roots. Therefore fragmentation of infested palm wood from a felled, mature plantation before subsequent replanting may provide inoculum. Failure of G. boninense to grow through non-sterile soil or organic debris from frond bases, suggests it is a poor competitor and that roots must contact inoculum directly. Severe disease occurred after 8 months on inoculated seedlings under shade, but not on seedlings exposed to sun. Soil temperatures in sunlight frequently rose above 40°C and reached 45°C, whereas in shade they never exceeded 32°C. Ganoderma boninense is probably inhibited in exposed soil since optimal growth in vitro was 25–30°C, and there was no recovery from 45°C. Soil temperature may explain why symptoms often first appear in mature plantations when canopy formation creates shade. Infection is not peculiar to senescing palms but can occur throughout the growth cycle.  相似文献
