The mammary elimination of four potentially toxic metabolites of Febantel has been investigated by HPLC methods in the cow after the administration of a 7,5 mg/kg single oral experimental dose of this anthelmintic. Gleditsia triacanthos L. pods were harvested at San Lorenzo del Escorial, Madrid, Spain, before and after seed desiccation. Maturing green pods, harvested from trees, contained soft light-green seeds, whereas fully mature brown pods, collected on the ground, contained hard dark-green seeds. Based on visual inspection of seeds, it was determined that the green color is located in the cotyledons and embryo of soft seeds but mainly in the seed coat of hard seeds. High performance liquid chromatographic analysis indicated that both hard and soft seeds contained the same set of photosynthetic pigments as fully developed leaves, but in different proportions. The hard and soft seeds mainly differed in their chlorophyll a to chlorophyll b ratio and in the composition of the xanthophyll cycle pool of pigments. Fluorescence at -196 degrees C revealed that the molecular organization of the pigment molecules in the seed coat of hard seeds differed from that in intact cotyledons of soft seeds and intact green leaves. The -196 degrees C fluorescence spectra also revealed the presence of a small heterogenous pool of non-photoactive protochlorophyll(ide)s, similar to those found in dark-grown tissues of gymnosperms and angiosperms. 相似文献

Ether lipids are composed of alkyl lipids with an ether bond at the sn-1 position of a glycerol backbone and alkenyl lipids, which possess a vinyl ether bond at the sn-1 position of the glycerol. These ether glycerolipids are present either as polar glycerophospholipids or neutral glycerolipids. Before studying the biological role of molecular species of ether glycerolipids, there is a need to separate and quantify total alkyl and alkenyl glycerolipids from biological samples in order to determine any variation depending on tissue or physiopathological conditions. Here, we detail the development of the first high-performance thin-layer chromatography method for the quantification of total alkyl and alkenyl glycerolipids thanks to the separation of their corresponding alkyl and alkenyl glycerols. This method starts with a reduction of all lipids after extraction, resulting in the reduction of neutral and polar ether glycerolipids into alkyl and alkenyl glycerols, followed by an appropriate purification and, finally, the linearly ascending development of alkyl and alkenyl glycerols on high-performance thin-layer chromatography plates, staining, carbonization and densitometric analysis. Calibration curves were obtained with commercial alkyl and alkenyl glycerol standards, enabling the quantification of alkyl and alkenyl glycerols in samples and thus directly obtaining the quantity of alkyl and alkenyl lipids present in the samples. Interestingly, we found a differential quantity of these lipids in shark liver oil compared to chimera. We quantified alkyl and alkenyl glycerolipids in periprostatic adipose tissues from human prostate cancer and showed the feasibility of this method in other biological matrices (muscle, tumor). 相似文献
Concern about the effects of pesticides on the environment and the desire to reduce purchased inputs are stimuli to reduce herbicide use. Field research was conducted for three growing seasons to compare the chemical contamination and effectiveness of three weed management practices. Weed control treatments included application of a herbicide mixture of 1.5 kg ai ha−1 of atrazine plus 0.9 kg ai ha−1 of pyridate, this mixture being broadcast alone or band-applied in combination with mechanical weeding, and mechanical weeding alone. The results show that it is possible to decrease the amount of atrazine residue in soil with the band-applied treatment. Such a method is very attractive because crop yields were not decreased, at least in the short run. However, we do not yet know the long-term efficiency of this method regarding groundwater quality, weed population dynamic and crop yield. Further work is therefore needed to assess the long-term impact of this weeding method. 相似文献
Basing decisions for the management of contaminated sediments on ecotoxicological data is still often met with skepticism by European stakeholders. These concerns are discussed as they pertain to bioassays to show how ecotoxicological data may provide added value for the sustainable management of sediment in aquatic systems.
Materials and methods
Five “concerns” are selected that are often raised by stakeholders. The ecotoxicological practice is discussed in light of the knowledge gained in recent decades and compared with chemical sediment analysis and chemical data.
Results and discussion
Common assumptions such as a higher uncertainty of biotest results for sediments compared to chemical analyses are not supported by interlaboratory comparisons. Some confusion also arises, because the meaning of biotest data is often misunderstood, questioning their significance in light of a limited number of organisms and altered test conditions in the lab. Because biotest results describe a sediment property, they should not be directly equated with an impact upon the biological community. To identify a hazard, however, the possibility of false-negative results due to the presence of contaminants that are not analyzed but are toxic is lower.
Conclusions
The cost of increased investment in ecotoxicological tests is, in our view, small compared with that of making false-negative assessments of sediment/dredged material that can ultimately have long-term environmental costs. As such, we conclude that ecotoxicological testing is an opportunity for sediment management decision-making that warrants more attention and confidence in Europe.
