Anadromous fishes are frequently restricted by artificial barriers to movement such as dams and culverts, so measuring dispersal helps identify sites where improved connectivity could promote range expansion and population viability. We used a combination of DNA‐based parentage analysis and mark–recapture techniques to evaluate dispersal by juvenile coho salmon (Oncorhynchus kisutch) in a population in the initial stages of colonisation following installation of fish passage structures at a previously impassable dam on the Cedar River, WA, USA. The spatial distribution of individuals within maternal families revealed that dispersal was common. Among the offspring of radio‐tagged mothers, 28% were collected outside the spawning reach and dispersed up to 6.3 km (median = 1.5 km). Most juveniles captured in a tributary (Rock Creek, where few adults spawned) had immigrated from the Cedar River and represented many different families. Juvenile dispersal therefore provided a secondary phase of spatial expansion following initial colonisation by adults. Consistent with the condition‐dependent dispersal hypothesis, juveniles that dispersed farther upstream in the tributary were larger than fish collected near the tributary mouth. Overall, the results demonstrated widespread dispersal in a system with low coho salmon densities, and this might increase the rate of population growth if it reduces the effects of local density dependence. By implication, juveniles can take advantage of rearing habitats reconnected through barrier removal, even when such areas are located several kilometres from adult breeding grounds.  相似文献

OBJECTIVE: To compare the elution characteristics of ceftiofur and liquid and powdered gentamicin and amikacin from polymethylmethacrylate (PMMA) and from hydroxyapatite cement (HAC). METHODS: PMMA and HAC beads in triplicate were impregnated with various amounts and formulations of antibiotics. Beads were immersed in 5 mL of phosphate buffered saline that was replaced at 1, 3, 6, and 12 hours, and 1, 2, 3, 5, 7, 10, 14, 18, 22, 26, and 30 days. The eluent was stored at -70 degrees C until assayed within 2 weeks by microbiological assay (gentamicin and amikacin) or capillary electrophoresis (ceftiofur). RESULTS: Rate of elution for all beads was greatest within the first 24 hours. Cumulative release of total antibiotic dose from beads over 30 days was significantly greater from HAC than PMMA. Antibiotic elution was directly related to the amount of antibiotic incorporated into the cement. Powdered and liquid forms of gentamicin had similar elution rates from PMMA. Elution of amikacin from PMMA beads was greater when the powdered form was used compared with liquid amikacin. Eluent concentrations of ceftiofur were similar to those of the aminoglycosides during the first 3 to 7 days but then decreased precipitously by comparison. CONCLUSIONS: Elution of antibiotics from HAC was greater than from PMMA. Gentamicin- and amikacin-impregnated PMMA and HAC released bactericidal concentrations of antibiotic for at least 30 days. Ceftiofur-impregnated PMMA or HAC is unlikely to provide long-term bactericidal concentrations. CLINICAL RELEVANCE: Gentamicin and amikacin elute effectively from PMMA and HAC.  相似文献

Neuropathic pain arises as a debilitating consequence of nerve injury. The etiology of such pain is poorly understood, and existing treatment is largely ineffective. We demonstrate here that glial cell line-derived neurotrophic factor (GDNF) both prevented and reversed sensory abnormalities that developed in neuropathic pain models, without affecting pain-related behavior in normal animals. GDNF reduces ectopic discharges within sensory neurons after nerve injury. This may arise as a consequence of the reversal by GDNF of the injury-induced plasticity of several sodium channel subunits. Together these findings provide a rational basis for the use of GDNF as a therapeutic treatment for neuropathic pain states.  相似文献

Transmembrane helices of integrin alpha and beta subunits have been implicated in the regulation of integrin activity. Two mutations, glycine-708 to asparagine-708 (G708N)and methionine-701 to asparagine-701, in the transmembrane helix of the beta3 subunit enabled integrin alphaIIbbeta3 to constitutively bind soluble fibrinogen. Further characterization of the G708N mutant revealed that it induced alphaIIbbeta3 clustering and constitutive phosphorylation of focal adhesion kinase. This mutation also enhanced the tendency of the transmembrane helix to form homotrimers. These results suggest that homomeric associations involving transmembrane domains provide a driving force for integrin activation. They also suggest a structural basis for the coincidence of integrin activation and clustering.  相似文献

The dye Procion Yellow M4RS crosses junctional membranes from cytoplasm to cytoplasm at electrotonic synapses between segments of the crayfish septate axon. The dye does not enter the cells from extracellular space. Thus permeability of junctional membranes is qualitatively different from that of nonjunctional membranes. Electron microscopy after fixation in the presence of lanthanum hydroxide indicates that these synapses are "gap junctions" and that there is a network of channels continuous with extracellular space between apposed junctional membranes. These channels must be interlaced with intercytoplasmic channels that are not open to extracellular space.  相似文献

Patent infections with Strongylus vulgaris were established in 6 of 8 helminth-free ponies given 41 to 101 adult worms via nasogastric tube. The parasites were removed from the cecum and ventral colon and transferred within 1 to 2 hours of the death of the donor horses. Eggs were found in the feces of the recipients in 2 or 3 days; egg counts reached maximum, 28 eggs per gram of feces, at 4 weeks after ponies were inoculated. In 6 ponies euthanatized 3 to 7 weeks after parasitic transfers were done, 28% of the inoculated worms were found alive at necropsy. A 7th pony was maintained as a donor for establishing infections for chemotherapy trials and, although never passing more than 6 eggs per gram of feces, shed infective larvae over a period of 2 years.  相似文献

A simulation model of litter size in swine based on ovulation rate, uterine capacity and potential embryo viability was compared to three genetic models to clarify its genetic characteristics. The simulation model is equivalent to independent culling based on fixed levels of potentially viable embryos and uterine capacity. Litter size also can be described by a combination of additive, additive x additive, mean environment x additive, random environment and additive x random environment effects. A third genetic model that can describe the simulation model is the associative effects model, in which litter size is the result of grouping two genotypes. The fixed independent culling levels model predicts that genetic parameters will change as the component means change. This genetic model also predicts that selection on an index of ovulation rate and uterine capacity would improve selection response for litter size. This genetic model predicts asymmetry of correlated responses in ovulation rate and uterine capacity when selecting for high and low litter size. The nonadditive genetic model predicts covariances among relatives that are different from their additive relationships; however, simulated results did not detect any differences. The nonadditive genetic model also predicts that heterosis for litter size will differ among crosses based on the mean environment and on additive x additive genetic interaction. The associative effects model predicts that selection for litter size will always lead to a positive response in litter size.  相似文献

A mathematical model of litter size in swine was developed from ovulation rate, potential embryonic viability and uterine capacity. The model assumed that ovulation rate was reduced to potentially viable embryos by factors innate to the ovum and embryo. Potentially viable embryos then could be further reduced to uterine capacity, the maximum number of fetuses that a female can carry to term. Consequently, litter size can be no greater than either ovulation rate or uterine capacity. Means and variances of ovulation rate and potential embryonic viability used in the model were based on experimental results. The mean and variance of uterine capacity were varied until the simulated mean and variance of litter size were equal to experimental results. Simulated results of relationships among ovulation rate, embryo survival and litter size were similar to observed experimental relationships. Heritabilities of simulated litter size and embryo survival were similar to literature values when the heritability of ovulation rate was set at .25 and the heritability of uterine capacity was set at either .15 or .20. Litter size was simulated for 25 combinations of average ovulation rate and uterine capacity to develop equations relating mean ovulation rate and uterine capacity to litter size, embryo survival and correlations among them. Results suggest that changing either ovulation rate or uterine capacity independently will not result in large changes in litter size. Consequently, the model suggests that a single gene, hormonal manipulation or nutritional change will not result in large increases in litter size and that combinations of factors will be needed to increase litter size.  相似文献

Choice of the optimal number and sequence of breeds in a periodic cross was determined by comparing the trade-off between increased utilization of breed differences and decreased utilization of heterosis. It was shown that the change in mean efficiency resulting from adding the next best breed to the best conventional n-breed rotation is always less than the change in efficiency predicted from the increase in heterosis. Periodic rotations were generally optimized by decreasing the proportion of poorer performing breeds in the rotation. However, efficiency of periodic rotations can exceed that of the better breed even when the difference in additive breed effects for efficiency is almost twice the effect of heterosis on efficiency. The periodic rotation that was optimal also tended to have the lowest inter-generational variance. It was suggested that inter-generational variances of component traits, which are not necessarily minimized when crosses are selected on a combined efficiency trait, can be considered by including inter-generational variance in an index or by introducing maximum thresholds.  相似文献
