Trichinellosis is a cosmopolitan zoonotic disease affecting a wide variety of animals, including man. Non-encapsulated and encapsulated species diverge with respect to their developmental strategies. Little is known at the molecular level about parasite-derived mediators responsible for host muscle cell transformation occurring during trichinellosis. In this context, host-parasite relationships in Trichinella-infected animals could be related to different host-immune and cell mediators, e.g. nitric oxide (NO). Here, we investigate the stimulatory/inhibitory role of L1 antigens from four encapsulated (T. spiralis, T. britovi, T. nelsoni and T. nativa) and one non-encapsulated (T. pseudospiralis) Trichinella species on NO production from rat macrophages in vitro. Our results demonstrate that encapsulated and non-encapsulated Trichinella species differ in their capacity to stimulate the secretion of NO from host macrophages. Biological significance of these differences should be further assessed in the available experimental models.  相似文献

Reduction in soil disturbance can stimulate soil microbial biomass and improve its metabolic efficiency, resulting in better soil quality, which in turn, can increase crop productivity. In this study we evaluated microbial biomass of C (MB-C) by the fumigation-extraction (FE) or fumigation-incubation (FI) method; microbial biomass of N (MB-N); basal respiration (BR) induced or not with sucrose; metabolic quotient (obtained by the ratio BR/MB-C) induced (qCO2(S)), or not with sucrose (qCO2); and crop productivity in a 14-year experiment in the state of Paraná, southern Brazil. The experiment consisted of three soil-tillage systems [no-tillage (NT), conventional tillage (CT) and no-tillage using a field cultivator every 3 years (FC)] and two cropping systems [a soybean–wheat-crop sequence (CS), and a soybean–wheat–white lupin–maize–black oat–radish crop rotation (CR)]. There were six samplings in the 14th year, starting at the end of the winter crop (wheat in the CS and lupin in the CR plots) and finishing at full flowering of the summer crop (soybean in the CS and maize in the CR). Differences in microbiological parameters were greater than those detected in the total C (TCS) and total N (TNS) contents of the soil organic matter (SOM). Major differences were attributed to tillage, and on average NT was higher than the CT in the following parameters: TCS (19%), TNS (21%), MB-C evaluated by FE (74%) and FI (107%), and MB-N (142%). The sensibility of the microbial community and processes to soil disturbance in the tropics was highlighted, as even a moderate soil disturbance every 3 years (FC) affected microbial parameters but not SOM. The BR was the parameter that most promptly responded to soil disturbance, and strong differences were perceived by the ratio of qCO2 evaluated with samples induced and non-induced with sucrose. At plowing, the qCO2(S):qCO2 was five times higher under CT, indicating a C-starving low-effective microbial population in the C-usage. In general, crop rotation had no effect on microbial parameters or SOM. Grain yield was affected by tillage and N was identified as a limiting nutrient. Linear regressions between grain yields and microbial parameters showed that soybean was benefited from improvements in the microbial biomass and metabolic efficiency, but with no significant effects observed for the maize crop. The results also indicate that the turnover of C and N in microbial communities in tropical soils is rapid, reinforcing the need to minimize soil disturbance and to balance inputs of N and C.  相似文献

Interferon-gamma (IFN-gamma) detection assay is being applied as an ancillary test to tuberculin tests in the diagnosis of bovine tuberculosis to detect the maximum number of infected animals. Among possible factors influencing the performance of tuberculosis-diagnostic tests, paratuberculosis, a widespread disease in Spain and other European countries, has been pointed out as a cause of false positive reactions. Still, its effect on the sensitivity of these tests in cattle has yet to be fully characterized. The impact of paratuberculosis in the apparent sensitivity of IFN-gamma assay was studied in a bullfighting cattle herd with a mixed tuberculosis-paratuberculosis infection, using culture of Mycobacterium bovis and Mycobacterium avium paratuberculosis as the gold standard to determine the infection status of every animal. A total of 218 animals were slaughtered and sampled for bacteriology after blood sampling. IFN-gamma assay showed a lower apparent sensitivity in animals with a mixed infection (50%) compared to all animals suffering tuberculosis (78.3%). This finding indicates that the presence of paratuberculosis in tuberculosis-infected herds could imply a serious impairment in the sensitivity of IFN-gamma detection test.  相似文献

Fusarium Head Blight (FHB), or “scab,” is a very destructive disease that affects wheat crops. Recent research has resulted in accurate weather-driven models that estimate the probability of an FHB epidemic based on experiments. However, these predictions ignore two crucial aspects of FHB epidemics: (1) An epidemic is very unlikely to occur unless the plants are flowering, and (2) FHB spreads by its spores, resulting in spatial and temporal dependence in risk. We develop a new approach that combines existing weather-based probabilities with information on flowering dates from survey data, while simultaneously accounting for spatial and temporal dependence. Our model combines two space-time processes, one associated with pure weather-based FHB risks and the other associated with flowering date probabilities. To allow for scalability, we model spatiotemporal dependence via a process convolutions approach. Our sample-based approach produces a realistic assessment of areas that are persistently at high risk (where the probability of an epidemic is elevated for extended time periods), along with associated estimates of uncertainty. We conclude with the application of our approach to a case study from North Dakota.  相似文献

Micro-fading spectrometry is a technique that combines visible reflectance spectroscopy and accelerated light aging testing. Therefore, it is a useful tool for determining the light-stability of dyes applied on textile substrates in a relatively short amount of time. Traditional accelerated light aging methods usually require controlling many variables such as time and intensity of illumination, correct positioning of the sample during irradiation, and reproducible spot selection for each subsequent spectrocolorimetric measurement. The use of micro-fade testing as an alternative way of conducting light-fastness assessments of dye/textile systems has been explored. The results indicate that the technique is suitable for testing the color stability of cultural heritage materials and industrially manufactured textiles since it is a direct and non-contact method that takes into account the aforementioned experimental variables.  相似文献

We compared the effects of medium light roast (MLR) and medium roast (MR) paper-filtered coffee on antioxidant capacity and lipid peroxidation in healthy volunteers. In a randomized crossover study, 20 volunteers consumed 482?±?61?ml/day of MLR or MR for four weeks. Plasma total antioxidant status (TAS), oxygen radical absorbance capacity (ORAC), oxidized LDL and 8-epi-prostaglandin F2α, erythrocyte superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activity were measured at baseline and after the interventions. MLR had higher chlorogenic acids-(CGA; 334?mg/150?mL) and less caffeine (231?mg/150?ml) than MR had (210 and 244?mg/150?ml, respectively). MLR also had fewer Maillard reaction products (MRP) than MR had. Compared with baseline, subjects had an increase of 21 and 26?% in TAS, 13 and 13?% in CAT, 52 and 75?% in SOD, and 62 and 49?% in GPx after MLR and MR consumption (P?相似文献

A comparison between traditional extraction techniques (hydrodistillation and organic solvent extraction) and supercritical fluid extraction was made for two different populations and crops of Artemisia absinthium L., cultivated in the field and aeroponically. The composition of the extracts, volatile and non volatile oils, was analyzed by GC-MS and HPLC-DAD, respectively. The antifeedant and phytotoxic activity of the extracts was tested on insect pests (Spodoptera littoralis, Myzus persicae and Rhopalosiphum padi) and plants (Lactuca sativa and Lolium perenne). The supercritical extracts exhibited stronger antifeedant effects than the traditional ones (up to 8 times more active) with moderate selective phytotoxic effects on L. perenne root growth (<50% inhibition).  相似文献

The fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae), is one of the most important pests of corn in Latin America. This insect presents two strains with behavioural and genetic differences. In Mexico, both strains are present, and at least two different FAW populations have been reported within the corn-strain. The objective of this study was to gather evidence of whether pheromonal communication varies among different S. frugiperda populations infesting corn in Mexico. First, we investigated any qualitative or quantitative difference in the composition of sex pheromones among populations; second, we studied whether male antennal responses to pheromone components vary among populations; and, finally, we investigated whether males from a region can discriminate between a synthetic pheromone blend characteristic of their region and blends formulated with the ratio of pheromone compounds emitted by females from other populations. Sex pheromone components were sampled by solid-phase microextraction and identified by gas chromatography coupled to mass spectrometry. Females from all populations consistently released three compounds: (Z)-9-tetradecenyl acetate (Z9-14:OAc), (Z)-11-hexadecenyl acetate (Z11-16:OAc) and (Z)-7-dodecenyl acetate (Z7-12:OAc). Our results showed that Z9-14:OAc and Z7-12:OAc evoked the highest and most variable antennal responses among the populations studied compared to Z11-16:OAc. However, males did not discriminate between local pheromone blends and those formulated from other populations in a field test. These results show that although there is geographic variation in the ratio of pheromone components and in the peripheral reception of them, males were not differentially attracted to different pheromone blends in the field.  相似文献

In Brazil, the first major invasion event of Bemisia tabaci was that of Middle East–Asia Minor 1 (MEAM1) species, formerly termed as B biotype, which commenced in the 1990s mainly by ornamental plants in São Paulo State. More than two decades after this invasion, the presence of the Mediterranean (MED) species of B. tabaci, formerly Q biotype, was reported in Rio Grande do Sul, the southernmost State of Brazil, and now in São Paulo and Paraná States, in southeastern Brazil. Specimens of whiteflies collected from commercial begonia, hydrangea, petunia and poinsettia greenhouses in São Paulo, and also from begonias and poinsettias collected in flower shops in Paraná, were all identified as belonging to MED species. Furthermore, the secondary endosymbionts Arsenophonus, Hamiltonella and Rickettsia of MED from São Paulo and Paraná were detected by PCR and their presence confirmed by sequencing and FISH analysis, and those results differed from MED detected in Rio Grande do Sul that harbored only Hamiltonella and Cardinium. Our results suggest a new MED invasion into Brazil and is associated with ornamental plants. The two MED populations are genetically different and suggest that they are separate invasions.  相似文献
