The effects of injection solvent and mobile phase composition on the reverse-phase liquid chromatographic determination of aflatoxin M1 (M1) were examined. M1 was converted to the more highly fluorescent derivative aflatoxin M2a (M2a). Using a C-18 column and a mobile phase of H2O-MeCN-MeOH (60 + 20 + 20) (MP-A), M2a was dissolved in various ratios of MeCN-H2O prior to injection. Chromatographic efficiency for the M2a peak varied from ca 2000 theoretical plates when injected in 30% aqueous MeCN to ca 9000 plates when injected in water alone. However, using the same C-18 column but with a mobile phase of H2O-IPA-MeCN (80 + 12 + 8) (MP-B), the M2a peak exhibited 25,000 plates when injected in 30% aqueous MeCN and 10,000 plates when injected in water alone.  相似文献

Twelve ruminally, duodenally, and ileally cannulated (average initial BW 313 +/- 20 kg) and 27 intact Hereford heifers (average initial BW 256 +/- 17 kg) were used in two experiments to evaluate dairy food wash water solids (WWS) as a protein source in medium-quality hay diets. Heifers received a basal diet of orchardgrass hay (7.4% CP) and were assigned to one of three supplement treatments: control (C; .9% CP), WWS (18.8% CP)-, and soybean meal (SBM; 19.1% CP)-based supplements (fed at 1.5 kg of DM/d). Supplements were formulated to have similar ME concentrations. Ruminal ammonia concentrations were greater (P less than .10) for WWS- and SBM-supplemented heifers than for C heifers at most sampling times. Moreover, WWS and SBM increased (P less than .10) total VFA (mM) and acetate (mol/100 mol) and lowered propionate (mol/100 mol) at several sampling times. Ruminal fluid volume (liters) was unchanged (P greater than .10) by treatment; however, fluid dilution and flow rate (liters/h) were less (P less than .10) in C heifers than in heifers fed SBM or WWS supplements. Wash water solids and SBM supplementation increased (P less than .10) OM, NDF, and ADF digestibilities compared with C heifers. Feeding WWS and SBM supplements increased BW at 84 d (P less than .10) compared with C-supplemented heifers. Forage intake at 54 and 84 d by heifers supplemented with SBM or WWS was greater (P less than .10) than by C heifers. Control-supplemented heifers had the least, WWS intermediate, and SBM the greatest ADG at 84 d (P less than .10; .14 vs .35 vs .48 kg/d, respectively). These data indicate that WWS may be used as a protein source without serious adverse effects in heifers consuming medium-quality hay for 84 d.  相似文献

? Washington State University, in Pullman, Washington, has recently begun plans to compost approximately 16,500 tons of the waste generated annually on campus. Animal manure from the university dairy and from Veterinary Sciences will contribute 10,000 tons. A food pulper has been installed in one of the dining halls and will contribute approximately 440 tons. Mixed waste paper that does not have a market, landscape trimmings and used potting soil will also be composted. In addition, Washington State University power plant generates approximately 1,000 tons of coal ash annually. This paper discuss a pilot project to determine the feasibility of including coal ash in the compost mixture.  相似文献

To evaluate effects of previous forage systems on feedlot performance, yearling Hereford steers (average initial weight of 249 kg) were grazed on tall fescue (TF), smooth bromegrass-red clover (BG-RC) or orchardgrass-red clover (OG-RC) pastures before finishing. Serial slaughter was utilized during the first 2 yr of this study to determine changes in carcass characteristics throughout finishing, while steers were slaughtered at approximately 29% body fat during the third year. Steers grazing TF entered the feedlot at lighter weights and maintained lighter weights throughout finishing (P less than .05) even though dry matter intakes and feed conversions were similar (P greater than .05) among treatments. Steers that previously grazed TF had less (P less than .05) body fat, body protein, fat thickness and marbling, smaller (P less than .05) ribeye areas and lower (P less than .05) USDA yield and quality grades than than those that grazed BG-RC and OG-RC. However, linear contrasts indicated that steers grazing TF were compensating in ribeye area, marbling and quality grade as days in feedlot increased. This was confirmed in the third-year study, since carcass characteristics were similar among all steers. During the third year, linear and quadratic contrasts indicated that steers that grazed TF partially compensated in body protein. As days in feedlot increased, live and hot carcass weights, body fat, fat thickness, kidney, pelvic and heart fat, marbling, and USDA yield and quality grades increased (P less than .05), while ribeye area and body protein plateaued. Although carcass characteristics were similar among treatments, steers backgrounded on TF entered the feedlot at lighter weights, and partially compensated in weight after 134 d of finishing.  相似文献

Paclitaxel (Taxol) was administered to 25 dogs with histologically confirmed malignant tumors at a dosage of 165 mg/m2 i.v. over 3-6 hours every 3 weeks. Dogs received premedication with antihistimines and corticosteroids to reduce hypersensitivity reactions. However, 64% of the dogs still experienced allergic reactions. Six dogs (24%) had grade 3 or 4 neutropenia, 6 dogs (24%) required hospitalization and 3 dogs (12%) died of sepsis. Five dogs (20%) had a partial response (osteosarcoma [2 dogs] mammary carcinoma [2 dogs] and malignant histiocytosis [1 dog]) for a median duration of 53 days. The overall toxicity was unacceptable at the 165 mg/m2 dose. Therefore, subsequent evaluations of paclitaxel in tumor-bearing dogs should a starting dose of 132 mg/m2 i.v. every 3 weeks.  相似文献

The biomechanical contribution of the interfragmentary Kirschner wire as a component of composite fixation for acetabular fracture repair was subjectively and objectively evaluated. Acetabular osteotomies were repaired using the screw/wire/polymethylmethacrylate (SWP) composite fixation with or without one of three configurations of Kirschner wire in 32 hemipelves obtained from 16 dogs. Reduction, assessed objectively and subjectively, was unaffected by Kirschner wire placement. Hemipelves repaired with Kirschner wire(s) were subjectively more stable prior to application of polymethylmethacrylate when manually assessed in multiple planes. Consistent incremental increases in stiffness, yield load, and maximum load sustained that were observed during biomechanical testing were not significant, with the exception that hemipelves repaired with two Kirschner wires had significantly greater yield loads than hemipelves repaired without Kirschner wires. The subjective results of this study support the use of at least one interfragmentary Kirschner wire to maintain reduction prior to polymethylmethacrylate application; however, fracture configuration and location may dictate the number and pattern of interfragmentary Kirschner wires used to maintain reduction prior to application of the polymethylmethacrylate. The objective results and observations made during biomechanical testing suggest that use of one or more interfragmentary Kirschner wires may enhance stability after polymethylmethacrylate application.  相似文献

Soil acidity and low nitrogen (N) and phosphorus (P) availability are the most important constraints for common bean (Phaseolus vulgaris L.) production in highly weathered soils of the humid tropics. The objective of this study was to identify bean lines capable of producing higher yields as the result of greater N and P use efficiency under conditions of intermediate soil N and P availability. Selection for adaptation to low soil fertility can be used by plant breeding programs to improve bean seed yields. The performance of 33 bean lines and the varieties ‘Morales’ and ‘Verano’ from Puerto Rico and ‘Salagnac 90’ from Haiti were evaluated under three fertilization regimes in an Oxisol (very-fine, kaolinitic, isohyperthermic Typic Eutrustox): (i) 50 kg N/ha (+N), 57 kg P2O5/ha (+P), 54 kg K2O/ha (+K), (ii) −N, +P and +K, and (iii) +N, −P and +K in two field plantings of June 2007 and January 2008. The black bean line PR0443-151 and red bean line PR0340-3-31 had consistently higher seed yields under nutrient non-limiting conditions (+N +P +K). The low N tolerant lines and those that consistently had the highest N use efficiencies were PR0443-151, VAX 3, RBF 11-33 and RBF 19-63. Lines that were both efficient in N utilization and responsive to N application were PR0443-151, IBC 309-23, and A774. PR0340-3-31, PR0443-151, A774 and VAX 3 had the highest seed yields under soil P limiting conditions and expressed high soil P use efficiency. The identification of the bean lines with high relative yields under both nutrient limiting and non-limiting conditions, high nutrient use efficiency, and tolerance to low soil N and P, can help breeding programs and eventual sustainability of bean production in the Caribbean.  相似文献
