I ndole acetic acid (IAA) production from tryptophan by cell suspensions of Rhizobium trifolii, R. leguminosarum, R. phaseoli and R. lupini was studied in the presence or in the absence of 2-ketoglutaric acid. In R. lupini, production of IAA was strongly enhanced by the ketoacid, but in fast growing rhizobia it was less enhanced or unaffected. On the other hand, glutamic acid inhibits IAA production by R. meliloti, but stimulates IAA production in both R. leguminosarum and R. phaseoli. Refined wheat flours commercially produced by five different U.S. and Mexican wheat blends intended for tortilla production were tested for quality and then processed into tortillas through the hot‐press forming procedure. Tortilla‐making qualities of the flour samples were evaluated during dough handling, hot pressing, baking, and the first five days on the shelf at room temperature. The predominant variables that affected the flour tortilla performance were wet gluten content, alveograph W (220–303) and P/L (0.70–0.94) parameters, farinograph water absorption (57%) and stability (10.8–18.7 min), starch damage (5.43–6.71%), and size distribution curves (uniform particle distribution). Flours produced from a blend of Dark Northern Spring (80%) and Mexican Rayon (20%) wheat had the highest water absorption, and tortillas obtained from this blend showed the highest diameter and lowest thickness. The whitest and best textured tortillas were obtained from the flour milled from three hard types of Mexican wheat blend. A Mixolab profile was generated from the best tortilla flours, those produced by mills 3 and 4. The Mixolab profile showed that a good flour for hot‐press tortillas had a relatively lower absorption and short dough mix time compared with a bread flour and should have a significantly higher gluten compared with an all‐purpose flour. Compared with bread flour, the tortilla flour had higher retrogradation and viscosity values. The Mixolab profile proved to be a good preliminary test to evaluate flours for hot‐press tortillas. 相似文献

To determine whether populations of Phytophthora infestans attacking wild and cultivated potatoes in the highlands of Peru are specialized on their hosts of origin, we characterized
isolates using several neutral markers, metalaxyl resistance and for aggressiveness in a detached leaf assay. One hundred
and fifty-three isolates were collected from the northern and central highlands of Peru from different potato cultivars (both
modern and native cultivars) and from different species of wild, tuber-bearing potatoes. All the isolates analyzed belonged
to one of four clonal lineages that had been described previously in Peru: EC-1, US-1, PE-3 and PE-7. The EC-1 lineage (n = 133) was dominant and present in similar frequencies on wild and cultivated potatoes. PE-3 (n = 14) was found primarily on cultivated potatoes, with only one isolate coming from a wild host. US-1 (n = 2) and PE-7 (n = 4) were rare; all but one (PE-7) occurred on wild potatoes. Isolates from the EC-1 lineage from modern cultivars were compared
in three separate detached leaf inoculation assays with EC-1 isolates from the wild potato species S. sogarandinum, S. bill-hookerii or S. huancabambense, respectively. No significant interactions between isolate type (from wild or cultivated potato) and host type (wild or cultivated)
were measured for any assay. It appears that the pathogen genotypes in the EC-1 lineage indiscriminately attack both wild
and cultivated tuber-bearing solanaceous hosts in Peru, and breeders should be able to select for resistance using the common

It is a dogma, that RB51 vaccination does not induce antibodies that interfere with Brucellosis diagnosis, therefore any animal positive to serological test is considered as an infected animal. To determine protection against Brucellosis virulent field strain, 35 pregnant cows from a free-Brucellosis herd, previously vaccinated as calves with 1 x 10(10) CFU of RB51, were revaccinated with RB51 reduced dose, and then introduced into a herd with an active outbreak. Seventeen cows resulted positive in card test after revaccination. All 35 pregnant revaccinated cows had normal parturition; nevertheless, RB51 vaccine strain was isolated from milk and vaginal exudates from two cows after delivery at day 120 post-revaccination. At 150 days post-revaccination, two cows were positives to card and rivanol test and the field virulent strain was isolated. Revaccination with a reduced dose of RB51 in endemic zones did not cause abortion and protected 94% of animals against field infection, but caused an atypical response to conventional serological tests. 相似文献

The aim of this work was to investigate the secretion of dehydroepiandrosterone (DHEA), testosterone (T), dihydrotestosterone (DHT) and oestradiol (E) as biological markers in response to illegal administration of testosterone, 19-nortestosterone (N) and oestradiol, either alone or in combination. Twenty male Friesian calves (age 13-14 months) were allotted to a control group (n = 5), and five experimental groups (n = 3) each. Each experimental animal was repeatedly injected with one of the following hormonal treatments: E, T, N, T+E and N+E. Circulating DHEA, T, DHT and E were determined by radioimmunoassay. The administration of T alone did not induce any variation in plasma DHEA, T, DHT and E, which were similar to those in the control group. In contrast, DHEA, T and DHT were on average significantly lower in the T+E and N-treated groups (p < 0.01), whereas the administration of N+E resulted in the reduction of plasma T and DHT without any modification of plasma DHEA. The administration of E alone or in combination increased circulating levels of E but did not affect androgen plasma profiles. The results indicate that plasma levels of T do not permit detection of illegal treatments because plasma androgens always remained within the physiological range. Illegal E treatment could be detected in blood samples when they were collected at least every 20 days. 相似文献

Small ruminant lentiviruses (SRLV = maedi-visna in sheep and caprine arthritis encephalitis in goats) are distributed throughout most countries of the world, particularly Europe. Laboratories from 16 European countries established collaborations within the framework of a COST (CO-operation in the field of Scientific and Technical Research) action sponsored by the European Union in order to (i) better organize their research programmes on SRLVs and (ii) to coordinate efforts to combat these two diseases. After five years, a consensus conference--the first one in the veterinary medicine field--concluded the work of this network of laboratories by reviewing the present position and discussing three important questions in the field of SRLVs: routes of transmission, consequences of infection and potential role of eradication programmes at either a European or local level, according to the situation in each country or region. The recovery of tartaric acid (TA) from distilled vinification lees coming from the white and red winemaking technology was optimized using response surface methodology and Statistica 5.0 software. The sequential treatment of dissolving TA and further calcium tartrate (CaT) precipitation could be used to recover up to 92.4% of the initial TA when distilled white lees were used. The residual lees were employed as economic nutrients for lactic acid production by Lactobacillus pentosus CECT-4023 using hemicellulosic vine shoot hydrolysates as carbon source. Distilled lees after TA extraction used as nutrients provided values of lactic acid (18.4-18.9 g/L), global volumetric productivities (0.82-0.84 g/L.h), and product yields (0.69-0.70 g/g) similar to those achieved when using the general medium for Lactobacilli (18.6 g/L, 1.11 g/L.h, and 0.62 g/g, respectively) or lees without TA extraction (16.4-17.2 g/L, 0.96-1.21 g/L.h, and 0.61-0.66 g/g, respectively). The in vitro antimicrobial activity of commercial coffee extracts and chemical compounds was investigated on nine strains of enterobacteria. The antimicrobial activity investigated by the disc diffusion method was observed in both the extracts and tested chemical compounds. Even though pH, color, and the contents of trigonelline, caffeine, and chlorogenic acids differed significantly among the coffee extracts, no significant differences were observed in their antimicrobial activity. Caffeic acid and trigonelline showed similar inhibitory effect against the growth of the microorganisms. Caffeine, chlorogenic acid, and protocatechuic acid showed particularly strong effect against Serratia marcescens and Enterobacter cloacae. The IC(50) and IC(90) for the compounds determined by the microtiter plate method indicated that trigonelline, caffeine, and protocatechuic acids are potential natural antimicrobial agents against Salmonella enterica. The concentrations of caffeine found in coffee extracts are enough to warrant 50% of the antimicrobial effect against S. enterica, which is relevant to human safety. 相似文献
