An experiment was carried out to determine how the late application of nitrogen (N) fertilizer affects the use of pre‐anthesis carbon reserves during the grain‐filling period of pot‐grown wheat with no water shortage. Increasing doses (equivalent to 0, 140, and 180 kg N ha–1) of N fertilizer were applied, either in two amendments (stages GS20 and GS30) or in three amendments (stages GS20, GS30, and GS37, according to Zadoks scale). The management of fertilizer by combining an increased N rate with late N application was able to stimulate canopy development, to raise photosynthetic capacity and carbohydrate accumulation during the vegetative stages, and to increase grain yield. Based on the dynamics of carbohydrate accumulation in the ear, three phases were differentiated during the grain‐filling period, whose temporary pattern remained stable regardless of the fertilizer management. The net remobilization of carbohydrates started 12 d after anthesis from the leaves and 28 d after anthesis from roots and stems. The increase of the N dose with late N application allowed on one hand a lower use of the pre‐anthesis carbon reserves in favor of greater de novo photosynthesis during the grain‐filling period, and on the other hand greater relative contribution of the leaf and ear C reserves to remobilization towards the grain. Further splitting the dose increased only the relative contribution of ear C reserves. Water deficit is a serious problem for most agricultural crops, especially in arid–semiarid regions, and limits sustainable development. Production can be improved by reducing evapotranspiration and loss of infiltrated water by the use of a plastic cover. We monitored soil moisture fortnightly over 1 yr using a neutron probe near four trees in an olive grove (Olea europaea, var. Arbequina), two of which had a plastic cover buried in the topsoil around them. These trees were monitored using three neutron probe access tubes per tree to compare the behaviour of soil moisture over time with two other identically instrumented and nearby trees with no cover. Analysis was based on the resultant moisture profiles. The plastic cover retained moisture and increased soil water residence time. During the dry season, the amount of water retained in the plots was at most 15–20% greater in the mid‐season and at least 5–6% greater at the end of season than in the central part of the plots near the trunk. The plastic cover was effective to ca. 50 cm with maximum water content near the soil surface. During the wet season, the cover did not affect soil water. Soil moisture was greater near the tree trunk as a result of stemflow and throughfall. 相似文献

The burrowing nematode, Radopholus similis (Cobb.) Thorne, causes the most damage to bananas. To minimize nematicide applications, cropping systems that use fallow, crop rotation and clean planting material have been developed in the French West Indies. In order to optimize the benefit of the intercropping period, we studied the survivorship of R. similis in different soil types and conditions. We monitored the survivorship of calibrated populations of R. similis in the laboratory on a Nitisol and on an Andosol, two soils derived from volcanic ashes and pumices. We studied water potentials ranging from 0 to ?700 kPa on undisturbed soil and on soil previously frozen to get rid of living nematodes. Mortality of adult R. similis decreased regularly, and was fairly well described by Teissier's model. In the previously frozen soils, R. similis survived longer in wet soils (half-life of 21–46 days at 0 to ?5 kPa) than in dry soils (half-life of less than 10 days between ?80 and ?250 kPa). In contrast, in undisturbed soils, R. similis survived longer in dry soils: half-lives ranged from 57 days at ?273 kPa to 17 days at water saturation in the Andosol, and 36 days at ?660 kPa to 14 days at water saturation in the Nitisol. These results are consistent with the absence of anhydrobiosis in R. similis, unlike Pratylenchus coffeae. P. coffeae survivorship curves over time do not follow a model derived from exponential decrease like Teissier's model. These results also show that the recommended one year host-free period required to sanitize soils cannot be shortened without risk, even if flooding the soil could improve it. 相似文献

The impact of detritivorous earthworms (Eisenia andrei) on microbial community structure and function in grape marc, a lignocellulosic enriched plant residue, was investigated in a mesocosm experiment. Analysis of carbon and nitrogen pools was also carried out in order to evaluate how changes in microbial communities affect plant residue decomposition. The grape marc was completely processed after fifteen days as a result of the high density of earthworms present and the rapid gut transit time. Eisenia andrei had a large impact on the structure of the microbial community, as revealed by phospholipid fatty acid analysis. Earthworm activity reduced the abundance of both bacterial (except for Gram-negative bacteria) and fungal PLFA biomarkers relative to the control values. Decreases in microbial activity and in protease and cellulase activities were also attributable to the presence of earthworms. Moreover, earthworms strongly modified the substrate utilization patterns of microbial communities, as revealed by BIOLOG analysis. The presence of earthworms led to an increase in the utilization of some amino acids and polymers, which reached a higher substrate diversity value than that in the control mesocosm. The differences in microbial communities were accompanied by a reduction in the total C content and the labile C pool, relative to the control, although there were no significant differences in either cellulose or hemicellulose contents. However, total N content increased in both mesocosms – with and without earthworms – and the concentration of NH4+ was also enhanced by earthworm activity. The results indicate that detritivorous earthworms play a key role in decomposing fresh plant residues in the short term via their intensive interactions with microbial communities. 相似文献

The impacts of tillage and cropping sequences on soil organic matter and nutrients have been frequently reported to affect the uppermost soil layers, but there is little published information concerning effects at greater depth. This article reports results on the distribution of soil organic carbon (SOC), active carbon (AC), N, Olsen‐P and extractable K within 100 cm in short (4 yr) and long (16 yr) term experiments under different tillage systems. Short (TT4) and long (TT16) traditional tillage are compared with conservation tillage, reduced (RT16) and non‐tillage (NT4). The results show more accumulation of SOC in the near‐surface under RT16 and NT4 in both experiments compared with traditional tillage. Moreover, greater C content occurs to 40 cm depth in the long‐term experiment. The results demonstrate the importance of time on C accumulation, not only in near‐surface layers but also at greater depths. Active C is an indicator of the increase in soil quality in the long‐term experiment. This trend is only apparent for the first 10 cm in the short‐term experiment. Patterns in N, Olsen‐P and extractable K are similar to that of SOC. However, only extractable K is significantly greater in soil under conservation tillage (RT16 and NT4) after short and long periods. Potassium availability is a good indicator of the changes caused by tillage. Our results indicate that studies of soils at depth could be very useful in long‐term experiments to demonstrate the effect of conservation tillage on C and nutrient distribution. 相似文献

Rationally planned structural modifications were carried out on benzylphenols and benzyl-1,3-benzodioxoles described as fly chemosterilants and as anti-juvenile hormones. The introduction of a prop-2-ynyloxy group at various sites of the molecule resulted in compounds with a moderate inhibitory action on cytochrome P-450 mono-oxygenases, as measured by aldrin epoxidation. One compound, 5-(4-methoxybenzyl)-6-prop-2-ynyloxy-1,3-benzodioxole, revealed chemosterilant activity on Phormia regina, but its activity was less than that of the parent compounds. 2,4-Di-tert-butyl-6-[4-(3-methoxy-3-methylbutoxy)benzyl]phenol, which possessed a juvenoid structure, revealed no juvenile hormone (JH) activity but showed a high sterilant effect against Dysdercus cingulatus. In contrast to the parent substances, none of the tested compounds showed a detectable anti-JH effect in the Galleria assay. 8-Methoxy-2,3-methylenedioxydibenz[b,e]oxepine, a hitherto undescribed fused heterocyclic ring system, was devoid of activity, indicating the importance of free rotation and/or molecular flexibility. In spite of the moderate activities of these compounds, the manifold biological potential of the quinone-methide mechanism justifies further research on these lines. 相似文献

OBJECTIVE: To determine whether nonthyroidal disease of various causes and severity is associated with abnormalities in baseline serum concentrations of total thyroxine (T4), triiodothyronine (T3), free T4, or thyrotropin (thyroid-stimulating hormone [TSH]) in dogs believed to be euthyroid. DESIGN: Case-control study. ANIMALS: 223 dogs with confirmed nonthyroidal diseases and presumptive normal thyroid function, and 150 clinically normal dogs. PROCEDURE: Serum total T4, total T3, free T4, and TSH concentrations were measured in dogs with confirmed nonthyroidal disease. Reference ranges for hormone concentrations were established on the basis of results from 150 clinically normal dogs. RESULTS: In dogs with nonthyroidal disease, median serum concentrations of total T4, total T3, and free T4 were significantly lower than those in clinically normal dogs. Median serum TSH concentration in sick dogs was significantly greater than that of clinically normal dogs. When stratified by severity of disease (ie, mild, moderate, and severe), dogs with severe disease had low serum concentrations of total T4, total T3, or free T4 more commonly than did dogs with mild disease. In contrast, serum TSH concentrations were more likely to remain within the reference range regardless of severity of disease. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicate that serum total T4, free T4, and total T3 concentrations may be low (ie, in the hypothyroid range) in dogs with moderate to severe nonthyroidal disease. Serum TSH concentrations are more likely to remain within the reference range in sick dogs. 相似文献

The aim of this study was to determine whether the sexual activity of local male Creole goats in subtropical Mexico can be induced during the non-breeding season by a long-day treatment followed by insertion of two melatonin implants. The experiment was carried out in the Laguna region in the State of Coahuila, Mexico (26 degrees N). Fourteen male goats were allocated to two balanced groups (n = 7 each) according to body and testicular weights. Males were kept together in two separate groups and fed lucerne hay for ad libitum intake and 300 g of commercial concentrate and had free access to water and mineral blocks. The control group remained in open sheds under natural photoperiod and ambient temperature conditions. The experimental group was placed in a light-proof building and exposed to 2.5 mo of long days (16 h of light/d) from November 1 to January 15. On January 16, each male received two s.c. melatonin implants and was exposed to natural photoperiodic changes in an open shed. In the control group, testicular weight exhibited seasonal variations; the highest value occurred on May 30 (146 +/- 10 g). Treated males reached maximum testicular weight earlier (March 15; 147 +/- 11 g), and sperm quality from January to March was higher than that observed in the control group (P < 0.05). Treatment caused an increase in LH pulse frequency (2.0 +/- 0.5 vs 0.3 +/- 0.2 pulse/8 h in February, 4.6 +/- 1.1 vs 0.1 +/- 0.1 pulse/8 h in March; experimental vs control group, respectively). In the control group, plasma testosterone remained low until mid-June and increased thereafter to remain elevated until the end of the study. In the experimental group, elevated plasma testosterone was observed from February to April and from July to November. Treating male goats in subtropical latitudes with artificial long days and melatonin can induce an intense sexual activity during the natural nonbreeding season. 相似文献

Nucleic acid (DNA) vaccination against tuberculosis in the European badger (Meles meles) is one approach to addressing the escalating problem of bovine tuberculosis in Great Britain. The aim of vaccination is to reduce the burden of tuberculosis within the badger population and the shedding of Mycobacterium bovis to levels that would break the transmission of infection to cattle. To this end, the vaccine would be required to limit the amount of disseminated tuberculosis in the badger, especially dissemination to the kidney from where M. bovis can be shed in the urine. A promising candidate DNA vaccine encoding a 26 kDa major antigen (MPB83) of M. bovis was evaluated in a mouse model of disseminated M. bovis infection. Using the DNA vaccine, protection against infection of the kidney was found to be greater than that achieved with the current live vaccine, Bacille Calmette-Guerin (BCG). Kidney tissue and skeletal muscle from the badger was used to derive primary cell cultures in which to examine the expression of MPB83 following transfection with the DNA vaccine. Kidney cortex gave rise to a monotypic culture of epithelial cells whilst the muscle gave rise to a mixed culture of fibroblasts and myoblasts. During culture the myoblasts differentiated into multinucleated myotubes, verified by immunofluorescent detection of mammalian desmin. Successful expression of MPB83 by transfected epithelial and myotube cells was confirmed by immunofluorescence using a monoclonal antibody specific to the protein. These observations fulfil the early requirements for the development of a DNA vaccine for badger tuberculosis. 相似文献
