Six German shorthaired pointer dogs (two females, four males) with exfoliative cutaneous lupus erythematosus (ECLE) were studied in a controlled setting until disease progression necessitated euthanasia. During investigations into the heredity of disease, five dogs received immunomodulatory drugs to alleviate clinical signs (lameness, erythema, scaling, erosions/ulcers). One dog served as a control and received only baths and oral fatty acids. Four dogs received ciclosporin (5–10 mg/kg once daily) for 4.5 months to 2 years. Ciclosporin decreased erythema and arthralgia, but did not halt worsening of lesions. Three dogs received hydroxychloroquine (5–10 mg/kg once daily) for 8 weeks, 7 months, and 9 months, respectively, with no side effects. Hydroxychloroquine appeared to slow clinical progression in two dogs on extended treatment and normalized globulin levels in all three dogs while receiving the drug. Four dogs, including the control dog, were euthanized between 1 and 4.5 years of age. Two remaining male dogs received a tumour necrosis factor (TNF)‐α antagonist, adalimumab, at 0.5 mg/kg every 2 weeks for 8 weeks then weekly for 8 weeks. Serum TNF‐α levels were not significantly altered nor were quantifiable changes seen in skin lesions or lameness. Subsequently, the dogs were maintained on hydroxychloroquine for another year. This is the first study to evaluate the use of a TNF‐α inhibitor for canine lupus and the first to address the safety of long‐term administration of hydroxychloroquine, albeit in a small number of dogs. The study documents the progression of ECLE and generally poor response to therapy.  相似文献

A newly-formulated, high protein high fibre (HPHF) diet has recently been shown to improve satiety in dogs. The current study examined its performance during weight loss in client-owned dogs with naturally-occurring obesity. Fifteen dogs were fed the HPHF diet, whilst a matched ‘control’ group of 27 dogs, received a high protein medium fibre diet (HPMF), with an equivalent caloric density. Baseline characteristics (signalment, percentage overweight, and body fat percentage) were not significantly different between groups. However, percentage weight loss was greater (median [range] 31.8% [12.0–41.2%] vs. 20.0% [5.9–45.0%], P = 0.016) and mean rate of weight loss faster (median [range] 1.0%/week [0.3–1.6%] vs. 0.7%/week [0.3–1.5%], P = 0.028) on HPHF compared with HPMF. Percentage body fat mass decrease (measured by dual-energy X-ray absorptiometry) was also greater in dogs fed the HPHF diet (median (range] 58% [32–85%) vs. 37% [15–72%), P = 0.002). Thus, a diet formulated to include high levels of both protein and fibre, improves outcome during weight loss in obese dogs.  相似文献

The partition of dry matter between leaf and bulb with sheath was studied in relation to the post-harvest physiology of cvs Bola and Robusta grown at three sites for two seasons and dried and stored under standardized conditions. Foliar fall-over was synchronous at all sites in each season but the effects of site showed that crops either partitioned dry matter to the bulb early, lost less fresh weight in drying and sprouted later in store, or partitioned dry matter late, lost more fresh weight in drying and sprouted sooner in store. Crops showing the former behaviour were associated with low rainfall and a high number of day degrees between 6° and 20°C during early growth. There was up to 10 days difference between the sites in 50% sprouting dates. Bulbs of cv Bola sprouted earlier than those of cv Robusta, and bulbs harvested when foliar fall-over was negligible sprouted before those from later harvests. There were no site effects on plant maturity, on bulb growth regulator content (cv Robusta one year only), on sugar content at harvest or during storage, on bulb respiration rates at the end of drying or in the spring, or on bulb weight loss during storage.  相似文献

Summary

The effect of high temperature stress (27ºC or 37ºC for 24 h) on total gene expression profiles in the annual-fruiting raspberry (Rubus idaeus L.) cultivars ‘Autumn Bliss’, ‘Autumn Treasure’, ‘Erika’, and ‘Polka’ were evaluated at the floral initiation stage using a customised Rubus microarray. Significantly affected genes were obtained by pairwise t-tests using ‘volcano plots’ for each cultivar × treatment. A 10ºC elevation in temperature altered levels of expression, in at least one cultivar, of 644 differentially expressed genes in total, with ‘Erika’ and ‘Autumn Treasure’ showing elevated expression of 38 genes compared to ‘Autumn Bliss’ and ‘Polka’. We identified 12 common candidate genes that were modulated differentially in ‘Autumn Bliss’ and ‘Erika’ at 37ºC compared to 27ºC. In addition, two aquaporin genes (PIP1 and TIP2) were down-regulated in ‘Autumn Bliss’, but up-regulated in ‘Autumn Treasure’, ‘Polka’, and ‘Erika’ at 37ºC. Other down-regulated genes from the list of 38 genes included those encoding major latex-like proteins, plasma membrane proteins, cysteine rich proteins, and other stress-related proteins. Validation by real-time quantitative RT-PCR (RT-qPCR) indicated subtle changes in differential gene expression, suggesting a mild response to heat stress. This study used molecular tools to increase our understanding of, and to identify candidate genes involved in, the heat stress response of four annual-fruiting raspberry cultivars.  相似文献

Many pet cats and dogs are fed dry extruded kibbled food by measuring cup, yet the precision and accuracy of this feeding strategy is not known. Over 12 studies, we assessed precision and accuracy of weighing out food portions, of various dry kibbled foods, by measuring cup. Poor precision was noted in all studies, with intra- and inter-subject coefficients of variation ranging from 2 to 13% and 2 to 28% respectively. Variable accuracy was also noted, which ranged from an 18% under-estimate to an 80% over-estimate in portion size. No specific factors were associated with imprecision, but the degree of inaccuracy was negatively associated with portion size (R = -0.67, p = 0.022), and positively associated with the number of subjects participating in the study (R = 0.60, p = 0.048). This is the first study to document imprecision and inaccuracy of using measuring cups to estimate portions of extruded dry kibbled food. Over time, such errors could contribute to insidious weight gain in companion animals, potentially contributing to the development of obesity. Imprecision in measuring food portions could also contribute to failure of weight management programmes for obese animals.  相似文献

Clusterin (CLU), also known as apolipoprotein J, is a widely expressed, heterodimeric, glycoprotein, important in tumourigenesis, apoptosis and immunoregulation. In humans, CLU expression has been associated with anaplastic large cell and Hodgkin's lymphoma. In this study, serum CLU levels in dogs with multicentric lymphoma (MLSA) were compared with healthy control dogs, using both western blot and enzyme‐linked immunosorbent assay (ELISA). Western blot confirmed the presence of CLU in dog sera at the predicted molecular weight and the relative levels detected correlated with the levels detected by ELISA. CLU level analysis by ELISA found treatment naïve dogs with MLSA had a significantly (P < .001) lower serum CLU level compared with healthy controls. However, there was no significant difference between MLSA dogs prior to treatment and in complete remission. The wide variation in serum CLU levels may limit its potential as a single candidate biomarker for MLSA, although any prognostic predictive value of serum CLU concentrations has yet to be assessed.  相似文献
