Seven FDA pesticide laboratories collaborated to develop and validate an LC-MS/MS method to determine 173 pesticides in <20 min. The average determination coefficient (r2) was >0.99 for all but two compounds tested. The limits of detection were <20 ng/mL for all compounds and <10 ng/mL for 363 of the 368 transitions reported. The method was used to determine pesticides in two AOAC sponsored proficiency samples. The LC-MS/MS determination was used for the analysis of oranges, carrots and spinach using the QuEChERS (Quick, Easy, Cheap, Effective, Rugged, Safe) method. Each matrix was fortified at 20, 100, 400, and 1000 ng/g. No false positive responses were detected in controls of the three matrices. 165 pesticides had recoveries between 70 and 130%, and 161 had minimum detection levels less than 10 ng/g. Recoveries of 169 compounds for the 1000 ng/g spikes were within 50-150%. A matrix effect study indicated all three matrices caused a small net suppressing effect, the most pronounced attributable to the citrus matrix. The procedure proved to be accurate, precise, linear, sensitive and rugged, and adds 100 pesticides to the scope of the FDA pesticide program. 相似文献

The objective of this study was to understand the effects of fumarate addition on methane (CH4) and VFA production in the rumen through a meta-analysis of its effects on ruminal batch cultures. Because the reduction of fumarate to succinate can draw electrons away from ruminal methanogenesis, fumarate has been studied as a potential feed additive to decrease CH4 production in ruminants. The average decrease in CH4 in batch cultures was of 0.037 micromol/micromol of added fumarate, which is considerably lower than 0.25 micromol/micromol, the decrease predicted from the stoichiometry of the reactions involved. One reason that fumarate was not effective at decreasing CH4 in batch cultures was that only an average of 48% of added fumarate appeared to be converted to propionate. Secondly, the incorporation of reducing equivalents in the conversion of fumarate to propionate was almost entirely offset by their release from an average of 20% of added fumarate that appeared to be converted to acetate. Thermodynamic calculations indicated that the conversion of added fumarate to both propionate and acetate was feasible. Fumarate appears to be more effective in decreasing CH4 production and increasing propionate in continuous culture than in batch culture. This suggests that microbial adaptation to fumarate metabolism can be important. Variation in populations of fumarate-reducers, methanogens, and protozoa could all be involved. Fumarate supplementation for an extended period may result in the amplification of otherwise small populations of fumarate-reducers. Addition of some of these organisms may be helpful to improve fumarate conversion to propionate. Strategies based on enhancing the rumen's capacity to convert fumarate to propionate by maintaining a low fumarate concentration have been effective. Thermodynamic considerations should be taken into account when designing strategies for CH4 abatement through the addition of external electron acceptors. 相似文献

The cost-effectiveness of adopting agri-environmental measures (AEMs) in Europe, which combine agricultural productions with reduced N losses, is debated due to poorly targeted site-specific funding that is allocated regardless of local variability. An integrated DAYCENT model-GIS platform was developed combining pedo-climatic and agricultural systems information. The aim was to evaluate best strategies to improve N fluxes of agro-ecosystems within a perspective of sustainable intensification. Indicators of agronomic efficiency and environmental quality were considered. The results showed that agronomic benefits were observed with a continuous soil cover (conservation agriculture and cover crops), which enhanced nitrogen use efficiency (+17%) and crop yields (+34%), although in some cases these might be overestimated due to modelling limitations. An overall environmental improvement was found with continuous soil cover and long-term change from mineral to organic inputs (NLeach < 10 kg ha?1 a?1, N-N2O emissions < 1 kg ha?1 a?1, soil C stock > 45 Mg ha?1), which were effective in the sandy soils of western and eastern Veneto with low SOM, improving the soil-water balance and nutrients availability over time. Present acid forming emissions to the atmosphere have the potential to alter significantly the chemistry of rain, snow, and surface water of weakly buffered lakes in the Upper Midwest. Average precipitation pH from field measurements during 1979–1983 declined from west to east from 4.8, 4.6, and 4.3 along a cross-section of sites in Minnesota, Wisconsin, and Michigan respectively where 990 lake and stream sampling sites were studied. Measurements of weakly buffered lakes show a parallel decline in lake water pH with the lowest values measured, 5.1, 4.6 and 4.4, respectively in the same regions. Correspondingly, the percentage of lakes sampled with little or no acid neutralizing capacity (ANC) was found to increase from 0 to 4 and 13%, respectively. The geographic patterns in ionic composition of airborne acids and bases, and the resultant surface water concentrations are compared. The acid forming capacity (AFC) from airborne inputs is calculated using mass balance and in-lake processes. Stoichiometric acid-base reactions are used to balance the observed chemical differences between airborne inputs and surface water composition considering nitrification, denitrification, other oxidation-reduction reactions, and the evaporation concentration process. Microbial activity in surface water can result in a net decrease in ionic strength from the conversion of most of the ammonium and nitrate to neutral compounds and biomass, but only a partial reduction of about 20% of the sulfate inputs to weakly buffered lakes. The resulting AFC of airborne inputs are calculated to range from 30 to 60, 50 to 90, and 80 to 130 μeq H+L-l, respectively, in northeastern Minnesota-Ontario, northcentral Wisconsin and northern Michigan-Ontario. The differences in AFC of airborne inputs from west to east, and differences in in-lake processes explain the observed acidity of weakly buffered lakes across the region. 相似文献

Fourier transform infrared spectroscopy (FT-IR, 4000-600 cm(-)(1)) was used to discriminate between intact and sonication-injured Listeria monocytogenes ATCC 19114 and to distinguish this strain from other selected Listeria strains (L. innocua ATCC 51742, L. innocua ATCC 33090, and L. monocytogenes ATCC 7644). FT-IR vibrational overtone and combination bands from mid-IR active components of intact and injured bacterial cells produced distinctive "fingerprints" at wavenumbers between 1500 and 800 cm(-)(1). Spectral data were analyzed by principal component analysis. Clear segregations of different intact and injured strains of Listeria were observed, suggesting that FT-IR can detect biochemical differences between intact and injured bacterial cells. This technique may provide a tool for the rapid assessment of cell viability and thereby the control of foodborne pathogens. 相似文献

Ethiopia has a large and diverse population of small ruminants, which contribute substantially to the livelihood and income of the rural poor and the country at large. However, the sector faces a number of challenges. Productivity per animal and flock offtake are both very low. Reasons attributed for the apparent low productivity are as follows: absence of appropriate breeding programmes, lack of technical capacity, inadequate and poor‐quality feeds, diseases leading to high lamb mortality, and underdeveloped markets in terms of infrastructure and information. Historically, sheep and goats have received little policy or investment attention. Genetic improvement of small ruminants could contribute to bridging the productivity gap. In the past, the government of Ethiopia has placed much emphasis on importing exotic genetics and cross‐breeding with local stock as a strategy for genetic improvement. However, this has not led to a significant productivity improvement and the programmes have generally been unsustainable. Currently, there is a change in approach and a recognition of the need to focus genetic improvement efforts on the local genetic resources that are well adapted to the diverse agro‐ecologies and production environments in the country. Community‐based breeding programmes (CBBPs), which focus on indigenous stock and consider farmers’ needs, views, decisions and active participation, from inception through to implementation, have been identified as programmes of choice. The Ethiopian government and the private sector need to invest in strategic areas around CBBPs to make the programme work for the poor and be sustainable in low‐input systems. 相似文献
