OBJECTIVE: To characterize generation of reactive oxygen species (ROS) by equine spermatozoa. SAMPLE POPULATION: Multiple semen samples collected from 9 stallions. PROCEDURE: Equine spermatozoa were separated from seminal plasma on a discontinuous polyvinylpyrrolidone (PVP)-coated silica gradient and resuspended in a modified Tyrode albumin-lactate-pyruvate medium. Amount of hydrogen peroxide (H2O2) generated was assayed by use of a 1-step fluorometric assay, using 10-acetyl-3,7-dihydroxyphenoxazine as a probe for detection of H2O2 in a microplate assay format. Concentration of H2O2 was determined by use of a fluorescence microplate reader. RESULTS: Amount of H2O2 generated increased significantly with time and spermatozoa concentration for live and flash-frozen spermatozoa, and amount of H2O2 generated was significantly greater for flash-frozen than for live spermatozoa. Addition of the reduced form of nicotinamide adenine dinucleotide phosphate (NADPH) significantly increased generation of H2O2 by live and flash-frozen spermatozoa. Addition of a calcium ionophore also significantly increased the amount of H2O2 generated by live spermatozoa but did not have an effect on amount of H2O2 generated by flash-frozen spermatozoa. Abnormal equine spermatozoa generated significantly greater amounts of H2O2 than did normal spermatozoa. CONCLUSIONS AND CLINICAL RELEVANCE: Equine spermatozoa generate ROS in vitro, possibly via a NADPH-oxidase reaction. Spermatozoa damaged during flash-freezing or morphologically abnormal spermatozoa generated significantly greater amounts of ROS than did live or morphologically normal spermatozoa. Damaged and abnormal spermatozoa generate greater amounts of ROS that may contribute to reduced fertility or problems related to semen preservation.  相似文献

Sharp peaks in nitrous oxide (N2O) fluxes under no-tillage in wet conditions appear to be related to near surface soil and crop cover conditions. Here we explored some of the factors influencing tillage effects on short-term variations in gas flux so that we could learn about the mechanisms involved. Field investigations revealed that a cumulative emission of 13 kg N2O–N ha−1 over a 12-week period was possible under no-tillage for spring barley. We investigated how reducing crop cover and changing the structural arrangement of the water-filled pore space (WFPS) by short-term laboratory compaction influenced N2O and carbon dioxide (CO2) fluxes in upward and downward directions in core samples from tilled and untilled soil. Increasing the downward flux of N2O within a soil profile by changing soil or moisture conditions may increase the likelihood of its further reduction to N2 or dissolution. We took undisturbed cores from 3 to 8 cm depth, equilibrated them to −1 or −6 kPa matric potential, incubated them and measured N2O and CO2 fluxes from the upper and lower surfaces in a purpose-designed apparatus before and after compaction in an uniaxial tester. We also measured WFPS, air permeability, bulk density and air-filled porosity before and after compaction. Spring barley was tested in 1999 and winter barley in 2000.Fluxes of N2O were from 1.5 to 35 times higher from no-tilled than ploughed even where the soil was of similar bulk density. Reduction of the crop cover increased CO2 flux and could reduce N2O flux. The effects of structural changes induced by laboratory compaction on the fluxes of N2O and CO2 were not influenced greatly by the tillage and crop cover treatments. Fluxes from the upper surfaces of cores (corresponding to 3 cm soil depth, upwards direction) could be up to 100 times greater (N2O) or 8 times (CO2) than from the lower surfaces (8 cm depth, downwards direction). These differences between surfaces were greatest when N2O fluxes were very high in no-tilled soil (4.2 mg N2O–N m−2 h−1) as occurred when WFPS exceeded 80% or became blocked with water, an effect that was increased by our compaction treatment. In general N2O fluxes increased with WFPS. The production and emission of N2O were strongly influenced by the soil physical environment, the magnitude of the water-filled pore space and continuity of the air-filled pore space in particular, produced in no-till versus plough cultivation.  相似文献

The effects of tillage on soil organic carbon content, carbohydrate content, monosaccharide composition, aggregate stability, compactibility and plasticity were investigated in a field experiment on a gleysol and on a cambisol under winter barley in South-East Scotland. Two long-term treatments (direct drilling and conventional mouldboard ploughing for 22 years) were compared with short-term direct drilling and broadcast sowing plus rotavation for 5 years. Carbohydrate released sequentially to cold water, hot water, 1.0 M HCl and 0.5 M NaOH was determined after hydrolysis as reducing sugar equivalent to glucose in both fresh and air-dried samples. All other measurements were made on dry soils only. About 3% of the soluble carbohydrate was extracted by cold water, 10% by hot water, 12% by HCl and 75% by NaOH from both the dry and fresh soils. The total reducing sugars of the fractions were proportional to the total organic carbon determined by dichromate oxidation or C analysis. Organic carbon and carbohydrates were concentrated near the surface of the direct drilled soil, but were more uniformly distributed with depth in the ploughed soil. The surface soil under direct drilling was more stable, less compactible and had greater plasticity limits than under ploughing. However, particle size distributions were unaffected by tillage so that differences in soil properties were attributed to differences in the quantity and quality of organic matter. Differences in compactibility, structural stability and plasticity limits between depths and tillage treatments correlated with total carbon and with total carbohydrates. The hot water extractable carbohydrate fraction correlated best with aggregate stability and the NaOH fraction correlated best with compactibility and plastic limit. Both fractions were greatest in the long-term direct drilled soil. The hot water fraction had a galactose plus mannose over arabinose plus xylose ratio of 1.0–1.6 in comparison to 0.4–0.7 in the NaOH fraction indicating that the microbial contribution within the hot water-soluble fraction was the greater. The hot-water fraction was likely to contain more exocellular microbial polysaccharides involved in the stabilizing of soil aggregates. The hot-water and NaOH carbohydrate fractions may be good indicators of soil organic matter quality relevant to the preservation of good soil physical conditions.  相似文献

Because of its requirement for signaling by multiple cytokines, Janus kinase 3 (JAK3) is an excellent target for clinical immunosuppression. We report the development of a specific, orally active inhibitor of JAK3, CP-690,550, that significantly prolonged survival in a murine model of heart transplantation and in cynomolgus monkeys receiving kidney transplants. CP-690,550 treatment was not associated with hypertension, hyperlipidemia, or lymphoproliferative disease. On the basis of these preclinical results, we believe JAK3 blockade by CP-690,550 has potential for therapeutically desirable immunosuppression in human organ transplantation and in other clinical settings.  相似文献

Effects of the endophyte Acremonium coenophialum in tall fescue on pregnant mares and foal viability were evaluated. Twenty-two mature pregnant mares were randomly chosen to graze either Kentucky-31 tall fescue that was free from A coenophialum (endophyte-free, EF) or tall fescue infected with A coenophialum (endophyte-present, EP) after the first 90 days of pregnancy through parturition. Concentrations of pyrrolizidine and ergopeptine alkaloids were significantly greater in EP grass, compared with EF pasture. Ten of 11 mares grazing EP pasture had obvious dystocia. Mean duration of gestation was significantly greater for the EP group, compared with the EF group. Foal survivability was severely reduced among mares grazing EP fescue with only 1 foal surviving the natal period. Udder development and lactation were low in mares grazing EP grass. The absence of clinical problems in mares grazing EF grass implicated the endophyte as the causative agent of reproductive problems and perinatal foal mortality in pregnant mares grazing endophyte-infected fescue grass. Caution should be exercised in allowing pregnant mares to graze pastures infected with the endophyte A coenophialum.  相似文献

After the observation of 2 horses with uveitis on a horse farm in the Minnesota River valley, 100 horses from this geographic area were given ophthalmologic examinations and were evaluated serologically for leptospirosis. A statistically significant (P less than 0.001) association was observed between the finding of antibodies against Leptospira interrogans serovar pomona and uveitis.  相似文献

Yersinia species use a variety of type III effector proteins to target eukaryotic signaling systems. The effector YopJ inhibits mitogen-activated protein kinase (MAPK) and the nuclear factor kappaB (NFkappaB) signaling pathways used in innate immune response by preventing activation of the family of MAPK kinases (MAPKK). We show that YopJ acted as an acetyltransferase, using acetyl-coenzyme A (CoA) to modify the critical serine and threonine residues in the activation loop of MAPKK6 and thereby blocking phosphorylation. The acetylation on MAPKK6 directly competed with phosphorylation, preventing activation of the modified protein. This covalent modification may be used as a general regulatory mechanism in biological signaling.  相似文献

In an effort to clarify the pathogenesis of pyometra, 20 cows with retained fetal membranes and 20 without, but with contemporary calving dates were studied. They were palpated and their uteri were subjected to sample collections for bacteriologic cultural examinations twice weekly for 4 weeks. Blood samples were obtained each day and evaluated for serum progesterone concentration. Three cows without and 3 with retained fetal membranes developed pyometra during the study, resulting in 3 groups designated control (CON), cows with retained fetal membranes (RFM), and cows with pyometra (PYO). Bacterial isolations occurred less frequently in the CON group than in the PYO and RFM groups. Growth patterns of bacteria also varied between groups. Coliform and incidental bacteria disappeared from the uterus of the PYO group by the end of the 3rd week. In contrast, heavy growth of Corynebacterium pyogenes and gram-negative anaerobic bacteria developed during this same period in the PYO group. In cows with pyometra, the significant persistent pathogenic bacteria recovered were C pyogenes and gram-negative anaerobic bacteria, especially Fusobacterium necrophorum and Bacteroides melaninogenicus. Anaerobic bacteria were isolated simultaneously with C pyogenes in most cows of the PYO group, but less often in CON and RFM groups, and highest growth levels were present near the time of ovulation. Clinically, pyometra usually developed about 10 days after observation of concurrent ovulation and high growth levels of C pyogenes and gram-negative anaerobic bacteria. A hypothesis is presented for development of pyometra in the cow.  相似文献
