Dough extensibility affects processing ease, gas retention, and loaf volume of finished products. The Kieffer dough extensibility test was developed to assess extensibility of small dough samples and is therefore adapted for use in breeding programs. Information is lacking on relationships between wheat growing environments and dough properties measured by the Kieffer dough extensibility test. This study documents the variability of dough extensibility (Ext), maximum resistance to extension (Rmax), and area under the extensibility curve (Area) in relation to breadmaking quality, and the effect of wheat growing environments. Mixograph, Kieffer dough extensibility, and bake tests were performed on flour milled from 19 hard red spring wheat (Triticum aestivum L.) genotypes grown during three growing seasons (2007‐2009) at six South Dakota locations. Although both genotype and environment had significant effects on Kieffer dough extensibility variables, environment represented the largest source of variation. Among genotype means, Area was most correlated (r = 0.63) with loaf volume, suggesting that by selecting lines with increased Area, loaf volume should improve. Rmax was positively correlated (r = 0.58) with loaf volume among genotype means but negatively correlated (r = –0.80) among environmental means. Ext was positively correlated (r = 0.90) with loaf volume among environmental means. Weather variables were correlated with Rmax, Ext and loaf volume and therefore could help predict end‐use quality.  相似文献

BACKGROUND: Pyrazosulfuron ethyl, a new rice herbicide belonging to the sulfonylurea group, has recently been registered in India for weed control in rice crops. Many field experiments revealed the bioefficacy of this herbicide; however, no information is available on the persistence of this herbicide in paddy soil under Indian tropical conditions. Therefore, a field experiment was undertaken to investigate the fate of pyrazosulfuron ethyl in soil and water of rice fields. Persistence studies were also carried out under laboratory conditions in sterile and non‐sterile soil to evaluate the microbial contribution to degradation. RESULTS: High‐performance liquid chromatography (HPLC) of pyrazosulfuron ethyl gave a single sharp peak at 3.41 min. The instrument detection limit (IDL) for pyrazosulfuron ethyl by HPLC was 0.1 µg mL?1, with a sensitivity of 2 ng. The estimated method detection limit (EMDL) was 0.001 µg mL?1 and 0.002 µg g?1 for water and soil respectively. Two applications at an interval of 10 days gave good weed control. The herbicide residues dissipated faster in water than in soil. In the present study, with a field‐soil pH of 8.2 and an organic matter content of 0.5%, the pyrazosulfuron ethyl residues dissipated with a half‐life of 5.4 and 0.9 days in soil and water respectively. Dissipation followed first‐order kinetics. Under laboratory conditions, degradation of pyrazosulfuron ethyl was faster in non‐sterile soil (t1/2 = 9.7 days) than in sterile soil (t1/2 = 16.9 days). CONCLUSION: Pyrazosulfuron ethyl is a short‐lived molecule, and it dissipated rapidly in field soil and water. Cashew apples and kernels from sixteen high-yielding varieties were chemically characterized. Protein, starch, amino acids and sodium contents in cashew kernels did not vary significantly among the high-yielding varieties. Reducing sugar content in the kernel was negligible compared to total sugar. Ascorbic acid, amino acid, phenol and tannin contents in cashew apple showed a significant variation among high-yielding varieties. Non-reducing sugar content in cashew apples was negligible. Varieties M 6/1, Bla 256-1, M 10/4 and M 44/3 appeared to be better for apple juice extraction because of their low tannin content. The study was conducted to assess the effects of salinity on growth and biochemical composition of freshwater catfish, Clarias batrachus. A static nonrenewable acute toxicity bioassay test was conducted and LC50 of salinity for 96-h exposure to the fingerling (14.5 cm) was 12.52 ‰. Based on these results, two sublethal salinity levels, viz. 4 and 8 ‰ were selected to study the long-term effects of salinity on C. batrachus for a period of 90 days. From the study, it was found that growth and survival rate were less in saline water (4 and 8 ‰). Maximum growth and survival were recorded in freshwater (0 ‰ salinity) and subsequently at 4 and 8 ‰. To assess the biochemical alteration, few important biomarkers were estimated. At the end of 90 days rearing period, glucose level in the brain and blood of C. batrachus was found to decrease with salinity. The level of liver and muscle glycogen in the fish reared at 4 ‰ was lower than that of control. Ascorbic acid in all organs under study was found to decrease with increasing salinity, which was attributed to stress mitigation effect of vitamin C. Acetylcholine esterase (AchE) activity recorded a gradual decrease with increasing salinity. Metabolic enzymes, alkaline phosphatase (ALP) activity and adenosine triphosphosphtase (ATPase) activity also reduced both in liver and muscle tissues with increasing salinity. From the present investigation, it can be concluded that exposure to higher salinity significantly (P < 0.01) affects the growth and physiological response of Clarias batrachus.  相似文献

The high growth-stimulating effect of plant extract has urged the plant biotechnologists to use natural supplements in the culture media instead of synthetic phytohormones. We advocated the effect of sprouted sorghum extract(SSE) on emergence, in vitro acclimatization, and genetic fidelity in coleoptile derived callus of indica rice variety ADT36. The use of SSE with Murashige Skoog medium efficiently acclimatized the root and shoot apical systems. A higher mat and seminal roots(3.4 g biomass) with an efficient shoot primordium elongation were observed with an increase in the concentration of SSE. Seeds treated with SSE medium showed higher germination and earlier coleoptile maturation about 48 h compared to untreated seeds, and there was a higher expression of e EF-1α with an increase in coleoptile length. B5 medium was effective on inducing embryogenic and nodular callus from 3-day-old coleoptile with 3.0 mg/L 2,4-dichlorophenoxyacetic acid and further proliferated effectively with 0.8 mg/L kinetin with a fresh weight of 180 mg. Highly significant regeneration was observed with combination of 2.5 mg/L 6-benzylamino purine and 3.0 mg/L α-naphthaleneacetic acid. The metabolic and genetic profiles of in vitro and directly cultivated plants were the same, examined through Fourier-transform infrared spectroscopy, random amplified polymorphic DNA(RAPD), inter-simple sequence repeat(ISSR) and R-ISSR(combination of RAPD and ISSR) markers, respectively, and thus confirming the significant efficacy of the SSE incorporated medium. Disarmed T-DNA was transformed to coleoptile derived callus through Agrobacterium tumefaciens LBA4404 and confirmed by GUS assay. The T-DNA integration was confirmed by DNA blot analysis using DNA from transient GUS-expressed explants. Thus, SSE can be used as a natural and organic supplement for organogenesis and efficient acclimatizations of shoot and root apical meristems in regenerated plants.  相似文献

Wild species offer a potential reservoir of genetic variation for crop improvement. Besides the valuable genes for disease resistance that the wild species have provided for rice improvement, recent studies have shown that these wild species could also provide favorable alleles for the improvement of yield and yield-related traits. The present study reports yet another potential of wild relatives of rice, which involves the improvement of seed protein content. A significant increase in seed protein content was observed in an interspecific hybrid between Oryza sativa ssp. indica and the wild species Oryza nivara. The hybrid showed a protein content of 12.4%, which was 28 and 18.2% higher than those of the parents O. nivara and IR 64, respectively. The increase in protein content was dependent on the genetic background of the rice variety used in the hybridization. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of seed storage proteins demonstrated that a significant increase in prolamins and glutelins was mainly responsible for the elevated protein content of the hybrid. Amino acid analysis of seed proteins revealed that the hybrid had net gains of 19.5% in lysine and 19.4% in threonine over the O. nivara parent on a seed dry weight basis. Molecular analysis indicated that the increase in protein content of the hybrid was not a result of chromosomal rearrangements or transposable element activation, at least in the chromosomal regions containing seed storage protein genes. A preliminary genetic analysis of the F 2 segregating population showed that the inheritance of the increased protein content was polygenic in nature. The development of this interspecific hybrid offers a great potential for selecting new rice cultivars that combine the high yield and superior cooking quality of IR 64 with improved seed protein content.  相似文献

Madagascar has 59 species of Coffea, of which 42 are listed as Critically Endangered, Endangered, or Vulnerable by criteria of the Red List Category system of the World Conservation Union. In an attempt to assess the conservation value of ex situ collections of Malagasy coffee species, a study was undertaken using the field genebank collections maintained at the Kianjavato Coffee Research Station. Three species were selected for this purpose, C. kianjavatensis, C. montis-sacri, and C. vatovavyensis, and for comparative purposes extant, in situ populations of the same species were studied. Parentage analyses of ex situ propagated offspring of C. kianjavatensis and C. montis-sacri were performed to assess if crossing with other Coffea species maintained in the field genebank is compromising the genetic integrity of the collection. For these three species, higher genetic diversity was observed in the ex situ populations compared to the in situ populations, highlighting the importance of preserving the plants currently in ex situ collections. Parentage analyses of seed-propagated offspring of C. kianjavatensis and C. montis-sacri revealed that cross contamination with pollen from other Coffea species in the ex situ field genebank is occurring. These results have significant implications for the conservation management of wild Coffea species and for the management of ex situ genebanks.  相似文献
