A unique wheat genotype carrying waxy‐type allelic composition at the Wx loci, Gunji‐1, was developed, and its starch properties were evaluated in comparison to parental waxy and wild‐type wheat varieties. Gunji‐1 was null in all three of the Wx genes but exhibited a lower level of Wx proteins than the wild‐type. Starch amylose content and cold water retention capacity were 10.1 and 70.5% for Gunji‐1, 4.2 and 76.6% for waxy, and 27.9 and 65.0% for wild‐type, respectively. No significant differences were observed in microstructure, granule size distribution, and X‐ray diffractograms of the starch granules isolated from Gunji‐1 compared with those of waxy and wild‐type wheat varieties. Starch pasting peak, breakdown, and setback viscosities and peak temperature of Gunji‐1 were intermediate between waxy and wild‐type wheat. In starch gel hardness, Gunji‐1 (1.1 N) was more similar to waxy wheat (0.5 N) than to the wild‐type variety (17.6 N). Swelling power, swelling volume, paste transmittance during storage, and gelatinization enthalpy of Gunji‐1 were lower than those of waxy wheat but greater than those of wild‐type wheat. Retrogradation of starch stored for one week at 4°C expressed with DSC endothermic enthalpy was absent in the waxy wheat variety, whereas Gunji‐1 exhibited both retrogradation of amylopectin and amylose‐lipid complex melting similar to the wild‐type parent, even though enthalpies of Gunji‐1 were much smaller than the wild‐type parent.  相似文献

The effects of organic versus conventional farming practices on wheat functional and nutritional characteristics were compared. Soft white winter wheat and hard red spring wheat were obtained from multiyear replicated field plots near Pullman, Washington, and Bozeman, Montana. Test weight, kernel weight, and kernel diameter tended to be greater in both soft and hard organic wheat than in conventional wheat in the Pullman studies. Phenolic content and total antioxidant capacity tended to be lower in organic than in conventional wheat. Flour ash, P, and Mg contents in whole wheat flour varied in parallel among cropping systems, but levels were not consistently associated with either organic or conventional cropping systems. Protein contents of whole wheat and refined flours were similar in organic and conventional wheat from Pullman when fertility levels were similar. Higher fertility was associated with higher protein content in both organic and conventional cropping systems. Soft wheat flour from a low‐fertility organic cropping system had lower sodium carbonate, lactic acid, and sucrose solvent retention capacities, lower protein content, and greater cookie diameter and cake volume than soft wheat flour from the higher fertility organic and conventional cropping systems; the change in end‐product quality was significant in one out of two crop years. In the Bozeman hard wheat studies, higher fertility in both organic and conventional cropping systems tended to increase protein content and bread loaf volume. Results indicated that neither organic nor conventional cropping systems were associated with substantially improved mineral and antioxidant nutritional properties, and end‐use quality of wheat was more strongly associated with fertility level than with organic versus conventional cropping systems.  相似文献

Background

Sled dogs performing endurance races have been reported to have a high incidence of gastric erosions or ulcerations and an increased risk of gastro intestinal bleeding leading to death in some cases. In addition, these dogs also become hypothyroid during training and exercise. Canine hypothyroidism has been shown to correlate with decreased von Willebrand factor antigen and potentially increased bleeding tendency. Whether increased gastro intestinal bleeding risk is exacerbated due to changes in the hemostatic balance is unknown. The aim of this study was to investigate the hemostatic balance in sled dogs before and after exercise and in addition evaluate any correlation to thyroid status. Twenty sled dogs have been assessed in untrained and trained condition and immediately after exercise. The first sample was collected in the autumn following a resting period, and subsequently the dogs were exposed to increased intensity of training. After four months the peak of physical condition was reached and a 68 km long sled pulling exercise was performed. Samples were collected before and immediately after the exercise. Evaluated parameters were: plasma thromboelastographic (TEG) R, SP, α and MA, activated partial thromboplastin time (aPTT), prothrombin time (PT), fibrinogen, von Willebrand factor (vWf), D-dimer, platelet number, thyroid hormones, hematocrit and C-reactive protein (CRP).

Results

Exercise induced an overall hypercoagulable state characterized by significant decreases of TEG R and SP and an increase of α, increased concentrations of plasma vWf and decreased aPTT. In addition, a proinflammatory status was seen by a significant increase of serum CRP concentrations. Thyroid status was confirmed to be hypothyroid as training and exercise induced significant decrease of thyroxin (T4), free thyroxin (fT4) and thyroxin stimulating hormone (TSH) concentrations. Fibrinogen decreased significantly and PT increased. The training-induced changes showed correlation between T4, fT4 and aPTT and correlation between TSH and fibrinogen. Exercise-induced changes showed correlation between T4 and PT.

Conclusions

Exercise was associated with a hypercoagulable state and an increase of vWf concentration in this group of sled dogs. Decreased thyroid hormone concentrations after training and exercise were confirmed, but were associated with increased and not decreased vWf in this group of sled dogs.  相似文献

This study was conducted to explore the influencing factors of ova in vitro fertilization (IVF) and transfer of the fertilized ova into the oviduct of recipient hens. The efficiency of fertilization was compared using three aspects: (i) the different time of ova collection and transfer, (ii) egg‐laying period of recipient hen; and (iii) semen volume. The following results are observed: 72%, 40% and 0% of ova were found in ovarian sac in 30~40 min, 50~60 min and more than 90 min post‐oviposition, respectively; 20%, 18%, 14% and 5.8% of ova were fertilized with 0.1, 0.2, 0.5 and 1.0 ml semen, respectively; and 33% and 100% of healthy chickens were hatched from fertile ova with 0.1 and 0.5 ml of semen, respectively. All oocytes obtained from ovary and mid‐oviduct were unfertilized. Embryos were transferred into recipient hens 30 min ± 10 min post‐oviposition, and 70% of shelled eggs were produced. There were no eggs produced in the other transfer times. This demonstrated that live chicken can be obtained by IVF of ova collected shortly after oviposition. It was important that the ovum was transferred into the oviduct infundibulum of recipient hens immediately or shortly after oviposition.  相似文献

The thyroid glands of two series of ninety-nine unselected dogs from consecutive autopsies in London and in Munich were examined histologically. The prevalence of goitre and of neoplasia was distinctly higher in dogs in the Munich series.

In acupuncture, the specificity of the point and the reproducibility of the location of the point are prerequisite to the specificity and reproducibility of research involving acupuncture stimulation. The transpositional method, which locates animal acupoints (AAs) on the surface of animal skin corresponding to the anatomic site of a human acupoint, has been generally adopted for research modeling. However, there remains a lack of consensus on the specific location of AA among researchers. The potential problems that the discrepancy in acupoint locating methods causes include the attempt to compare research results. This report is a conceptual study that calls attention to the problems of inconsistency in AA location, and proposes a transpositional 121 AA system in a mouse and rat model. Further discussion, and the establishment of reproducible transpositional AA systems, will prompt further quantitative research and exchange of scientific ideas.  相似文献

We report the preparation of polybenzoxazole (PBO) fiber from polyhydroxyamide (PHA) precursor fiber which is free from strong acid such as polyphosphoric acid. We prepared the PHA fibers with different spin-draw ratios (SDRs) using a wet-spinning method and the PBO fibers with an SDR of 3.5 (SDR-3.5 PBO fibers) were prepared by various heat-treatment temperatures, and investigated their morphology, crystalline structure, and mechanical properties. The simultaneous thermogravimetric analysis-mass spectrometry (STA-MS) and field-emission scanning electron microscopy (FE-SEM) results confirmed that the diameter of the SDR-3.5 PBO fiber was much smaller than that of the SDR-3.5 PHA fiber, due to the release of water during the thermal cyclization reaction which forms the PBO structure. The wide-angle Xray diffraction (WAXD) pattern of the SDR-3.5 PBO fiber heat-treated at 350 °C (SDR-3.5 PBO 350 fiber) showed two peaks, at 2θ=14.83 ° and 24.38 °, and the diffraction angles dropped with increasing heat-treatment temperature. In addition, the initial modulus and tensile strength of the SDR-3.5 PBO fiber heat-treated at 550 °C (SDR-3.5 PBO 550 fiber) were found to be 19.1 GPa and 449.2 MPa, which were much higher than those of the SDR-3.5 PHA fiber, 9.3 GPa and 227.0 MPa, respectively.  相似文献
