Zoonotic disease surveillance is typically triggered after animal pathogens have already infected humans. Are there ways to identify high‐risk viruses before they emerge in humans? If so, then how and where can identifications be made and by what methods? These were the fundamental questions driving a workshop to examine the future of predictive surveillance for viruses that might jump from animals to infect humans. Virologists, ecologists and computational biologists from academia, federal government and non‐governmental organizations discussed opportunities as well as obstacles to the prediction of species jumps using genetic and ecological data from viruses and their hosts, vectors and reservoirs. This workshop marked an important first step towards envisioning both scientific and organizational frameworks for this future capability. Canine parvoviruses as well as seasonal H3N2 and pandemic H1N1 influenza viruses are discussed as exemplars that suggest what to look for in anticipating species jumps. To answer the question of where to look, prospects for discovering emerging viruses among wildlife, bats, rodents, arthropod vectors and occupationally exposed humans are discussed. Finally, opportunities and obstacles are identified and accompanied by suggestions for how to look for species jumps. Taken together, these findings constitute the beginnings of a conceptual framework for achieving a virus surveillance capability that could predict future species jumps.  相似文献

AIM: To test the hypothesis that a single strategic treatment with a new class of anthelmintic could slow the development of resistance to existing classes of anthelmintic.

METHODS: An existing model was used to simulate nematode parasite dynamics and the development of anthelmintic resistance. Variations on a five-drench preventive programme of treatments for lambs, in which either zero, the first, third or fifth treatment was substituted with a different class of drug, were compared for the time to reach treatment failure (defined as efficacy <95%). The sensitivity to variations in the death rate of adult worms, that varied from 1 to 5%, and the dominance of resistance genes were also assessed.

RESULTS: Replacing one of the five treatments with a different class of anthelmintic almost always slowed the development of resistance, and was never worse than using the same drug for all treatments. Further, there were large differences in the relative time to treatment failure depending on which treatment was substituted. Changing the first treatment always had the least benefit, whereas changing the fifth treatment always had the greatest. This pattern was independent of the daily death rate of adult worms, and was not influenced by the dominance of resistance under treatment.

CONCLUSIONS: The results indicated that strategic substitution of a single treatment with a new class of anthelmintic, at the end of a series of preventive treatments to lambs using an existing class, could slow the further development of resistance to the latter. This strategic use of a new anthelmintic class has the potential to greatly extend the life of existing anthelmintics if these are still effective.  相似文献

Recent legislation in the European Union requires regulators of member states to carry out risk assessments using data for actual or potential operator exposure, or estimates of exposure from models. However, the existing models have few datasets from studies carried out on greenhouse or indoor crops, particularly in southern Europe. In this study potential dermal and inhalatory exposures were measured in two trials in Italian greenhouses. The total potential dermal operator exposure of the applicator, measured with a whole-body passive dosimetry method, was 15.4 and 37.1 mL/h of the diluted pesticide mixture. The majority of the contamination was on the hands and on the lower part of the coverall. Approximately 0. 003% of the active ingredient (ai) applied to the crop area contaminated the coverall worn by the operator. The potential dermal exposure during the mixing and loading phase accounted for 6-8% of the total potential dermal exposure during the whole process. Inhalation exposure accounted for only 0.05-0.07% of the total potential operator exposure. Model predictions of the potential operator exposure using a modified version of the German model overestimate the mixing-loading exposure while underestimating the application exposure. These data are evidence that the estimation coefficient set for hand-held application to ornamental and horticultural crops may be inadequate for the agronomic conditions of southern Europe.  相似文献

Effects of varying the proportions of NO3 and NH4+ in the growth medium on seedling growth and tomato fruit yield (Lycopersicon esculentum L. cv. Trust F1) were investigated in greenhouse hydroponic experiments. The presence of NH4+ as the sole N source (11 mM) was toxic: it curtailed growth and decreased chlorophyll content of the leaves. However, at low concentration (10 % of total N), the presence of NH4+, with or without added dissolved inorganic carbon (DIC), increased vegetative growth and fruit yield by ˜ 15 %, and enhanced taste/flavor of the fruits. In DIC‐enriched treatment, pH was maintained at 5.8 by addition of KHCO3 or as CaCO3. The presence of NH4+, at 10 % of total N, inhibited NO3 uptake rates by ˜ 27 %. The rates of uptake of NO3 and NH4+ were comparable (13.3 and 14.2 mmol plant—1 d—1, respectively, in the presence of DIC, and 14.7 and 14.0 mmol plant—1 d—1, respectively, in the absence of DIC), despite such a large difference in their concentrations in the nutrient feed solution. A higher proportion of NH4+ (up to 50 % of total N) had no further significant effect upon early vegetative growth, but in a long‐term experiment resulted in a high incidence of blossom end‐rot (BER) disease, thereby severely curtailing fruit yield. The presence of even 1.1 mM NH4+ reduced Ca2+ and Mg2+ accumulation in the leaves as well as in fruits.  相似文献

We review some of the unique features of biogeochemical cycling in forests of the eastern Sierra Nevada Mountains, USA. As is the case for most arid and semi-arid ecosystems, spatial and temporal variability in nutrient contents and fluxes are quite high. “Islands of fertility” are common in these forests, a result of spatial variations in both litterfall and decomposition rates. Dry summer conditions greatly inhibit biological activity in the O horizon, and thus most annual litter decomposition takes place beneath the snowpack when moisture is available. Snowmelt duration is shortened near tree boles because of local warming, resulting in earlier drying of the O horizon, significantly lower decomposition rates, and increased O horizon mass. Water and nutrient fluxes vary spatially because of snowdrift in winter and surface runoff over hydrophobic soils in summer and fall. Moisture variability in the vertical as well as the horizontal dimension has significant consequences for nutrient fluxes. Because of the very dry summers, rooting in the O horizons is absent in these forests, and thus competition between microbes and trees for nutrients in that horizon is non-existent. Nutrients mineralized from the O horizon and not taken up by plants enrich runoff through the O horizons over hydrophobic mineral soils, resulting in very high concentrations of inorganic N and P in runoff waters. Substantial temporal variations in water and nutrient fluxes occur on a seasonal (with snowmelt being the dominant hydrologic event of the year), annual, and decadal basis. The most significant temporal variation is due to periodic fire, which we estimate causes annualized N losses that are two orders of magnitude greater than those associated with leaching and runoff. We hypothesize that fire suppression during the 20th century may have contributed to the deterioration of nearby Lake Tahoe by allowing buildups of N and P in O horizons which could subsequently leach from the terrestrial ecosystem to the Lake in runoff. In general, we conclude that biogeochemical cycling in these forests is characterized by greater spatial and temporal variability than in more mesic forest ecosystems.  相似文献

以麦秸纤维为增强材料、聚丙烯为基体物质、马来酸酐接枝聚丙烯(MAPP)为改性剂,制备麦秸纤维-聚丙烯复合材料。利用DMA、DSC、TG和SEM,探讨了MAPP的添加量(质量百分比1%、2%、5%、10%)和麦秸纤维形态(9、9~28、28~35、35目)对麦秸纤维 聚丙烯复合材料的热力学性能和结晶性能的影响。结果表明:①当MAPP的添加量为2%时,麦秸纤维-聚丙烯复合材料的储能弹性模量减小;当MAPP的添加量增加到5%、10%时,复合材料的储能弹性模量增加。②在麦秸纤维-聚丙烯体系内添加MAPP后,麦秸纤维 聚丙烯复合材料的结晶温度提高约1℃,结晶度增加了4%~8%;麦秸纤维以28~35目的形态作为聚丙烯基体的增强材料时,其复合材料的结晶温度为122.7℃,结晶率达到45.8%。③麦秸纤维-聚丙烯复合材料的热分解峰温分别为355和460℃。④麦秸纤维以纤维束的形态分布在基体聚丙烯中起增强作用,在整个体系内,麦秸纤维局部团聚且断裂明显。添加MAPP后,有利于基体物质在麦秸纤维表面的均匀覆盖。   相似文献

Infection and transformation of cells of the bovine immune system by Theileria annulata and T. parva were compared. Preliminary experiments with mammary gland macrophages indicated that they were permissive to infection by T. annulata but only to a limited extent by T. parva. Further experiments involved several purified subpopulations of bovine cells including bovine monocytes, T cells and MHC class II positive and negative populations. These subpopulations were incubated with T. annulata or T. parva sporozoites in limiting dilution cultures. T. annulata preferentially infected macrophage type cells and also MHC class II positive cells, whereas the frequency of MHC class II negative cells infected by this parasite was negligible. T cells also showed a very low level of infection. In complete contrast, T. parva preferentially infected T cells and did not infect cells phenotypically defined as monocytes at all. These results suggested that class II expression was necessary for T. annulata infection and not necessary for, though not a barrier to T. parva infection. T. annulata infected cell lines all expressed class II molecules to varying degrees. Other available phenotypic markers were only expressed at very low levels or no longer expressed. The immunological significance of the different cell preferences and phenotypes of infected cell lines of T. annulata and T. parva is discussed.  相似文献

Severe dermal necrosis caused by Pasteurella multocida Serotype 1 was diagnosed in three dressed turkey carcasses and two live turkeys from a commercial flock. The dressed carcasses were among several condemned at a processing plant. The isolate, P. multocida Serotype 1, produced progressive dermal necrosis when experimentally inoculated into injured skin of turkeys. The organism was reisolated from the dermal lesions. The turkey houses were found to be infested by mice; the skin injury and infection with P. multocida probably originated from mouse bites.  相似文献
